Eggshell quality is among the most important factors affecting hatchability in broiler breeders, and therefore several methods for its assessment are available in the poultry industry. Among them, eggshell translucency has received special attention in recent years due to its connection with ultrastructural disorganization of the shell layers. However, there is very limited data on the impact of translucency on hatching eggs and on the possible links between this trait and specific gravity (SG) or shell color. Thus, our study investigated associations and interactions between eggshell translucency, SG, and color on incubation parameters of eggs from the same breeding flock (Ross 308AP, 51 wk of age). To this end, light and dark eggs within 5 different SG categories (≥1.065, 1.070, 1.075, 1.080, and ≤1.085) were selected from 15,976 eggs, graded into 3 translucency scores, and later incubated to evaluate egg weight loss, hatchability and embryonic mortalities. In general, translucency scores were evenly distributed within SG categories (χ2 [8, N = 1,138] = 13.67, P = 0.090) and color (χ2 [2, N = 1,138] = 4.93, P = 0.084). No interactions between eggshell translucency and SG or between translucency and color were found for the analyzed variables. An interaction was observed between SG and eggshell color for the variable egg weight loss, where the light-shelled eggs, in most SG categories lost more weight throughout incubation than dark eggs. Eggshell translucency affected egg weight loss, hatchability, and embryonic mortality on 11 to 18 d of incubation, with highly translucent eggs showing the worst results. At the same time, eggs with SG lower than 1.070 displayed the greatest weight loss, lowest hatchability, and highest contamination. We found no influence of eggshell color on weight loss or hatchability, but light-shelled eggs exhibited higher late embryonic mortality. Together, these data suggest that despite its effects on certain hatching parameters, shell translucency bears no relationship to SG or color.

We assessed the extent to which CO2 levels altered different hatching and chick parameters. In Experiments 1 and 2, a total of 16,184 eggs from Cobb 500 breeders were incubated in single stage incubators under three different conditions: (a) standard ventilated incubator (CON, Exp.1 and 2); (b) increasing CO2 levels during the first 10 days of incubation until 0.7% (V7000, Exp. 1) and (c) until 0.8% (V8000, Exp. 2). High levels of CO2 improved hatchability, possibly due to lower embryo mortality from ED18 to ED21. Internal and external pipping in experiment V8000 started later than in CON; nevertheless, the hatch still occurred before in V8000 as a result of the shorter durations of external pipping and hatch. In Experiment 3, a total of 12,138 eggs from Cobb 500 were incubated in single stage incubators under three different conditions: (a) standard ventilated incubator (CON); (b) increasing CO2 levels until 1.0% with ventilation (V10000); and (c) increasing CO2 levels until 1.0% without ventilation (NV10000). Hypercapnic conditions led to better hatchability and lower embryo mortality from ED18 to ED21. Internal pipping started earlier in NV10000, but only V10000 differed from CON in terms of the average time for hatch. Hypercapnic groups also showed shorter durations of external pipping and hatch when compared to CON. Post-hatch analysis revealed no differences among incubation conditions in terms of body weight gain, feed conversion ratio, mortality by sudden death syndrome, and production factor. Nevertheless, V10000 showed a lower mortality by ascites and a better viability when compared to CON, while NV10000 presented a higher mortality by other causes. Altogether, our findings indicate that in addition to not being detrimental to embryo survival, high CO2 levels reduce embryonic mortality at 18-21 days of incubation and increase hatchability.(AU)

Hatchery efficiency is based on hatchability and the number of salable chicks. The hatchery sector has been seeking new alternatives to optimize production rates, including the use of different systems (multistage [MS] or single-stage [SS] machines) to improve incubation conditions. The present study aimed to compare results for hatchability, chick quality, and broiler performance of chicks from 2 incubator systems-MS and SS. The experimental design for hatchability, hatch window, egg weight loss, and chick performance variables was completely randomized with 2 treatments (MS and SS). Performance variables were analyzed as a 2 x 2 factorial arrangement (incubator type x chick sex). Egg weight loss between incubation and transfer was higher for eggs incubated in MS (P < 0.05). Hatchability was higher for eggs incubated in SS (P < 0.05), and chicks in SS had a longer hatch window (P < 0.05). Embryo diagnosis revealed higher final mortality for embryos incubated in MS (P < 0.05), as well as higher percentages of alive and dead pipped and cracked eggs (P < 0.05). Physical quality was better for chicks from SS (P < 0.05). There was no interaction between the studied factors for performance results (P > 0.05). Incubator type did not affect broiler performance for any of the studied ages (P > 0.05), whereas male broilers had better performance than females (P < 0.05). The SS incubation system proved better than the MS system at meeting embryo requirements during embryo development, with better hatching rates and chick quality, although performance variables were not influenced by incubation type.

The data mining technique (DM) was applied to analyses in a classification task, using the type of breeder and house system for delineating classes. The database was analyzed in three different ways: original database, attribute selection, and expert analysis. Models were selected on the basis of model precision and class accuracy. The data mining technique allowed for the classification of hatchery fertile eggs from different genetic groups, as well as hatching rates and the percentage of fertile eggs (the attributes with the greatest classification power). Broiler breeders showed higher fertility (> 95 %), but higher embryonic mortality between the third and seventh day post-hatching (> 0.5 %) when compared to laying hen breeders eggs. In conclusion, applying data mining to the hatchery process, selection of attributes and strategies based on the experience of experts can improve model performance.

About 33.3% of chicken life is conditioned by the result of embryonic development. Therefore, understanding and improving the early phase of the broilers life is critical to guarantee their well-being. Thus, the objective of this research was to evaluate the relationship between the breeders age, breed and the incubator types on quality and animal welfare in the hatchery. The experimental design was completely randomized in a 2 x 2 x 3 factorial arrangement (incubator, breed and age), with 12 treatments and 6 replicates each. The eggs derived of Cobb 500 and Rosss breeders at the beginning of the laying period, middle age and at the end of the laying period. It is concluded that the size of the eggs is related to the breeders age and breed, which influences directly the number of bled eggs. In addition, well-being in the hatchery can be affected by decreased hatching and contamination that is higher in older breeders. Cobb eggs also need more attention because they are larger, therefore, they have a higher risk of breakage and they are more sensitive to the low concentration of CO2 at the hatcher.

The aim of this study was to determine the optimum timing of thermal manipulation during early, mid, late and long lasting embryogenesis that may result in improvement of body performance and myofiber development (fiber diameter and thickness of perimysium) of pectoral and thigh muscles by a comparative morphometrical investigation on post-hatch day 35. 1440 fertile Ross broiler eggs were divided randomly and equally into 5 treatment groups: control (no TM), TM1, TM2 TM3 and TM4 were thermally subjected to 39ºC for 18h with 65% relative humidity daily during embryonic days ED 7-11, ED 11-15, ED 15-18 and ED 7-18 respectively. Out of TM conditions that were investigated, TM1 resulted in a significant improvement of hatchability rate (95.14%)and body performance when compared to the control. On post-hatch day 35, there were significant differences in BW, skinned carcass, breast, thigh and gut weight for all groups when compared to each other with a higher significant in TM1 and TM2 when compared to those of the control. FCR in this study was lower in TM groups when compared to the control group. In TM3 and TM4 the chicks show a significantly lower value of body performance when compared with those of the control group, TM1 and TM2.The histological evaluation of Pectoral muscle revealed myofibers of different diameter (38.39-40.27 µm) and perimysial thickening (17-30.5 µm), while myofibers diameter of thigh muscle ranged between (29-32.9 µm) and the perimysial thickening ranged between (14.36-22.15 µm). Thus, these results indicate that TM duringED7-11 improved muscle growth and body performance and this finding may be applied by commercial breeders to produce more enhanced broiler chickens.

This study aimed to evaluate the effect of post-hatch fasting time on the weights of body and digestive organs of chicks. Fertile eggs from 62-week-old broiler breeders (Cobb Fast lineage) were incubated and, after hatching, female chicks were randomly divided into six treatments, which corresponded to fasting periods (0, 6, 12, 24, 48, and 72 hours), with 40 chicks per treatment. At 01, 03, 06, and 10 days after hatching, measurements of body weight; residual yolk weight; relative weights of proventriculus + gizzard, intestine + pancreas and liver; and intestine total length were made. At 6 days of age, the chicks submitted to post-hatch fasting, for up to 12 hours, demonstrated greater development, with body weights higher than the other birds. Yet, when fed, no compensatory gain was observed and, at 10 days of age, the birds submitted to 48- and 72-h fasting remained with a lower body development. Intestine growth was also compromised by post-hatch fasting, being reduced in both weight and length. A post-hatching fasting of up to 24 hours did not interfere with the weights of body and digestive organs of 10-day-old female broiler chickens. However, 48- and 72-h post-hatch fastings affected adversely the weight and growth of digestive organs in the birds.


O efeito dos períodos de jejum pós-eclosão foi avaliado sobre o peso inicial das aves e de seus órgãosdigestórios. Ovos férteis de matrizes de frango de corte (linhagem Cobb Fast) de 62 semanas de idadeforam incubados e após a eclosão os pintinhos (fêmeas) foram divididos em seis tratamentos de 40aves cada, correspondentes a tempos diferentes de jejum (0, 6, 12, 24, 48 e 72 horas). Com 1, 3, 6e 10 dias de idade, o peso corporal das aves e do vitelo residual, pesos relativos do proventrículo +moela, intestino + pâncreas, fígado, e o comprimento total do intestino foram analisados. Aos 6 dias deidade, os animais que foram submetidos ao jejum pós-eclosão de até 12 horas demonstraram melhordesenvolvimento com maior peso corporal em relação aos demais. Essas aves não apresentaram ganhocompensatório quando alimentadas; no entanto, aos 10 dias de idade, os animais submetidos a 48 e72 horas de jejum, permaneceram com menor desenvolvimento corporal. O crescimento do intestinotambém foi comprometido em função do jejum pós-eclosão, com redução de comprimento e peso. Ojejum pós-eclosão de até 24 horas não afetou o peso corporal e o peso de órgãos digestórios em fêmeas de frango de corte criadas até 10 dias. Já jejum de 48 e 72 horas afeta de forma negativa o peso corporal e o crescimento dos órgãos digestórios.

Este estudo objetivou avaliar os efeitos da inoculação de proteína isolada de soja (P.I.S.) em ovos embrionados de matrizes semipesadas com diferentes idades. Foram utilizados 320 ovos embrionados de matrizes semipesadas da linhagem Rhode Island Red com 35 e 70 semanas de idade. Ao 17º dia de incubação, os ovos foram inoculados com 0,5mL de solução diretamente na cavidade alantoide. O delineamento experimental utilizado foi o inteiramente ao acaso, em esquema fatorial (2 x 4), constituído por dois fatores: idades das matrizes (matrizes jovens e matrizes velhas) e soluções inoculadas (ovo íntegro; 0,5% de solução salina; 1% de P.I.S. + 0,5% de solução salina; e 2% de P.I.S. + 0,5% de solução salina), totalizando oito tratamentos contendo 40 ovos cada. Os dados coletados foram analisados pelo teste de Tukey a 5% de significância. A inoculação de proteína isolada de soja apresentou efeito positivo sobre a relação pinto-ovo, porém sem proporcionar melhora nos rendimentos de incubação, no desenvolvimento dos órgãos e no desempenho de pintos na fase pré-inicial. A idade da matriz influenciou diretamente as variáveis avaliadas, em que ovos oriundos de aves velhas apresentaram maior peso, maior mortalidade embrionária na fase tardia, pintos mais pesados e pior conversão alimentar.(AU)


This study aimed to evaluate the influence of inoculation of isolated soy protein (P.I.S.) in embryonated eggs of semi-heavy with different ages. 320 embryonated eggs were used from semi-heavy breeders of Rhode Island Red lineage with 35 and 70 weeks of age. In the 17th day of incubation, the eggs were inoculated with 0.5 ml of solution in the allantoic cavity. The experimental method was completely randomized in factorial scheme (2 x 4), consisting of two factors: matrices ages (young and old matrices) and inoculation solutions (non-inoculated eggs; 0.5% of saline solution; 1% of P.I.S. + 0.5% of saline solution; and 2% of P.I.S. + 0.5% of saline solution, totalling eight treatments containing 40 eggs each. The inoculation of isolated soy protein showed positive effect on the chick-egg relation, albeit without improving incubation yields, organ development and chicks's performance in the pre-initial phase. The age of the breeder directly influenced the variables evaluated, with eggs from older breeder showing better weight, higher embryonic mortality in late phase, heavier chicks and worse feed conversion.(AU)

A inoculação de proteína isolada de soja apresentou efeito positivo sobre a relação pinto-ovo, porém sem proporcionar melhora nos rendimentos de incubação, no desenvolvimento dos órgãos e no desempenho de pintos na fase pré-inicial. A idade da matriz influenciou diretamente as variáveis avaliadas, em que ovos oriundos de aves velhas apresentaram maior peso, maior mortalidade embrionária na fase tardia, pintos mais pesados e pior conversão alimentar.(AU)


This study aimed to evaluate the influence of inoculation of isolated soy protein (P.I.S.) in embryonated eggs of semi-heavy with different ages. 320 embryonated eggs were used from semi-heavy breeders of Rhode Island Red lineage with 35 and 70 weeks of age. In the 17th day of incubation, the eggs were inoculated with 0.5 ml of solution in the allantoic cavity. The experimental method was completely randomized in factorial scheme (2 x 4), consisting of two factors: matrices ages (young and old matrices) and inoculation solutions (non-inoculated eggs; 0.5% of saline solution; 1% of P.I.S. + 0.5% of saline solution; and 2% of P.I.S. + 0.5% of saline solution, totalling eight treatments containing 40 eggs each. The inoculation of isolated soy protein showed positive effect on the chick-egg relation, albeit without improving incubation yields, organ development and chicks's performance in the pre-initial phase. The age of the breeder directly influenced the variables evaluated, with eggs from older breeder showing better weight, higher embryonic mortality in late phase, heavier chicks and worse feed conversion.(AU)

Calmodulin is vital for chick embryos morphogenesis in the incubation time 48-66 h when the rudimentary C-shaped heart attains an S-shaped pattern and the optic vesicles develop into optic cups. Melatonin is in the extraembryonic yolk sac of the avian egg; melatonin binds calmodulin. The aim of this study was to investigate the function of melatonin in the formation of the chick embryo optic cups and S-shaped heart, by pharmacological methods and immunoassays. Mel1a melatonin receptor immunofluorescence was distributed in the optic cups and rudimentary hearts. We separated embryonated chicken eggs at 48 h of incubation into basal, control and drug-treated groups, with treatment applied in the egg air sac. At 66 h of incubation, embryos were excised from the eggs and analyzed. Embryos from the basal, control (distilled water), melatonin and 6-chloromelatonin (melatonin receptor agonist) groups had regular optic cups and an S-shaped heart, while those from the calmidazolium (calmodulin inhibitor) group did not. Embryos from the luzindole (melatonin receptor antagonist) and prazosin (Mel1c melatonin receptor antagonist) groups did not have regular optic cups. Embryos from the 4-P-PDOT (Mel1b melatonin receptor antagonist) group did not have an S-shaped heart. Previous application of the melatonin, 6-chloromelatonin or forskolin (adenylate cyclase enhancer) prevented the abnormal appearance of chick embryos from the calmidazolium, luzindole, prazosin and 4-P-PDOT groups. However, 6-chloromelatonin and forskolin only partially prevented the development of defective eye cups in embryos from the calmidazolium group. The results suggested that melatonin modulates chick embryo morphogenesis via calmodulin and membrane receptors.

Incubation management can have direct effects on neonate health and consequently affect post-hatching development. The effects of incubation in multiple and single stage incubators with different concentrations of CO2 were evaluated in terms of the vessel density in the chorioallantoic membrane, hatching, heart morphology, and body development of the neonate up to the tenth day. A total of 2,520 fertile eggs were used and distributed in a completely randomized design with 4 levels of CO2 in 4 single-stage incubators (4,000; 6,000; 8,000; and 10,000 ppm) and a control treatment based on multiple-stage incubation, totaling 5 treatments. The levels of CO2 were used during the first 10 d of the incubation period, and after this period, all eggs were submitted to the same level of CO2 (4,000 ppm). Eggs that were incubated in multiple-stage incubators presented a lower percentage of vessels in the chorioallantoic membrane, lower yolk absorption by the embryo, wall depth of the right ventricle, and greater humidity losses in the eggs when compared to eggs in the single-stage incubators. The eggs submitted to hypercapnia, between 5,000 and 6,000 ppm of CO2, had a higher percentage of vessels in the chorioallantoic membrane; the embryos originating from these eggs had higher weight, with higher relative weight of the liver. However, the same levels reduced the yolk absorption. Single-stage incubation with moderate levels of hypercapnia is an efficient tool to be adopted by the hatcheries when attempting to improve chick quality.

Long storage periods may increase embryo mortality. The objective of this study was to evaluate the effects of storage period on the weight loss, eggshell surface temperature, hatchability, and embryonic mortality of Japanese quail eggs. Two hundred fertile eggs were collected from a flock of 30-week-old Japanese quails (Coturnix coturnix japonica). The eggs were collected for 10 consecutive days after lay, and immediately incubated. A completely randomized experimental design with 10 treatments, corresponding to the number of days eggs were stored between egg collection and setting, with 20 replicates each, was applied. Egg weight loss increased with storage period duration, starting on day 6 (2.1%, on average) and reached 3.26%, on average, in eggs stored for 10 days. The highest hatchability (p>0.05) was obtained in eggs stored for two days, which also lost the least weight (1.20%). Storage period did not influence eggshell surface temperature (p>0.05) during incubation, but higher temperatures (p 0.05) were measured on days 10 and 15 of incubation compared with day 5. Eggs stored for ten days presented the highest weight loss, and therefore, a storage period of up to five days is recommended to maintain the quality of incubated Japanese quail eggs. Furthermore, egg surface temperature increases during the second half of the incubation period as a result of increasing embryonic metabolic rate.

Two experiments were conducted to evaluate the effect of broiler breeder age and incubator type on hatching parameters, hatch window, embryo diagnosis results, and hatchling physical quality. The treatments consisted of a combination of three broiler breeder ages (29, 35 and 59 weeks of age) and two incubator types (single stage, SS; or and multiple stage, MS). A completely randomized design in a 3x2 factorial arrangement was applied. In Experiment I, 1,896 eggs were used and 360 eggs in Experiment II. There was an interaction between breeder age and incubator type only for hatchling physical quality score. Independently of incubator type, hatchability rate, late embryo mortality, and egg contamination were higher in the eggs laid by older breeders (59-wk-old). Early mortality (0-4 days) was higher in the embryos from young breeders (29-wk-old). A shorter hatch window birth was obtained in the SS incubator, resulting in higher hatchling body weight relative to egg weight, and better hatchling physical quality score. Both types of incubators provide good conditions for embryo development; however, the physical quality of chicks derived from eggs from intermediate-aged breeders (35-wk-old) is better when eggs are incubated in SS incubators.

The water content of the chicken eggshell has a major influence on gas (CO2 and O2) permeability. Inappropriate water loss during the incubation period increases embryo mortality and decreases chick quality. So far only the procedures that enable to determine the total water content in the eggshell have been described and developed. Our analysis of the 1H MAS NMR spectra of the chicken eggshell samples revealed three signals, differing significantly in the chemical shift and relaxation times (T1) parameters. In this work we have assigned those signals and described the changes in their intensities that occur during the incubation period. Using 1H MAS NMR it is possible to distinguish two types of water reservoirs in the chicken eggshell. This approach can be used for more detailed analysis of the water content in the eggshells.

Adequate environmental temperature control is essential for incubation efficiency. Layer breeder eggs (n=360) were weighed and distributed in a completely randomized experimental design with two treatments, consisting of two incubation temperatures (T1=37.5 °C, control; and T2=39.0 °C, hot), with two incubators per temperature, and 90 eggs per incubator, totaling 360 eggs. Hatchability, embryo mortality, and chick cloacal and body surface temperatures were not affected by incubation temperature. Eggs incubated at the hot temperature presented greater egg mass loss and higher eggshell conductance than those incubated at the control temperature. Layer chicks derived from eggs incubated at control temperature showed greater absolute weight, yolk-free egg mass, and heavier hearts than those from eggs submitted to heat stress during incubation. The control group presented lower base excess and ionized calcium blood levels. Incubating eggs at temperatures higher than those recommended compromises body and heart development of layer chicks and negatively affects blood ionized calcium availability, and therefore, bone mineralization during embryo development. Efficient temperature control during the incubation of fertile eggs is essential to obtain good quality layer chicks.
