Planning for effective conservation demands an accurate understanding of the ecological aspects of species, particularly their distribution and habitat preferences. This is even more critical in the case of data-poor, rare, and threatened species, such as seahorses, mainly when they inhabit vulnerable ecosystems like estuaries. Given the importance of better understanding these parameters to design seahorse conservation strategies, we mapped the distribution and assessed habitat preferences of longsnout seahorses (Hippocampus reidi) in a mangrove estuary in a Brazilian protected area. Using generalised linear mixed-effects models we found that dense mangrove cover macro-habitats and shallow depths predicted seahorse sightings and higher densities. Furthermore, the selective index of micro-habitats used by seahorses showed that seahorses exhibited a preference for mangrove structures as holdfasts (i.e., fallen branches). Due to the significant importance of mangroves in providing suitable habitats for H. reidi in estuaries, it is crucial to enforce the protection of these ecosystems in conservation and management strategies for the species.

The seahorse is a marine teleost fish member of the Syngnathidae family that displays a complex variety of morphological and reproductive behavior innovations and has been recognized for its medicinal importance. In the Brazilian ichthyofauna, the seahorse Hippocampus reidi is among the three fish species most used by the population in traditional medicine. In this study, a protocol was performed based on fast heat inactivation of proteases plus liquid chromatography coupled to mass spectrometry to identify native peptides in gills of seahorse H. reidi. The MS/MS spectra obtained from gills allowed the identification of 1080 peptides, of which 1013 peptides were present in all samples and 67 peptide sequences were identified in an additional LC-MS/MS run from an alkylated and reduced pool of samples. The majority of peptides were fragments of the internal region of the amino acid sequence of the precursor proteins (67%), and N- and C-terminal represented 18% and 15%, respectively. Many peptide sequences presented ribosomal proteins, histones and hemoglobin as precursor proteins. In addition, peptide fragments from moronecidin-like protein, described with antimicrobial activity, were found in all gill samples of H. reidi. The identified sequences may reveal new bioactive peptides.

STUDY QUESTION: Does oxidative stress (OS) activate autophagy in human sperm? SUMMARY ANSWER: Human spermatozoa subjected to OS activate an autophagic response. WHAT IS KNOWN ALREADY: Autophagy is a regulated pathway of lysosomal degradation which helps eukaryotic cells to maintain or restore homeostasis, being a cellular stress response mechanism. OS is a main cause of impaired sperm function and is linked to male infertility; however, whether OS activates autophagy in human spermatozoa is unknown. STUDY DESIGN, SIZE, DURATION: Human spermatozoa were exposed separately to ionomycin and hydrogen peroxide in order to induce OS. An untreated control group was included. Sperm cells under OS were then exposed to chloroquine in order to block autophagy. An untreated control and a control incubated only with the OS inducer were included in each experimental setting. PARTICIPANTS/MATERIALS, SETTING, METHODS: For this study, semen samples from normozoospermic donors were used and motile sperm cells were selected by the swim up technique. First, the generation of OS under our experimental conditions was demonstrated by analyzing sperm parameters including viability, reactive oxygen species (ROS) production, mitochondrial membrane potential (ΔΨm) motility and thiol oxidation. Then, proteins involved in autophagy, including the microtubule-associated protein light chain 3 (LC3), particularly LC3-I and LC3-II, autophagy-related 5 (ATG5) and autophagy-related 16 (ATG16) proteins as well as the phosphorylated form of AMP-activated protein kinase (pAMPK) were evaluated in spermatozoa exposed to OS and compared to the untreated control. Finally, the impact of autophagy blocking by chloroquine treatment on sperm quality, metabolic parameters, including glycolysis and oxidative phosphorylation, as well as the cell death markers phosphatidylserine externalization and caspase activation was analyzed. Sperm quality parameters, cell death markers and autophagy-related proteins were analyzed by flow cytometry. Motility was evaluated by the computer-assisted sperm analysis system and metabolic parameters were analyzed using an extracellular flux analyzer. MAIN RESULTS AND THE ROLE OF CHANCE: Exposure to ionomycin and hydrogen peroxide promotes OS resulting in increased ROS production and decreased viability, ΔΨm and motility, while increasing thiol oxidation. These alterations were accompanied by a decrease in LC3-I, indicating that autophagy was activated upon OS exposure. Ionomycin also caused an increase in LC3-II, ATG5, ATG16 and pAMPK content. Autophagy blocking of sperm exposed to OS caused deterioration in sperm quality and metabolic parameters as well as an increase in cell death markers. LARGE SCALE DATA: N/A. LIMITATIONS, REASONS FOR CAUTION: The study was carried out in vitro using motile sperm from normozoospermic donors; tests on sperm from infertile patients were not carried out. The autophagy blocking plus OS might generate a non-specific response to a highly stressful situation leading to the induction of cell death. WIDER IMPLICATIONS OF THE FINDINGS: Human spermatozoa subjected to OS activate an autophagic response and its blockage results in increased oxidative damage and commits spermatozoa to cell death. These results suggest a crucial role of autophagy as a stress response by male gametes, which contributes to maintaining the functionality and lifespan of ejaculated sperm cells. Detection of autophagy activation in sperm cells ex vivo could be included in semen analysis as a marker of OS, especially in men displaying high levels of seminal ROS. Novel strategies that aim to activate this cellular stress response could improve sperm quality/functionality under natural ejaculate conditions in which increased ROS levels are expected. Finding new alternatives to traditional live preys such as Artemia and rotifers, which do not always promote optimal fish growth and survival, is required for the successful aquaculture of highly specialized predatory species, including seahorses. The present study assessed the nutritional value of an interesting marine amphipod (Parhyale hawaiensis), and evaluates through a feeding trial its potential use as a natural prey for 10-months lined seahorse, Hippocampus erectus. P. hawaiensis showed high levels of valuable lipids (20.4-26.7% on dry matter basis) and polyunsaturated fatty acids (PUFAs) ( 26.4-41% of total FAs), including the long-chain PUFAs (LC-PUFAs) arachidonic acid (ARA) (2.9-7.7%), eicosapentaenoic acid (EPA) (4.3-6.5%) and docosahexaenoic acid (DHA) (2.1-6.2%). A comparison between wild-captured and cultured amphipods revealed a significant improvement of the amphipod FA profile in terms of DHA%, total omega-3 (n3) FAs and n3/n6 ratio when employing both a conventional amphipod culture based on a commercial shrimp diet, and, to a lesser extent, a large (3,500 L) biofloc system. Seahorses fed with frozen/wild amphipods, either singly or in combination with Artemia enriched with Super Selco® (INVE Aquaculture, Belgium) for 57 days, substantially improved seahorse growth and FA profiles in terms of ARA, EPA and DHA%, including indices associated to marine sources, such as Σn3 and n3/n6, compared to a diet based solely on enriched Artemia. These results support the use of marine amphipods as an alternative food organism for juvenile H. erectus and suggest a potential use for general marine aquaculture.

This study was carried out using hydro-acoustic technology in a water tank environment. Data were obtained using the single-beam scientific echo-sounder SIMRAD EK-15 at a frequency of 200 kHz and analyzed via the Sonar-4 software. The measurement result of the TS (mean±S.E.M.) of Hippocampus kuda female, male and pregnant male seahorses were -56.24±0.047, -57.25±0.032, and -58.26±0.06, respectively. There was a significant difference in the mean TS value of H. kuda based on the reproductive state (P < 0.05). Furthermore, the response to the mean TS value of pregnant H. kuda male (the presence of a brood pouch) was lowly significant compared to the female (absence of a brood pouch) (P< 0.05). The results showed a possibility of finding a pregnant H. kuda male in a water column through active acoustic methods in the future.

NUAK1 is an AMPK-related kinase located in the cytosol and the nucleus, whose expression associates with tumor malignancy and poor patient prognosis in several cancers. Accordingly, NUAK1 was associated with metastasis because it promotes cell migration and invasion in different cancer cells. Besides, NUAK1 supports cancer cell survival under metabolic stress and maintains ATP levels in hepatocarcinoma cells, suggesting a role in energy metabolism in cancer. However, the underlying mechanism for this metabolic function, as well as its link to NUAK1 subcellular localization, is unclear. We demonstrated that cytosolic NUAK1 increases ATP levels, which associates with increased mitochondrial respiration, supporting that cytosolic NUAK1 is involved in mitochondrial function regulation in cancer cells. NUAK1 inhibition led to the formation of "donut-like" structures, providing evidence of NUAK1-dependent mitochondrial morphology regulation. Additionally, our results indicated that cytosolic NUAK1 increases the glycolytic capacity of cancer cells under mitochondrial inhibition. Nuclear NUAK1 seems to be involved in the metabolic switch to glycolysis. Altogether, our results suggest that cytosolic NUAK1 participates in mitochondrial ATP production and the maintenance of proper glycolysis in cancer cells. Our current studies support the role of NUAK1 in bioenergetics, mitochondrial homeostasis, glycolysis and metabolic capacities. They suggest different metabolic outcomes depending on its subcellular localization. The identified roles of NUAK1 in cancer metabolism provide a potential mechanism relevant for tumor progression and its association with poor patient prognosis in several cancers. Further studies could shed light on the molecular mechanisms involved in the identified metabolic NUAK1 functions.

The central nervous system (CNS) of Teleostei is a complex system of self-governance and its morphology is reflected in the physiological and reproductive behaviors. The Indo-Pacific seahorse, Hippocampus barbouri Jordan & Richard son, 1908, is a new candidate species for aquaculture in Thailand. In this study, we investigated the brain morphology of H. barbouri across various developmental windows. Light microscopic observations of adult brains revealed a large optic tectum in the mesencephalon, whereas the cerebral hemispheres and the cerebellum are of medium size. The detailed brain structures were generally similar to those of other teleosts; however, only five distinct layers were present in the optic tectum, including the stratum marginale, stratum opticum, stratum album central, stratum griseum central, and stratum periventriculae, versus six layers observed in other fish. One day after birth (1 DAB) the brain was a packed structure without any clear sub-structures. The number of capillaries in the optic tectum began to increase at 6 DAB, and at 14 DAB several features, including small blood vessels in the optic tectum and Purkinje cells, became noticeable. By 35 DAB, the optic tectum became highly vascularized and included five layers. Additionally, large Purkinje cells were developed in the cerebellum. Based on the brain development pattern, we speculate that the predatory ability of this fish starts to develop from 6 to 14 days after birth.

This study explored the suitability of the seahorse Hippocampus reidi (Ginsburg, 1933) for assessing biomarkers of genotoxic effects and its use as a sentinel organism to detect the effects of acute exposure to petroleum hydrocarbons. Fish were exposed to three concentrations of crude oil (10, 20 and 30 g/kg) for 96 h, and the activity of phase II biotransformation enzyme glutathione S-transferase (GST) was measured. In addition, we performed genotoxicity assays, such as comet assay, micronucleus (MN) test and nuclear abnormalities (NA) induction, on the erythrocytes of the fish species. Our results revealed that the inhibition of hepatic GST activity in H. reidi was dependent on increasing crude oil concentrations. In contrast, an increase in the damage index (DI) and MN frequency were observed with increased crude oil concentrations. These results indicate that the alkaline comet assay and micronucleus test were suitable and useful in the evaluation of the genotoxicity of crude oil, which could improve determinations of the impact of oil spills on fish populations. In addition, H. reidi is a promising "sentinel organism" to detect the genotoxic impact of petroleum hydrocarbons.

The aim of this study was to identify the times of anesthetic induction and recovery in slender seahorses (Hippocampus reidi) that were exposed to the essential oil of Lippia alba (EO), as well as the efficacy of EO as a stress-reducing agent in the transport of this species. Slender seahorses were placed in 1-L aquaria containing different concentrations of EO (0, 10, 20, 50, 150, 300 and 450 µL L-1), and after induction, fish were transferred to aquaria that were free of anesthetic to evaluate their recovery time. In an additional experiment, slender seahorses were transported in plastic bags with 15 µL L-1 of EO for 4 or 24 h. The increased concentration of EO proportionally decreased the time required for the induction of anesthesia. EO treatment (15 µL L-1) inhibited the increase in blood glucose levels that was provoked by transportation for 4 or 24 h. Transportation for 24 h also decreased the number of lymphocytes and increased the neutrophil count, and these effects were avoided with the addition of EO to the water. These results demonstrate that EO was effective as an anesthetic at concentrations of 10-20 µL L-1 for slight sedation and transport and at 150 µL L-1 for deep anesthesia in the slender seahorse.(AU)
O objetivo deste estudo foi identificar os tempos da indução e recuperação anestésica em cavalos marinhos (Hippocampus reidi) expostos ao óleo essencial de Lippia alba (OE), assim como a eficácia do OE como um agente redutor de estresse no transporte desta espécie. Os cavalos marinhos foram colocados em aquários contendo 1 litro de água e diferentes concentrações de OE (0, 10, 20, 50, 150, 300 e 450 µL L-1), após a indução, os peixes foram transferidos à aquários livre de anestésico para avaliar o tempo de recuperação. Em um outro experimento os cavalos marinhos foram transportados em sacos plásticos contendo 15 µL L-1 do OE por 4 ou 24h. A concentração crescente do OE diminuiu proporcionalmente o tempo exigido para a indução da anestesia. O óleo essencial (15 µL L-1) inibiu o aumento nos níveis de glicose sanguínea provocada pelo transporte por 4 ou 24 h. O transporte por 24 h igualmente diminuiu o número de linfócitos e aumentou o número de neutrófilos, estas alterações foram evitadas com o uso do OE na água. Estes resultados demonstram que no cavalo marinho o OE é eficaz para a sedação e transporte na faixa de 10-20 µL L-1 e para a anestesia profunda recomenda-se 150 µL L-1.(AU)

This study was carried out to fill the gaps that remain under Hippocampus reidi biology. Analysis of variations of sex ratio, density, breeding season, distribution and home range of a population of the endangered Brazilian seahorse H. reidi from a rocky shore on Araçatiba beach, Ilha Grande, Brazil were carried out. Araçatiba beach is a tourist Environmental Protected Area, suffering antropic pressure. A fixed population of H. reidi was studied, where al lthe individuals were visually tagged and sex, reproductive state and location on site were identified from December 2002 to November 2004. A total of 20 individuals were visually tagged with a mean density of 0.18 m-2. Sex ratios were skewed, with more females than males. All the males brooded during 13 months and presented smaller home range than the females during the breeding season. The highest densities were found on shallowest areas.
Este estudo foi realizado com o objetivo de preencher algumas lacunas sobre a biologia natural de Hippocampus reidi. Foram analisadas as variações na proporção sexual e densidade, período reprodutivo, distribuição e área de deslocamento de uma população do cavalo marinho brasileiro ameaçado de extinção Hippocampus reidi de um costão rochoso da praia de Araçatiba, Ilha Grande, Brasil. A praia de Araçatiba é uma Área de Proteção Ambiental turística, a qual sofre com a pressão antropogênica. Foi estudada uma população fixa de H. reidi, sendo que todos os indivíduos foram marcados visualmente e foram determinados o sexo, o período reprodutivo e a localização na área de novembro de 2002 a novembro de 2004. Um total de 20 indivíduos foram marcados com uma densidade média na área de 0,18 m-2. A proporção sexual variou de forma alternada com maior número de fêmeas que de machos. Durante 13 meses todos os machos encontrados estavam incubando. Os cavalos marinhos apresentaram área de deslocamento menor que as fêmeas durante o período reprodutivo. As maiores densidades foram encontradas em áreas mais rasas da área de estudo.
RESUMEN
The vertebral system in Hippocampus hippocampus is highly specialised because of the vertical locomotion and tail prehensility. The vertebral elements represent a special case of morphological changes, being the metameric structures organised along a natural functional series. We investigated the shape changes along the vertebral spine in H. hippocampus through geometric morphometrics, in order to describe functional and structural patterns. Actually, the dorso-ventral tail bending ability in the genus Hippocampus is one of the most impressive morphological modifications in the evolutionary history of fishes. Vertebrae were analysed using a 2D configuration from the left lateral view. The variation along the vertebral series suggests the identification of cervical, abdominal, dorsal, and caudal groups. The first three (cervical) elements and the lOth (supra-dorsal) structure show peculiar morphologies and local adaptations, associated with neck angulation and dorsal fin muscles, respectively. The vertebral size decreases from the anteriormost element backward, with some local variation at the dorsal area. Major changes are related to allometric variation at the neural región. The caudal elements are characterised by a marked size decrease, with consequent allometric shape changes involving the rotation of the posterior vertebral opening. This allometric trajectory leads to a natural ventral bending of the tail, promoting its prehensile function. This morphological survey suggests an interesting structural network between posture, locomotion, and vertebral anatomy, underlying the main functional changes in the Hippocampus biomechanics. Geometric morphometrics is rather suitable to approach metameric studies in terms of serial variation and functional adaptations.
El sistema vertebral en Hippocampus hippocampus es altamente especializado, debido a la locomoción vertical y la cola prehensible. Los elementos vertebrales representan un caso especial de cambios morfológicos, siendo estructuras metaméricas organizadas a lo largo de una serie funcional natural. Se investigaron los cambios en la forma, a lo largo de la columna vertebral en H. hippocampus a través de morfometría geométrica, con el fin de describir sus patrones funcionales y estructurales. Actualmente, la capacidad de flexión dorso-ventral de la cola en el género Hippocampus es una de las más impresionantes modificaciones morfológicas en la historia evolutiva de los peces. Las vértebras se analizaron mediante una configuración 2D de la vista lateral izquierda. La variación a lo largo de la serie vertebral sugiere la identificación de grupos cervicales, abdominales, dorsales, caudales. Los tres primeros elementos (cervicales) y la 10ª (supra-dorsal) muestran una estructura morfológica peculiar y adaptaciones locales, asociadas con la angulación del cuello y los músculos de la aleta dorsal, respectivamente. El tamaño vertebral disminuye desde elemento más anterior hacia atrás, con algunas variaciones locales en la zona dorsal. Los principales cambios están relacionados con la variación alométrica en la región neural. Los elementos caudales se caracterizan por una marcada disminución de tamaño, con los consiguientes cambios en la formas aloméricas de rotación de la abertura vertebral posterior. Esta trayectoria alométrica conduce a una flexión ventral natural de la cola, promoviendo su función prensil. Este estudio morfológico sugiere una interesante red estructural entre postura, locomoción y la anatomía vertebral junto a los principales cambios funcionales en la biomecánica de Hippocampus. La morfometría geométrica es bastante adecuada para los estudios con enfoques metaméricos, en términos de variación de serie y adaptaciones funcionales.

This paper provides a case study of a threatened seahorse species, Hippocampus reidi, highlighting the importance of using ecological information to assist conservation and management initiatives. Underwater visual sighting data (50 x 2m transect) gathered along the NE, SE and S portions of the Brazilian coast revealed an unequal distribution across localities, perhaps related to harvesting pressure, and a mean density of 0.026 ind.m-2. Our findings suggest some restricted spatial use by H. reidi, which was consistent with its estimated home range, and with re-sighting of specimens. Reproduction was recorded year-round, however productive peaks may exist. Components of habitat structure mostly used as anchoring points were mangrove plants, macroalgae, cnidarians, seagrass, sponges, and bryozoans. Conservation recommendations include: further characterization and mapping of habitats; assessment of availability and condition of microhabitats in selected areas, and studies on dispersal routes during initial life stages
Neste artigo apresenta-se um estudo de caso de uma espécie ameaçada de cavalo-marinho, Hippocampus reidi, enfatizando a importância de utilizar informações ecológicas nas ações de conservação e manejo. Dados de censo visual (transectos de 50 x 2m) obtidos nas porções NE, SE e S da costa brasileira revelaram uma distribuição heterogênea entre as localidades, talvez relacionada à pressão de coleta, e uma densidade média de 0.026 ind.m-2. Os resultados sugerem limitação no uso do espaço por H. reidi (consistente com a área vital estimada e com a re-avistagem de exemplares) e que a reprodução ocorre o ano inteiro, embora com possíveis picos. Os componentes da estrutura do habitat mais freqüentemente utilizados como pontos de ancoragem pelos cavalos-marinhos foram vegetação de mangue, macroalgas, cnidários, capim marinho e briozoários. Recomendações conservacionistas incluem caracterização adicional e mapeamento de habitats; avaliação da disponibilidade e condição dos microhabitats em áreas selecionadas, e estudos sobre rotas de dispersão durante as fases iniciais de vida

We studied fecundity, survival, and growth of the seahorse Hippocampus ingens under semi-controlled conditions. Three wild brood stock mature males of 14.8, 24.5, and 32.0 g released 1,598, 1,703, and 1,658 juveniles. Juvenile stocking densities of 12 were settled in 60-1 aquariums in groups of 1, 12, and 20 days old organisms. The rate of survival was 21.5, 61.9, and 59.0%, respectively, in 35 days. Juveniles were fed a mix diet of rotifers B. plicatilis and Artemia nauplii, then they were transferred to a cement tank of 100,000 1 at a density of 50/1,000 1 and fed with live adult Artemia for 60 days more. They grew from an average of 0.7, 1.5, and 2.7 to 4.5, 5.4, and 6.7 cm, respectively, in 95 days. The seawater temperature varied from 17 to 23 degrees C.
Estudiamos la fecundidad, supervivencia, y crecimiento del caballito de mar, Hippocampus ingens en condiciones semi-controladas. Tres machos reproductores silvestres de 14.8, 24.5 y 32.0 g produjeron 1 598, 1 703 y 1 658 jóvenes. La densidad utilizada fue de 12 jóvenes por acuario de 60 l. Se agruparon en 1, 12 y 20 días de nacidos por acuario. La supervivencia fue de 78.5, 38.1 y 41.0 % en 35 días. Se les alimentó con una mezcla de rotíferos B. plicatilis y nauplios de Artemia para después transferirlos a estanques de 100 000 l a una densidad de 50/1 000 l, donde se les alimentó con Artemia adulta durante 60 días más. Crecieron de un promedio de 0.7, 1.5, y 2.7 a 4.5, 5.4 y 6.7 cm, respectivamente, en 95 días. La temperatura del agua marina utilizada varió de 17 a 23 oC.
