RESUMEN

An efficient procedure for in vitro propagation of Herreria salsaparrilha Martius was established from single-node explants (fourth and fifth nodes from apex to the base) derived from donor plants maintained under shading-house conditions. After surface sterilization, explants are inoculated in test tubes containing 15 mL of Murashige and Skoog (MS) medium without growth regulators. Cultures are maintained under 35 µmol m-2 s-1 irradiance, a 16/8-h light/dark light regime, at 26 ± 2 °C. The subcultures are carried out under the same conditions, adding 6-benzyladenine 1.0 mg/L and Phytagel® 2.8 g/L. Shoots are elongated and rooted by transferring individual shoots to half-strength MS medium without growth regulators. After 25-30 days, elongated rooted shoots are transferred to plastic pots containing 25-30 mL of sterile distilled water, covered with a transparent plastic bag, and kept under the same growth room conditions for 2 days. Plants are transferred to cups containing autoclaved and washed sand and kept in a shading house (50% light interception) for acclimatization. True-to-type adult plants were successfully recovered under ex vitro conditions.

Smilax brasiliensis Sprengel is a monocotyledon of the Smilacaceae family, native to the Brazilian Cerrado, popularly known as "salsaparrilha" or "japecanga". In this study, the ethanol extract (EE) and the hexane (HEXF), dichloromethane (DCMF), ethyl acetate (ACF), and hydroethanol (HEF) fractions of the stems were obtained. The chemical composition was determined, the contents of phenolic compounds and flavonoids were quantified, and the antioxidant potential and the cytotoxic effect on Artemia salina were evaluated. Fatty acid esters, hydrocarbons, and phytosterols were identified in the HEXF analyzed by gas chromatography - mass spectrometry (GC-MS). The EE and DCMF, ACF, and HEF were analyzed by liquid chromatography coupled to a diode array detector and mass spectrometer (LC-DAD-MS), and the identified constituents included glycosylated (rutin, 3-O-ß-galactopyranosyl quercetin, 3-O-ß-glucopyranosyl quercetin, O-deoxyhexosyl-hexosyl quercetin, O-deoxyhexosyl-hexosyl kaempferol, O-deoxyhexosyl-hexosyl O-methyl quercetin, and others), and non-glycosylated (quercetin) flavonoids, phenylpropanoids (3-O-E-caffeoyl quinic acid, 5-O-E-caffeoyl quinic acid, O-caffeoyl shikimic acid, and others), neolignan, steroidal saponin (dioscin), and N-feruloyltyramine. The EE, DCMF, and ACF showed high total contents of phenolic compounds (112.99, 175.71, and 524.02 µg of GAE/mg, respectively), and in the ACF and DCMF a great content of flavonoids was also quantified (50.08 and 31.49 µg of QE/mg, respectively). The EE, DCMF, ACF, and HEF exhibited great antioxidant potential by DPPH (IC50 1.71 - 32.83 µg/mL) and FRAP (IC50 0.63 - 6,71 µg/mL) assays. A maximum cytotoxic activity on A. salina of 60% was observed for the DCMF (LC50 = 856.17 µg/mL). This study contributes to the phytochemical study of S. brasiliensis since these compounds were identified for the first time in the stems of this species. The S. brasiliensis stems demonstrated to be a rich source of polyphenols compounds and exhibited high antioxidant potential without toxicity. Thus, extract and fractions obtained from the S. brasiliensis stems can be used in food supplements or as natural antioxidants in the food industry.

AbstractThe species of the genus Smilax, popularly known as sarsaparilla, are widely used in folk medicine due to the antirheumatic properties of its underground structures. Smilax fluminensis and S. syphilitica occur in forested areas and form thickened stems called rhizophores from which adventitious roots grow. To provide information for more accurate identification of the commercialised product and for elucidating the process of stem thickening, a morphology and anatomy study of the underground organs of the two species was conducted. The adventitious roots differ in colour and diameter depending on the stage of development. They are white and have a larger diameter in the early stages of development, but as they grow, the adventitious roots become brown and have a smaller diameter due to the disintegration of the epidermis and virtually the entire cortex. In brown roots, the covering function is then performed by the lignified endodermis and the remaining walls of the cells from the last parenchyma cortical layer. These results are similar to those found in studies of other Smilax and suggest that the anatomy of the roots can be useful for identifying fraud in commercialised materials. The thickening process of the nodal regions of the rhizophores in both species involves the activity of axillary buds and pericyclic layers.


ResumoAs espécies de Smilax, conhecidas popularmente como salsaparrilha, são amplamente utilizadas na medicina tradicional devido às propriedades antirreumáticas das estruturas subterrâneas. Smilax fluminensis e S. syphilitica ocorrem em áreas florestais e formam caules espessados denominados rizóforos a partir dos quais são emitidas raízes adventícias. Com o intuito de fornecer informações para a identificação mais precisa do material comercializado e no entendimento do processo de espessamento do caule, foi realizado o estudo morfológico e anatômico dos órgãos subterrâneos das duas espécies. As raízes adventícias apresentam diferenças na coloração e no diâmetro dependendo da fase de desenvolvimento. As raízes nas fases iniciais do desenvolvimento são brancas e possuem diâmetro maior, porém com o desenvolvimento, devido à desintegração da epiderme e de praticamente todo o córtex, as raízes tornam-se marrons e de diâmetro menor. Nas raízes marrons, a função de revestimento passa a ser exercida pela endoderme lignificada e pelas paredes remanescentes das células da penúltima camada cortical. Os resultados são semelhantes aos encontrados nos estudos de outras Smilax e sugerem que a anatomia das raízes pode ser útil na identificação de fraudes em materiais comercializados. O processo de espessamento das regiões nodais dos rizóforos nas duas espécies envolve a atividade das gemas axilares e de camadas pericíclicas.

Roots of Smilax species (Smilacaceae), named as salsaparrilha, have been used for centuries in Asia and Americas as depurative (=for "cleaning blood"), diuretic and sudorific. In southeast of Brazil, roots of Herreria salsaparrilha Mart. (Agavaceae) are also named as salsaparrilha and are used for the same purpose. In this study, we have evaluated the antihyperlipidemic and antihyperglycemic effects of extracts from roots of Smilax brasiliensis and H. salsaparrilha in mice fed with high-refined carbohydrate diet (HC). The chemical composition of the products was determined by LC-DAD and LC-MS. Groups of mice that received the HC diet showed an increase in plasma concentrations of glucose, triglycerides and total cholesterol, compared to control group, without treatment (p<0.05). Triglycerides were reduced significantly (p<0.05) in HC diet group that received 100 and 200mg/kg BW/day of both salsaparrilha extracts. Glucose and total cholesterol levels were reduced significantly (p<0.05) in the groups that received the higher doses (200mg/kg BW/day) of both extracts of salsaparrilha. Extracts of S. brasiliensis, at this dose also showed a higher reduction in triglycerides levels (p<0.001) and promoted a significant reduction in the adipocyte area (p<0.05). Both extracts showed the presence of saponins in LC analysis but S. brasiliensis has a higher concentration of phenolics, mainly chlorogenic acid. The presence of steroidal saponins might be responsible for the reduction of the cholesterol levels, while phenolics in S. brasiliensis by the metabolism of triglycerides and better fat distribution. The result is according with the traditional use of these plants and shown their potential for use as functional foods.

Smilax L. species, popularly known in Brazil as salsaparrilha, have been used in folk medicine as tonic against rheumatism and as anti-syphilitic since the sixteenth century; however, until today, their roots have been explored in an extractive way. The aim of this study was to analyze the propagation of Smilax fluminensis aiming to meet the demand and to propose procedures for its sustainable management. It was analyzed the percentage of seed germination, rooting of aerial and subterranean stem cuttings and plant resprouting capacity after removal of the shoots. The monitoring lasted one year. Seeds were subjected to temperatures: 25°C, 30°C, 20-30°C and 20-35°C under a daily photoperiod of eight hours and in the absence of light. For rooting the stem cuttings, aerial and subterranean stems about 20cm long with two nodal regions were subjected to treatment with distilled water (control) and indolbutyric acid (IBA) at 100ppm. The best germination percentages obtained for S. fluminensis were 80 percent at 20-30°C under light and 85 percent at 30°C in the dark. Only subterranean stem cuttings showed significant difference concerning fresh and dry matter of roots with higher values in treatments with hormone compared to the control.


As espécies do gênero Smilax L., conhecidas popularmente como salsaparrilha, são empregadas na medicina popular como fortificante contra o reumatismo e antissifilítico desde o século XVI, porém até hoje as suas raízes são exploradas de maneira extrativista. O objetivo deste estudo foi analisar a propagação de Smilax fluminensis visando a auxiliar no atendimento da demanda e apontar propostas para o seu manejo sustentável. Foram analisados a porcentagem de germinação de sementes, o enraizamento de estacas de ramos aéreos e subterrâneos e o acompanhamento ao longo de um ano da capacidade de rebrotamento das plantas após a retirada dos ramos aéreos. As sementes foram submetidas às temperaturas: 25, 30, 20-30 e 20-35°C, sob fotoperíodo diário de oito horas e na ausência de luz. Para o enraizamento de estacas, foram utilizados ramos aéreos e subterrâneos com aproximadamente 20cm com duas regiões nodais submetidas ao tratamento com água destilada (controle) e com ácido indolbutírico (AIB) a 100ppm. As melhores porcentagens de germinação obtidas para S. fluminensis, 80 por cento em 20-30°C no claro e 85 por cento a 30°C no escuro. Apenas as estacas de ramos subterrâneos enraizaram, houve diferença significativa em relação à massa fresca e seca de raízes com valores maiores nos tratamentos com hormônio em relação ao controle.

Brazilians have been using the underground organs of Smilax species in alternative medicine since the 19th century because of their anti-rheumatic qualities. However, even nowadays, these species are explored only by extractivism. Studies on seed germination and development of these organs could be useful to preserve these plants. After germination, seedling development of Smilax polyantha was analyzed to understand underground stem formation. Furthermore, to analyze the ontogenesis of the underground system, seedlings aged from one to twelve months were sectioned. One of the most striking features of this species is the presence of two stem branching systems. The plumule gives rise to the first stem branching system with negative geotropism. Its first underground axillary bud sprouted into the other caulinar axis with positive geotropism. The horizontal growth and the subsequent thickening of this underground organ depended on the development of axillary buds from basal nodes of the previous branches. The cotyledonary bud did not play a role in the underground formation, as previously described in the literature for this genus, but the buds of the basal cataphylls built the second stem branching system. In this study we discuss the terminology and suggest calling this second stem branching system a rhizophore.


As espécies de Smilax L. são utilizadas na medicina popular brasileira desde o século 19 devido às propriedades anti-reumáticas atribuídas aos órgãos subterrâneos de todas as espécies. No entanto, ainda hoje, essas espécies são exploradas apenas por extrativismo. Estudos sobre a germinação e o desenvolvimento dos órgãos subterrâneos podem ser úteis para preservar essas plantas. Após a germinação, o desenvolvimento de plântulas de Smilax polyantha foi analisado para compreender a formação do sistema subterrâneo. Para a análise da ontogênese do sistema subterrâneo foram seccionadas plantas em diferentes estágios de desenvolvimento entre um e doze meses. Uma das características mais marcantes desta espécie é a presença de dois sistemas de ramificação caulinar. A plúmula dá origem ao primeiro sistema caulinar de ramificações com geotropismo negativo. As gemas axilares subterrâneas desse primeiro eixo caulinar originam o segundo eixo caulinar com geotropismo positivo. O crescimento horizontal e o espessamento do órgão subterrâneo dependem do desenvolvimento de gemas axilares de nós basais dos ramos anteriores. A gema cotiledonar não participa da formação do caule subterrâneo, como descrito anteriormente na literatura para este gênero, mas as gemas axilares basais dos ramos caulinares aéreos originam o segundo eixo de ramificação caulinar subterrâneo. Neste estudo, após ser discutida a terminologia mais adequada, sugere-se chamar o caule subterrâneo dessas espécies de rizóforo.

Anacardium occidentale L. espécie nativa do continente centro e sul-americano, é planta amplamente utilizada, empregada na alimentação humana e animal, bem como na medicina popular. Neste trabalho objetivamos o reconhecimento das principais características diagnósticas das folhas e dos receptáculos carnosos. Detalhes como o tipo de cutícula, paredes celulares, anexos epidérmicos e inclusões celulares são destacados nas descrições e nas figuras. As folhas encerram flavonóides, saponinas,compostos fenólicos e óleo essencial.


Anacardium occidentale L., a native species of Central and South America, is a plant used as human and animal food as well as in popular medicine. The purpose of this paper is to show the most important anatomical characteristics of leaves and edible receptacles (pseudo-fruits) for identification purposes. Details like the type of cuticle, stomata, epidermis, annexes and celular inclusions were emphasized in the descriptions and on the pictures. Phytochemical reactions for flavonoids, saponins, phenolic compounds and essential oil complement the identification.
