RESUMEN

Counting growth layers in dentine and/or secondary cementum is widely used for age determination in wild mammals but the underlying seasonal changes in the structure and degree of mineralisation of dental tissue have not been well characterised. We embedded first (m1) and second (m2) mandibular permanent molar teeth from a 12-year-old female Svalbard reindeer (Rangifer tarandus platyrhynchus) in PolyMethylMethAcrylate (PMMA), prepared cut and polished surfaces coated with evaporated carbon and used 20 kV back-scattered electron imaging in a scanning electron microscope (BSE-SEM) to study aspects of dental tissue structure which depend on the degree of mineralisation at the micron and sub-micron scale. BSE-SEM revealed differences between the mineral content of growth layers (annulations) in the secondary cementum and the primary and secondary dentine, the latter, incidentally, still forming at death in m1. Wide bands of less well mineralised tissue formed in the cementum during active appositional phases. Thin, denser bands formed by maturation-mineralisation of existing tissue when growth slowed in winter. This maturation mimics the processes seen in lamellar bone and articular cartilage. Counter to previous suggestions, there was evidence of substantial resorption and repair of the secondary cementum and of formation of dentine throughout life. Secondary dentine is layered by mineral content like cementum. In the crown, this was mainly tubular dentine with well-marked interglobular dentine layers. In the lower pulp chamber and root, it was largely without tubules. Substantial non-mineralised spaces found at the cement-dentine junction in the root apical regions in m2 represent inclusions of the Hertwig's Epithelial Root Sheath (HERS) or the Epithelial Rests of Malassez (ERM) between the two tissues, a phenomenon which has previously only been identified in Muridae. The anatomical changes which result in the formation of the incremental lines (annulations) in dental tissues of reindeer, identified here for the first time at the micrometre level, are likely to be common across most if not all long-lived species of mammals living in seasonal environments.

Site fidelity-the tendency to reuse familiar spaces-is expected to improve fitness. Familiarity with the local environment is particularly crucial when resource demands or predation risk are high. Consequently, site fidelity often peaks during reproduction when energetic costs are high and offspring are vulnerable. For many species, the environment they experience is not solely a function of geography but also of the social environment. Social fidelity, the selection for familiar social environments, could constitute an independent or parallel strategy to spatial fidelity when considering behaviour at the spatial-social interface. Using global positioning system locations from caribou across Newfoundland, we tested whether females selected calving sites based on proximity to familiar conspecifics, in addition to geographical (spatial) fidelity. These strategies were synergistic, not alternative, and correlated across the population but more variable within individuals. We also tested whether either form of fidelity affected reproductive success. We failed to detect an effect of spatial or social fidelity on reproductive success in this population. Nevertheless, given the association between social and spatial fidelity and the demonstrated fitness consequences of site fidelity in other systems, familiar conspecifics and the potential benefits these social partners provide may be an underappreciated component driving site fidelity.This article is part of the theme issue 'The spatial-social interface: a theoretical and empirical integration'.

For prey, movement synchrony represents a potent antipredator strategy. Prey, however, must balance the costs and benefits of using conspecifics to mediate risk. Thus, the emergent patterns of risk-driven sociality depend on variation in space and in the predators and prey themselves. We applied the concept of predator-prey habitat domain, the space in which animals acquire food resources, to test the conditions under which individuals synchronize their movements relative to predator and prey habitat domains. We tested the response of movement synchrony of prey to predator-prey domains in two populations of ungulates that vary in their gregariousness and predator community: (i) elk, which are preyed on by wolves; and (ii) caribou, which are preyed on by coyotes and black bears. Prey in both communities responded to cursorial predators by increasing synchrony during seasons of greater predation pressure. Elk moved more synchronously in the wolf habitat domain during winter and caribou moved more synchronously in the coyote habitat domains during spring. In the winter, caribou increased movement synchrony when coyote and caribou domains overlapped. By integrating habitat domains with movement ecology, we provide a compelling argument for social behaviours and collective movement as an antipredator response. Abundance estimation is frequently an objective of conservation and monitoring initiatives for threatened and other managed populations. While abundance estimation via capture-mark-recapture or spatially explicit capture-recapture is now common, such approaches are logistically challenging and expensive for species such as boreal caribou (Rangifer tarandus), which inhabit remote regions, are widely dispersed, and exist at low densities. Fortunately, the recently developed 'close-kin mark-recapture' (CKMR) framework, which uses the number of kin pairs obtained within a sample to generate an abundance estimate, eliminates the need for multiple sampling events. As a result, some caribou managers are interested in using this method to generate an abundance estimate from a single, non-invasive sampling event for caribou populations. We conducted a simulation study using realistic boreal caribou demographic rates and population sizes to assess how population size and the proportion of the population surveyed impact the accuracy and precision of single-survey CKMR-based abundance estimates. Our results indicated that abundance estimates were biased and highly imprecise when very small proportions of the population were sampled, regardless of the population size. However, the larger the population size, the smaller the required proportion of the population surveyed to generate both accurate and reasonably precise estimates. Additionally, we also present a case study in which we used the CKMR framework to generate annual female abundance estimates for a small caribou population in Jasper National Park, Alberta, Canada, from 2006 to 2015 and compared them to existing published capture-mark-recapture-based estimates. Both the accuracy and precision of the annual CKMR-based abundance estimates varied across years and were sensitive to the proportion of pairwise kinship comparisons which yielded a mother-offspring pair. Taken together, our study demonstrates that it is possible to generate CKMR-based abundance estimates from a single sampling event for small caribou populations, so long as a sufficient sampling intensity can be achieved.

During a survey for helminths in reindeer (Rangifer tarandus) across the Palearctic region, eggs were found in zoo reindeer feces. These were identified as eggs of Fasciola hepatica based on their morphology, morphometrics, location, and analysis of their partial sequence of ITS rDNA region. Some of the eggs had an appendage, previously unreported. Additionally, adults of F. hepatica were studied. Eggs derived from their uteri were also appendaged. Diagnostic morphological traits of F. hepatica eggs (abopercular appendage, knob, egg shell thickening, and egg shape) are discussed in this article. Three dimensional models of F. hepatica eggs were created to demonstrate the eggs features as best as possible. Since fecal examination remains gold standard in diagnosing fasciolosis in humans and animals worldwide, our findings may contribute to improved diagnostics. This research has also shown that reindeer can be a final host for F. hepatica. We also discuss whether the Novaya Zemlya archipelago might be the northernmost site of fasciolosis.

For decades, researchers have employed sound to study the biology of wildlife, with the aim to better understand their ecology and behaviour. By utilizing on-animal recorders to capture audio from freely moving animals, scientists can decipher the vocalizations and glean insights into their behaviour and ecosystem dynamics through advanced signal processing. However, the laborious task of sorting through extensive audio recordings has been a major bottleneck. To expedite this process, researchers have turned to machine learning techniques, specifically neural networks, to streamline the analysis of data. Nevertheless, much of the existing research has focused predominantly on stationary recording devices, overlooking the potential benefits of employing on-animal recorders in conjunction with machine learning. To showcase the synergy of on-animal recorders and machine learning, we conducted a study at the Kutuharju research station in Kaamanen, Finland, where the vocalizations of rutting reindeer were recorded during their mating season. By attaching recorders to seven male reindeer during the rutting periods of 2019 and 2020, we trained convolutional neural networks to distinguish reindeer grunts with a 95% accuracy rate. This high level of accuracy allowed us to examine the reindeers' grunting behaviour, revealing patterns indicating that older, heavier males vocalized more compared to their younger, lighter counterparts. The success of this study underscores the potential of on-animal acoustic recorders coupled with machine learning techniques as powerful tools for wildlife research, hinting at their broader applications with further advancement and optimization.

Individuals that isolate themselves to give birth can use more than one strategy in choosing birth sites to maximize reproductive success. Previous research has focused on the consistency in the use of the same birth-site across years (i.e., spatial fidelity), but individuals alternatively may use similar habitat conditions across years (i.e., habitat fidelity). Using GPS telemetry, we determined whether woodland caribou expressed spatial or habitat fidelity during calving, and evaluated intrinsic and extrinsic factors associated with expressing either type of fidelity. We identified 56 individuals with ≥2 putative birth events, via a movement-based model, across northern Ontario between 2010 and 2014. Individuals were classified as expressing (1) spatial fidelity by comparing sequential calving locations to a random spatial distribution of available calving locations, (2) habitat fidelity using a logistic use model compared to a null (intercept only) model, (3) no fidelity (neither criterion met), or (4) both spatial and habitat fidelity (both criteria met). Across all individuals, 37% expressed no fidelity (36 of 98), 15% expressed only spatial fidelity (15 of 99), 35% expressed only habitat fidelity (34 of 98), and 14% expressed both spatial and habitat fidelity (14 of 98). Older individuals were more likely to express spatial fidelity, whereas lower availability of upland and lowland conifer forests without linear features increased the probability an individual expressed habitat fidelity. Our results indicate that managing for caribou calving needs to consider protecting both specific, known birthing sites, but also broad-scale areas of preferred habitat for calving. Understanding the mechanisms that influence caribou expressing calving fidelity, and associated fitness costs, is crucial for the conservation of the species.

We investigated Alongshan virus infection in reindeer in northeastern China. We found that 4.8% of the animals were viral RNA-positive, 33.3% tested positive for IgG, and 19.1% displayed neutralizing antibodies. These findings suggest reindeer could serve as sentinel animal species for the epidemiologic surveillance of Alongshan virus infection.

Strong disturbances may induce ecosystem transitions into new alternative states that sustain through plant-soil interactions, such as the transition of dwarf shrub-dominated into graminoid-dominated vegetation by herbivory in tundra. Little evidence exists on soil microbial communities in alternative states, and along the slow process of ecosystem return into the predisturbance state. We analysed vegetation, soil microbial communities and activities as well as soil physico-chemical properties in historical reindeer enclosures in northernmost Finland in the following plot types: control heaths in the surrounding tundra; graminoid-dominated; 'shifting'; and recovered dwarf shrub-dominated vegetation inside enclosures. Soil fungal communities followed changes in vegetation, whereas bacterial communities were more affected by soil physico-chemical properties. Graminoid plots were characterized by moulds, pathotrophs and dark septate endophytes. Ericoid mycorrhizal and saprotrophic fungi were typical for control and recovered plots. Soil microbial communities inside the enclosures showed historical contingency, as their spatial variation was high in recovered plots despite the vegetation being more homogeneous. Self-maintaining feedback loops between plant functional types, soil microbial communities, and carbon and nutrient mineralization act effectively to stabilize alternative vegetation states, but once predisturbance vegetation reestablishes itself, soil microbial communities and physico-chemical properties return back towards their predisturbance state.

Bone extracellular matrix (ECM) proteins play a key role in bone formation and regeneration, including structural and regulatory functions. The Phylobone database consists of 255 ECM protein groups from 39 species and can be used to support bone research. Here, we gathered bone ECM proteins from reindeer (Rangifer tarandus), a member of the Cervidae family. The importance of reindeer lies in their ability to regenerate their antlers, in both male and female individuals. Protein sequences were extracted from the National Center for Biotechnology Information's repository and selected by homology searches. We identified 215 proteins and their corresponding functional domains, which are putatively present in the bone ECM of reindeer. Protein sequence alignments have shown a high degree of conservation between R. tarandus and other members of the Cervidae family. This update expands the Phylobone database and shows that it is a useful resource for the preliminary annotation of bone ECM proteins in novel proteomes.

Harvesting and culling are methods used to monitor and manage wildlife diseases. An important consequence of these practices is a change in the genetic dynamics of affected populations that may threaten their long-term viability. The effective population size (N e) is a fundamental parameter for describing such changes as it determines the amount of genetic drift in a population. Here, we estimate N e of a harvested wild reindeer population in Norway. Then we use simulations to investigate the genetic consequences of management efforts for handling a recent spread of chronic wasting disease, including increased adult male harvest and population decimation. The N e/N ratio in this population was found to be 0.124 at the end of the study period, compared to 0.239 in the preceding 14 years period. The difference was caused by increased harvest rates with a high proportion of adult males (older than 2.5 years) being shot (15.2% in 2005-2018 and 44.8% in 2021). Increased harvest rates decreased N e in the simulations, but less sex biased harvest strategies had a lower negative impact. For harvest strategies that yield stable population dynamics, shifting the harvest from calves to adult males and females increased N e. Population decimation always resulted in decreased genetic variation in the population, with higher loss of heterozygosity and rare alleles with more severe decimation or longer periods of low population size. A very high proportion of males in the harvest had the most severe consequences for the loss of genetic variation. This study clearly shows how the effects of harvest strategies and changes in population size interact to determine the genetic drift of a managed population. The long-term genetic viability of wildlife populations subject to a disease will also depend on population impacts of the disease and how these interact with management actions.

Annual antler growth begins in the spring and is completed by late summer for male caribou (Rangifer tarandus groenlandicus) from the Qamanirjuaq herd (Nunavut, Canada), aligned with both the spring migration and a seasonal dietary shift. Antlers may provide a non-lethal means of studying short- and long-term changes in caribou ecology through incorporated isotopes of carbon (δ13C) and nitrogen (δ15N). We sampled the antlers of 12 male caribou from the Qamanirjuaq herd culled in September 1967. We predicted that serial sampling of antlers would reflect the known seasonal dietary change from lichen to grass-like and shrub diet based on rumen contents from individuals culled during the same period. The δ13C and δ15N were analyzed in food sources and every 3 cm along each antler's length. The carbon isotope compositions of collagen (δ13Ccol) varied by ~0.5‰ among individuals and within antlers, while the carbon isotope compositions of antler bioapatite (δ13CCO3) increased by 1-1.5‰ from pedicle to tip. Values of δ15Ncol increased within antlers by 1-3‰ from pedicle to tip and varied by 3‰ among the individuals sampled. Antler collagen was lower in δ15Ncol by ~1‰ relative to bone collagen. Bayesian mixing models were conducted to test for changes in dietary proportions from antler isotope compositions. Mixing models did not indicate significant dietary shifts for any individual during antler formation, showing consistently mixed diets of fungi, horsetail, lichen, and woody plants. Increases in δ15Ncol in antler tissue could, therefore, correspond to subtle seasonal dietary changes and/or the physiological stress of antler tissue development.

In northern Sweden, improvements of grazing conditions are necessary for the continuation of traditional, natural pasture-based reindeer husbandry. Ground and tree lichen constitute the main fodder resource for reindeer during winter but have reached critically low levels. Using a forest decision support system, we prescribe adapted forest management to improve the preconditions for reindeer husbandry and compare outcomes with the continuation of current forest management. We found that adapted management increases the forest area with ground lichen habitat by 22% already within 15 years, while a continuation of current management would result in a further decrease in ground lichen. Tree lichen habitat can be retained and increased in all scenarios, which is important in a changing climate. Compared to a continuation of current practices, adapted management with significantly improved conditions for lichen resulted in a decrease in net revenues from wood production by 11-22%.

Assessing the effects of industrial development on wildlife is a key objective of managers and conservation practitioners. However, wildlife responses are often only investigated with respect to the footprint of infrastructure, even though human activity can strongly mediate development impacts. In Arctic Alaska, there is substantial interest in expanding energy development, raising concerns about the potential effects on barren-ground caribou (Rangifer tarandus granti). While caribou generally avoid industrial infrastructure, little is known about the role of human activity in moderating their responses, and whether managing activity levels could minimize development effects. To address this uncertainty, we examined the influence of traffic volume on caribou summer space use and road crossings in the Central Arctic Herd within the Kuparuk and Milne Point oil fields on the North Slope of Alaska. We first modeled spatiotemporal variation in hourly traffic volumes across the road system from traffic counter data using gradient-boosted regression trees. We then used generalized additive models to estimate nonlinear step selection functions and road-crossing probabilities from collared female caribou during the post-calving and insect harassment seasons, when they primarily interact with roads. Step selection analyses revealed that caribou selected areas further from roads (~1-3 km) during the post-calving and mosquito seasons and selected areas with lower traffic volumes during all seasons, with selection probabilities peaking when traffic was <5 vehicles/h. Using road-crossing models, we found that caribou were less likely to cross roads during the insect seasons as traffic increased, but that response dissipated as insect harassment became more severe. Past studies suggested that caribou exhibit behavioral responses when traffic exceeds 15 vehicles/h, but our results demonstrate behavioral responses at much lower traffic levels. Our results illustrate that vehicle activity mediates caribou responses to road infrastructure, information that can be used in future land-use planning to minimize the behavioral responses of caribou to industrial development in sensitive Arctic landscapes.

The contraction of species range is one of the most significant symptoms of biodiversity loss worldwide. While anthropogenic activities and habitat alteration are major threats for several species, climate change should also be considered. For species at risk, differentiating the effects of human disturbances and climate change on past and current range transformations is an important step towards improved conservation strategies. We paired historical range maps with global atmospheric reanalyses from different sources to assess the potential effects of recent climate change on the observed northward contraction of the range of boreal populations of woodland caribou (Rangifer tarandus caribou) in Quebec (Canada) since 1850. We quantified these effects by highlighting the discrepancies between different southern limits of the caribou's range (used as references) observed in the past and reconstitutions obtained through the hindcasting of the climate conditions within which caribou are currently found. Hindcasted southern limits moved ~105 km north over time under all reanalysis datasets, a trend drastically different from the ~620 km reported for observed southern limits since 1850. The differences in latitudinal shift through time between the observed and hindcasted southern limits of distribution suggest that caribou range recession should have been only 17% of what has been observed since 1850 if recent climate change had been the only disturbance driver. This relatively limited impact of climate reinforces the scientific consensus stating that caribou range recession in Quebec is mainly caused by anthropogenic drivers (i.e. logging, development of the road network, agriculture, urbanization) that have modified the structure and composition of the forest over the past 160 years, paving the way for habitat-mediated apparent competition and overharvesting. Our results also call for a reconsideration of past ranges in models aiming at projecting future distributions, especially for endangered species.

The decline of most caribou (Rangifer tarandus) populations underlines the need to understand the determinants of key demographic parameters. In migratory caribou, we have limited information on rates and drivers of pre-weaning mortality. We fitted 60 pregnant females of the Rivière-aux-Feuilles caribou herd with GPS camera collars to track the survival of calves from birth to weaning in 2016-2018. Over the three years, calf survival rate before weaning, i.e. to 01-Sep, approximately three months of age, was 0.63 (CI 0.50-0.77). Summer mortality risk was mainly influenced by calf birth date, with calves born earlier in the calving season having a lower mortality risk than those born later. Mortality also increased when calves experienced low or high temperature during calving. This study provides the first estimates of pre-weaning survival of migratory caribou calves in this herd, illustrating the value of new technologies to collect data otherwise difficult to obtain in widely distributed migratory populations. This approach can easily be extended to other large herbivores and predators. Our study brings new insights on how climate change may affect summer juvenile survival given the increased temperatures and faster changes in plant phenology expected in the future.

For the first time, information is provided on the size of the home range of Siberian forest reindeer (Rangifer tarandus valentinae) of the Altai-Sayan population of Kuznetsk Alatau, the minimum distance that the deer passes per day, their changes, and changes of habitats during the annual cycle. Possible reasons for the cyclical use of the habitat by forest reindeer are discussed.

The aim of the present work was to provide an overview of management and feeding practices, and the prevalence of endoparasite infections in captive Swiss reindeer. On two visits to eight farms or zoos, a standardized questionnaire was completed. A total of 67 reindeer were weighed, and fecal samples were collected. The primary management concerns voiced by owners/managers were feeding and successful breeding. All reindeer were fed roughage ad libitum and supplementary feed for reindeer or other browsers, with different compositions in each herd. Males over two years of age weighed from 60 kg up to 127.5 kg, whereas females had a body weight from 53.5 kg to 86.5 kg. The prevalence of gastrointestinal strongyles was 68.6% (46/67), with reindeer in zoos having a lower prevalence (36%; 9/25) than reindeer from private farms (88%; 37/42). Capillaria sp., Strongyloides sp., and Trichuris sp. were detected in lower prevalences (<24%) and were also more frequent in private farms. Intestinal protozoa, as well as fluke and tapeworms, were not detected in any herd. This study provides an overview on husbandry, feeding, and endoparasite prevalence in reindeer in Switzerland and should be of help for breeders and veterinarians dealing with this animal species.

BACKGROUND: Genome assembly into chromosomes facilitates several analyses including cytogenetics, genomics and phylogenetics. Despite rapid development in bioinformatics, however, assembly beyond scaffolds remains challenging, especially in species without closely related well-assembled and available reference genomes. So far, four draft genomes of Rangifer tarandus (caribou or reindeer, a circumpolar distributed cervid species) have been published, but none with chromosome-level assembly. This emblematic northern species is of high interest in ecological studies and conservation since most populations are declining. RESULTS: We have designed specific probes based on Oligopaint FISH technology to upgrade the latest published reindeer and caribou chromosome-level genomes. Using this oligonucleotide-based method, we found six mis-assembled scaffolds and physically mapped 68 of the largest scaffolds representing 78% of the most recent R. tarandus genome assembly. Combining physical mapping and comparative genomics, it was possible to document chromosomal evolution among Cervidae and closely related bovids. CONCLUSIONS: Our results provide validation for the current chromosome-level genome assembly as well as resources to use chromosome banding in studies of Rangifer tarandus.

Attraction sites are important for environmental pathogen transmission and spillover. Yet, their role in wildlife disease dynamics is often poorly substantiated. Herein, we study the role of salt licks as potential attraction sites for the spillover of gastrointestinal parasites from domestic sheep to wild reindeer. Eggs from the introduced sheep nematode Nematodirus battus were found in faecal samples of both species, suggestive of spillover. DNA metabarcoding of soil, collected at salt licks, revealed that N. battus, in addition to Teladorsagia circumcincta, were the most frequently occurring parasitic nematodes, with a significantly higher prevalence of nematodal DNA in salt lick soil compared to soil from control sites nearby. The finding of similar DNA haplotypes of N. battus in sheep, reindeer, and salt lick soil supports the hypothesis of spillover to reindeer via salt licks. More detailed investigation of the genetic diversity of N. battus across these hosts is needed to draw firm conclusions. Infection with these sheep nematodes could potentially explain a recently observed decline in the calf recruitment rate of the Knutshø reindeer herd. This study also supports the hypothesized role of artificial salt licks as hot spots for the transmission of environmentally persistent pathogens and illustrates the importance of knowledge about such attraction points in the study of disease in free-roaming animals.
