In the present study, we estimated the levels of infestation of the main fish species that are hosts for two Triaenophorus species: T. crassus and T. nodulosus. The prevalence of T. crassus and T. nodulosus infestations in the intestine of their definitive host-pike Esox lucius was similar (71.0% and 77.4%, respectively). At the same time, the prevalence of T. crassus infestation in muscle tissue was significantly different between the second intermediate hosts, Coregonus lavaretus pidschian (31.4%) and Cor. l. pravdinellus (91.2%), due to considerable differences in their diets. For T. nodulosus, we found significant variations in the levels of prevalence among the second intermediate hosts-100% for Lota lota, 81.8% for Cottus sibiricus 31.9% for Thymallus arcticus, and 24.5% for Perca fluviatilis-that we also explained using different diets. Moreover, analysis of the symmetry of parasite infestations did not reveal any asymmetry between the number of cysts in the left and right body surfaces of the "planktivorous" form/species of whitefish, whereas in the ''benthivorous", an asymmetry of parasite infestations was found.

Determining the physiological effects of parasites and characterizing genes involved in host responses to infections are essential to improving our understanding of host-parasite interactions and their ecological and evolutionary consequences. This task, however, is complicated by high diversity and complex life histories of many parasite species. The use of transcriptomics in the context of wild-caught specimens can help ameliorate this by providing both qualitative and quantitative information on gene expression patterns in response to parasites in specific host organs and tissues. Here, we evaluated the physiological impact of the widespread parasite, the pike tapeworm (Triaenophorus nodulosus), on its second intermediate host, the Eurasian perch (Perca fluviatilis). We used an RNAseq approach to analyse gene expression in the liver, the target organ of T. nodulosus plerocercoids, and spleen which is one of the main immune organs in teleost fishes. We compared perch collected from multiple lakes consisting of individuals with (n = 8) and without (n = 6) T. nodulosus plerocercoids in the liver. Results revealed a small number of differentially expressed genes (DEGs, adjusted p-value ≤0.05) in both spleen (n = 22) and liver (n = 10). DEGs in spleen consisted of mostly upregulated immune related genes (e.g., JUN, SIK1, THSB1), while those in the liver were often linked to metabolic functions (e.g., FABP1, CADM4, CDAB). However, Gene Ontology (GO) analysis showed lack of functional enrichment among DEGs. This study demonstrates that Eurasian perch displays a subtle response at a gene expression level to T. nodulosus plerocercoid infection. Given that plerocercoids are low-metabolic activity transmission stages, our results suggest that moderate T. nodulosus plerocercoid infection most likely does not provoke an extensive host immune response and have relatively low physiological costs for the host. Our findings illustrate that not all conspicuous infections have severe effects on host gene regulation.

PURPOSE: Studies of proteins expressed in the morphological structures of the parasite are necessary for elucidating the biological functions of unknown proteins and understanding the molecular basis of parasitism. The research aim was to investigate the spatial distribution of major proteins in scolex, immature and gravid proglottids of Triaenophorus nodulosus and Triaenophorus crassus. METHODS: Protein extracts of worm body parts were analyzed using two-dimensional difference gel electrophoresis (DIGE) and mass spectrometry. RESULTS: Comparison of the protein repertoire of the adult worm and the encysted plerocercoid revealed differences between the worm body parts, life stages and parasite species. The content of proteins associated with the cytoskeleton and musculature (actin, myosin regulatory light chain, and tropomyosin 2) decreased with distance from the scolex. Mature proglottids were rich in transforming growth factor-beta-induced protein, propionyl-CoA carboxylase, glutamate dehydrogenase and beta-tubulin. Interspecific variation in T. nodulosus and T. crassus was found in the content of the myosin, paramyosin, the major vault protein and an uncharacterized secreted protein TRINITY_DN24645. Differential expression of TRINITY_DN24645, paramyosin and tropomyosin 2 was found between plerocercoids and adult worms. CONCLUSION: The present study provides the first characteristics of the spatial distribution of the major proteins of T. crassus and T. nodulosus. Comparison of the protein composition of plerocercoids and adult parasites indicates a significant similarity in the proteomic organization of Triaenophorus sp. in the second intermediate and final hosts. The gradual change in the morphological organization of tapeworms in the longitudinal direction coincided with the expression of some structural and metabolic proteins.

Lake Teletskoye (West Siberia, Russia) is inhabited by a sympatric pair of whitefish, with each member of the pair being characterized by different feeding habits. Coregonus lavaretus pidschian (Gmelin, 1789) is a large 'benthivorous' form, while C. l. pravdinellus (Dulkeit, 1949) is a small 'planktivorous' form. Fish were collected from the end of August to the middle of September in 2017 and 2019-2020 in the north part of Lake Teletskoye. For the 'benthivorous' form the prevalence, intensity and abundance of T. crassus ranged from 22.4% to 51.9%, 1.9-2.8 and 0.4-1.3, respectively, whereas the same indices for the 'planktivorous' form ranged from 94.7% to 97.5%, 4.2-4.8 and 4.0-4.7, respectively. The level of prevalence of infection and abundance of T. crassus in muscle was relatively stable among studied years for both forms. The level of prevalence was higher in the years 2019 and 2020 than in 2017 for the 'benthivorous' form, whereas for the 'planktivorous' form this index did not change during the studied years. For the first time, a partial sequencing of the cox1 gene (593 bp) for T. crassus was sequenced. All 15 plerocercoids of T. crassus were represented by four haplotypes.

The European perch Perca fluviatilis L. serves as a host of different endohelminths of Trematoda, Cestoda, Nematoda, and Acanthocephala. Its natural range covers freshwater basins throughout much of Europe, including the Danube. Since information about endohelminths of European perch from this international river basin has been rather sporadic, the parasitological examinations of 700 perch from the central region of the Danube river basin in Slovakia were performed in October 2017 and April 2018. The larval stages of Triaenophorus nodulosus (Cestoda) were found in cysts located in the perch liver and adults of Proteocephalus percae (Cestoda) were isolated from the intestine. The larval stages of Eustrongylides sp. (Nematoda) and metacercariae of Clinostomum complanatum (Trematoda), both potential causative agents of fish-borne zoonoses, were found in the musculature. Spatial and seasonal differences in the occurrence of currently detected helminths were discussed with data on biological and environmental conditions of particular sampling site.

Natural infection of 2 to 6-year-old perch with the cestode parasites Triaenophorus nodulosus was shown to have minor effects on the studied components of the antioxidant defense system, nucleic acids degradation, and carbohydrate metabolism enzymes in the liver of the fish. The level of infection of 1-4 parasite larvae per fish observed in wild population of perch was shown to be moderate in terms of its effect on the health of the host fish. The activity of hepatic enzymes ß-galactosidase, ß-glucosidase, cathepsin D, and glutathione S-transferase showed different responses in infected males and females, which indicates different potential resistance of fish to the stress exposure between genders.

Although parasites are expected to affect their host's fitness, quantitative proof for impacts of parasitism on wild populations is hampered by confounding environmental factors, including dietary resource. Herein, we evaluate whether the physiological conditions of European perch (Perca fluviatilis) in three large peri-alpine lakes (Geneva, Annecy, and Bourget) depend on (a) the nutritional status of the juvenile fish, as revealed by stable isotope and fatty acid compositions, (b) the prevalence of the tapeworm Triaenophorus nodulosus, a parasite transmitted to perch through copepod preys, or (c) interactive effects of both factors. At the scale of lake populations, the deficit in growth and fat storage of juvenile perch during their first summer coincides with a high parasite prevalence and also a low quality of dietary resource. Yet, at the individual level, parasites had no evident effect on the growth of the juvenile perch, while impacts on fat storage appeared only at the highest prevalence of the most infected lake. Fatty acid and stable isotope analyses of fish tissue do not reveal any impact of T. nodulosus on diet, physiology, and feeding behaviour of fish within lakes. Overall, we found a low impact of parasitism on the physiological condition and trophic status of juvenile perch at the end of their first summer. We find instead that juvenile perch growth and fat storage, both factors tied to their winter survival, are under strong nutritional constraints. However, the coinciding nutritional constraints and parasite prevalence of perch juveniles in these three lakes may result from the indirect effect of lake nutrient concentrations, which, as a major control of zooplankton communities, simultaneously regulate both the dietary quality of fish prey and the host-parasite encounter rates.

The unusual yellow-finned morph of European perch Perca fluviatilis found in Lake Constance suffers more severely from macroparasite infections, including the tapeworm Triaenophorus nodulosus and the gill worm Ancyrocephalus percae, than conspecifics elsewhere. Microsatellite analysis of yellow-finned P. fluviatilis and red-finned variant recently discovered in Lake Constance revealed significant genetic differentiation. Red-finned P. fluviatilis and fish with mixed fin colour, suggested backcrosses between red and yellow-finned colour morphs, exhibit better resilience to parasite infection, suggesting that the inability of the yellow-finned morph to reject macroparasites may have a genetic basis.
