RESUMEN

The objective of this study was to evaluate the enzymatic activity of plasma cholinesterase in Chelonia mydas marine turtles belonging to two populations, according to their capture sites, under the absence and probable influence of anthropic effects. A total of 74 animals were used and later divided into two groups, based on the capture site. Blood samples were collected from all captured animals, which were then released into the sea at the site of capture. A descriptive statistical analysis of the plasma cholinesterase activity values and an analysis comparing these values based on the capture site were performed. Samples of heparinized plasma from animals captured at the two different sites were analyzed. Plasma cholinesterase activity ranged from 121 to 248U/L, with a mean and standard deviation of 186.1±30.68U/L. When comparing plasma cholinesterase activity values in individuals based on the capture site, a significant difference was observed. Establishing reference values for different sea turtle populations is necessary to interpret future sampling results and to allow sea turtles to be used as sentinels of ecosystem health. Future studies are needed to evaluate other populations and the activity of plasma cholinesterase in juvenile marine turtles, in relation to environmental contamination.(AU)


O objetivo desse estudo foi avaliar a atividade enzimática da colinesterase plasmática em tartarugas marinhas da espécie Chelonia mydas em duas populações de acordo com o local de captura, sob ausência e provável influência de efeito antrópico. Foi utilizado um total de 74 animais e posteriormente divididos em dois grupos de acordo com o local de captura. Foram coletadas amostras de sangue de todos os animais capturados e em seguida liberados ao mar no mesmo local. Foi realizada uma análise estatística descritiva dos valores da atividade plasmática de colinesterase do total de animais e análise comparando os valores de acordo com o local de captura. Foram analisadas amostras de plasma heparinizado de animais capturados em dois locais distintos. Os valores da atividade plasmática de colinesterase variaram de 121 a 248U/L, com média e desvio padrão de 186.1±30.7U/L. Quando comparados os valores de atividade plasmática da colinesterase nos indivíduos de acordo com o local de captura, foi observada diferença significativa. O estabelecimento de valores de referência para diferentes populações de tartarugas marinhas são necessários para interpretar os futuros resultados amostrais e permitir que as tartarugas marinhas sejam usadas como sentinelas da saúde do ecossistema. Estudos futuros são necessários para avaliar outras populações e a atividade da colinesterase plasmática de tartarugas marinhas juvenis em relação à contaminação ambiental.(AU)

Biomarkers are frequently used in ecotoxicology as they allow to study toxicant effects happening at low concentrations of exposure. However, most sublethal studies only evaluate cellular biomarkers which lack evident ecological relevance. We used a multibiomarker approach to estimate the toxic effects of ethoprophos, an organophosphate insecticide commonly used in banana plantations, on the tropical fish Astyanax aeneus (Characidae). We measured biomarkers at sub-individual (cellular) and individual (metabolism, behavior) levels and examined relationships among these responses. A sublethal exposure to ethoprophos caused a significant (54%) reduction of brain Cholinesterase (ChE) activity, reflecting the pesticide's high neurotoxicity. However, other biomarkers like oxidative stress, biotransformation reactions, and resting metabolic rate were not affected. Exposure to ethoprophos modified antipredator behaviors such as escape response and detection avoidance (light/dark preference): exposed fish escaped slower from a simulated attack and preferred brighter areas in a novel tank. The relationship between ChE activity and reaction time suggests that pesticide-induced ChE inhibition reduces escape ability in fish. Our results provide evidence that impacts of organophosphate pesticides on fish ecological fitness can occur even with short exposures at very low concentrations.

Background: Nitrate and nitrite poisoning is associated with pasture intake that has high nitrate levels and leads to acute methemoglobinemia. Pasture may accumulate nitrate under certain conditions, such as excessively fertilized soil or environmental conditions that enhance the N absorption (rain preceded by a period of drought). After ingestion of plants, this substrate reaches the rumen and, in physiological conditions, is reduced to nitrite and afterward to ammonia. The aim of this study was to evaluate changes in cholinesterase activities and oxidative stress caused by subclinical poisoning for nitrate and nitrite in cattle fed with Pennisetum glaucum in three different fertilization schemes.Materials, Methods & Results: In order to perform the experimental poisoning, the pasture was cultivated in three different paddocks: with nitrogen topdressing (urea; group 1), organic fertilizer (group 2) or without fertilizer (group 3; control). Nitrate accumulation in forage was evaluated by the diphenylamine test. After food fasting of 12 h, nine bovine were randomly allocated to one of the experimental groups and fed with fresh forage (ad libitum) from respective paddock. In different time points from beginning of pasture intake (0, 2, 4, 6 and 9 h) heart rate and respiratory frequency were assessed, as well as mucous membrane color and behavioral changes. Blood samples from jugular vein into vials with and without anticoagulant were collected. From blood samples, serum nitrite levels, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzyme activity were evaluated, as well as oxidative stress through the following parameters: levels of nitrate/nitrite (NOx ), thiobarbituric acid reactive substances (TBARS) and reactive oxygen species (ROS), beyond the antioxidant system by enzyme activity measurement of catalase (CAT) and superoxide dismutase (SOD).[...]

The aim of this study was to evaluate the cholinesterase activity in serum, whole blood, and lymphocytes, as well as to verify its relation to immune response in rats experimentally infected by Sporothrix schenckii. For this study, 63 Wistar rats (Rattus norvegicus), male, adult were divided into three groups: the negative control group (GC: n = 21), the group infected subcutaneously (GSC: n = 21), and the group infected intraperitoneally (GIP: n = 21). The groups were divided into subgroups and the following variables were evaluated at 15, 30, and 40 days post-infection (PI): acetylcholinesterase (AChE) activity in lymphocytes and whole blood, butyrylcholinesterase (BChE) activity in serum, cytokines levels (IL-1, IL-6, TNFα, and INF-γ), immunoglobulins levels (IgA, IgG, IgM, and IgE), and protein profile by electrophoresis. Both infected groups showed increased levels of inflammatory parameters (P < 0.05) in tissue and inflammatory infiltrates. The activities of AChE in lymphocytes and BChE in serum increased (P < 0.05) significantly in animals from the GSC group on day 40 PI compared to the GC group. Regarding the GIP, there was a marked increase in the AChE activity in lymphocytes on days 30 and 40 PI, and in whole blood on days 15, 30, and 40 PI compared to GC. Furthermore, IL-10, an anti-inflammatory cytokine, was also present in high levels during chronic systemic S. schenckii infections in animals. Therefore, it is concluded that cholinesterase has an important modulatory role in the immune response during granulomatous infection by S. schenckii.

Biomarcadores podem ser usados de forma preditiva, permitindo que sejam tomadas ações de controle antes que ocorram danos ambientais irreversíveis com consequências ecológicas severas, no entanto, espécies sentinelas são necessárias para avaliação desses marcadores. As tartarugas marinhas são consideradas espécies sentinelas quando acometidas por fibropapilomas, sendo sinalizadora do desequilíbrio ambiental marinho nas suas áreas de ocorrência. Com o objetivo de propor a determinação da atividade da colinesterase plasmática em tartarugas verdes (Chelonia mydas) como biomarcador, procedeu-se a determinação da atividade enzimática em animais saudáveis e em localidade de baixo impacto antrópico (Arquipélago de Fernando de Noronha, Pernambuco, Brasil) para servir como referência para comparação com animais capturados em locais de maior impacto antrópico. Ao todo foram analisadas amostras de plasma heparinizado de 35 animais capturados. Todas as amostras analisadas apresentaram alguma atividade enzimática de colinesterase plasmática. Os valores obtidos de colinesterase variaram de 162 a 379 UI/L, com média e desvio padrão de 216,4 ± 51,4 UI/L. Nos estudos de repetibilidade e reprodutibilidade obtiveram-se coeficientes de variação menor que 5% em todas as análises, portanto a metodologia analítica utilizada se mostrou confiável. A longevidade das tartarugas marinhas da espécie C. mydas, o comportamento alimentar, juntamente com o fato de possuirem atividade enzimática detectável podem indicar essa espécie como bioindicadora de exposição a poluentes que influenciam na atividade da colinesterase plasmática.(AU)

Sea turtles affected by fibropapillomas are considered sentinel species, signaling the marine environmental imbalance in their areas of occurrence. With the objective to use the plasma cholinesterase activity in green turtles (Chelonia mydas) as a biomarker, the measurement of enzyme activity was evaluated in healthy animals and in location of low pollution impact (Fernando de Noronha, Pernambuco, Brazil) to serve as a reference for comparison with animals trapped in places with higher pollution impact. Were analyzed heparinized plasma samples from 35 captured animals. All samples analyzed showed enzymatic activity of plasma cholinesterase. The values obtained ranged between 162-379 IU/L, with mean and standard deviation of 216.4 ± 51.4 IU/L. In studies of repeatability and reproducibility were obtained variance coefficients of less than 5% in all analyzes, so the analytical methodology proved successful. The longevity of marine turtles of the species C. mydas, feeding behavior, along with the fact possess detectable enzyme activity may indicate this species as a bioindicator of exposure to pollutants that influence plasma cholinesterase activity.(AU)

Cholinesterase (ChE) activity has been used for many years as a biomarker of exposure to organophosphate and carbamate pesticides. Recent studies have demonstrated that there could be biological factors that determine ChE type and levels; thus, juvenile Sergeant major (Abudefduf saxatilis) ChE enzymes were biochemically characterized. ChE enzymes found in the head and trunk were evaluated for their substrate preference and sensitivity to selective inhibitors. The use of the head and trunk was chosen as a strategy to reduce dissection time and to ensure sample uniformity between stations. The results indicated that there are two types of ChE enzymes in the head: acetylcholinesterase (AChE) and atypical butyrylcholinesterase (BChE) that exhibits intermediate characteristics of human AChE and BChE activities. Atypical BChE is predominantly found in the trunk. The results also indicated that the ChE activity found in A. saxatilis may be used as a biomarker in studies monitoring the Mexican Caribbean.

Se realizó un estudio comparativo y experimental para determinar el nivel plasmático de colinesterasa (con la técnica de Ensayo del reactivo de Ellman) en 51 mujeres embarazadas y 52 no embarazadas, expuestas en forma crónica a insecticidas organofosforados y carbamatos en la zona del páramo merideño y que acudieron a control y/o consulta en el Hospital tipo I de Mucuchíes (Mérida) con la finalidad de establecer la prevalencia de sus efectos en la zona mencionada y la ulterior consecuencia en el producto de la gestación. Los datos se analizaron mediante el software estadístico SPSS versión 15 para Windows. Los resultados mostraron que las pacientes embarazadas con o sin antecedentes patológicos, sus niveles de colinesterasa tienden a disminuir en cierta magnitud, se encontró diferencia estadísticamente significativa entre los niveles de colinesterasa promedio de mujeres embarazadas con antecedentes con respecto a las que no tenían antecedentes, en las mujeres no embarazadas y sin antecedentes que vivían cerca de zonas de cultivo con exposición a plaguicidas el nivel promedio de colinesterasa fue menor al nivel promedio de las mujeres en edad reproductiva no expuestas y una disminución de los valores de colinesterasa promedio en las gestantes menores de 39 años de edad. Se concluyo que las mujeres embarazadas con antecedentes patológicos presentan inhibición de niveles de colinesterasa plasmática con respeto a las no embarazadas y sin antecedentes.


A study was conducted to determine comparative and experimental plasma cholinesterase level (with the assay technique of Ellman's reagent) in 51 pregnant women and 52 non-pregnant, chronically exposed to organophosphate and carbamate wilderness area in Merida and who came to control and / or reference in the type I Mucuchíes Hospital Merida state in order to establish the prevalence of its effects on the area mentioned and further result in the product of gestation. The data were analyzed using SPSS statistical software version 15 for Windows. The results showed that pregnant patients with or without pathological history cholinesterase levels tend to decrease to some extent, statistically significant difference was found between the average cholinesterase levels of pregnant women with a history over which they had no background in nonpregnant women with no history of living near farming areas with pesticide exposure cholinesterase level was lower average to average level of reproductive age women unexposed and decreased average cholinesterase values in pregnant women under 39 years of age. It was concluded that pregnant women with medical history presented inhibition of plasma cholinesterase levels with respect to the non-pregnant and no history.

Organophosphates (OPs) are widely used as pesticides, and its urinary metabolites as well as the blood cholinesterases (ChEs) activity have been reported as possible biomarkers for the assessment of this pesticide exposure. Moreover, the OPs can induce mutagenesis, and the bone marrow micronucleus test is an efficient way to assess this chromosomal damage. This paper reports a study carried out to verify the correlation among the disulfoton exposure, blood ChEs activity, urinary diethyl thiophosphate (DETP), and diethyl dithiophosphate (DEDTP), as well as micronucleated polychromatic erythrocytes (MNPCEs) frequency. Four groups of rats (n=12) were exposed to disulfoton at 0, 2.8, 4.7, and 6.6 mg kg-1 body weight. The blood ChEs activity, urinary DETP and DEDTP concentrations, and MNPCEs frequency were determined. It was observed that the plasmatic and erythrocytary ChEs activity decreased from 2.9% to 0.5% and from 35.9 to 3.3%, respectively, when the disulfoton dose was increased from 0 to 6.6 mg kg-1 (correlation of 0.99). Urinary DETP and DEDTP concentrations, as well as the MNPCEs frequency, increased from 0 to 6.58 µg mL-1, from 0 to 0.04 µg mL-1, and from 0 to 1.4%, respectively, when the disulfoton dose was increased from 0 to 6.58 mg kg-1 body weight.


Os organofosforados (OPs) são amplamente usados como praguicidas e a atividade da colinesterase sanguínea bem como os metabólitos urinários desses praguicidas têm sido reportados como biomarcadores eficazes para avaliar casos de exposição. Além disso, os OPs podem induzir mutagênese e o teste de micronúcleo de medula óssea é uma boa alternativa para avaliar os danos cromossômicos. Esse artigo reporta um estudo sobre a correlação entre a exposição a dissulfoton, a atividade da colinesterase sanguínea, a excreção urinária de dietil tiofosfato e dietil ditiofosfato e a frequência de micronúcleos em eritrócitos policromáticos. Quatro grupos de ratos (n=12) foram expostos a dissulfotom nas doses de 0, 2,8, 4,7, e 6,6 mg kg-1 de peso corpóreo. A atividade da colinesterase sanguínea as concentrações urinárias de dietil tiofosfato e dietil ditiofosfato e a frequência de micronúcleos foram determinadas. Os resultados demonstraram que as atividades da colinesterase plasmática e eritrocitária diminuíram de 2,9 para 0,5% e de 35,9 para 3,3% , respectivamente, quando a dose de dissulfoton foi aumentada de 0 para 6,6 mg kg-1 (correlação de 0,99). As concentrações urinárias de dietil tiofosfato e dietil ditiofosfato bem como a frequência de micronúcleos aumentaram de 0 a 6,56 µg mL-1, 0 a 0.04 µg mL-1 e de 0 a 1.4%, respectivamente, quando a dose de dissulfotom foi aumentada de 0 a 6,58 mg kg-1.

Background: The cholinergic system is involved in many biological functions in mammals and is associated with pathogenesis of infectious diseases, as has participation in transmission of nerve impulses in cholinergic synapses, haematopoiesis, regulation of inflammatory markers, production and coordination of movement, and memory. Rangelia vitalii is a parasite endemic to south of Brazil. This parasite multiplies in the blood and can be visualized in plasma in its free form and/or within leukocytes and erythrocytes, causing various pathologies. Therefore, the purpose of this study was to investigate the activity of cholinergic system enzymes in dogs experimentally infected with R. vitalii. Materials, Methods & Results: Twelve dogs were used, divided into two groups: control group (n = 5), consisting of healthy animals, and infected group with R. vitalii (n = 7). Fresh blood samples of these infected animals were inoculated in seven dogs (2 mL/dog through the jugular vein). Blood samples were collected on days 0, 10 and 20 post-infection (PI). Butyrylcholinesterase (BChE) activity was measured in serum and acetylcholinesterase (AChE) in lymphocytes and whole blood. Boold samples were diluted 1:50 (v/v) in lysis solution (0.1 mmol/L potassium/sodium phosphate buffer containing 0.03% Triton X-100) and frozen (-20 ºC by 7 days) to determine AChE activity in whole blood. Lymphocytes were also obtained from whole blood with EDTA by gradient separation using Ficoll-Histopaque™ plus to AChE activity this cell. After analysis of the samples, was observed that the dogs infected with R. vitalii presented a signifi cant (P < 0.01) increase in AChE activity in whole blood on days 10 and 20 PI. However, the infected group showed a reduced activity in AChE in lymphocytes (P < 0.01) and BChE in serum (P < 0.05) on day 20 PI. Discussion: According to the literature, infected dogs R. vitalii develop regenerative anemia evidenced by an increase in the erytroid precursors in bone marrow associated with alterations of leucogram as leukopenia, neutropenia, eosinopenia, lymphocytosis and monocytosis. Furthermore, it was observed severe thrombocytopenia, with alteration in platelet aggregation and activity of enzymes involved in the control of ATP, ADP and adenosine levels on platelets, thereby influencing hemostasis and contributing to the typical bleeding disease. AChE activity in whole blood was increased in dogs parasitized by R. vitalii observed in this study. This increase may be a compensatory effect to severe anemia caused by the parasite infection, because this enzyme is involved in the maturation of erythrocytes and in the regulation of hematopoiesis. In the present study, we found that the reduction in AChE activity in lymphocytes is associated to lymphocytosis; and it is known that ACh is produced within lymphocytes and has the ability to negatively modulate the immune response, acting directly on the inhibition of inflammatory mediators. Therefore, the decrease of AChE activity may have an anti-inflammatory action in order to have more free ACh to bind lymphocytes and inhibit inflammation. The enzyme BChE can also act as an inflammatory marker in various diseases, similar to AChE, because the enzyme can hydrolyze acetylcholine when AChE is inhibited. In conclusion, our results indicate that canine rangeliosis alters the activity of cholinesterase's, which may be involved in the pathogenesis of the disease, as well as various pathological conditions.

O objetivo deste trabalho foi avaliar a eficácia de uma coleira impregnada com Flumetrina (2,5%) e Propoxur (10%) sobre adultos de Ctenocephalides felis felis e Rhipicephalus sanguineus em cães Beagle com avaliação de possíveis alterações da colinesterase sérica (butirilcolinesterase) após o uso da coleira. Para isso foram utilizados 20 cães da raça Beagle, divididos em dois grupos, onde em 10 animais foram colocadas as coleiras impregnadas e os outros 10 sem nenhum tratamento (controle). Cada cão foi infestado com 50 casais de C. felis felis e 25 casais de R. sanguineus semanalmente no primeiro mês e a partir do segundo mês quinzenalmente até o dia +210. Quarenta e oito horas após as infestações foram realizadas avaliações com contagem de pulgas e carrapatos vivos recuperados nos animais. Para dosagem da colinesterase sérica foram coletadas amostras de sangue dos animais nos dias -14, -7, +1, +5, +10, +30, +45, +60, +75, +90, +105, +120, +135, +150, +165, +180, +195 e +210. Todos os animais foram avaliados semanalmente em relação a sinais clínicos compatíveis com intoxicação. Posteriormente ao término do experimento de eficácia as coleiras foram removidas e após sete dias foi realizada nova dosagem da colinesterase. A eficácia para C. felis felis foi superior a 80% até o dia +168 após tratamento. A eficácia para R. sanguineus foi superior a 80% até o dia +182. Foi verificado que a coleira não diminuiu significativamente (p?0,05) os níveis séricos de butirilcolinesterase nos cães comparando-se as médias antes e após o tratamento, no entanto, a atividade sérica da colinesterase aumentou significativamente (p?0,05) após a retirada da coleira. Evidenciou-se que a coleira a base de propoxur e flumetrina apresenta eficácia superior a cinco meses sobre C. felis felis e R. sanguineus. A alteração da colinesterase sérica dos animais pelo propoxur não foi relevante e nenhum sinal de toxicidade foi observado durante o período experimental

In this study, the acute toxicity and the in vivo effects of commercial chlorpyrifos, carbofuran and glyphosate formulations on cholinesterase (ChE), glutathione S-transferase (GST) and lactate dehydrogenase (LDH) activities of the mosquitofish (Gambusia yucatana) were investigated. In a first phase of the study, head and muscle ChE were characterized with different substrates (acetylthiocholine iodide, s-butyrylthiocholine iodide and propionylthiocholine iodide) and the selective inhibitors eserine hemisulfate, 1,5-bis(4-allyldimethylammoniumphenyl)-pentan-3-one dibromide (BW284C51), and N,N'-diisopropylphosphorodiamic acid (iso-OMPA). The results obtained suggest that the enzyme present in both head and muscle of G. yucatana is mainly acetylcholinesterase (AChE). Acute toxicity was evaluated by exposing fish to several concentrations of single pesticides and of a mixture of chlorpyrifos/glyphosate. LC50 values were determined after 96 h of exposure, except in the case of carbofuran for which LC50 was calculated after 24 h since almost all the fish died within this period. LC50 values were 0.085 mg/l for chlorpyrifos, 17.79 mg/l for glyphosate, 0.636 mg/l for carbofuran and 0.011 mg/l for the chlorpyrifos/glyphosate mixture. A Toxic Unit approach was used to compare the toxicity of chlorpyrifos and glyphosate when occurring in a mixture with their toxicities as single compounds. Synergistic effects of chlorpyrifos and glyphosate when present in a mixture were found. At the end of each bioassay (24 h for carbofuran, 96 for the other substances/mixture), effects on biomarkers were analyzed. Muscle LDH activity was not altered by any of the three pesticides tested. Gill GST activity was significantly inhibited (40%) by carbofuran after 24 h of exposure to concentrations equal or higher than 0.06 mg/l. ChE muscle and head activity were significantly inhibited (50% and 30%, respectively) by carbofuran at concentrations equal or higher than 0.25 mg/l. Chlorpyrifos induced a significant inhibition of both muscle and head ChE (80% and 50%, respectively) after 96 h of exposure to concentrations equal or higher than 0.05 mg/l. Carbofuran did not induce significant alterations of fish ChE. The ChE EC50 determined for chlorpyrifos/glyphosate mixture (0.070 mg/l) was higher than the correspondent value calculated for chlorpyrifos alone (0.011 mg/l) suggesting an antagonistic effect of glyphosate on ChE inhibition by chlorpyrifos. ChE activity of G. yucatana seems to be a good biomarker to diagnose the exposure of wild populations of this species exposed to anticholinesterase pesticides.

Field methods are needed to detect and monitor the organophosphate pesticide exposure of young children. Twenty children, aged 11 to 18 mo, living in an agricultural community along the United States/Mexico border were enrolled in a pilot study investigating methods to detect pesticide exposure. Healthy children were recruited at pediatric clinics with the informed consent of their parents. Venous blood samples were collected from children twice, 4 wk apart. Cholinesterase activity was compared in whole heparinized venous blood with venous blood samples dried on filter paper. Although the amount of activity in the dried blood was consistently less than in the heparinized blood, the activity was significantly correlated: Spearman r = .6 (p = 0.01). This dried blood method may be used during field studies to evaluate changes in cholinesterase values in children over time.

The initial sampling in the Marine Monitoring Program (MOMAM), coordinated by the Ministry of Marine Affairs (IEAPM), was performed along the southeast coast of Brazil. Orthopristis ruber samples were collected at Guanabara, Sepetiba and Ilha Grande Bays. Microsomal CYP1A levels and cytosolic cholinesterase (ChE), catalase (CAT) and glutathione S-transferase (GST) activities were measured in the liver of these fish according to established procedures. CAT activity and CYP1A content were significantly higher (P < or = 0.05) in fish caught at Guanabara Bay, which might be due to higher levels of peroxisome proliferators and Ah receptor agonists, respectively, at this site compared to the other sites. Also, lower GST activity was observed in fish from this site, which may possibly be related to the presence of oxidative-stress inducing compounds.

In central nervous system, acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) hydrolyse acetylcholine. Diminished cholinesterase activity is known to alter several mental and psychomotor functions. The symptoms of cholinergic crisis and those observed during acute attacks of acute intermittent porphyria are very similar. The aim of this study was to investigate if there could be a link between the action of some porphyrinogenic drugs on brain and the alteration of the cholinergic system. To this end, AChE and BuChE activities were assayed in whole and different brain areas. Muscarinic acetylcholine receptor (mAChR) levels were also measured. Results obtained indicate that the porphyrinogenic drugs tested affect central cholinergic transmission. Quantification of mAChR gave quite different levels depending on the xenobiotic. Veronal administration inhibited 50% BuChE activity in whole brain, cortex and hippocampus; concomitantly cortex mAChR was 30% reduced. Acute and chronic isoflurane anaesthesia diminished BuChE activity by 70-90% in whole brain instead cerebellum and hippocampus mAChR levels were only altered by chronic enflurane anaesthesia. Differential inhibition of cholinesterases in the brain regions and their consequent effects may be of importance to the knowledge of the mechanisms of neurotoxicity of porphyrinogenic drugs.

Se evaluó la estabilidad de la colinesterasa plasmática y la colinesterasa eritrocítica a temperatura ambiente, 4§C y -20 §C, durante un periodo de tres meses. Se determinó que la colinesterasa plasmática es estable en refrigeración y congelación. A temperatura ambiente la actividad de esta enzima disminuye significativamente en una semana. Por su parte la colinesterasa eritrocítica es estable únicamente en refrigeración. El proceso de congelamiento y descongelamiento provoca disminución importante en la actividad de la enzima. En las muestras conservadas por un día a -20 §C, la actividad de la enzima eritrocitaria fue enre 10 y 20 por ciento más baja que la actividad basal. A temperatura ambiente la actividad de esta enzima disminuye en forma importante en una semana. Se describe la modificación de un método para la cuantificación de colinesterasa plasmática mediante monitoreo continuo empleando ferricianuro como indicador. La tiocolina liberada de la propioniltiocolina reduce el ferricianuro a ferrocianuro y la disminición de absorbancia a 405 nm es proporcional a la actividad de la enzima. Las precisiones día a día para muestras con valores de colinesterasa bajos y altos mostraron coeficientes de variación de 3,2 y 1,2 por ciento y en un mismo día de 1,1 y 0,52 por ciento respectivamente. La bilirrubina no presenta una interferencia importante y cada 50mg/dL de hemoglobina producen una interferencia de -4 por ciento. El reactivo de ferricianuro almacenado en botella ámbar es estable por al menos 3 meses a temperatura ambiente y 6 meses a 4-8 §C. Al comparar los resultados con un método que emplea la reacción de Ellman se obtuvo una ecuación de regresión lineal de Y = 1,22 (X) - 732, con un coeficiente de correlación (r) de 0,988 y una desviación estándar sobre la línea de regresión (Sy/x) de 378 U/L. OBJECTIVES: To explore whether symptoms resulted from pesticide spray drift on residentially exposed populations in rural Nicaragua. METHODS: 100 residents, each 10 years of age or older, were randomly selected from a Nicaraguan community surrounded by actively sprayed cotton fields (the exposed community) and from a socioeconomically similar community far from agricultural spraying (the control community). Subjects working with pesticides were excluded, and the study was conducted at the end of the 1990 cotton spraying season (August-December). Demographic information, exposure questions, and prevalence of 11 acute symptoms and 17 chronic symptoms were gathered from a structured interview. Finger stick erythrocyte cholinesterase (AChE) was measured with a portable colorimeter. Acute symptoms were grouped according to their previously known associations with cholinesterase (ChE) inhibitors into four ordinal categories (asymptomatic, non-specific, possible, probable). RESULTS: Residents from the exposed community were significantly more likely to report recently sighting a spray plane near their community, exposure to pesticide from drift, crossing recently sprayed fields, eating home grown food, and feeling ill after drift exposure. The mean AChE value was significantly lower for residents of the exposed community (4.9 v 5.3 IU/dl). The proportion of subjects complaining of one or more chronic or acute symptoms was significantly higher for the exposed community (87%) than for the controls (53%). Odds ratios for residents in the exposed community, by symptom categories, were non-specific 1.6 (95% confidence interval (95% CI) 0-8 to 3.2), possible 4.1 (95% CI 1.7 to 10.2), and probable 9.93 (95% CI 2-9 to 34.4). CONCLUSION: These findings indicate a strong association between exposure to aerial pesticides and symptoms. This study should be replicated with more quantitative exposure measures, for if confirmed, the results have relevance for millions in rural communities worldwide.
