RESUMEN
IgY antibodies are found in the blood and yolk of eggs. Several studies show the feasibility of utilising IgY for immunotherapy and immunodiagnosis. These antibodies have been studied because they fulfil the current needs for reducing, replacing, and improving the use of animals. Affinity and avidity represent the strength of the antigen-antibody interaction and directly influence antibody action. The aim of this review was to examine the factors that influence the affinity and avidity of IgY antibodies and the methodologies used to determine these variables. In birds, there are few studies on the maturation of antibody affinity and avidity, and these studies suggest that the use of an adjuvant-type of antigen, the animal lineage, the number of immunisations, and the time interfered with the affinity and avidity of IgY antibodies. Regarding the methodologies, most studies use chaotropic agents to determine the avidity index. Studies involving the solution phase and equilibrium titration reactions are also described. These results demonstrate the need for the standardisation of methodologies for the determination of affinity and avidity so that further studies can be performed to optimise the production of high avidity IgY antibodies.

INTRODUCTION: Sleeve gastrectomy is one of the main techniques used to treat severe obesity. The study of the immunohistochemical expression of ghrelin in the gastric mucosa has already been related to weight loss and can be a promising method to predict the surgical outcome. PURPOSE: To analyze the immunohistochemical expression of ghrelin in the gastric mucosa and its correlation with weight loss, comorbidities, and inflammatory changes after sleeve gastrectomy. METHODS: Thirty-five patients submitted to sleeve gastrectomy were evaluated, 29 of whom were female (82.9%), with a mean age of 35.2 years and an average body mass index of 38.1 kg/m2. Endoscopic samples of the mucosa were collected, whose ghrelin expression was evaluated in a semi-quantitative way through the stained antibody area. These data were correlated with weight loss at 3, 6, and 12 months and with the control of comorbidities, and inflammatory alterations. RESULTS: The average total weight loss (TWL%) was 17.7, 26.4, and 32.1%, respectively, at 3, 6, and 12 months. A negative correlation was found between the immunohistochemical expression of ghrelin in the endoscopic biopsy of the fundus and weight loss at 3 (s = - 0.536; p = 0.001) and 6 months (s = - 0.339; p = 0.047). CONCLUSION: The immunohistochemical expression of ghrelin in the mucosa of the gastric fundus was negatively correlated with early weight loss after sleeve gastrectomy.

In the present work, venoms from five species of the genus Agkistrodon were evaluated in terms of their enzymatic (Phospholipase A2 and caseinolytic) and biological (edema forming, hemorrhagic, procoagulant and lethal) effects. Horses were used to produce monovalent hyperimmune sera against each of three venoms (A. bilineatus, A. contortrix and A. piscivorus) and their neutralizing potency, expressed as Median Effective Dose (ED50), was determined against the venoms of all five species. In terms of PLA2 and caseinolytic activities, all venoms are extremely homogeneous. PLA2 activity is high, while caseinolytic activity is low when in contrast with that of the rattlesnake Crotalus simus. On the other hand, biological activities showed marked interspecific differences, particularly between the species from Mexico and those from the United States. Mexican species displayed higher edema-forming, hemorrhagic and lethal effects than US species, while none of the species studied presented procoagulant activity. All three monovalent hyperimmune sera showed good neutralizing potency against the analyzed venoms. Nonetheless, we observed relevant immunochemical differences among the venoms using ELISA and Western Blot assays. We conclude that the venoms of A. piscivorus (USA) and A. bilineatus would be ideal to use as immunogens for the production of a polyvalent antivenom with good neutralizing potency against the venoms of all the species of the genus.

SUMMARY Primary osteosarcoma of the breast (POB) is an extremely rare and aggressive tumor. Differential diagnosis of POB includes osteosarcoma of the chest wall and metaplastic breast carcinoma. Imaging tests that exclude the existence of a direct connection between the tumor and chest wall, as well as histopathological and immunohistochemical studies that rule out the presence of an epithelial component are required for the diagnosis of POB. We report a case of a 69-year old woman with POB. Imaging and pathological findings are presented. Therapeutic approach is discussed in the light of current knowledge, including potential complications.
A abordagem terapêutica é discutida à luz do conhecimento atual, incluindo potenciais complicações.

Objectives: To evaluate the expression of sphingosine kinase 1 (SPK1) in the bladder wall in patients with neurogenic lower urinary tract dysfunction and its association with clinical, urodynamic and pathological features. Materials and Methods: The expression of SPK1 was studied in bladder wall specimens obtained from cystectomy using immunohistochemistry in ten patients with spinal cord injury (n=8) or multiple sclerosis (n=2) with urodynamically proven neuropathic bladder dysfunction, and in controls (n=5). Inflammation and fibrosis were analysed with histological criteria and SPK1 expression was determined by individual immunohistochemical staining. Results: Significant increased SPK1 urothelial immunoreactivity was shown in patients compared to control group (p=0.03). By contrast, SPK1 immunoreactivity in patients was significantly decreased in the sub-urothelium, muscles and nerves, p=0.02; 0.01 and 0.003, respectively. Patients with neurogenic detrusor overactivity (NDO) had higher SPK1 urothelium expression than those without any DO (p=0.04). Conclusions: SPK1 is expressed in the human bladder wall, specifically the urothelium, in bladder specimens from patients with NDO. The role of SPK1 in the pathophysiology of NDO needs further elucidation.

ABSTRACT Introduction: Stromal cells interact with tumor cells and play an important role in cancer invasion and metastasis. Tumor-associated macrophages (TAMs) can exhibit M1 phenotype, important immune effector cells, or M2 phenotype, able to suppress the function of M1 macrophages and influence angiogenesis and tissue repair. The CD68 antibody recognizes M1 and M2 macrophages, whereas CD163 antibody is specific only to identify M2 macrophages. Objective: To investigate the presence of TAMs in a number of cases squamous cell carcinoma (SCC) of the tongue by associating it with the occurrence of metastasis. Material and methods: Immunohistochemistry was used to evaluate the immunopositivity for CD68 and CD163 in 27 cases of SCC of the tongue. Results: The percentage of CD68 positive macrophages was higher than CD163 positive macrophages in all specimens studied. Comparing CD163 and CD68 immunostaining in the studied groups, it was observed that cases without metastasis had a higher percentage of CD68 positive cells in relation to CD163 positive cells, which was statistically significant. Conclusion: Considering these results, there was a predominance of M1 macrophages in SCC of the tongue cases without metastasis, suggesting the influence of these cells in clinical behavior of the lesion.
OBJECTIVE: CD20 expression was reported at different rates in patients with multiple myeloma. The importance of this B-cell antigen for plasma cells is still unknown. This study aimed to investigate CD20 expression of myeloma cells in bone marrow, and any relationship between the stage of disease, isotype and clinical features. METHODS: Sixty-one patients who were admitted to the hematology clinic of the Adnan Menderes Medical School with the diagnosis of multiple myeloma according to the criteria of the "International Myeloma Working Group" were enrolled in this study. Age, gender, Durie-Salmon stage, history of autologous hematopoietic stem cell transplantation, and the distribution pattern and positivity of CD20 expression on multiple myeloma cells in bone marrow were evaluated. The Mann-Whitney U and chi-square tests were used for statistical analysis with a p-value<0.05 being accepted as statistically significant. RESULTS: Thirty patients (48.9%) had positive scores for CD20 with the distribution pattern being most likely interstitial in 55.6% of the cases. There was no statistically significant difference between immunohistochemical positivity for CD20 expression on multiple myeloma cells, immunoglobulin type, and the stage of disease. CONCLUSION: The combination of immunohistochemical studies with flow cytometry may reveal the importance of CD20 positivity in patients with multiple myeloma more clearly.

Desenho do estudo: Estudo prospectivo experimental. .
Introduction: Some clinical trials revealed a correlation between increased serum estrogen and nasal symptoms or inflammatory changes in nasal mucosa. Estrogen receptors tend to be controlled by a negative feedback, to avoid a deleterious stimulus over several body functions while in hyperestrogenic periods. This study proposes a hypothesis where mechanisms regulating expression of estradiol receptors in nasal mucosa are absent in some patients, and their concentration remains steady even in periods of high serum hormonal concentration, potentially leading to local estrogenic symptoms in nasal mucosa. Study design: This was an experimental prospective study. Aim: To determine whether estrogen levels induce the reduction of the number of estrogen receptors in the nasal mucosa. Methods: In the present study, 30 adult male guinea pigs were subjected to a biopsy of the middle nasal turbinate and received 0.5 mL of estradiol cypionate intraperitoneally for 30 consecutive days. Afterwards, samples from contralateral middle turbinate were obtained. Immunohistochemical analysis of estrogen receptors were performed pre- and post-treatment. Results: The post-treatment group showed reduction of receptor expression when compared to the pre-treatment group. (p = 5.2726-5). Conclusion: A reduction in the expression of the nasal estrogen receptor was observed after 30 days of estradiol administration. .

Partial hepatectomy (PH) alters serum concentrations of substances involved in cellular proliferation, leading to the compensatory liver hyperplasia. Furthermore, angiogenesis is mainly stimulated by vascular endothelial growth factor (VEGF) and is a fundamental requirement either in liver regeneration or in tumours growth. This study looks at the expression of VEGF, DNA synthesis (DNAs) and mitotic activity (MA) in hepatectomised (H) and hepatectomised-tumour bearing (HTB) mice throughout a 24 h period. Adult male mice were sacrificed every 4 h from 26 to 50 h post-hepatectomy. H mice show a circadian rhythm in VEGF expression with a maximum value of 2.6 ± 0.1 at 08/46 h of day/hours posthepatectomy (HD/HPH); in DNAs, the maximum value was 3.4 ± 0.3 at 16/30 (HD/HPH) and in MA it was 2.3 ± 0.01 at 12/50 (HD/HPH). In HTB animals the peak of VEGF expression appears at 16/30 (HD/HPH) with a maximum value of 3.7 ± 0.1, the peak of DNAs was at 00/38 (HD/HPH) with a value of 4.6 ± 0.3 and the maximum value of MA of 08/46 (HD/HPH) with a value of 3.01 ± 0.3. We can conclude that the presence of the tumour induces modifications in the intensity and the temporal distribution of the circadian curves of VEGF expression, DNAs and MA of hepatectomised animals.

To evaluate the process of alveolar repair in ovariectomized rats after implantation with Biosilicate. Materials and Methods: Sixty rats were divided into four groups (n = 15) according to treatment: Group 1 - rats underwent sham surgery with alveoli filled with blood clot; Group 2 - rats underwent sham surgery with alveoli filled with crystalline Biosilicate; Group 3 - ovariectomized rats with alveoli filled with blood clot; Group 4 - ovariectomized rats with alveoli filled with crystalline Biosilicate. After 7, 14 and 28 days the animals were sacrificed and histomorphometric analysis (histology and histometric) and immunohistochemical (Osteopontin and Osteocalcin) were measured . The comparison of the results was performed using the statistical program Assistat 2013 for Windows by Analysis of Variance and Mann-Whitney tests, with significant results being found when p ≤ 0:05 . Results: During the 7- day period, Groups 1 and 3 showed better and significantly higher results in bone formation than Groups 2 and 4 (p<0.05). After 14 days the statistical differences remained between Groups 1 and 3 and Groups 2 and 4 (p<0,05). At 28 days, the statically difference remained significant between the same Groups, while the groups implanted with Biosilicate presented about 60% of the socket filled by new bone. Immunohistochemistry revealed expression of proteins osteocalcin and osteopontin, especially in animals subjected to ovariectomy surgery. Conclusions: Groups 1 and 3 were those with the best results throughout the study, with the crystalline Biosilicate presenting favorable result in bone repair without bioglass resorption. The immunohistochemical analysis showed that animals subjected to ovariectomy had a higher signaling in Ostecalcina and Osteopontin proteins...

We describe a rare case of primary pleural synovial sarcoma in a 27-year-old man with a 4-month history of dry cough and left-sided chest pain. A CT scan showed a large cystic mass in the left pleural cavity. The patient underwent two video-assisted thoracoscopic biopsies and the diagnosis of synovial sarcoma of the pleura was established. After neoadjuvant chemotherapy, which resulted in a partial response, the tumor was completely resected with extrapleural pneumonectomy. Pathological findings showed less than 5% of viable cancer and free surgical margins. The patient is clinically well 24 months after surgery, with no evidence of recurrent disease.
Problem: To date, there are only three reports from Colombia about the malaria diagnostic testNow ICT Malaria Pf / Pv ® (NowICT). The results from these studies showed major differences insensitivity and specificity. Objective: To evaluate the diagnostic performance of NowICT compared to thick smear for the diagnosis of Plasmodium infection in matched blood samples from mothers (maternal peripheral blood), umbilical cord and placenta. Methods: We used a closed (blinded/masked) and parallel design for the evaluation of a diagnostic test. The sample size was calculated with statistical and epidemiological parameters; this consisted of 131 thick smears from maternal peripheral blood. Blood samples from placenta and umbilical cord were also studied (386 samples tested in total). Results: The sensitivity of Now ICT for detection of P.vivax was below 70% in any of the samples (maternal blood, placental blood or cord blood). The specificity was greater than 99%. Values for P. falciparum infection were not calculated since too few cases were detected. Conclusions: Now ICT Malaria Pf / Pv ® is not a useful diagnostic tool in Colombia since the sensitivity for the most frequent species in the country, P. vivax, is poor. This interpretation is consistent with the WHOs general conclusions about the state of development of rapid diagnostic tests for malaria.

Quanto ...
OBJECTIVE: To compare the clinical and histological outcomes of the repair of induced abdominal wall defects with Parietex® and Optilene Mesh Elastic® + Safil® in direct contact with abdominal viscera (intraperitoneal position, IPOM). METHODS: Sixteen rabbits were allocated into two groups with 8 animals each, corresponding to evaluation on the 30th and 60th postoperative days. All animals were submitted to two standardized symmetric abdominal wall defects, comprising all muscular layers and the peritoneum, followed by repair through the intraperitoneal placement of two different meshes. The experimental design allowed each animal to be its own control. Macroscopic aspects evaluated were: surgical site infection, mesh erosion, suture insufficiency, fistulae, intra-abdominal infection and adhesion formation. Microscopic parameters analyzed consisted of collagen deposition and the immunohistochemical healing process. RESULTS: The formation of intraperitoneal adhesions and the other macroscopic outcome measures evaluated did not present significant statistical differences between the two meshes, neither did type I and III collagen deposition and MMP-1 and MMP-8 antibody expression. MMP-13 antibody exhibited significantly higher expression on the 30th postoperative day with Optilene+Safil and on the 60th day with Parietex. CONCLUSION: Due to the similar results obtained, both macroscopically and microscopically, the meshes can be considered equivalent with respect to the healing of surgical wounds in abdominal wall defects in rabbits.
