RESUMEN

Rhizoctonia solani compromises the production of lima bean, an alternative and low-input food source in many tropical regions. Inoculation of bacterial strains has been used, but research on their biocontrol and growth promotion potential on lima bean is scarce. The objective of this study was to evaluate the effects of inoculation with rhizobacterial strains of the genera Bacillus, Brevibacillus, Paenibacillus, Burkholderia, Pseudomonas, and Rhizobium in combination or not with N2-fixing Rhizobium tropici on the control of damping-off disease and growth promotion in lima bean plants. Greenhouse experiments were conducted to evaluate the inoculation with bacterial strains with biocontrol potential in combination or not with R. tropici in substrate infected with R. solani CML 1846. Growth promotion of these strains was also assessed. Strains of Brevibacillus (UFLA 02-286), Pseudomonas (UFLA 02-281 and UFLA 04-885), Rhizobium (UFLA 04-195), and Burkholderia (UFLA 04-227) co-inoculated with the strain CIAT 899 (Rhizobium tropici) were the most effective in controlling R. solani, reducing the disease incidence in 47-60% on lima bean. The promising strains used in the biocontrol assays were also responsive in promoting growth of lima bean under disease and sterile conditions. A positive synergistic effect of co-inoculation of different genera contributed to plant growth, and these outcomes are important first steps to improve lima bean production.

Disease stress caused by plant pathogens impacts the functioning of the photosynthetic apparatus, and the symptoms caused by the degree of severity of the disease can generally be observed in different plant parts. The accurate assessment of plant symptoms can be used as a proxy indicator for managing disease incidence, estimating yield loss, and developing genotypes with disease resistance. The objective of this work was to determine the response of the photosynthetic apparatus to the increased disease severity caused by web blight Thanatephorus cucumeris (Frank) Donk on the common bean (Phaseolus vulgaris L.) leaves under acidic soil and the humid tropical conditions of the Colombian Amazon. Differences in chlorophyll fluorescence parameters, including Fv/Fm, Y(II), Y(NPQ), Y(NO), ETR, qP, and qN in leaves with different levels of severity of web blight in an elite line (BFS 10) of common bean were evaluated under field conditions. A significant effect of web blight on the photosynthetic apparatus was found. A reduction of up to 50% of energy use dedicated to the photosynthetic machinery was observed, even at the severity scale score of 2 (5% surface incidence). The results from this study indicate that the use of fluorescence imaging not only allows for the quantifying of the impact of web blight on photosynthetic performance, but also for detecting the incidence of disease earlier, before severe symptoms occur on the leaves.

In an experiment on organic production of okra (Abelmoschus esculentus (L.) Moench) that was carried out from September 2013 to January 2014, in Manaus, Amazonas state, Brazil, we observed large chlorotic, necrotic, helical, discontinuous, dark or light-brown lesions with partial detachment of the injured area on the adaxial surface of leaves located in the median and basal portions of the plants. A whitish mycelium mantle covers the lesions on the leaves at the abaxial surface at high moisture conditions. Using morphological characteristics, Kochs postulates, and phylogenetic analyses of the ITS-5.8S rDNA region, we identified that the fungus causing the lesions on the okra leaves was Thanatephorus cucumeris (Frank) Donk (asexual stage of Rhizoctonia solani Kuhn of the anastomosis group AG-1 ID). This is the first report of T. cucumeris causing web blight on okra in Brazil, and probably in the world. So far, T. cucumeris was described on okra only on post-harvest pods rotting and seedlings damping off.


Em um experimento sobre a produção orgânica do quiabeiro (Abelmoschus esculentus (L.) Moench), que foi instalado em Manaus, Amazonas, Brasil, no período de setembro de 2013 a janeiro de 2014, observou-se, na face adaxial do limbo foliar das folhas medianas e baixeiras, a ocorrência de lesões cloróticas e necróticas grandes, helicoidais, de coloração marrom escuro ou marrom claro e descontínuas, com desprendimento parcial da área lesionada. Na face abaxial, sobre as manchas, em condições de alta umidade, constatou-se a presença de um manto micelial esbranquiçado do patógeno, facilmente visível, recobrindo a área colonizada. Por meio da análise de características morfológicas, postulados de Koch e análise filogenética da região ITS-5.8S do rDNA do fungo isolado, identificou-se Thanatephorus cucumeris (Frank) Donk (fase assexuada Rhizoctonia solani Kuhn grupo de anastomose AG-1 ID) como o agente causal da doença. Este é o primeiro relato de T. cucumeris causando mancha foliar em quiabeiro no Brasil e, provavelmente, no mundo. Até então, sua ocorrência em quiabeiro estava restrita à podridão pós-colheita em frutos e tombamento de mudas.
