RESUMEN
The number of food products with genetically modified (GM) crops on the global market has increased due to advancements in genetic engineering technology. Legislation regulating the labeling and use of GM crops has increased considerably worldwide to provide consumers with health and safety assurance. It is still unclear whether genetically modified organisms (GMOs) are present in the food market of the Kingdom of Saudi Arabia due to a lack of scientific studies. This work was planned to detect GM rice and GM potatoes in the Saudi food market. One hundred non-labeled rice and rice product samples and 50 potato and potato samples were collected randomly from different market sites of Makkah, Riyadh and Jeddah during 2022-2023. The cetyl trimethyl ammonium bromide (CTAB) method was used to extract DNA. Viviants DNA extraction kit was used to extract DNA from rice starch and potato chips. To find GMOs in samples, CMOScreen 35S and NOS test kits were utilized. DNA-based qualitative and quantitative approaches were used to screen targets for PCR detection of GM rice sequences. The results indicated that 32 (32%) rice samples were positive for CaMV 35S promoter, while no positive result was detected for the NOS terminator. Besides, 30% of potato samples were positive for the CaMV 35S promoter, and the same samples were positive for the presence of the Cry V gene. It could be concluded that there were GM rice and potatoes in the Kingdom of Saudi Arabia's food markets. Establishing strong regulations and certified laboratories to monitor genetically modified foods (GMF) or crops in the Saudi market is recommended.

The study of extremophilic microorganisms has sparked interest in understanding extraterrestrial microbial life. Such organisms are fundamental for investigating life forms on Saturn's icy moons, such as Enceladus, which is characterized by potentially habitable saline and alkaline niches. Our study focused on the salt-alkaline soil of the Al Wahbah crater in Saudi Arabia, where we identified microorganisms that could be used as biological models to understand potential life on Enceladus. The search involved isolating 48 bacterial strains, sequencing the genomes of two thermo-haloalkaliphilic strains, and characterizing them for astrobiological application. A deeper understanding of the genetic composition and functional capabilities of the two novel strains of Halalkalibacterium halodurans provided valuable insights into their survival strategies and the presence of coding genes and pathways related to adaptations to environmental stressors. We also used mass spectrometry with a molecular network approach, highlighting various classes of molecules, such as phospholipids and nonproteinogenic amino acids, as potential biosignatures. These are essential features for understanding life's adaptability under extreme conditions and could be used as targets for biosignatures in upcoming missions exploring Enceladus' orbit. Furthermore, our study reinforces the need to look at new extreme environments on Earth that might contribute to the astrobiology field.

OBJECTIVES: Disease surveillance is an essential component of public health and a core function of National Public Health Institutes (NPHIs), including to better prepare and respond to infectious diseases outbreaks. Strengthening NPHIs in their efforts to establish and maintain efficient surveillance systems is an opportunity to ensure future outbreak preparedness and response; yet, guidance on how to increase and prioritise capacity building efforts is limited. This study sought to investigate approaches to capacity building and training for disease surveillance at national level and understand the potential role of NPHIs. STUDY DESIGN: Qualitative study. METHODS: This is a qualitative study, based on a literature review and interviews undertaken between June and November 2022. Fifty seven in-depth interviews were conducted in five countries: Côte d'Ivoire, Ecuador, Madagascar, Namibia, and the Kingdom of Saudi Arabia. Participants included a range of professionals from government, NPHIs, academic institutions and the private sector. Interviews were thematically analysed. RESULTS: Selected countries varied in terms of their disease surveillance capacities, as well as in the structure of their surveillance systems and decision-making. Research identified shared priority areas for action at national level, identifying common challenges and opportunities: 1) capacity building, here specifically the need for a training agenda at national level to ensure sustainability and guide donor funded training offers; 2) data tools and technology-to help decision-makers select the best software tool to address countries' identified need; 3) data sharing-the need for clear data sharing standards and norms for national to international data sharing; and 4) genomic sequencing-the need for national genomic surveillance strategies and reporting guidelines. CONCLUSION: Addressing challenges and using opportunities to strengthen disease surveillance at national level is an important step to build capacity in this area and to help prevent future epidemic and pandemics globally. The findings of this study help decision-makers to identify priority areas for capacity building and understand the potential role and significance of NPHIs.

We report the first record of the occurrence of the banana weevil, Cosmopolites sordidus (Germar, 1823) (Coleoptera: Curculionidae), an economically important pest of bananas (Musa spp.), from Fifa Mountains in Saudi Arabia. Moreover, we recorded the first observation of damage caused to bananas by C. sordidus in a banana farm in Jazan Province, southwestern Saudi Arabia, in March 2022. Molecular characterization using DNA sequences of the mitochondrial COI gene confirmed the morphological identification of C. sordidus. This discovery is considered a warning notice to prevent the potential establishment and spread of this dangerous pest in the banana cultivation regions in Saudi Arabia. Therefore, it is recommended that detection and monitoring of banana weevil should be undertaken in Saudi banana farms in order to restrict the dissemination of this weevil to other banana cultivation areas.

This study explores the influence of salinity on some physiological and biochemical pathways of four facultative halophytes (Abutilon pannosum, Indigofera oblongifolia, Senna italica, and Tetraena coccinea) along the southwest coast of Jeddah Governorate. Through a comparative analysis of these plants in both saline and non-saline environments, the study investigates chlorophyll levels, ion concentrations within the plants, the correlation with the SOS1 gene, and the impact of salinity on metabolic compounds. The overarching goal is to gain insights into the adaptive mechanisms of these specific plants to salt stress, providing valuable information for addressing global agricultural challenges associated with salinity. Throughout the study, metabolic, ionic, and molecular responses of these plants were scrutinized in both environments. The findings revealed elevated levels of Na+, K+, Ca2+, and Mg2+ in saline habitats, except for Na+ in I. oblongifolia. Despite increased concentrations of Chl b, variations were noted in Chl a and carotenoids in plants exposed to salt. Osmoregulatory patterns in A. pannosum and I. oblongifolia exhibited reversible changes, including heightened protein and proline levels in A. pannosum and decreased levels in I. oblongifolia, accompanied by alterations in amino acids and soluble carbohydrates. Senna italica displayed higher levels of osmolytes, excluding proline, compared to salinized environments, while T. coccinea exhibited lower levels of amino acids. The accumulation of Na+ emerged as the primary mechanism for ionic homeostasis in these plants, with non-significant decreases observed in K+, Mg2+, and Ca2+. Notably, an overexpression of the SOS1 gene (plasma membrane Na+/H+ antiporter) was observed as a response to maintaining ionic balance. Understanding these halophytes will be critical in addressing salinity challenges and enhancing crop tolerance to salinity.

The potential of Alhagi maurorum (Boiss.) aqueous extract (AME), used in traditional medicine for treatment or prevention of urolithiasis, to dissolve calcium oxalate stones in vitro was evaluated. In order to determine the litholytic potential of the extract, Calcium oxalate urinary stones were incubated during 12 weeks under continuous shaking in the presence of AME, Rowanix or NaCl 9 g/mL solution were used as controls. After the incubation period, the residual weight of the treated calculi was determined and the rate of dissolution was calculated. The medium pH variation was measured and changes in the calcium oxalate crystals at the stone surface were assessed using a scanning electron microscope (SEM). The results showed a significant dissolution effect for the extract on the kidney calculi during the experimentation period. At the end of the experiment, the percentages of calculi weight decrease were 41.23, 4.97 and 55.67% for the extract, NaCl solution and Rowanix, respectively. Gas Chromatography analysis revealed mainly the presence of the following phyto-compounds: Cyclopropenone, 2,3-diphenyl; 1-Nonadecanol; methyl-alpha-D-mannopyranoside; cis-9-Hexadecenal. These compounds unarguably play crucial roles in the health care system especially in cancer treatment and many other diseases including urolithiasis. The urinary stone dissolution, independent of medium pH, could be attributed to formation of complexes between the phytochemical compounds in the extract and the calculi.

Abstract Several reasons may underlie the dramatic increase in type2 diabetes mellitus. One of these reasons is the genetic basis and variations. Vitamin D receptor polymorphisms are associated with different diseases such as rheumatoid arthritis and diabetes. The aim of this study is to investigate the possible association of two identified mutations ApaI (rs7975232) and TaqI (rs731236). Eighty-nine healthy individuals and Fifty-six Type 2 Diabetic (T2D) patients were investigated using RFLP technique for genotyping and haplotyping as well. The distribution of Apal genotypes was not statistically significant among the control (P=0.65) as well as for diabetic patients (P=0.58). For Taql allele frequencies of T allele was 0.61 where of G allele was 0.39. The frequency distribution of Taql genotypes was not statistically significant among the control (P=0.26) as well as diabetic patients (P=0.17). Relative risk of the allele T of Apa1 gene is 1.28 and the odds ratio of the same allele is 1.53, while both estimates were < 1.0 of the allele G. Similarly, with the Taq1 gene the relative risk and the odds ratio values for the allele T are 1.09 and 1.27 respectively and both estimates of the allele C were 0.86 for the relative risk and 0.79 for the odds ratio. The pairwise linkage disequilibrium between the two SNPs Taq1/apa1 was statistically significant in control group (D = 0.218, D' = 0.925 and P value < 0.001) and similar data in diabetic groups (D = 0.2, D' = 0.875 and P value < 0.001). These data suggest that the T allele of both genes Apa1 and Taq1 is associated with the increased risk of type 2 diabetes. We think that we need a larger number of volunteers to reach a more accurate conclusion.

Specimens of Cnemidocarpa amphora were collected monthly from the Arabian Gulf from September 2017 to August 2018. Parts of their gonads were prepared for histological testing. The gonads' diameters varied by month. Each gonad contained many ovarian follicles with different morphologies and was surrounded by several testicular follicles. The ovarian and testicular follicles were separate, although the latter were always present near the former. Repeated measures ANOVA tests were conducted to investigate monthly changes in oocyte stages. In squirts measuring 12-13 cm in length, the gonads measured 30-50 mm from July to August; 20-25 mm from September to October; 15-20 mm from November to February; and 25-40 mm from March to June. Oogonia budded from the germinal epithelium with diameters of 20-30 µm. Previtellogenic oocytes measuring 70-120 µm were characterized by the deposition of small granules of protein around the nucleus, a continuous layer of follicular cuboidal epithelium, and scattered vacuoles in the ooplasm. The measurement of gonads and oocyte diameters were performed by image analysis (Image scope 2.3, Image Line, Inc.) and stage micrometer. The vitellogenic oocytes measured 130-220 µm and the follicular epithelium consisted of flattened and cuboidal layers. Beneath the vitelline membrane, scattered test cells appeared in the ooplasm and different granules of protein and MPS were deposited in the ooplasm. In the later phase, lipid droplets began to appear in the ooplasm. Yolk bodies formed after the impregnation of various granules together and the oocyte was ready to be shed. Before spawning, a yolk membrane appeared above the ooplasm. Post-vitellogenic oocytes, in which the homogeneity of ooplasm was restored, underwent gradual lysis and entered the atretic phase. Different stages of sperm development were present year-round in different follicles of the same squirt; hence, the testes were always mature.

Fish are considered one of the important sources of protein which are invaded by different parasites. This study aimed to shed light on monogenean parasites that infect fish within the family Sparidae in Saudi Arabia. A total of 30 Argyrops filamentosus specimens were collected from the Red Sea, the city of Jeddah (Saudi Arabia), and then examined for the presence of monogenean parasites. Parasitic species were isolated and studied morphologically using light microscopic examination and molecularly via the partial sequencing of the 28S rRNA gene. Only a monogenean parasitic species has been identified. This parasite is morphologically and morphometric compatible with previously Acleotrema maculatus Morsy, El-Fayoumi & Fahmy (2014), identified from Plectropomus maculatus in the Red Sea, Egypt. Phylogeny revealed that this putative diplectanid species nested well within a clade clustering Diplectanidae species, which along with morphological data, suggests it is a member of the genus Acleotrema. Query sequences showed identities of 98.92% for 28S rRNA (AF026118.1) of Acleotrema sp. This study reflects the first account of this genus as endoparasite taxa of the examined sparid fish, as well as providing novel DNA data for this species.

The Laughing Dove (Streptopelia senegalensis) is a columbid bird commonly found throughout most of Africa, the Middle East, Central Asia, India and Australia. Its preferred habitat includes scrubland, agricultural lands, and human settlements. Despite this species' extensive breeding range, very little information is available regarding its breeding ecology. The main objective of this study was to investigate the breeding features of the Laughing Dove in Taif City, Saudi Arabia between March and May 2018 and December 2018 and May 2019. The results indicate that this species constructs its nests at a mean height of 2.74 ± 0.1 m from the ground. The average number of nesting materials used for building a Laughing Dove nest was 109.3 ± 11.25. This study recorded that this species starts breeding in the middle of February. A clutch of two eggs comprised 72% of all the nests, with a mean of 1.75 ± 0.06 eggs per clutch. The average egg size was 25.27 ± 0.43 mm x 20.25 ± 0.19 mm, with an average egg volume of 5.01 ± 0.13 cm3. In total, 39.5% of the eggs hatched, and 64.7% of hatchlings reached the fledgling stage, resulting in an overall breeding success of 25.6%. This study suggested that nest desertion and predation were the major factors contributing to nest failure.

The olive leaf extract and olive leaf indicated a high potential for application in food additives and foodstuffs. It could be these bio-products useful and important in condition therapy related with oxidative stress and can use it to develop functional foods and to improve the food's shelf life. The olive leaf chemical composition of Oleaeuropaea L. grown from eljouf in Saudi Arabia, using solvents of increasing polarity cyclohexane, dichloromethane, chloroform, ethyl acetate, methanol and ethanol was determined using by GC/MS. Furthermore, the antioxidant activity (diphenylpicrylhydrazyl (DPPH), anti-aging, and anti-tuberculosis of olive leaf extracts were evaluated. The results indicated that extract of Oleaeuropaea L. has a considerable contains in polyphenols (hydroxytyrosol, oleuropein and their derivatives) regarding its antioxidant effects, the major components were detected by GC/MS in Olea dichloromethane extract are Hexadecanoic acid (15.82%), 7(4Dimethylaminophenyl)3,3,12trimethyl3,12dihydro6 Hpyrano[2,3c]acridin 6 one (11.21%), and in Olea chloroform extract are Hexatriacontane (12.68%), nTetratr iacontane (10.95%). The results concluded that the plant extract of chloroform showed no anti-aging activities and the lower anti-aging activities for cyclohexane extract, while, the Olea dichloromethane extract was the most active extract. The obtained data confirmed that the most active extract of anti-tubercolisis was for chloroform and ethyl acetate extract, while, anti-tubercolisis activity of ethanolic extract was the lower. The extract amount as well as the solvent polarity influence the inhibitory activity. A favorable connection was demonstrated inter alia the leaf extracts antioxidant activity and the content of total phenol.

Grewia tenax (Forssk.) Fiori (Malvaceae) grows in the Arabian Peninsula and is used for several medicinal purposes. To characterize the dermatological bioactivities of G. tenax in terms of its antimelanoma, antityrosinase and antioxidant activities. Cytotoxicity was assessed by cell proliferation and mitochondrial viability assays. Ability to inhibit mushroom tyrosinase and scavenge free radicals were evaluated by an enzymatic and DPPH scavenging microtiter assay, respectively. Phytochemical analyses were carried out using TLC, HPLC-UV and NMR. The chloroform extract shown significant cytotoxic activity in terms of mitochondrial viability (43 ± 14 µg/mL). We identified lupeol and b-sitosterol as the main active components for the tyrosinase inhibitory activity of the hexane extract. Scavenging activity of the DPPH· radical was confined to the water extract. Extracts from this plant have the potential to be used as a base in the development of cosmeceutical products intended to whiten skin or to combat radical-induced physiopathological processes.

Infectious Bronchitis Virus (IBV) is a highly contagious pathogen that causes a serious illness with global circulation. While there is extensive data available on the virus's existence and transmission in commercial chickens in Saudi Arabia, there is a lack of such information regarding guineafowls. Therefore, this study aimed to investigate possible IBV infection among guineafowls in the Al-Hassa Governorate of the Eastern Province, Saudi Arabia. Oropharyngeal and cloacal swabs were collected from several unvaccinated flocks of guinea fowls without respiratory clinical symptoms in November and December 2022, totaling 350 samples. Total RNA was extracted from the swab samples, and a conventional reverse transcription-polymerase chain reaction was employed to detect IBV. The results revealed varying amounts of IBV in oropharyngeal and cloacal swabs at different points in time, suggesting that IBV may be widely distributed among guineafowls without exhibiting any symptoms. These findings indicate that guineafowls could act as reservoirs, influencing the ecology and epidemiology of the disease. Notably, this study reports the first occurrence of IBV in the province of Al-Ahsa, highlighting that guineafowls have been naturally exposed to the virus. To support the development of effective vaccination techniques and control measures for the disease in Saudi Arabia, the recommendation for future research endeavors is conducting ongoing surveillance, viral isolation, sequencing, phylogenetic tree analysis, and serotype characterization of IBV in guineafowls.(AU)
