RESUMEN

Efficient poultry production can be accomplished using combined technologies. A combination of in ovo and ethno-veterinary technologies can deliver significant benefits, including reduced labour and production costs. This study evaluated the effect of in ovo administration of soursop leaf extract (SLE) on the hatching performance and physiology of Noiler chicks. A total of 550 eggs were incubated, and 460 fertile eggs were randomly distributed into 4 groups with 3 replicates on the 18th d of incubation. The eggs were then injected with 0.75 µg SLE, 1.5 µg SLE, and saline solution (negative control) at a rate of 0.2 ml in the air space. The noninjected group served as the positive control. At the end of the hatching period, the various groups were evaluated for embryo mortality, hatchability, hatch duration, organ weight, serum biochemistry, and chick quality. The results showed no significant differences (P > 0.05) in embryonic mortality, hatchability, organ weight, total protein, albumin, globulin, aspartate aminotransferase, alanine aminotransferase, and glucose among the treatment groups. However, chick weight, chick quality, and serum triglyceride levels were significantly (P < 0.05) higher in the extract-injected group. Additionally, incubation and hatch times were significantly lower (P < 0.05) in the SLE group compared to the other groups. In ovo administration of soursop leaf extract resulted in reduced incubation duration, hatch time, and embryo mortality. In conclusion, the in ovo injection of SLE improved hatch performance and chick quality.

Leaves of Annona muricata have medicinal potential which has gained attention from researchers around the world. This study has an objective to screen the antioxidant and cytotoxicity of ethyl acetate extract from A. muricata leaves and its fraction. The fine powder of A. muricata was macerated in methanol and further partitioned using two different solvents, namely n-hexane and ethyl acetate. In this article, we reported the screening results for ethyl acetate extract. Fractionation was then performed on the extract by means of column chromatography by gradient elution resulting in five combined fractions. Brine shrimp lethality test and 1-diphenyl-2-pycrilhidrazil (DPPH) assays were employed to evaluate the cytotoxicity and antioxidant of the extract, respectively. Characterization using gas chromatography-mass spectroscopy (GC-MS) was then conducted. The cytotoxicity of the samples was indicated by median lethal concentration50 values ranging from 28.84 to 1023.3 ppm. As for the antioxidant activity, the DPPH median inhibitory concentration50 values ranged from 4.12 to 180.66 ppm. GC-MS analysis on the most bioactive fraction revealed the predominating phytochemical contents of neophytadiene, palmitic acid, and phytol. In conclusion, the fraction of ethyl acetate extract from A. muricata leaves could potentially act as a strong antioxidant and moderate cytotoxic agent.

Soursop (Annona muricata L.) fruit tea is a health-beneficial product that promotes economic development and addresses the issue of excessive agricultural waste. Prolonging the shelf-life of soursop fruit tea has been of scientific interest currently. This study evaluated the effects of three types of packaging materials of soursop fruit tea (e.g., paper, paper-combined Polyetylen (PE), and aluminum-combined PE) and different storage temperatures (5, 15, 30, and 45°C) on various product characteristics, total polyphenol content (TPC), total flavonoid content (TFC), total ascorbic acid (TAA), and 2,2-diphenyl-1-picryl hydrazyl (DPPH)/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) free radical scavenging capacity during 4 weeks of storage. The results revealed that the sample stored in aluminum-combined PE packaging at 30°C retained most of the product's characteristics and nutritional values. This was evidenced by the moisture content of 2.49%, TAA of 3.9 ± 1.4 mg/100 g dry weight, TPC of 12.89 ± 0.47 mgGAE/g, TFC of 0.54 ± 0.004 mgQE/g, DPPH scavenging activity of 4.06 ± 0.02 mgAA/g, and ABTS scavenging activity of 13.34 ± 0.32 mgAA/g. Additionally, the microbiological quality of the sample met the standard of TCVN 9740:2013. Overall, the study highlights the importance of packaging materials and storage temperatures to maintain the nutritional quality of soursop fruit tea. It provides valuable insights into the suitable storage conditions for preserving the quality and health-promoting effects of this product.

The obtained seeds from fruit processing are considered by-products containing proteins that could be utilized as ingredients in food manufacturing. However, in the specific case of soursop seeds, their usage for the preparation of protein isolates is limited. In this investigation a protein isolate from soursop seeds (SSPI) was obtained by alkaline extraction and isoelectric precipitation methods. The SSPI was sonicated at 200, 400 and 600 W during 15 and 30 min and its effect on the physicochemical, functional, biochemical, and structural properties was evaluated. Ultrasound increased (p < 0.05) up to 5 % protein content, 261 % protein solubility, 60.7 % foaming capacity, 30.2 % foaming stability, 86 % emulsifying activity index, 4.1 % emulsifying stability index, 85.4 % in vitro protein digestibility, 423.4 % albumin content, 83 % total sulfhydryl content, 316 % free sulfhydryl content, 236 % α-helix, 46 % ß-sheet, and 43 % ß-turn of SSPI, in comparison with the control treatment without ultrasound. Furthermore, ultrasound decreased (p < 0.05) up to 50 % particle size, 37 % molecular flexibility, 68 % surface hydrophobicity, 41 % intrinsic florescence spectrum, and 60 % random coil content. Scanning electron microscopy analysis revealed smooth structures of the SSPI with molecular weights ranging from 12 kDa to 65 kDa. The increase of albumins content in the SSPI by ultrasound was highly correlated (r = 0.962; p < 0.01) with the protein solubility. Improving the physicochemical, functional, biochemical and structural properties of SSPI by ultrasound could contribute to its utilization as ingredient in food industry.

Innovations for product preservation have attracted interest as they may increase the shelf-life of items when stored properly. In this study, the effects of various storage conditions, including four types of packaging (paper packaging, paper combined PE packaging, aluminum combined PE packaging, and plastic jar packaging) and temperatures (5, 15, 30, and 45 °C) on the quality of dried soursop were evaluated. The results demonstrated that the combination of plastic jar packaging and a storage temperature of 15 °C retained a significant portion of the initial total ascorbic acid content, total polyphenol content, and total flavonoid content. After four weeks of storage, the dried soursop preserve packaged in a plastic jar and stored at 15 °C exhibited a moisture content of 22.977 ± 0.093 %, total ascorbic acid content of 9.7 ± 0.46 mg/100gDW, total polyphenol content of 8.12 ± 0.06 mgGAE/gDW, total flavonoid content of 0.18 ± 0.02 mgQE/gDW, DPPH and ABTS scavenging activity of 0.69 ± 0.01 mgAA/gDW and 0.82 ± 0.01 mgAA/gDW, respectively. Moreover, the product meets the requirements of decision 46/2007/QD-BYT regulating the limits on biological and chemical contamination in food. The study offers valuable insights for the food industry in optimizing packaging and storage conditions to ensure the storage of quality and health-beneficial properties of this product.

Shrimp has been known for its delicacy, but it undergoes rapid deterioration induced by biochemical and microbiological reactions. Melanosis is a major cause of discoloration associated with consumer rejection. All ethanolic extracts from different leaves including soursop, noni, and Jik leaves were dechlorophyllized via the "Green" sedimentation method before being used. The inhibitory activity against polyphenoloxidase (PPO) from Pacific white shrimp (Litopeneous vannamei) and the copper-chelating properties of varying extracts were compared. Soursop leaf extract (SLE) showed higher PPO inhibitory activity and copper-chelating ability than others (p < 0.05). Based on LC-MS, aempferol-3-O-rutinoside was identified as the most abundant compound, followed by catechin and neocholorigenic acid. The efficacy of SLE at different levels (0.25-1%) for inhibiting melanosis and preserving the quality of Pacific white shrimp was evaluated during refrigerated storage at 4 °C for 12 days in comparison with that of a 1.25% sodium metabisulfite (SMS)-treated sample. SLE at a level of 1% effectively retarded melanosis and bacterial growth, in which the total viable count did not exceed the microbial limit within 12 days. In addition, 1% SLE treatment impeded autolysis, reduced protein degradation and decomposition, and minimized lipid oxidation, as witnessed by the lower increases in pH, TVB-N, and TBARS values. Sensory evaluation indicated higher likeness scores and overall acceptability for SLE-1% and SMS-1.25% shrimps than those of the control and other samples. Therefore, SLE could be used as a natural alternative that effectively lowered the melanosis and quality loss of shrimp during refrigerated storage.

Pharmacological and nutritional benefits of plant leaf extracts can be harnessed to address the problem of poor chick quality, performance and high mortality, which affect both hatchery and farm managers. This study sought to evaluate the in ovo effect of Soursop leaf extract (SLE) on the hatching and post-hatch performance of Noiler chicks. A total of 640 fertile eggs were randomly distributed into four groups of five replicates: (0.25 µg SLE), (0.5 µg SLE), (0.75 µg SLE) and a non-injected control. On day 18 of incubation, 0.2 ml of SLE was injected into the airspace of the eggs. At hatch, embryonic mortality, hatchability, and chick quality were evaluated. Hatched chicks were reared to assess feed intake, weight gain, feed conversion ratio (FCR) and mortality. Results showed no significant difference (p > 0.05) in embryonic mortality, hatchability and weight gain among the treatment groups. Chick weight and feed intake were significantly higher (p < 0.05) in SLE groups, while FCR and mortality were significantly lower (p < 0.05) in SLE groups. It was concluded that in ovo injection of SLE improved chick weights, feed conversion and livability of Noiler chicks.

Graviola (Annona muricata) is a tropical plant with many traditional ethnobotanic uses and pharmacologic applications. A metabolomic study of both aqueous and DMSO extracts from Annona muricata leaves recently allowed us to identify dozens of bioactive compounds. In the present study, we use a proteomic approach to detect altered patterns in proteins on both conditioned media and extracts of HT-1080 fibrosarcoma cells under treatment conditions, revealing new potential bioactivities of Annona muricata extracts. Our results reveal the complete sets of deregulated proteins after treatment with aqueous and DMSO extracts from Annona muricata leaves. Functional enrichment analysis of proteomic data suggests deregulation of cell cycle and iron metabolism, which are experimentally validated in vitro. Additional experimental data reveal that DMSO extracts protect HT-1080 fibrosarcoma cells and HMEC-1 endothelial cells from ferroptosis. This work set out to, first, assess the role of soursop flower extracts (SFE) in limiting palm olein oxidation during the production of plantain chips, before ascertaining the effect of these soursop-flower-enriched fried palm olein on some biochemical and hematological parameters of rats. The extracts were added to 1.5 kg of oil at 1000, 1400, and 1800 ppm, while BHT at 200 ppm served as a positive control (PO+BHT), and the oil without additives was the negative control (PO). The samples were subjected to 15 frying cycles. Total oxidation values varied between 5.94 ± 0.0 and 31.58 ± 0.37; 8.08 ± 0.25 and 28.24 ± 0.00 and 13.71 ± 0.24 and 42.71 ± 0.40 respectively for palm olein enriched with SFE, for PO+BHT and for PO. Twenty-one groups each comprising five rats received, through dietary supplementation, oils subjected to 0, and 5, 10 and 15 frying cycles for a duration of 30 days. The alanine transaminase and aspartate transaminase of rats fed with oils enriched with SFE at fresh states and at 5 frying cycles was comparable to that of the neutral control group (23.45 ± 2.65 and 93.10 ± 3.53 U/L) and lower than that of the negative control group (52.15 ± 2.01 and 124.07 ± 1.89 U/L). The HDL cholesterol of these animals was also comparable to that of the neutral control group (67.82 ± 4.06 mg/dl) and higher than that of the negative control group (50.25 ± 5.20 mg/dl). White blood cells and mean corpuscular volume of rats fed with fried olein previously enriched with SFE were lower than those fed with fried olein without additives. These extracts are recommended as natural antioxidants for the stabilization of palm olein.

Soursop fruits are widely used in the folk medicine to treat a variety of health conditions. Once the chemical structure of dietary fibers from fruits is closely related to its biological functions in the human body, we aimed to explore structural features and biological activity of dietary fibers from soursop. Polysaccharides that constitute the soluble and insoluble fibers were extracted and further analyzed using monosaccharide composition, methylation, molecular weight determination and 13C NMR data. Soursop soluble fibers (SWa fraction) were characterized as having type II arabinogalactan and a highly methyl esterified homogalacturonan, while non-cellulosic insoluble fibers (SSKa fraction) were mainly composed by a pectic arabinan, a xylan-xyloglucan complex and a glucuronoxylan. The oral pre-treatment with SWa and SSKa promoted antinociception in mice writhing test, reducing the number of pain-like behaviors (in 84.2 % and 46.9 %, respectively, at 10 mg/kg) and peritoneal leucocyte migration (55.4 % and 59.1 %, at 10 mg/kg), effects possibly associated with the pectins present in fruit pulp extractions. SWa also significantly inhibited the plasmatic extravasation of Evans blue dye in 39.6 % at 10 mg/kg. This paper describes for the first time the structural features of soursop dietary fibers that may be of biological significance in future.

Tomatoes are very important vegetable crops in the world but with a perishable nature. Due to its highly perishable nature, various methods have been investigated to increase its shelf life while still preserving its qualities. Therefore, this study investigated the potentials of the parts of Annona muricata and the calyces of Hibiscus sabdariffa in the preservation of tomato fruits. Tomato fruits were washed and treated with powdered, aqueous and ethanolic extracts of the leaves, seeds, bark of the Annona muricata and calyces of Hibiscus sabdariffa at different concentrations of 3%, 5%, 6%, 9% and 12% w/v. The tomato fruits were placed in well-aerated baskets for a period of 35 days during which organoleptic and microbial analysis were carried out. The different treatments with Annona muricata and Hibiscus sabdariffa had significant effects on the preservation of the tomato fruits at p < 0.05. The leaves of Annona muricata proved most effective preserving up to 50% of the tomatoes after the monitoring period. Moreover, 6% (w/v) of the aqueous extract of the leaves of Annona muricata resulted in a preservation rate of 75% of the tomato fruits. The spoilage microorganisms isolated from the tomato fruits are Candida krusei, Candida sp., Bacillus subtilis and Bacillus sp. The results of this research carried out shows that the extracts of Annona muricata and Hibiscus sabdariffa had significant preservative activities on the tomato fruits (p < 0.05), thus minimising wastes and economic loss to the farmers and country in general.

Annona muricata leaves are traditionally used as an anticancer plant in the world. The aim of this study was to evaluate the ameliorative effect of the essential oil from Annona muricata leaves (EOAm) in an experimental model of breast cancer and to determine the volatile constituents with gas chromatography-mass spectrometry (GC-MS). Thirty female rats were assigned to five groups: the control group; the DMBA (7,12-dimethylbenz[α]anthracene) group; and three groups received daily EOAm doses of 50, 100, and 200 mg/kg/day, plus DMBA, respectively. After 13 weeks of treatment, tumors were analyzed pathologically and biochemical markers in serum were noted. As a result, in GC-MS analysis, 40 compounds were identified and 4 of them were abundant: Z-caryophyllene (40.22%), followed by α-selinene (9.94%), ß-pinene (8.92%), and ß-elemene (7.48%). Furthermore, EOAm in a dose-dependent form produced a reduction in tumor frequency and the accumulated tumor volume was reduced by 50% and 71% with doses of 100 and 200 mg/kg, respectively. Serum levels of reduced glutathione (GSH) increased and malondialdehyde (MDA) decreased significantly compared to the DMBA group. Serum levels of vascular endothelial growth factor (VEGF) decreased significantly from 70.75 ± 7.15 pg/mL in the DMBA group to 46.50 ± 9.00 and 34.13 ± 11.50 pg/mL in groups treated with doses of 100 and 200 mg/kg, respectively. This study concludes that the EOAm leaves showed an ameliorative effect in a murine model of breast cancer.

Soursop leaves are a source of phytochemical compounds, such as phenolic acids, flavonoids, hydrolyzable tannins, and acetogenins. These compounds can have several types of biological activities. Lactic acid bacteria can uptake phenolic compounds present in plants or fruits. The aim of the present work was to investigate the in vitro effect of hexane, acetone, methanolic, and aqueous extracts of soursop leaves (Annona muricata L.) on the growth, motility, and biofilm formation of Lactobacillus casei, and to determine compounds related to growth. The minimum concentration promoting growth, motility (swimming, swarming, and twitching), and biofilm-forming capacity (crystal violet) were evaluated. The results showed the growth-promoting capacity of acetone and aqueous extracts at low doses 25-50 mg/L, and an inhibition in the four extracts at higher doses of 100 mg/L. The L. casei growth is related to ellagic acid, quercetin rhamnoside, kaempferol dihexoside, quercetin hexoside, secoisolariciresinol, and kaempferol hexoside-rhamnoside. Hexane extract increased the three types of motility, while aqueous maintained swimming and twitching motility similar to control. The four extracts inhibited the biofilm formation capacity.

Soursop (Annona muricata Lin.) is a plant belonging to the Annonaceae family that has been widely used globally as a traditional medicine for many diseases. In this review, we discuss the traditional use, chemical content, and pharmacological activities of A.muricata. From 49 research articles that were obtained from 1981 to 2021, A.muricata's activities were shown to include anticancer (25%), antiulcer (17%), antidiabetic (14%), antiprotozoal (10%), antidiarrhea (8%), antibacterial (8%), antiviral (8%), antihypertensive (6%), and wound healing (4%). Several biological activities and the general mechanisms underlying the effects of A.muricata have been tested both in vitro and in vivo. A.muricata contains chemicals such as acetogenins (annomuricins and annonacin), alkaloids (coreximine and reticuline), flavonoids (quercetin), and vitamins, which are predicted to be responsible for the biological activity of A.muricata.

Bioactive compounds extracted from plants such as antimicrobials have attracted the attention of consumers and the food industry. This study aimed to determine the antimicrobial activity and chemical composition of Annona muricata leaf oleoresin obtained by supercritical CO2 extraction. The oleoresin was obtained by supercritical CO2 extraction and the chemical identification by gas chromatography coupled to mass spectrometry. Antimicrobial activity was evaluated by broth microdilution method against 14 foodborne fungi and bacteria. The oleoresin major chemical class was phytosterols (22.7%) and the major compounds were γ-sitosterol (15.7%), α-tocopherol (13.7%), phytol (13.1%), and hexadecanoic acid (11.5%). Minimum inhibitory concentration against bacteria ranged from 0.0025 to 0.010 mg mL-1. The oleoresin had high bactericidal activity against all bacteria, mainly Enterobacter cloacae and Pseudomonas aeruginosa with 0.005 mg mL-1 minimum bactericidal concentration. However, it had low fungicidal activity. The leaf oleoresin of A. muricata has promising applications in food, cosmetic, and pharmaceutical industries.

The tropical plant Annona muricata has been widely used for traditional ethnobotanic and pharmacologic applications. Extracts from different parts of this plant have been shown to have a wide range of biological activities. In the present study, we carry out a metabolomic study of both aqueous and DMSO extracts from Annona muricata leaves that has allowed us to identify 33 bioactive compounds. Furthermore, we have shown that aqueous extracts are able to inhibit endothelial cell migration and both aqueous and DMSO extracts inhibit the formation of tubule-like structures by endothelial cells cultured on Matrigel. We conclude that extracts of Annona muricata leaves have great potential as anti-angiogenic natural combinations of bioactive compounds.

Heat induced stress associated with dry climatic conditions in the tropics does have adverse effects reproduction in rabbits, and this in-turn impacts negatively on the income of rabbit farmers. However, natural products might prove to be a reliable, safe and cheap remedy for ameliorating reproductive such anomalies in rabbits. The potential of soursop for mitigating heat induced reproductive deficiency in rabbit bucks was investigated during the peak of dry climatic conditions in southwestern Nigeria. Sixty mixed breed (New Zealand white x Chinchilla) adult rabbit bucks were allotted to five treatments of four replicates (3 bucks per replicate) each in a completely randomized design. Soursop (Annona muricata) fruit was processed into juice using standard procedures and was designated as soursop juice. The juice was administered orally daily per kg body weight (BW), 0.55ml/kgBW distilled water (control), 0.55 ml/kgBW soursop juice, 1.11 ml/kgBW soursop juice, 1.67 ml/kgBW soursop juice and 2.22 ml/kgBW soursop juice to designated treatments 1 to 5, respectively for 56 days. Semen samples were collected with an improvised artificial vagina on the 28th and 56th day of the study, semen quality and seminal oxidative status were evaluated using standard procedures. Results showed that rabbit buck exposed to heat stress had lower semen quality, seminal antioxidants and increased seminal lipid peroxidation. However, the consumption of soursop juice lowered lipid peroxidation and enhanced (p < 0.05) antioxidant production in the seminal fluid of heat-stressed bucks than bucks on control group. Bucks' semen quality and antioxidant status peaks in heat-stressed bucks gavaged 2.22 ml/kgBW soursop juice and gives 100% recovery from the effects of heat induced stress. It can be concluded that 2.22 ml/kgBW soursop juice administered for 56 days did enhance spermatozoa quality and mitigated lipid peroxidation by improving antioxidant capacity of male rabbits in a dose dependent manner on extremely dry climatic conditions.

Soursop seeds present a potential source of edible oil production. This work was aimed at determining the effect of oven and sun drying on the chemical properties and lipid profile of soursop seed oil as well as the functional properties of the defatted seed flour. The chemical properties, lipid profiles, and functional properties of soursop seeds dried for 0, 6, 12, 18, 24, and 30 hr, and 0, 1, 3, and 5 days, respectively, in the oven and on the sun using time T0 as the control sample were determined using oil quality indices, gas phase chromatography, and functionality tests for flours, respectively, with a view of highlighting the potentials of the defatted seed. The result of the study revealed that the chemical properties of oils for oven-dried and sun-dried seeds changed with drying technique and time, with iodine value being the more affected parameter, and peroxide value (PV) being the least. The control exhibited the highest free fatty acids (FFAs), peroxide value (PV), thiobarbituric acid (TBA) value, and saponification value compared with dried samples. The fatty acid profiling showed that the predominant fatty acids were C18:1n-9, C18:2n-6, and C16:0 and that unsaturated fatty acids (UFA), monounsaturated fatty acids (MUFA), polyunsaturated fatty acids (PUFA), and omega-fats were not significantly affected by the oven drying time. The mean PUFA content ranged from 31.72% with sundried seeds to 30.92% after 30 hr of oven-drying and was not significantly affected by the drying technique. The oils contained more n-6 (30.60%) fatty acids than n-3 (1.12%). The PUFA/SFA ratios [1.14-1.37] as well as the Atherogenic index (AI) [0.25-0.27] were acceptable because of the recommended range of FAO/WHO. PUFA/SFA, n-6/n-3, and Atherogenic index (AI) did not change much with the sun-drying technique compared with oven-drying. Flours from sun-dried seeds had better functional properties than oven-dried and more than 3 different types of proteins (based on isoelectric points of proteins). It can be concluded that soursop seed contains good quality oil, which can be exploited to improve nutrition. Manufacturers of animal feeds should explore the agro-industrial use of its oil and defatted seed flour.

INTRODUCTION: Breast cancer is the second most common cancer in women globally, and the incidence rate has increased annually. Traditional medicine is frequently used as a cancer treatment, and soursop or Annona muricata L (A. muricata) is a traditional medicinal plant that has been widely used as an anticancer treatment and requires more thorough study. METHODS: In this research, we prepared ethanol extract and three solvents, ie, ethyl acetate, n-hexane and water fractions of A. muricata leaves and assessed their antiproliferation and cytotoxic activity on MCF7 breast cancer cells compared with that on CV1 normal kidney cells; observation of cell morphology by stained with mixture of propidium iodide and 4',6-diamidino-2-phenylindole indicated that this treatment induced an ongoing process of apoptotic cell death in MCF7 cells. To clarify the cell death mechanism via apoptosis, we assessed the mRNA expression in the caspase cascade of caspase-9, caspase-3, and PARP-1, and anti-apoptotic, Bcl-2 which mediated cytotoxic activity of extracts and ethyl acetate fractions of A. muricata leaves against MCF7 cells. RESULTS: The ethanol extract, ethyl acetate, n-hexane, and water fractions of A. muricata leaves had IC50 values of 5.3, 2.86, 3.08, and 48.31 µg/mL, respectively, in MCF7 cells but had no activity in CV1 cells. The high cytotoxic activity of A. muricata leaves was reflected by changes in the morphology of cancer cells that appeared after 6 h exposure to A. muricata leaf extract and ethyl acetate fraction; the membrane and nucleus of cells undergoing apoptosis were characterized by the rupture and loss of membranes and nuclei. The mechanism that mediates this cytotoxic activity in MCF7 cells was mediated through a decrease in the expression of Bcl-2 mRNA and an increase in caspase-9 and caspase-3 mRNA expression. CONCLUSION: Therefore, the leaves of the medicinal plant A. muricata contained compounds that on extraction exerted a highly effective activity as an anticancer treatment for breast cancer via induced apoptotic cell death.

ETHNOPHARMACOLOGICAL RELEVANCE: Breast cancer is a serious threat in low-income as well as developed countries. To face this, many herbal preparations are prescribed by traditional healers in Cameroon, among which is Anonidium mannii commonly called "wild soursop". AIM: This study was undertaken to assess the anti-tumor effect of A. mannii ethanolic extract on cancer cell growth and against DMBA-induced mammary tumors in rats. MATERIALS AND METHODS: The well characterized MTT bioassay was used to assess the cytotoxic potential of A. mannii ethanolic extract in liver (HepG2), prostate (DU145 & PC3) and breast (MCF-7) cancer cell lines. Considering the fact that breast cells were the most sensitive to the extract, a 7,12-dimethylbenz(a)anthracene (DMBA)-induced breast tumor rat model was used to assess the possible anticancer effect of A. mannii extract. Indeed, rats were treated with either tamoxifen (3.3 mg/kg BW) or A. mannii extract (16.5, 50 and 150 mg/kg BW) or vehicle (2% ethanol) for 20 weeks. Tumor incidence, tumor mass and volume, oxidative stress status in tumor as well as tumor histoarchitecture were evaluated. RESULTS: A 24 h incubation of tested cells with the A. mannii extract significantly slowed cell growth in a concentration-dependent manner with an interesting effect in breast cells (IC50 ~61.5 µg/mL). As compared to the DMBA rats, those treated with A. mannii extract (50 and 150 mg/kg) showed reduced breast tumor incidence (28%), tumor burden (95.34% at 50 mg/kg and 99.14% at 150 mg/kg) and tumor volume (~92%). A. mannii extract counteracted the high proliferation of terminal mammary ducts induced by DMBA, mainly at 50 mg/kg. Furthermore, the extract decreased MDA and nitrite levels but increased SOD activity in the mammary gland. High Performance Liquid Chromatography coupled with Mass Spectrometry (HPLC-MS) analysis detected potential anticancer and antioxidant alkaloids in A. manni extract, which are close to those found in Annona muricata. CONCLUSION: These results provide evidence on the in vitro and in vivo anticancer effects of A. mannii, and therefore support its use in traditional medicine system to fight against cancer.
