RESUMEN

In this study, the concentrations of trace elements (TEs) in Uroteuthis edulis caught from the East China Sea were determined. There were significant differences between TE concentrations in different body parts. Cu, Zn, and Cd were the most concentrated in the digestive glands and the concentrations of Cr and Co were highest in the gills. No significant differences in concentrations were shown between these tissues. In the four tissues analyzed, the mantle recorded the highest proportion of elemental load, while the digestive glands and gills had the lowest proportions. After maturity, TEs in the mantle showed no significant differences. In the digestive gland, the concentrations of all elements, except Zn, were significantly increased. The gonads illustrated apparent increases in the concentrations of Cr, Cu, and As. In the gills, the concentrations of Co and As were markedly increased.

This study reveals early prey eating by a short-finned pilot whale (Globicephala macrorhynchus Gray, 1846, Cetacea: Delphinidae) in the Canary Islands. Stomach contents, trophic markers, skin isotopic ratios of nitrogen (δ15N:15N/14N) and carbon (δ13C:13C/12C), and fatty acid profiles of the blubber of a short-finned pilot whale of 213 cm size euthanized in free-ranging conditions were analyzed. A total of 15 species of oegopsid squids, mostly diel vertical mesopelagic migrant species of the families Enoploteuthidae, Ommastrephidae, and Histioteuthidae, as well as mother's milk, were identified in the stomach contents. Asperoteuthis acanthoderma (Lu, 1977, Cephalopoda: Chiroteuthidae) was found as first time in this area, suggesting the possibility of its presence on both sides of the subtropical Atlantic, extending its current known distribution. The δ15N value (11.55‰) was higher than expected based on the size range of squid ingested, but lower than that of adult pilot whales, suggesting that mother's milk intake has a significant effect on these values in calves. Similarly, the δ13C values (-17.99‰) were shifted to those of adult pilot whales rather than the ingested squids, also due to the ingestion of high-fat breast milk. The fatty acid (FA) composition of blubber showed a clear stratification. Long-chain polyunsaturated fatty acids (LC-PUFA) were mainly present in the inner layer, while most relevant ≤C20 monounsaturated fatty acids (MUFA) were more abundant in the outer layer.

Maceration with sodium salts is applied to irmprove water holding capacity in squid-based products. The aim of this work was to determine how the mineral content of squid flesh is affected by maceration and further vacuum-cooking. Atlantic squids (Loligo vulgaris) from two fisheries (FAO areas No. 47 and 34) were sampled. Macrominerals (g/100 g) present in raw flesh were Na, Mg, P, S, K and Ca, while microminerals accounting for >1 mg/kg were Zn, Si, Sr, Fe, Cu, Al and Mn. As a result of maceration (3 + 1.5% w: w NaCl+ Na citrate) and vacuum-cooking (at 65 °C for 20 min), some squid minerals was removed. The levels of Cd and As were reduced by half, while Na content increased from 0.28 to 0.49 g/100 g. Maceration with sodium salts generally led to minerals leaching (except for Na) with the medium. Further cooking produced additional losses of most of the minerals present in macerated squid (except Pb and Cd). Squid microminerals were hardly removed with the cooking juice. The consumption of macerated-cooked squid covered > 10% of the recommended dietary intake for Na, P, Zn, Mg and Mn, while health risks were almost negligible and mainly concerned Cd (up to 14% of the probable tolerable weekly intake). The combination of both treatments involves certain loss of most of the essential minerals but also contributes to reducing toxicological risks related to mineral intake through squid products.

Trace elements and δ15N values were analysed in micronekton (crustaceans, fishes and squids) sampled in the south-western Indian Ocean. Myctophids were associated with high concentrations of arsenic at La Pérouse and MAD-Ridge seamounts, and with lead and manganese at MAD-Ridge and in the Mozambique Channel. The difference in cadmium, copper and zinc concentrations between micronekton broad categories reflected differing metabolic and storage processes. When significant, negative relationships were found between micronekton body size and trace element concentrations, which can possibly be attributed to differing metabolic activity in young and old individuals, dietary shifts and/or dilution effect of growth. No relationships were found between trace element concentrations and δ15N values of micronekton (except cobalt which decreased with increasing δ15N values), since most trace elements are not biomagnified in food webs due to regulation and excretion processes within organisms. All trace element pairs were positively correlated in fishes suggesting regulation processes.

Microplastics are a relevant environmental concern in marine ecosystems due to their ubiquity. However, knowledge on their dispersion patterns within the ocean basin and the interaction with biota are scarce and mostly limited to surface waters. This study investigated microplastic contamination in two species of deep-sea cephalopods from the southwestern Atlantic with different ecological behaviour: the vampire squid (Vampyroteuthis infernalis) and the midwater squid (Abralia veranyi). Microplastic contaminated most of the evaluated specimens. V. infernalis showed higher levels of contamination (9.58 ± 8.25 particles individual-1; p < 0.05) than A. veranyi (2.37 ± 2.13 part. ind.-1), likely due to the feeding strategy of V. infernalis as a faecal pellets feeder. The size of extracted microplastics was inversely proportional to the depth of foraging. The microplastics were highly heterogeneous in composition (shape, colour and polymer type). Our results provide information regarding microplastic interaction with deep-sea organisms and evidence of the biological influence in the microplastic sinking mechanism.

OBJECTIVE: Conventional MEG provides an unsurpassed ability to, non-invasively, detect epileptic activity. However, highly resolved information on small neuronal populations required in epilepsy diagnostics is lost and can be detected only intracranially. Next-generation on-scalp magnetencephalography (MEG) sensors aim to retrieve information unavailable to conventional non-invasive brain imaging techniques. To evaluate the benefits of on-scalp MEG in epilepsy, we performed the first-ever such measurement on an epilepsy patient. METHODS: Conducted as a benchmarking study focusing on interictal epileptiform discharge (IED) detectability, an on-scalp high-temperature superconducting quantum interference device magnetometer (high-Tc SQUID) system was compared to a conventional, low-temperature SQUID system. Co-registration of electroencephalopraphy (EEG) was performed. A novel machine learning-based IED-detection algorithm was developed to aid identification of on-scalp MEG unique IEDs. RESULTS: Conventional MEG contained 24 IEDs. On-scalp MEG revealed 47 IEDs (16 co-registered by EEG, 31 unique to the on-scalp MEG recording). CONCLUSION: Our results indicate that on-scalp MEG might capture IEDs not seen by other non-invasive modalities. SIGNIFICANCE: On-scalp MEG has the potential of improving non-invasive epilepsy evaluation.

In the present article, we present the experimental results concerning the fine-tuning and optimization of superconducting quantum interference device (SQUID) parameters by thermal annealing. This treatment allows for the modification of the parameters in order to meet a specific application or to adjust the device parameters to prevent the increase of magnetic field noise and work instability conditions due to a different critical current with respect to the design value. In particular, we report the sensor critical current, the voltage-flux (V-Φ) characteristics and the spectral density of the magnetic field of SQUID magnetometers for different annealing temperatures. The measurements demonstrate that it is possible to achieve a fine control of the most important device parameters. In particular, we show that thermal annealing allows for the reduction of SQUID noise by more than a factor of 5 and makes the device working operations very stable. These results are very useful in view of quantum technology applications related to superconducting quantum computing where the correct functioning of the quantum bit depends on the fine control of the superconducting quantum device parameters and selectable annealing is possible by using a suitable laser as a thermal source.

The oegopsid squid family Onychoteuthidae was recently revised based on morphology, but sufficient material for a complementary molecular analysis has not been available until now. In the present study, over 250 sequences of cytochrome c oxidase subunit I (COI) and 16S rRNA for 222 individuals were analysed to create a combined phylogeny for the family. Results support monophyly for the family and all seven onychoteuthid genera (including Moroteuthopsis, established herein as the senior genus name for species formerly attributed to Kondakovia); 29 genetically distinct species were recovered, with the BIN (Barcode Index Number) analysis for COI showing good congruence overall with morphological species groupings. No sequences were available for five additional known species, making the total family diversity likely to exceed 34 species. Seven of the BINs formed in this study appear to represent undescribed taxa, suggesting that even in this relatively well-studied family, much additional work remains before a comprehensive understanding of the diversity and evolutionary relationships for the Onychoteuthidae can be achieved.

We review stationary and mobile systems that are used for the nondestructive evaluation of room temperature objects and are based on superconducting quantum interference devices (SQUIDs). The systems are optimized for samples whose dimensions are between 10 micrometers and several meters. Stray magnetic fields from small samples (10 µm-10 cm) are studied using a SQUID microscope equipped with a magnetic flux antenna, which is fed through the walls of liquid nitrogen cryostat and a hole in the SQUID's pick-up loop and returned sidewards from the SQUID back to the sample. The SQUID microscope does not disturb the magnetization of the sample during image recording due to the decoupling of the magnetic flux antenna from the modulation and feedback coil. For larger samples, we use a hand-held mobile liquid nitrogen minicryostat with a first order planar gradiometric SQUID sensor. Low-Tc DC SQUID systems that are designed for NDE measurements of bio-objects are able to operate with sufficient resolution in a magnetically unshielded environment. High-Tc DC SQUID magnetometers that are operated in a magnetic shield demonstrate a magnetic field resolution of ~4 fT/√Hz at 77 K. This sensitivity is improved to ~2 fT/√Hz at 77 K by using a soft magnetic flux antenna.

Arsenic and Cadmium concentrations in Todarodes pacificus (4 males and 4 females), Sepia longipes (1 male and 3 unknown), five Sepia madokai caught in 2012 (1 male and 4 females) and five S. madokai caught in 2014 (5 females) were collected from the East China Sea (ECS) during intervals from 2010 Oct. through to 2014 Oct. The internal organs were vacuum dried and the elements were measured by ICP-MS. Among the toxic elements, As, Cd, and Hg in the digestive gland, gonads and branchial hearts of squids and cuttlefishes were measured separately. The squid T. pacificus practically inhabits offshore, whereas cuttlefishes, such as S. longipes and S. madokai, inhabit bottom coastal waters in the ECS. Cd concentration in digestive gland of T. pacificus and S. madokai is higher than similar species from other regions. This high concentration seems to be derived from anthropogenic activities and geological conditions present in the ECS. The squid and cuttlefish also accumulate high amounts of As, not only in their digestive gland but also in other organs. Arsenic could be derived from high volcanic activities. In the case of Hg, the muscles were reported to contain the highest concentration in the species studied, but unlike concentration of Cd in digestive gland, it was high compared to other studies. These elements can be utilized as indicators for environmental pollution in marine systems. For monitoring of harmful elements in squids and cuttlefish, for example Cd and As in the digestive gland and Hg in mantle should be measured.

Cephalopods in nature undergo highly dynamic skin coloration changes that allow rapid camouflage and intraspecies communication. The optic lobe is thought to play a key role in controlling the expansion of the chromatophores that generate these diverse body patterns. However, the functional organization of the optic lobe and neural control of the various body patterns by the optic lobe are largely unknown. We applied electrical stimulation within the optic lobe to investigate the neural basis of body patterning in the oval squid, Sepioteuthis lessoniana Most areas in the optic lobe mediated predominately ipsilateral expansion of chromatophores present on the mantle, but not on the head and arms; furthermore, the expanded areas after electrical stimulation were positively correlated with an increase in stimulating voltage and stimulation depth. These results suggest a unilaterally dominant and vertically converged organization of the optic lobe. Furthermore, analyzing 14 of the elicited body pattern components and their corresponding stimulation sites revealed that the same components can be elicited by stimulating different parts of the optic lobe and that various subsets of these components can be coactivated by stimulating the same area. These findings suggest that many body pattern components may have multiple motor units in the optic lobe and that these are organized in a mosaic manner. The multiplicity associated with the nature of the neural controls of these components in the cephalopod brain thus reflects the versatility of the individual components during the generation of diverse body patterns. SIGNIFICANCE STATEMENT: Neural control of the dynamic body patterning of cephalopods for camouflage and intraspecies communication is a fascinating research topic. Previous studies have shown that the optic lobe is the motor command center for dynamic body patterning. However, little is known about its neural organization and the mechanisms underlying its control of body pattern generation. By electrically stimulating the optic lobe of the oval squids and observing their body pattern changes, surprisingly, we found that there is no somatotopic organization of motor units. Instead, many of these components have multiple motor units within the optic lobe and are organized in a mosaic manner. The present work reveals a novel neural control of dynamic body patterning for communication in cephalopods.

Practical temperature measurements in accordance with the international system of units require traceability to the international temperature scales currently in force. Along with the awaited redefinition of the unit of temperature, the kelvin, on the basis of the Boltzmann constant, in future its mise en pratique will allow the use of approved methods of primary thermometry for the realization and dissemination of the kelvin. To support this process, we have developed a DC superconducting quantum interference device-based noise thermometer especially designed for measurements of thermodynamic temperature in a broad temperature range from 5 K down to below 1 mK. In this paper, we describe in detail the primary magnetic field fluctuation thermometer and the underlying model applied for the temperature determination. Experimental measurement results are presented for a comparison with the Provisional Low Temperature Scale 2000 between 0.7 K and 16 mK including an uncertainty budget for the measured thermodynamic temperatures. In this set-up, the relative combined standard uncertainty is equal to 0.6%.

This study investigated the effect of single- and two-cycle high hydrostatic pressure (HHP) treatments on water properties, physicochemical, and microbial qualities of squids (Todarodes pacificus) during 4 °C storage for up to 10 d. Single-cycle treatments were applied at 200, 400, or 600 MPa for 20 min (S-200, S-400, and S-600), and two-cycle treatments consisted of two 10 min cycles at 200, 400, or 600 MPa, respectively (T-200, T-400, and T-600). HHP-treated samples had higher (P < 0.05) content of P2b (immobilized water) and P21 (myofibril water), but lower P22 (free water) than those of control. The single- and two-cycle HHP treatments at the same pressure level caused no significant difference in water state of squids. The two-cycle HHP treatment was more effective in controlling total volatile basic nitrogen, pH, and total plate counts (TPC) of squids during storage, in which TPC of S-600 and T-600 was 2.9 and 1.8 log CFU/g at 10 d, respectively, compared with 7.5 log CFU/g in control. HHP treatments delayed browning discoloration of the squids during storage, and the higher pressure level and two-cycle HHP were more effective. Water properties highly corresponded with color and texture indices of squids. This study demonstrated that the two-cycle HHP treatment was more effective in controlling microbial growth and quality deterioration while having similar impact on the physicochemical and water properties of squids in comparison with the single-cycle treatment, thus more desirable for extending shelf-life of fresh squids.

The oegopsid squids Abraliopsis and Pterygioteuthis are abundant and diverse genera with taxonomic and distributional problems. Identification and distribution of species in the Mexican Pacific has been somewhat controversial. Here are provided a large series of new records for Abraliopsis affinis, Abraliopsis falco, Pterygioteuthis gemmata, Pterygioteuthis giardi and Pterygioteuthis hoylei from the Gulf of California and off the SW coast of Mexico. All five species were collected in the central or the southern Gulf of California, or in both. Abraliopsis affinis was found in seven samples with a total of 48 specimens, from 21°59' to 24°53'12"N. Abraliopsis falco was much less represented in the samples (14 specimens) but it was found in 10 localities, four of which correspond to the central-southern Gulf of California (north to 27°44'53"N) and six to SW Mexico (south to 16°49'18"N). In the case of Pterygioteuthis gemmata, only two records (three specimens) were obtained, both in the SW Gulf of California, while Pterygioteuthis giardi (nine specimens) records were all from the central Gulf of California (27°44'53" to 25°39'59"N). In the case of Pterygioteuthis hoylei (nine specimens), material was obtained in six localities, also in a restricted latitudinal range (24°23'48" to 25°56'56"N).

The most consumed squid species worldwide were characterized regarding their concentrations of minerals, fatty acids, cholesterol and vitamin E. Interspecific comparisons were assessed among species and geographical origin. The health benefits derived from squid consumption were assessed based on daily minerals intake and on nutritional lipid quality indexes. Squids contribute significantly to daily intake of several macro (Na, K, Mg and P) and micronutrients (Cu, Zn and Ni). Despite their low fat concentration, they are rich in long-chain omega-3 fatty acids, particularly docosahexaenoic (DHA) and eicosapentanoic (EPA) acids, with highly favorable ω-3/ω-6 ratios (from 5.7 to 17.7), reducing the significance of their high cholesterol concentration (140-549 mg/100g ww). Assessment of potential health risks based on minerals intake, non-carcinogenic and carcinogenic risks indicated that Loligo gahi (from Atlantic Ocean), Loligo opalescens (from Pacific Ocean) and Loligo duvaucelii (from Indic Ocean) should be eaten with moderation due to the high concentrations of Cu and/or Cd. Canonical discriminant analysis identified the major fatty acids (C14:0, C18:0, C18:1, C18:3ω-3, C20:4ω-6 and C22:5ω-6), P, K, Cu and vitamin E as chemical discriminators for the selected species. These elements and compounds exhibited the potential to prove authenticity of the commercially relevant squid species.

The concentrations of 18 polycyclic aromatic hydrocarbons (PAHs) were determined in five commercially valuable squid species from different geographical origins (Atlantic, Indic and Pacific Oceans). Out of the 18 quantified PAHs (the 16 PAHs considered by US EPA as priority pollutants, dibenzo(a,l)pyrene and benzo(j)fluoranthene) only dibenz(a,h)anthracene was not detected. The total concentrations of PAHs varied by a factor of more than 100-fold, from 0.22 (Loligo gahi) to 60.9 µg/kg ww (Loligo reynaudii). Intra- and inter-specific variability of PAH levels was statistically assessed. Nine carcinogenic (probable/possible) PAHs accounted for 1% (L. reynaudii) to 26% (Loligo opalescens) of the total PAHs content being the main contributors naphthalene (in Loligo duvaucelii, L. reynaudii and Loligo vulgaris species), chrysene (in L. opalescens) and indeno(1,2,3-cd)pyrene (in L. gahi). PAHs source analysis indicated that four of the five zones of capture of the different squid species are significantly affected by both petrogenic and pyrolytic sources. Assessment of the target carcinogenic risks, established by the US EPA, suggested that L. gahi (Atlantic Ocean) and L. opalescens (from Pacific Ocean) may pose additional risks for consumers, if not eaten in moderation, derived from benzo(a)pyrene ingestion.
