RESUMEN

During scaling of fermentations, choosing a bioreactor is fundamental to ensure the product's quality. This study aims to produce bioherbicides using Trichoderma koningiopsis fermentation, evaluating process parameters in an Airlift bioreactor. As a response, we quantified the production of enzymes involved in the bioherbicide activity (amylase, cellulase, laccase, lipase, and peroxidase). In addition, it evaluated the agronomic efficiency of the fermented extract optimized through tests that promoted soybean growth and nodulation, soybean seed germination, and in vitro phytopathogen control. As a result of optimizing the scaling bioprocess, it was possible to obtain an adequate fermentation condition, which, when applied to soybean seeds, had beneficial effects on their growth. It allowed the production of an enzyme cocktail. These results add a crucial biotechnological potential factor for the success of the optimized formulation in the Airlift bioreactor, in addition to presenting relevant results for the scientific community.

Abstract Background: Identifying risk factors in cardiovascular surgery assists in predictability, resulting in optimization of outcomes and cost reduction. Objective: This study aimed to identify preoperative and intraoperative risk predictors for prolonged hospitalization after coronary artery bypass grafting (CABG) surgery in the state of São Paulo, Brazil. Methods: A cross-sectional analysis using data from the REPLICCAR II database, a prospective, consecutive, multicenter registry that included CABG surgeries performed between August 2017 and July 2019. The primary outcome was a prolonged hospital stay (PHS), defined as a postoperative period exceeding 14 days. Univariate and multivariate logistic regression analyses were performed to identify the predictors with significance set at p <0.05. Results: The median age was 63 (57-70) years and 26.55% of patients were female. Among the 3703 patients analyzed, 228 (6.16%) had a PHS after CABG, with a median hospital stay of 17 (16-20) days. Predictors of PHS after CABG included age >60 years (OR 2.05; 95% CI 1.43-2.87; p<0.001); renal failure (OR 1.73; 95% CI 1.29-2.32; p <0.001) and intraoperative red blood cell transfusion (OR 1.32; 95% CI 1.07-2.06; p=0.01). Conclusion: Age >60 years, renal failure, and intraoperative red blood cell transfusion were independent predictors of PHS after CABG. The identification of these variables can help in multiprofessional strategic planning aimed to enhance results and resource utilization in the state of São Paulo.

RESUMEN

In this work, the Honduras Mahogany (Swietenia macropnylla King, SMK) seed husk was used as a novel biomass resource for producing activated carbon by physical activation. The texture characteristics and chemical characterization of resulting products were investigated in correlation with the process parameters. Based on the thermochemical properties of the SMK biomass, the process conditions were set to a rate of about 10 °C/min under nitrogen (N2) flow of 500 cm3/min heated to 500 °C, then switched to carbon dioxide (CO2) flow of 100 cm3/min in the specified activation conditions (i.e., temperature of 700-850 °C for holding times of 0-60 min). Our findings showed that the texture characteristics (i.e., surface area and pore volume) increased with an activation temperature increase from 700 to 800 °C for a holding time of 30 min but gradually decreased as the temperature increased thereafter. Similarly, the texture characteristics also indicated an increasing trend with the residence time extending from 0 min to 30 min but slightly decreased as the time was extended to 60 min. Therefore, the optimal activation conditions for producing SMK-based activated carbon should be set at 800 °C for a holding time of 30 min to obtain the maximal texture characteristics (i.e., BET surface area of 966 m2/g and total pore volume of 0.43 cm3/g). On the other hand, the chemical characteristics were analyzed by energy dispersive X-ray spectroscopy (EDS) and Fourier Transform infrared spectroscopy (FTIR), showing oxygen complexes contained on the hydrophilic surface of the resulting activated carbon.

RESUMEN

Due to the high content of phenolics and anthocyanins of Hibiscus sabdariffa L. tea and the sensibility of these bioactive compounds, this work aimed to optimize the obtention of microcapsules by spray-drying, using inulin as a carrier agent. Using a Box-Behnken Design, the effects of inlet temperature (130, 150, and 170 °C), feed flow rate (5, 10, and 15 mL min-1), and inulin concentration (5, 10, and 15 g L-1) were evaluated. It was possible to obtain pale-rose, slightly sweet instant powders with good total polyphenol content (1.12 mgGAE g-1) and anthocyanins encapsulation efficiency (32.3-60.6%), besides moisture (4.61-17.79%) and water activity (0.221-0.501), indicating physico-chemical and microbiological stability of the microcapsules. A simultaneous optimization with the desirability function was performed to maximize all the response variables analyzed, and the optimum conditions of 5 g L-1 of inulin, inlet temperature of 170 °C, and feed flow rate of 83 mL min-1 were found.

RESUMEN

Curcumin (CUR) is one natural bioactive compound acknowledged for diverse therapeutic activities, but its use is hindered by its poor bioavailability, fast metabolism, and susceptibility to pH variations and light exposure. Thus, the encapsulation in poly(lactic-co-glycolic acid), or PLGA, has been successfully used to protect and enhance CUR absorption in the organism, making CUR-loaded PLGA nanoparticles (NPs) promising drug delivery systems. However, few studies have focused beyond CUR bioavailability, on the environmental variables involved in the encapsulation process, and whether they could help obtain NPs of superior performance. Our study evaluated pH (3.0 or 7.0), temperature (15 or 35 °C), light exposure, and inert atmosphere (N2) incidence in the encapsulation of CUR. The best outcome was at pH 3.0, 15 °C, without light incidence, and without N2 usage. This best nanoformulation showed NP size, zeta potential, and encapsulation efficiency (EE) of 297 nm, -21 mV, and 72%, respectively. Moreover, the CUR in vitro release at pH values 5.5 and 7.4 suggested different potential applications for these NPs, one of which was demonstrated by the effective inhibition of multiple bacteria (i.e., Gram-negative, Gram-positive, and multi-resistant) in the minimal inhibition concentration assay. Besides, statistical analyses confirmed a significant impact of temperature on the NP size; in addition, temperature, light, and N2 affected the EE of CUR. Thus, the selection and control of process variables resulted in higher CUR encapsulation and customizable outcomes, ultimately enabling more economical processes and providing future scale-up guidelines.

RESUMEN

BACKGROUND: Four commercial starter cultures containing Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus (S1, S2, S3 and S4; S3 also contained Limosilactobacillus fermentum) were compared for fermentation, volatile flavor compounds, physicochemical parameters and microbiology, in yogurt prepared from three milk base formulations with increased protein (B1, B2 and B3). RESULTS: The fermentation patterns differed among starters, with Yoflex Mild 1.0 (S4) and SLB95 (S2) showing the longest fermentation time, depending on the formulation. At 21 days, S. thermophilus counts were similar among starters and higher than 8.52 log CFU mL-1 , for all yogurts. The highest counts (6.86 log CFU mL-1 ) for L. delbrueckii subsp. bulgaricus was found for S2 yogurts made from whey protein hydrolysate (B3). Minor water-holding capacity was detected for YF-L811 (S1) yogurts. Yoflex Harmony 1.0 (S3) starter containing Lim. fermentum produced a distinctive volatile profile characterized by aldehydes with respect to yogurts prepared with S1, S2 and S4, which were characterized by ketones. CONCLUSION: Results indicate the usefulness of carrying out studies similar to the present one to select the most appropriate process conditions depending on the desired product. © 2022 Society of Chemical Industry.

RESUMEN

With the evolution of percutaneous coronary intervention techniques and their greater safety, the use of radial access, the greater need to optimize resources in health management and the greater demand from patients for an agile service to solve their problems, the discharge on the same day after percutaneous coronary interventions have been gaining ground in the global scenario as a feasible strategy. The objective of this article was to demonstrate that there is already a scientific basis for this, which extends from the financial aspect, and the patient's experience, to the technical details related to the safety of the strategy. A structured protocol based on evidence and on the most recent recommendations from two major medical societies is also proposed. Selecting patients well, training the multidisciplinary team and educating the patient are fundamental parts for the success of the strategy.

RESUMEN

Anaerobic digestion (AD) is widely adopted for remediating diverse organic wastes with simultaneous production of renewable energy and nutrient-rich digestate. AD process, however, suffers from instability, thereby adversely affecting biogas production. There have been significant efforts in developing strategies to control the AD process to maintain process stability and predict AD performance. Among these strategies, machine learning (ML) has gained significant interest in recent years in AD process optimization, prediction of uncertain parameters, detection of perturbations, and real-time monitoring. ML uses inductive inference to generalize correlations between input and output data, subsequently used to make informed decisions in new circumstances. This review aims to critically examine ML as applied to the AD process and provides an in-depth assessment of important algorithms (ANN, ANFIS, SVM, RF, GA, and PSO) and their applications in AD modeling. The review also outlines some challenges and perspectives of ML, and highlights future research directions.

RESUMEN

The newly isolated Burkholderia gladioli BRM58833 strain was shown to secrete an alkaline lipase highly active and stable in organic solvents. Lipase production was optimized through the cultivation of the strain by solid-state fermentation in wheat bran. The lipase extraction conditions were also optimized. The low-cost extract obtained has shown a high hydrolytic activity of 1096.7 ± 39.3 U·gds-1 (units per gram of dry solids) against pNPP and 374.2 ± 20.4 U·gds-1 against triolein. Proteomic analysis revealed the optimized extract is composed of two esterases and three true lipases, showing a preference for long-chain substrates. The highest activity was obtained at 50 °C and pH 9. However, the extract maintained more than 50% of its maximum activity between pH 8.0 and 10.0 and throughout the whole temperature range evaluated (32-70 °C). The enzymes were inhibited by SDS, EDTA, ZnSO4 and FeCl3 and activated by FeSO4, MgCl2 and BaCl2. The lipases conserved their activity when incubated in solvents as acetonitrile, diethyl ether, n-heptane n-hexane, toluene, methanol and t-butanol. The resistance of these lipases to solvents and expressive thermostability when compared to other lipases, reveal their potential both in hydrolysis reactions and in synthesis of esters.

RESUMEN

The hydrothermal carbonization (HTC) optimization of oat husk was performed using a response surface methodology. Furthermore, anaerobic digestion (AD) of spent liquor and hydrochar addition were evaluated in the biomethane potential (BMP) test. Results found that temperature influences the most in the studied responses (i.e., mass yield (MY) and higher heating value (HHV)). Optimal hydrochar MY (53.8%) and HHV (21.5 MJ/kg) were obtained for 219.2 °C, 30 min, and 0.08 of biomass/water ratio. A successful prediction capability of the optimization approach was observed, archiving an error < 1% between predicted and validated responses. The BMP experiment showed the feasibility of spent liquor as a potential substrate to be treated by AD (144 NmLCH4/gCOD). Hydrochar boosted the methane production of spent liquor increasing up to 17% compared to digestion with no hydrochar addition. These findings provide new insights regarding oat husk valorization by integrating HTC and AD for energy production.

RESUMEN

As an essential trace element for animals, copper significantly contributes to the growth and health of animals. Compared to inorganic trace elements, organic trace elements are better supplements; notably, they are acquired through microbial transformation. Therefore, we screened for copper-enriched microorganisms from high copper content soil to obtain organic copper. Sodium diethyldithio carbamate trihydrate was applied as a chromogenic agent for determining micro amounts of intracellular copper through spectrophotometry. In total, 50 fungi were isolated after the successful application of the screening platform for copper-rich microbes. Following morphological and molecular biology analyses, the N-2 strain, identified as Aspergillus niger sp. demonstrated showed better copper enrichment potential than others. Notably, the strain tolerance to copper was nearly thrice that of Saccharomyces cerevisiae, up to 1600mg/L. The content of the organic bound copper was 22.84mg Cu/g dry cell. Using the Central Composite Design (CCD) response surface method, we optimized the fermentation condition (inoculation amount, 13%; temperature, 28(C; pH, 5.0). Compared to the original strain results under the single factor fermentation condition, we reported an increase by 24.18% under the optimized conditions. Collectively, these findings provide a reference for uncovering new and low-cost organic copper additives.(AU)

This study investigated the phycoremediation process from swine digestate integrated with photosynthetic biomass and biogas production in the context of circular economy. Effects of total ammonia nitrogen (TAN) and pH on biomass productivity and nutrients removal, using a central rotational composite design, were evaluated. pH showed a significant effect on biomass productivity and phosphate removal. The strain Chlorella sorokiniana (LBA#39) was able to tolerate up to 1300 mg TAN L-1 at neutral pH, with maximum biomass productivity of 198 mg DW L-1 d-1 and removal of 90 and 70 (%) of phosphate and nitrogen, respectively. The biomass harvested after phycoremediation from digestate showed high content of volatile solids (95.4%) and proteins (59.5%). Biochemical methane potential (BMP) from microalgae monodigestion was 292 ± 10 mLNCH4 gVSadd-1. The use microalgae biomass addition in the biodigestion process increased up to 32.1% in biogas production. It is an attractive approach to integrating raw materials into existing agro-industrial facilities and improving biogas production, adopting the concept of circular economy and mitigating greenhouse gas emissions.

ABSTRACT Polyaluminium chloride (PAC) is a pre-hydrolyzed inorganic polymeric coagulant that is being used widely in the treatment of water supply and wastewater by forming a hard and heavy Al(OH)3* precipitate, in addition to having a smaller effect on pH. The literature, however, is still scarce in correlating the chemistry and physics of the coagulant with process optimizations. The objective of this paper is to use PAC in studies of water supply treatability through coagulation, flocculation, and sedimentation. For this purpose, a mathematical modeling was initially performed to adjust turbidity and pH, and then remaining turbidity and apparent color coagulation diagrams were constructed with a pH of 4.0 to 9.0 and a dosage of 10 to 120 mg.L-1 of PAC (0.6 to 6.9 mg.L-1 of Al3+). Different gradients of velocity and mixing times for coagulation and flocculation were also studied. The coagulation diagrams indicated satisfactory results in the pH range between 7 and 9 with a dosage higher than 30 mg.L-1 of PAC (1.7 mg.L-1 of Al3+), and it is related to the minimum solubility of Al(OH)3*. Coagulation and flocculation optimizations achieved an improvement of up to 79% in the remaining turbidity, a reduction of up to 83% in the settler area or a reduction of up to 67% in PAC dosage, due to an increase in the size of the floc produced. An in-depth knowledge of the chemistry and physics of the process related to the use of PAC can, therefore, bring benefits to its use.

RESUMEN

Lovastatin, and its semisynthetic derivative simvastatine, has great medical and economic importance, besides great potential for other uses. In the last years, a deeper and more complex view of secondary metabolism regulation has emerged, with the incorporation of cluster-specific and global transcription factors, and their relation to signaling cascades, as well as the new level of epigenetic regulation. Recently, a new mechanism, which regulates lovastatin biosynthesis, at transcriptional level, has been discovered: reactive oxygen species (ROS) regulation; also new unexpected environmental stimuli have been identified, which induce the synthesis of lovastatin, like quorum sensing-type molecules and support stimuli. The present review describes this new panorama and uses this information, together with the knowledge on lovastatin biosynthesis and genomics, as the foundation to analyze literature on optimization of fermentation parameters and medium composition, and also to fully understand new strategies for strain genetic improvement. This new knowledge has been applied to the development of more effective culture media, with the addition of molecules like butyrolactone I, oxylipins, and spermidine, or with addition of ROS-generating molecules to increase internal ROS levels in the cell. It has also been applied to the development of new strategies to generate overproducing strains of Aspergillus terreus, including engineering of the cluster-specific transcription factor (lovE), global transcription factors like the ones implicated in ROS regulation (or even mitochondrial alternative respiration aox gen), or the global regulator LaeA. Moreover, there is potential to apply some of these findings to the development of novel unconventional production systems. KEY POINTS: • New findings in regulation of lovastatin biosynthesis, like ROS regulation. • Induction by unexpected stimuli: autoinducer molecules and support stimuli. • Recent reports on culture medium and process optimization from this stand point. • Applications to molecular genetic strain improvement methods and production systems.

RESUMEN

In the present paper, a computational methodology for the development of microdevices and microreactors are presented. The methodology was based on Computational Fluid Dynamics (CFD), i.e., the solution of transport equations governing the flow field. The methodology presents a step-by-step tutorial for modeling and simulation of such microdevices that can be used by beginner or experienced users. The proposed methodology was employed in two study cases: fluid mixing and fluid mixing accompanied by chemical reaction. Two new geometry designs were evaluated: a micrometric scale channel with triangular cross section (MT) and a millimeter range scaled channel (MTB). It is expect that the reported methodology contributes to the popularization of CFD usage among researchers, scientists and Microfluidic enthusiasts. Also, it can motivate future studies to focus firstly on geometry optimization by numerical simulations, providing a faster and economical way to develop microdevices. •A CFD-based methodology was presented for the development of microdevices and microreactors•The methodology can be used in distinct fluid mixing and chemical reaction systems•Numerical simulation allows a faster microdevice development procedure with costs reductions.

RESUMEN

A novel tannase and gallic acid-producing Penicillium rolfsii (CCMB 714) was isolated from cocoa leaves from the South of Bahia. The influence of nutritional sources and the simultaneous effect of parameters involved in the fermentation process were available. Tannase (9.97 U mL-1) and gallic acid (9 mg mL-1) production were obtained in 48 h by submerged fermentation in non-optimized conditions. Among the carbon sources, tested gallic acid and tannic acid showed the highest tannase production (p<.05) when compared with methyl gallate and glucose. After optimization using the temperature and tannic acid concentration as variables with the Central Compound Rotational Design (CCRD), the maximal tannase production (25.6 U mL-1) was obtained at 29.8 °C and 12.7%, respectively, which represents an increase of 2.56 times in relation to the initial activity. The parameters optimized for the maximum production of gallic acid (21.51 mg mL-1) were 30 °C and 10% tannic acid. P. rolfsii CCMB 714 is a new strain with a high tannase and gallic acid production and the gallic acid produced is very important, mainly for its applications in the food and pharmaceutical industry.
