RESUMEN

Gold nanoparticles (AuNPs) are biocompatible nanomaterials with potential application in the food industry. The safety of AuNPs oral consumption remains inconclusive, and information on possible long-term toxicity is limited. The current study aimed to evaluate the subchronic oral toxicity of AuNPs in male and female Institute of Cancer Research (ICR) mice. Citrate-coated spherical AuNPs with 53 nm diameters were prepared and orally administered to the mice. No mortality or clinical abnormalities were observed following daily administration of AuNPs at the dosages of 0.2, 2, and 20 mg/kg for 90 days. There was no significant difference in body weight or the relative organs' weights between the control and AuNPs-treated mice. No gross abnormalities or histopathological changes were observed except that the male mice treated with high dose (20 mg/kg AuNPs) showed minor infiltration in the kidneys, and female mice showed a reduced A/G ratio and elevated platelet indices. Overall, the 90-day long-term oral consumption of AuNPs did not cause significant toxicity in mice.

The gold standard of histopathology for the diagnosis of Barrett's esophagus (BE) is hindered by inter-observer variability among gastrointestinal pathologists. Deep learning-based approaches have shown promising results in the analysis of whole-slide tissue histopathology images (WSIs). We performed a comparative study to elucidate the characteristics and behaviors of different deep learning-based feature representation approaches for the WSI-based diagnosis of diseased esophageal architectures, namely, dysplastic and non-dysplastic BE. The results showed that if appropriate settings are chosen, the unsupervised feature representation approach is capable of extracting more relevant image features from WSIs to classify and locate the precursors of esophageal cancer compared to weakly supervised and fully supervised approaches.

Preparations of palytoxin (PLTX, derived from Japanese Palythoa tuberculosa) and the congeners 42-OH-PLTX (from Hawaiian P. toxica) and ovatoxin-a (isolated from a Japanese strain of Ostreopsis ovata), as well as a 50:50 mixture of PLTX and 42-OH-PLTX derived from Hawaiian P. tuberculosa were characterized as to their concentration, composition, in-vitro potency and interaction with an anti-PLTX monoclonal antibody (mAb), after which they were evaluated for lethality and tissue histopathology after intraperitoneal (IP) and aerosol administration to rats. Once each preparation was characterized as to its toxin composition by LC-HRMS and normalized to a total PLTX/OVTX concentration using HPLC-UV, all four preparations showed similar potency towards mouse erythrocytes in the erythrocyte hemolysis assay and interactions with the anti-PLTX mAb. The IP LD50 values derived from these experiments (0.92, 1.93, 1.81 and 3.26 µg/kg, for the 50:50 mix, 42-OH-PLTX, PLTX, and ovatoxin-a, respectively) were consistent with published values, although some differences from the published literature were seen. The aerosol LD50 values (0.063, 0.045, 0.041, and 0.031 µg/kg for the 50:50 mix, 42-OH PLTX, PLTX, and ovatoxin-a, respectively) confirmed the exquisite potency of PLTX suggested by the literature. The tissue histopathology of the different toxin preparations by IP and aerosol administration were similar, albeit with some differences. Most commonly affected tissues were the lungs, liver, heart, salivary glands, and adrenal glands. Despite some differences, these results suggest commonalities in potency and mechanism of action among these PLTX congeners.

The health of Largescale Sucker (Catostomus macrocheilus) in the lower Columbia River (USA) was evaluated using morphometric and histopathological approaches, and its association with organic contaminants accumulated in liver was evaluated in males. Fish were sampled from three sites along a contaminant gradient. In 2009, body length and mass, condition factor, gonadosomatic index, and hematocrit were measured in males and females; liver and gonad tissue were collected from males for histological analyses; and organ composites were analyzed for contaminant content in males. In 2010, additional data were collected for males and females, including external fish condition assessment, histopathologies of spleen, kidney and gill and, for males, liver contaminant content. Multivariate analysis of variance indicated that biological traits in males, but not females, differed among sites in 2009 and 2010. Discriminant function analysis indicated that site-related differences among male populations were relatively small in 2009, but in 2010, when more variables were analyzed, males differed among sites in regards to kidney, spleen, and liver histopathologies and gill parasites. Kidney tubular hyperplasia, liver and spleen macrophage aggregations, and gill parasites were generally more severe in the downstream sites compared to the reference location. The contaminant content of male livers was also generally higher downstream, and the legacy pesticide hexachlorobenzene and flame retardants BDE-47 and BDE-154 were the primary drivers for site discrimination. However, bivariate correlations between biological variables and liver contaminants retained in the discriminant models failed to reveal associations between the two variable sets. In conclusion, whereas certain non-reproductive biological traits and liver contaminant contents of male Largescale Sucker differed according to an upstream-downstream gradient in the lower Columbia River, results from this study did not reveal the specific environmental factors responsible for the differences in health status among fish populations.
