RESUMEN

Metal nanoparticles, especially silver, have been used in various medical scenarios, due to their excellent antimicrobial effects. Recent studies have shown that AgNPs do not exert mutagenic effects on target bacteria, but the degree to which they compromise eukaryotic genomes remains unclear. To study this, we evaluated the mutagenic effects of AgNPs on the fission yeast Schizosaccharomyces pombe ATCC-16979, of which ∼23% genes are homologous to human ones, at single-nucleotide resolution, and whole-genome scale by running 283 mutation accumulation lines for ∼260,000 cell divisions in total. We also explored the action and mutagenesis mechanisms using differential gene-expression analysis based on RNAseq. Upon AgNPs treatment, the genomic base-substitution mutation rate of S. pombe at four-fold degenerate sites increased by 3.46×, and small indels were prone to occur in genomic regions that are not simple sequence repeats. The G:C → T:A transversion rate was also significantly increased, likely mostly from oxidative damage. Thus, in addition to their antimicrobial potency, AgNPs might pose slight genotoxicity threats to eukaryotic and possibly human genomes, though at a low magnitude.

RESUMEN

After successfully launching two space laboratories, namely, Tiangong-1 and Tiangong-2, China has announced her next plan of constructing the Chinese Space Station (CSS) in 2022. The CSS will provide not only platforms for Chinese scientists to carry out experimental studies in outer space but also opportunities for open international cooperation. In this article, we review the development of China's manned space exploration missions and the preliminary plan for CSS. Additionally, China has initiated space radiation research decades ago with both ground-based simulation research platform and space vehicles and has made noticeable progresses in several aspects. These include studies on human health risk assessment using mammalian cell cultures and animals as models. Furthermore, there have been numerous studies on assessing the space environment in plant breeding.

In the present study, sodium azide (SA) toxicity and the anti-mutagenic effects of different algal extracts at 0.1% and 0.2% concentrations were studied on the mitotic index (MI), chromosomal and nuclear aberrations using Allium cepa L. root assay. Moreover, phytochemical screening of photosynthetic pigments, antioxidants compounds, total antioxidant, DPPH scavenging activity, polysaccharides, and phenolic contents were done for two red seaweeds (Laurencia obtusa (Hudson) Lamouroux and Polysiphonia morrowii Harvey) and for one brown seaweed (Dictyopteris delicatula Lamouroux). Treatment with 300 µg/ml sodium azide (SA) induced the highest number of aberrations in A. cepa root. A highly significant decrease in the MI appeared after treatment with SA, whereas its value increased following different algal extracts treatments. The highest anti-mutagenic inhibition activity of Dictyopteris delicatula added at 0.2% concentration was 72.96%, 69.84%, 56.89% and 43.59% with the algal polyphenol, polysaccharide, aqueous and methanol extract treatments, respectively. The different algal extracts minimized the genotoxicity and exhibited anti-mutagenic potential against SA in a dose-dependent manner. Phytochemical studies showed that Dictyopteris delicatula contained the highest total phenol, chlorophyll-a and carotenoid quantity. Moreover it exhibited the highest total antioxidant and DPPH scavenging activities. Total polysaccharides and the weight percentage of sulphated polysaccharides were relatively higher in Polysiphonia morrowii followed by Laurencia obtusa. Hydroquinone and bromophenol were detected only in the studied brown and red seaweeds, respectively. Polysiphonia morrowii and Laurencia obtusa contained the highest quantity of galactose, rhmnose and xylose, while Dictyopteris delicatula contained fucose and mannitol as main monosaccharide units. In conclusion, the studied seaweeds may be considered as rich sources of natural antioxidants. Meanwhile the investigated different algal extracts can minimize the genotoxicity in a dose-dependent manner and exhibit anti-mutagenic potential against the mutagenic substance sodium azide.

CONTEXT: Insecticides are used to control pests. Cypermethrin and fenvalerate are widely used pirethroid insecticides in the world. Rosa canina L. (Rosaceae) is used as a traditional medicinal plant against viral infections and disorders of the kidneys and urinary tract due to its high vitamin C level. OBJECTIVE: The genotoxic effects of cypermethrin and fenvalerate were examined with the micronucleus (MN) test. Then, we determined the ability of the water (RC(wtr)) and ethanol (RC(eta)) extracts of rosehip (R. canina) to overcome the possible genotoxic effects of the insecticides. MATERIALS AND METHODS: Preliminary studies determined that the application concentrations were 20, 30, 40, and 50 ppm for cypermethrin, 25, 50, 75, and 100 ppm for fenvalerate, and 100 ppm for rosehip extracts. DMSO (dimethyl sulphoxide) (1%) and 1 mM EMS (ethyl methanesulfonate) were used as negative and positive control groups, respectively. The application groups belonging to insecticides and plant extracts were added to culture tubes including chromosome B medium and peripheral blood for MN test. RESULTS: The MN frequencies were found 0.725 in the negative control group, 2.700 in the positive control groups, 1.275 in the highest application group of cypermethrin, and 1.600 in the highest application group of fenvalerate. The MN frequencies in cypermethrin + RC(wtr), cypermethrin + RC(eta), fenvalerate + RC(wtr), and fenvalerate + RC(eta) application groups were, respectively, determined as 1.000, 1.075, 1.225, and 1.275. CONCLUSION: According to the results, cypermethrin and fenvalerate have genotoxic effects, the water and ethanol extracts of rosehip reduced the genotoxicity of the both insecticides.

We report a case of AML-M1 with 5q aberration at diagnosis. The patient was treated with high-dose chemotherapy (HDCT). After remission induction, he received allogenic peripheral blood stem cell transplantation (PBSCT) from an HLA-match donor brother. The successive follow-up conventional cytogenetics investigations in remission after HDCT and PBSCT revealed cytogenetic remission. The most interesting observation in this case is that relapsed marrow revealed the emergence of an entirely new, highly aberrant, unrelated clone with unusual translocations t(6;17)(p23;p11.2),+8,der(8)dup inv(8)(q23qter), t(10;19)(q26;q13.3) 4½ months after PBSCT. Our findings suggest the possibility of a mutagenic effect of HDCT and myeloablative intense chemotherapy before PBSCT that could have induced a genetic lesion in the recipient's genetically unstable stem cells in an environment of immunosuppression. The highly complex nature of the clone and the rapid clonal evolution indicates the possibility of selective pressure with proliferative advantage.
