RESUMEN Objetivos. Determinar la alimentación del Aedes aegypti en brotes de dengue de dos zonas rurales del Perú durante el ciclón Yaku y El Niño Global del 2023. Material y métodos. Se analizaron ocho muestras de sangre (8 pooles) obtenidas del abdomen de 80 especímenes Aedes aegypti capturados en los distritos rurales de Querecotillo y Marcavelica durante brotes de dengue acontecidos en el ciclón Yaku y en El Niño Global. Se extrajo ADN de las muestras analizadas, se llevó a cabo una PCR dirigida al gen CytB como marcador genético y los productos PCR fueron digeridos enzimáticamente con las restrictasas Hae III y Mwo I. Los productos PCR-RFLP fueron visualizados por electroforesis en gel de agarosa al 4%. Resultados. Se obtuvo ADN de todas las muestras y como producto PCR un amplicón de 358 pb. Así mismo, el único RFLP en Hae III observado fue el de Homo sapiens sapiens (233 y 125 pb). No se observó RFLP en Hae III de Gallus gallus y RFLP en Mwo I de Canis familiaris y Mus musculus. Conclusión. En brotes de dengue de zonas rurales, durante el ciclón Yaku y en El Niño Global, el Aedes aegypti presentó un comportamiento alimenticio antropofílico conservado.
ABSTRACT Objective. To determine the feeding behavior of Aedes aegypti in dengue outbreaks in two rural areas of Peru during the Yaku cyclone and El Niño phenomenon of 2023. Material and methods. Eight blood samples (8 pools) were obtained from the abdomen of 80 Aedes aegypti specimens captured in the rural districts of Querecotillo and Marcavelica during the Yaku cyclone and El Niño dengue outbreaks. DNA was extracted from the analyzed samples, then a PCR was directed at the CytB gene as a genetic marker and the PCR products were enzymatically digested with the restrictases Hae III and Mwo I. The PCR-RFLP products were visualized by agarose gel electrophoresis at 4%. Results. DNA was obtained from all samples and a 358 bp amplicon was obtained as a PCR product. Likewise, the only RFLP found in Hae III was from Homo sapiens sapiens (233 and 125 bp). RFLP was not found in Hae III of Gallus gallus and RFLP in Mwo I of Canis familiaris and Mus musculus. Conclusion. Aedes aegypti showed conserved anthropophilic feeding behavior in dengue outbreaks in rural areas during the Yaku cyclone and El Niño.

ABSTRACT BACKGROUND: Gastric cancer is the fourth most common cause of worldwide cancer. Also in contrast to the huge advances in curing, the chance of living is very low even in surgery cases. Having a genetic predisposition plays an important role in cancer development. The association between Metallothionein-2A gene polymorphisms and the risk of adenocarcinoma has been widely studied, yet there is only one study on stomach diseases. OBJECTIVE: In this study, we aimed to investigate the association between 2 (MT-2A) polymorphisms and adenocarcinoma. METHODS: This cross-sectional case control study was performed between Mach 2014 and January 2015 at the Tuba Hospital of Sari, Iran. Peripheral blood samples were collected in EDTA tube. DNA extraction was performed using the spin column procedure. The MT-2A polymorphisms MT-2A (rs1610216), (rs28366003) were determined by polymerase chain reaction-restriction fragment length polymorphism analysis in 95 a topic adenocarcinoma patients and 90 healthy individuals from Iranian population. RESULTS: The MT-2A rs1610216 polymorphism increased the risk of adeno carcinoma in our Iranian population [OR: 3.8533; 95%CI, 1.3155-11.2869; P=0.0139] and rs28366003 [OR: 4.0978; 95%CI, 1.2521-13.4108; P=0.0197]. CONCLUSION: The MT-2A gene polymorphism was associated with the risk of adenocarcinoma in the Iranian population.
RESUMO CONTEXTO: O câncer gástrico é a quarta causa mais comum de câncer em todo o mundo. Também em contraste com os enormes avanços na cura, a chance de viver é muito baixa, mesmo em casos de cirurgia. Ter uma predisposição genética desempenha um papel importante no desenvolvimento do câncer. A associação entre polimorfismos do gene metalotioneína-2A e o risco de adenocarcinoma tem sido amplamente estudada, mas há apenas um estudo sobre doenças estomacais. OBJETIVO: Neste estudo, objetivou-se investigar a associação entre 2 (MT-2A) polimorfismos e adenocarcinoma. MÉTODOS: Um estudo de controle de caso transversal foi realizado entre março de 2014 e janeiro de 2015 no hospital Tuba, Sari, Irã. Amostras de sangue periférico foram coletadas em tubo EDTA. A extração do ADN foi executada usando o procedimento da coluna da rotação. Os polimorfismos MT-2a MT-2A (rs1610216), (rs28366003) foram determinados pela análise do polimorfismo do comprimento do fragmento da reação-limitação de cadeia da polimerase em 95 pacientes com adenocarcinoma tópico e em 90 indivíduos saudáveis da população iraniana. RESULTADOS: O polimorfismo MT-2A rs1610216 aumentou o risco de adenocarcinoma de em nossa população iraniana. [OR: 3,8533; 95%CI, 1,3155-11,2869; P=0,0139] e rs28366003 [OR: 4,0978; 95%CI, 1,2521-13,4108; P=0,0197]. OBJECTIVE: To establish the concordance between the two tests as identifying methods for circulating species in Colombia. MATERIALS AND METHODS: A total of 96 isolates from patients with cutaneous or mucosal leishmaniasis were selected and identified by MLEE and PCR-RFLP with miniexon and hsp70 as the molecular targets, which were used sequentially. Restriction enzymes HaeIII and BccI were similarly applied. Cohen's kappa coefficient and the 95% confidence interval (CI) were calculated. RESULTS: The kappa coefficient and the 95% CI between MLEE and PCR-RFLP displayed "very good" concordance with a coefficient of 0.98 (CI95%: 0.98 to 1.00). The identified species were Leishmania Viannia braziliensis, Leishmania Viannia panamensis, Leishmania Viannia guyanensis and Leishmania Leishmania amazonensis. A total of 80 of the 96 isolates were sequenced and the results obtained by PCR-RFLP were confirmed. CONCLUSION: Due to the concordance obtained between tests results with the amplification of the genes miniexon and hsp70, PCR-RFLP is proposed as an alternative for identifying circulating Leishmania species in Colombia.

Abstract Introducción. La electroforesis de enzimas multilocus (Multilocus Enzyme Electrophoresis, MLEE) es el estándar de referencia para la tipificación de las especies de Leishmania. La prueba está restringida a laboratorios especializados por su complejidad técnica, sus costos y el tiempo necesario para obtener resultados. La PCR-RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism) se utiliza para tipificar especies de Leishmania. Objetivo. Establecer la concordancia entre las dos pruebas como métodos de tipificación de las especies circulantes en Colombia. Materiales y métodos. Se seleccionaron 96 aislamientos de pacientes con leishmaniasis cutánea o mucocutánea y se tipificaron mediante MLEE y PCR-RFLP con los blancos moleculares miniexon y hsp70 usados en serie. Las enzimas de restricción aplicadas fueron la HaeIII y la BccI, respectivamente. Se calculó el coeficiente kappa y un intervalo de confianza (IC) de 95 %. Resultados. Se determinó que la concordancia fue "muy buena" al obtener un coeficiente de 0,98 (IC95%: 0,98-1,00). Las especies identificadas fueron: Leishmania Viannia braziliensis, L. (V.) panamensis, L. (V.) guyanensis y L. (L,) amazonensis. De los 96 aislamientos, 80 se enviaron a secuenciación y se confirmaron los resultados obtenidos mediante PCR-RFLP. Conclusión. Dada la concordancia obtenida con la PCR-RFLP amplificando los genes miniexon y hsp70, se propone esta prueba como alternativa para la tipificación de especies de Leishmania circulantes en Colombia.

Fundamento: la principal causa para el cáncer cervico uterino es el papilomavirus humano de alto riesgo. No existen antecedentes de estudios moleculares para la tipificación de papilomavirus humano en la población de Camagüey. La reacción en cadena de la polimerasa es una técnica de Biología Molecular que se ha usado desde siempre para el diagnóstico clínico; esta permite confirmar la presencia del ADN del Papilomavirus en el ADN total extraído a partir de muestras de pacientes con cáncer de cuello uterino. Objetivo: demostrar por primera vez los genotipos papilomavirus humano de alto riesgo circulantes, que causan cáncer de cuello uterino en la población femenina de Camagüey, Cuba. Métodos: se realizó un estudio analítico prospectivo donde se estudiaron 22 pacientes femeninas de la provincia de Camagüey, que fueron atendidas en la consulta de Patología de cuello del Hospital Ginecoobstétrico. La identificación y tipificación de los genotipos papilomavirus humano se realizó mediante el procedimiento molecular polimorfismo de longitud en los fragmentos de restricción. Resultados: el 63, 6 % de los pacientes presentaron lesiones tipo exofítica, el 4, 5 % endofítica y el 31, 8 % de otros tipos. Este estudio confirmó que los genotipos papilomavirus humanos de alto riesgo circulantes en la provincia Camagüey son los genotipos 16 y 31, donde el más frecuente fue el genotipo 16. Conclusiones: la presente investigación constituye el primer reporte de un estudio molecular de papilomavirus humanos a partir de muestras de pacientes con cáncer de cuello uterino en la provincia de Camagüey, Cuba. Estos resultados, junto a los obtenidos por otros autores, tienen una contribución importante en el diseño de preparados vacunales preventivos o terapéuticos, cada vez más efectivo hacia una solución anticipada para el cáncer de cuello uterino en Cuba.
Background: it is demonstrated that the main cause of cervical cancer is high risk humanp virus. There is no precedent of molecular studies for the typing of Human Papilloma Virus in the population of Camagüey. Polymerase chain reaction is Molecular Biology technique that has been used traditionally for the clinical diagnosis and other purposes. This technique allows confirming the presence of papillomavirus´DNA in the total extracted DNA, from samples of patients with cervical cancer. Objective: to demonstrate for the first time existing high-risk human papilloma virus genotypes that cause cervical cancer in female population of Camagüey, Cuba. Methods: a prospective analytic study was conducted, in which 22 female patients of the province of Camagüey were studied. They received medical attention at Ana Betancourt Hospital. Identification and typing of the Human Papilloma Virus genotypes was carried through the molecular procedure Restriction Fragment Length Polymorphism. Results: patients who presented exophytic lesions accounted for 63, 6%, 4, 5 % had endophytic type, and 31, 8 % presented other types. This study confirmed that high-risk Human Papilloma Virus genotypes existing in the province of Camagüey are genotypes 16 and 31, and the most frequent is 16. Conclusions: this research is the first report of a molecular study of Human Papilloma Virus from samples of patients with cervical cancer in the province of Camagüey, Cuba. These results, along with the ones obtained by other authors, make an important contribution in the design of the increasingly effective therapeutic and preventive vaccine to an anticipated solution to cervical cancer in Cuba.

INTRODUCTION: Ten viral genotypes (A-J) distributed in all continents have been described for hepatitis B virus (HBV). One of the methodologies for determining the viral genotype is the restriction fragment length polymorphism (RFLP) technique, a simple and relatively inexpensive method, albeit with some limitations. OBJECTIVE: The initial objective of the project was to identify the HBV genotypes by RFLP in serum samples obtained from patients and blood donors. However, due to the discrepancies of RFLP patterns it was also necessary to perform phylogenetic genotyping and in silico analysis of HBV sequences. MATERIALS AND METHODS: We obtained 56 serum samples. DNA extraction was followed by PCR amplification of a fragment of HBV ORF S. We analyzed PCR products by RFLP with AlwI, BsrI, CfrI, HpaII and StyI, and we sequenced some. We compared the patterns obtained with those in previous reports. We also performed RFLP analysis in silico since we found differences between the patterns expected and those obtainedResults: We identified genotypes A and F, subgenotype F3, in the samples. This result is in agreement with those of previous studies carried out in Colombia; indeed, subgenotype F3 is the most frequent in the Andean region of the country, while genotype A is the most frequent HBV genotype in the western region (department of Chocó). Based on the in silico analysis of 229 HBV sequences from GenBank and 11 sequences of this study, we identified the RLFP pattern for genotype F, subgenotype F3, and we described some modifications of genotype A RFLP patterns. CONCLUSIONS: We identified the single nucleotide polymorphism pattern for genotype F, subgenotype F3, by in silico analysis and sequencing. Further robust in silico analyses are necessary to validate the RFLP patterns of HBV genotype and subgenotypes.

INTRODUÇÃO: A incidência de tuberculose (TB) em Santos (SP) situa-se em 73/100.000 habitantes-ano. A prevalência média de coinfecção TB/HIV é de 16%, taxas de cura e abandono de tratamento, entre casos novos são, respectivamente, 71% e 12%. Tais indicadores sugerem elevado risco para TB multidroga resistente (TBMR) no município, com incidência estimada em 1,9/100.000 habitantes-ano. OBJETIVO: Descrever e analisar o perfil de sensibilidade às drogas (TS) de primeira e segunda linha de tratamento entre pacientes com TB pulmonar (TBP), estimar a incidência de TBMR e a proporção de TB extensivamente resistente (TBXR); analisar aspectos moleculares, epidemiológicos e institucionais dos casos de TBP resistentes em Santos (SP). MÉTODOS: Estudo descritivo de uma coorte de pacientes de TBP, com início de tratamento ou retratamento entre 01 de janeiro de 2011 a 31 de dezembro de 2012. Definiu-se como caso de TBP, indivíduos com 15 anos ou mais, de ambos os sexos, residentes no município de Santos, com manifestações clínicas compatíveis com TBP e confirmação por cultura com isolamento de Mycobacterium tuberculosis. As variáveis de interesse para o estudo foram as características sociodemográficas, história atual e pregressa de TB, aspectos relativos ao tratamento, co-morbidades, ao diagnóstico e resistência a drogas. Para as análises comparativas entre proporções foram usados os testes qui-quadrado de Pearson e o Exato de Fisher e para variáveis contínuas o teste T de Student ou o de Kruskal - Wallis. Os perfis genéticos dos isoladas resistentes a ao menos uma droga foram obtidos pela técnica RLFP e analisados pelo programa Bionumerics versão 5.0 (Applied Maths - Bélgica). A descrição da distribuição espacial da TB resistentes e clusters foram feitas mediante a inserção dos casos no mapa de Santos, por endereço de residência, segundo o índice de vulnerabilidade social. INTRODUCTION: The incidence of tuberculosis (TB) in Santos (SP) is located around 73 / 100,000-year, approximately double that found on average in the country. The average prevalence of TB / HIV is 16% cure rates and treatment dropout among new cases are, respectively, 71% and 12%. Such indicators suggest high risk for multidrug-resistant TB (MR-TB) in the city, with the incidence estimated at 1.9 / 100,000-year. OBJECTIVE: To describe and analyze the sensitivity to drugs of first and second line treatment of patients with pulmonary TB (PTB) to estimate the incidence of MR-TB and extensively drugresistant TB (TBXR), describe molecular and institutional aspects, spatial distribution, epidemiological PTB resistant cases in the city of Santos (SP). METHODS: A descriptive study of a cohort of patients with PTB residing in the city who started treatment or retreatment in the period January 2011 to December 31, 2012. The case definition PTB individuals 15 years or more, both sexes, living in the city of Santos (SP), who present clinical manifestations compatible with PTB and whose confirmation was made by culture with isolation of M. tuberculosis. The variables of interest for the study were: bacteriological / laboratory socio-demographic characteristics, current and previous history of TB, aspects related to treatment, and comorbidities. For comparative analyzes of proportions the chi-squared tests and Fisher's exact were used for continuous variables and the Student t test or the Kruskal - Wallis. The genetic profiles of isolates resistant to at least one drug were obtained by RFLP (length polymorphism restriction fragment) and analyzed using version BioNumerics 5.0 (Applied Maths - Belgium) software. The description of the spatial distribution of resistant TB and the clusters was made by inserting the cases in Santos map, by address of residence, which was according to the index of social vulnerability. BACKGROUND: Dombrock blood group system genotyping has revealed various rearrangements of the Dombrock gene and identified new variant alleles in Brazil (i.e., DO*A-SH, DO*A-WL and DO*B-WL). Because of the high heterogeneity of the Brazilian population, interregional differences are expected during the investigation of Dombrock genotypes. OBJECTIVE: The present study aims to determine the frequencies of Dombrock genotypes in blood donors from Minas Gerais and compare the frequencies of the HY and JO alleles to those of another population in Brazil. METHODS: The frequencies of the DO alleles in Minas Gerais, a southeastern state of Brazil, were determined from the genotyping of 270 blood donors. Genotyping involved polymerase chain reaction and restriction fragment length polymorphism analysis to identify the 323G>T, 350C>T, 793A>G, and 898C>G mutations, which are related to the HY, JO, DO*A/DO*B, and DO*A-WL/DO*B-WL alleles, respectively. Moreover, the frequencies of rare HY and JO alleles were statistically compared using the chi-square test with data from another Brazilian region. RESULTS: The HY allele frequency in Minas Gerais (2.4%) was almost twice that of the JO allele (1.5%). The frequency of the HY allele was significantly higher (p-value = 0.001) than that in another Brazilian population and includes a rare homozygous donor with the Hy- phenotype. In addition, the DO*A-WL and DO*B-WL alleles, which were first identified in Brazil, were found in the state of Minas Gerais. CONCLUSIONS: The data confirm that the frequencies of DO alleles differ between regions in Brazil. The population of Minas Gerais could be targeted in a screening strategy to identify the Hy- phenotype in order to develop a rare blood bank.

CONTEXT AND OBJECTIVE: Atherosclerotic disease is the leading cause of death in Brazil. It is a complex disease and its prevention involves identification and control of risk factors. Moderately increased plasma homocysteine concentration (hyperhomocysteinemia) has been considered to be a risk factor for several vascular diseases. Mutations in the methylenetetrahydrofolate reductase (MTHFR) enzyme, which is involved in homocysteine metabolism, have been investigated as potential vascular disease risk factors. G1793A polymorphism was described in 2002 and there are few studies analyzing its involvement in diseases. The objective of this study was to investigate the prevalence of G1793A polymorphism in subjects with early coronary artery disease (CAD). DESIGN AND SETTING: Cross-sectional study with control group conducted at a private cardiology clinic and a molecular biology laboratory (Universidade do Vale do Itajaí). METHODS: We studied 74 early-onset CAD+ patients and 40 CAD- individuals with normal angiography results. DNA was extracted from blood samples. Molecular data were obtained via PCR/RFLP and agarose gel electrophoresis. RESULTS: The occurrence of G1793A heterozygotes was similar in the control (5%) and test (6.25%) groups, thus showing that in the population studied there was no correlation between the marker and occurrences of early CAD. There was also no association between the polymorphism and the risk factors for atherosclerosis. CONCLUSIONS: The frequency of the 1793A allele in the test group (3.4%) was similar to what was found in the control individuals (2.5%). There was no correlation between G1793A polymorphism and occurrences of early CAD in this population. .
CONTEXTO E OBJETIVO: A doença aterosclerótica é a principal causa de morte no Brasil. Trata-se de doença multifatorial e sua prevenção passa pela identificação e controle dos fatores de risco. A concentração plasmática moderadamente aumentada de homocisteína (hiperhomocisteinemia) tem sido considerada importante fator de risco para várias doenças vasculares. Mutações na enzima metilenotetrahidrofolato redutase (MTHFR), envolvida no metabolismo de homocisteína, têm sido investigadas como fatores de risco para doenças vasculares. O polimorfismo G1793A foi descrito em 2002 e existem poucos estudos sobre sua implicação em doenças. O objetivo do presente trabalho foi verificar a prevalência do polimorfismo MTHFR G1793A em indivíduos portadores de doença arteriocoronariana (DAC) precoce. TIPO DE ESTUDO E LOCAL: Estudo transversal com grupo controle realizado em Clínica Cardiológica Particular e Laboratório de Biologia Molecular (Universidade do Vale do Itajaí). MÉTODOS: Foram estudados 74 pacientes DAC+ precoce e 40 DAC- com resultado angiográfico normal. O DNA foi extraído de amostras de sangue. Dados moleculares foram obtidos através de PCR/RFLP e eletroforese em gel de agarose. RESULTADOS: A ocorrência de heterozigotos G1793A foi similar em ambos os grupos controle (5%) e teste (6,25%), mostrando que na população estudada não existiu correlação entre o marcador e a ocorrência de DAC precoce. Não houve associação entre o polimorfismo e os fatores de risco para aterosclerose. CONCLUSÕES: A frequência do alelo 1793A no grupo teste (3,4%) foi parecida com a encontrada nos indivíduos do controle (2,5%). Background: Dombrock blood group system genotyping has revealed various rearrangements of the Dombrock gene and identified new variant alleles in Brazil (i.e., DO*A-SH, DO*A-WL and DO*B-WL). Because of the high heterogeneity of the Brazilian population, interregional differences are expected during the investigation of Dombrock genotypes. Objective: The present study aims to determine the frequencies of Dombrock genotypes in blood donors from Minas Gerais and compare the frequencies of the HY and JO alleles to those of another population in Brazil. Methods: The frequencies of the DO alleles in Minas Gerais, a southeastern state of Brazil, were determined from the genotyping of 270 blood donors. Genotyping involved polymerase chain reaction and restriction fragment length polymorphism analysis to identify the 323G>T, 350C>T, 793A>G, and 898C>G mutations, which are related to the HY, JO, DO*A/DO*B, and DO*A-WL/DO*B-WL alleles, respectively. Moreover, the frequencies of rare HY and JO alleles were statistically compared using the chi-square test with data from another Brazilian region. Results: The HY allele frequency in Minas Gerais (2.4%) was almost twice that of the JO allele (1.5%). The frequency of the HY allele was significantly higher (p-value = 0.001) than that in another Brazilian population and includes a rare homozygous donor with the Hy- phenotype. In addition, the DO*A-WL and DO*B-WL alleles, which were first identified in Brazil, were found in the state of Minas Gerais. Conclusions: The data confirm that the frequencies of DO alleles differ between regions in Brazil. The population of Minas Gerais could be targeted in a screening strategy to identify the Hy- phenotype in order to develop a rare blood bank. .

OBJETIVO: avaliar a frequência do polimorfismo Fok1 do gene do receptor da vitamina D (VDR) em mulheres inférteis com endometriose e Grupo Controle, e sua associação com a doença. MÉTODOS: estudo caso-controle que incluiu 147 mulheres inférteis com endometriose e 154 mulheres férteis sem endometriose como Controle. O polimorfismo Fok1 (rs10735810, T2C), que promove uma troca de T/C no éxon 2 do gene VDR, foi identificado por PCR-RFLP (análise de polimorfismos de fragmentos de restrição), que envolve a combinação de amplificação por PCR (reação em cadeia da polimerase) e digestão com endonuclease de restrição. O teste do χ2 foi utilizado para comparar as frequências dos genótipos e alelos entre os grupos. Todos os valores de p foram bicaudais, e o nível de significância considerado foi 0,05 (α<0,05). RESULTADOS: os genótipos TT, TC e CC do polimorfismo FokI do gene VDR apresentaram frequência de 44,2 por cento, 46,9 por cento e 8,9 por cento nas mulheres inférteis com endometriose e 41,6 por cento, 50 por cento e 8,4 por cento no Grupo Controle, e não apresentaram diferença significante quando comparados (p=0,8), mesmo quando as pacientes com endometriose foram subdivididas de acordo com o estadiamento da endometriose (p=0,3 para endometriose mínima e leve e p=0,2 para endometriose moderada e grave). Em relação aos alelos, T e C estavam presentes, respectivamente, em 67,6 por cento e 32,3 por cento das mulheres inférteis com endometriose (p=0,8), em 63,5 por cento e 36,5 por cento das mulheres com endometriose mínima/leve (p=0,5), em 72,5 por cento e 27,5 por cento das mulheres com endometriose moderada/grave (p=0,2), e em 66,6 por cento e 33,4 por cento das mulheres do Grupo Controle. PURPOSE: to evaluate the frequency of VDR gene polymorphism Fok1 in infertile women with endometriosis and Control and its relation to the disease. METHODS: a case-control study that included 147 infertile women with endometriosis and 154 fertile women without endometriosis as Control. Fok1 polymorphism (rs10735810, T2C), which promotes a T/C exchange in exon 2 of the VDR gene, was identified by the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), that involves the combination of amplification by PCR and digestion with restriction endonuclease. The χ2 test was used to compare allele and genotype frequencies between groups. All p-values were two-tailed and a p-value < 0.05 was considered statistically significant. RESULTS: the TT, TC and CC genotype frequencies of VDR Fok1 polymorphism were 44.2 percent, 46.9 percent and 8.9 percent in infertile women with endometriosis and 41.6 percent, 50 percent and 8.4 percent in the Control Group. No significant difference was found (p=0.8), even when the patients were subdivided according to the stage of endometriosis (p=0.3 for minimal and mild endometriosis and p=0.2 for moderate and severe endometriosis). Alleles T and C were present, respectively, in 67.6 percent and 32.3 percent of infertile women with endometriosis (p=0.8), in 63.5 percent and 36.5 percent of women with minimal/mild endometriosis (p=0.5), in 72.5 percent and 27.5 percent of women with moderate/severe endometriosis (p=0.2), and in 66.6 percent and 33.4 percent of the Control Group. OBJECTIVE: Adenosine may play a role in the pathophysiology of schizophrenia, since it modulates the release of several neurotransmitters such as glutamate, dopamine, serotonin and acetylcholine, decreases neuronal activity by pos-synaptic hyperpolarization and inhibits dopaminergic activity. Adenosine deaminase participates in purine metabolism by converting adenosine into inosine. The most frequent functional polymorphism of adenosine deaminase (22G→A) (ADA1*2) exhibits 20-30 percent lower enzymatic activity in individuals with the G/A genotype than individuals with the G/G genotype. The aim of this study was to evaluate the ADA polymorphism 22G→A (ADA1*2) in schizophrenic patients and healthy controls. METHOD: The genotypes of the ADA 22G→A were identified with allele-specific PCR strategy in 152 schizophrenic patients and 111 healthy individuals. RESULTS: A significant decrease in the frequency of the G/A genotype was seen in schizophrenic patients (7/152 - 4.6 percent) relative to controls (13/111 - 11.7 percent, p = 0.032, OR = 2.6). CONCLUSION: These results suggest that the G/A genotype associated with low adenosine deaminase activity and, supposingly, with higher adenosine levels is less frequent among schizophrenic patients.
OBJETIVO: A adenosina pode ter um papel importante na fisiopatologia da esquizofrenia, uma vez que modula a liberação de vários neurotransmissores, tais como glutamato, dopamina, serotonina e acetilcolina, diminui a atividade neuronal por hiperpolarização pós-sináptica e inibe a atividade dopaminérgica. A adenosina desaminase participa do metabolismo das purinas pela conversão de adenosina em inosina. O mais frequente polimorfismo funcional da adenosina desaminase (22G →A) (ADA1*2) exibe uma diminuição de 20-30 por cento da atividade funcional em indivíduos com genótipo G/A quando comparados com indivíduos com o genótipo G/G. O objetivo deste estudo foi avaliar o polimorfismo 22G→A (ADA1*2) em pacientes esquizofrênicos e em controles saudáveis. MÉTODO: Os genótipos da ADA 22G →A foram identificados através de uma estratégia de PCR alelo-específica em 152 pacientes esquizofrênicos e 111 controles saudáveis. RESULTADOS: Foi observada uma diminuição significativa na frequência do genótipo G/A em pacientes esquizofrênicos (7 - 4,6 por cento) em relação ao grupo controle (13 - 11,7 por cento, p = 0,032, OR = 2,6). CONCLUSÃO: Estes resultados sugerem que o genótipo G/A associado com baixa atividade de adenosina desaminase, e potencialmente com níveis aumentados de adenosina, é menos frequente entre pacientes esquizofrênicos.

Mycobacterium tuberculosis é um dos mais bem sucedidos patógenos do homem. As cepas virulentas são mais facilmente transmitidas, induzindo respostas imunes variáveis. Avaliamos a resposta celular tipo Th1, através da produção de IFN-γ, como resposta a cepas com padrões diversos em voluntários sadios. Nossos resultados mostraram que indivíduos com teste tuberculínico (TT) negativo já tiveram contato com algumas das cepas testadas, ao passo que indivíduos com TT positivo não responderam a todas as cepas testadas. Cepas resistentes induziram uma média menor de produção de IFN-γ que aquelas sensíveis. Uma possível aplicação prática disto seria que a produção de IFN-γ, em relação a uma cepa isolada específica, poderia auxiliar na previsão da resposta ao tratamento dos pacientes.
Mycobacterium tuberculosis is one of the most successful human pathogens. Highly virulent strains, which are more easily transmitted than are less virulent strains, elicit variable immune responses. We evaluated the Th1 responses (IFN-γ production) in healthy volunteers after stimulation with various strains. Our results show that the individuals with negative tuberculin skin test (TST) results were not necessarily naive to all of the strains tested, whereas individuals with positive TST results did not respond to all of the strains tested. Drug-resistant strains induced a lower mean level of IFN-γ production than did drug-sensitive strains. One possible practical application of this finding would be for the prediction of responses to treatment, in which it might be advantageous to have knowledge of the estimated IFN-γ production elicited by a specific isolated strain.

Introducción. Varios estudios sugieren que algunos polimorfismos del gen de la interleucina-1beta humana (IL-1beta), como -511, -31 y +3954, están asociados al cáncer gástrico, debido al efecto inhibidor que esta citocina tiene sobre la secreción ácida del estómago, lo cual facilita la colonización e infección por agentes como Helicobacter pylori, así como la génesis de estados preneoplásicos que pueden conducir al desarrollo de cáncer. Objetivo. Genotipificar los polimorfismos +3954,-511 y -31 de la IL-1beta y establecer sus frecuencias en una población de pacientes con diferente sintomatología gástrica. Materiales y métodos. Se analizaron 111 biopsias del antro gástrico obtenidas de pacientes con sintomatología de alguna alteración gástrica. La detección de H. pylori en las muestras se realizó mediante PCR empleando iniciadores específicos para cada región y la genotipificación de las regiones polimórficas de la IL-1beta se realizó por RFLP empleando las enzimas Aval, Alul y Taql para -511, -31 y +3954, respectivamente. Resultados. Se detectó H. pylori en 59,5% de las biopsias gástricas, mientras que el estudio histopatológico reveló que 82,9% de los pacientes padecía alguna enfermedad. La caracterización de las regiones polimórficas del gen de la IL-1beta, seguida de la tipificación por RFLP, permitió evidenciar los tres posibles genotipos de cada uno de los polimorfismos en la población. En los pacientes infectados por H. pylori se encontró con mayor frecuencia (28,6%) el genotipo CC en la región polimórfica -31. Conclusión. No se encontraron diferencias significativas en los genotipos de los individuos infectados y los no infectados por H. pylori, a excepción del genotipo CC en la región polimórfica -31, el cual se encontró con mayor frecuencia en los pacientes con enfermedades benignas.
Introduction. The human interleukin-1beta gen (IL-1 beta) polymorphisms such as -511, -31 and +3954 have been associated with the presence of gastric cancer, due to the inhibitor effect that this protein has on acid secretion in the stomach. Thisfacility can enhance the colonization and infection by agents like Helicobactor. pylori and the genesis of preneoplastic states that can lead to cancer development. Objective. Three polymorphisms of IL-1beta (+3954, -511 and -31) will be genetically characterized and their frequencies established in a population of patients with gastric symptoms. Materials and methods. Gastric antrum biopsies were obtained from 111 patients that showed signs of gastric disorder. A PCR was done to detect the H. pylori presence; a PCR using designed primers for specific regions was done to define the three polymorphic regions of IL-1beta, and a RFLP was carried out using Aval, Alul and TaqI for the position -511, -231 and +3954 for each case. Results. Helicobacter pylori was detected in 59.5% of the evaluated gastric while the histopathology study revealed that 82.9% of patients had some pathology. Characterization of polymorphic regions of IL-1beta gen were joined to RFLP typing evidenced that all descfribed genotypes were present in the study population. However, patients with benign pathologies infected with H. pylori had a high frequency of the CC genotype (28.6%) in the -31 polymorphic regions.Conclusion. No significant differences were found between the genotype frequenciess of the H. pylori-infected and the non-infected populations with one exception. The CC genotype in the -31 polymorphic region was associated with benign pathologies.

Context and objective: Genetic investigation of central nervous system (CNS) tumors provides valuable information about the genes regulating proliferation, differentiation, angiogenesis, migration and apoptosis in the CNS. The aim of our study was to determine the prevalence of genetic polymorphisms (codon 31 and 3' untranslated region, 3'UTR) and protein expression of the cyclin-dependent kinase inhibitor 1A (CDKN1A) gene in patients with and without CNS tumors. Design and setting: Analytical cross-sectional study with a control group, at the Molecular Biology Laboratory, Pediatric Oncology Department, Hospital das Clínicas de Ribeirão Preto. Methods: 41 patients with CNS tumors and a control group of 161 subjects without cancer and paires for sex, age and ethnicity were genotyped using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). Protein analysis was performed on 36 patients with CNS tumors, using the Western Blotting technique. Results: The frequencies of the heterozygote (Ser/Arg) and polymorphic homozygote (Arg/Arg) genotypes of codon 31 in the control subjects were 28.0 percent and 1.2 percent, respectively. However, the 3'UTR site presented frequencies of 24.2 percent (C/T) and 0.6 percent (T/T). These frequencies were not statistically different (P > 0.05) from those seen in the patients with CNS tumors (19.4 percent and 0.0 percent, codon 31; 15.8 percent and 2.6 percent, 3'UTR site). Regarding the protein expression in ependymomas, 66.67 percent did not express the protein CDKN1A. The results for medulloblastomas and astrocytomas were similar: neither of them expressed the protein (57.14 percent and 61.54 percent, respectively). Conclusion: No significant differences in protein expression patterns or polymorphisms of CDKN1A in relation to the three types of CNS tumors were observed among Brazilian subjects.
Contexto e objetivo: A investigação genética dos tumores do sistema nervoso central (SNC) provê valiosa informação sobre os genes que regulam a proliferação, diferenciação, angiogênese, migração e apoptose. O objetivo deste estudo é determinar a prevalência entre os polimorfismos genéticos (códon 31 e da região 3' não traduzida, 3'UTR) e a expressão protéica do gene inibidor de quinase dependente de ciclina 1A (CDKN1A) em pacientes com e sem tumor do SNC. Tipo de estudo e local: Estudo transversal analítico com grupo controle, desenvolvido no Laboratório de Biologia Molecular do Departamento de Oncologia Pediátrica do Hospital das Clínicas de Ribeirão Preto. Métodos: 41 pacientes com tumor do SNC e um grupo controle de 161 indivíduos sem câncer pareados por idade, sexo e etnia foram genotipados mediante uma reação de polimorfismo no comprimento de fragmentos de restrição (RFLP). A análise das proteínas foi realizada em 36 pacientes com tumor de SNC mediante Western Blotting. Resultados: A frequência do genótipo heterozigoto (Ser/Arg) e do homozigoto polimórfico (Arg/Arg) do códon 31 nos controles foi 28,0 por cento e 1,2 por cento, respectivamente. Entretanto, o sítio 3'UTR apresentou uma frequência de 24,2 por cento (C/T) e 0,6 por cento (T/T). Estas frequências não são significativamente diferentes (P > 0,05) daquelas observadas no grupo dos pacientes com tumor de SNC (19,4 por cento e 0,0 por cento, códon 31; 15,8 por cento e 2,6 por cento, sítio 3'UTR). Com respeito à expressão protéica, nos ependimomas, 66,67 por cento não expressaram a proteína CDKN1A. This retrospective molecular study involving restriction fragment length polymorphism, using insertion sequence 6110 as a marker, was conducted in order to provide an initial insight into the genetic diversity of Mycobacterium tuberculosis strains isolated in the slums of the Complexo de Manguinhos, located in the city of Rio de Janeiro, Brazil. Of the 67 strains evaluated, 23 (34.3 percent) were found to belong to clusters (total clusters, 10). Household and social chains of transmission were associated with clustering, in 20 percent and 60 percent, respectively. Living in the Conjunto Habitacional Programado 2 slum was associated with clustering. Although not significant, it is relevant that 26 percent of the clustered strains presented primary resistance. These findings, although possibly underestimating the prevalence due to the failure to analyze all strains, could help improve the local tuberculosis control program.
Este estudo retrospectivo envolvendo polimorfismo de fragmento de restrição e utilizando como marcador a seqüência de inserção 6110, foi realizado para fornecer informações iniciais quanto à diversidade genética das cepas de Mycobacterium tuberculosis isoladas em favelas do Complexo de Manguinhos, na cidade do Rio de Janeiro. Das 67 cepas isoladas, 23 (34,3 por cento) foram agrupadas em clusters (total de clusters, 10). A transmissão entre comunicantes domiciliares e extra-domicialiares estave associada a 20 por cento e 60 por cento dos clusters, respectivamente. Ser morador do Conjunto Habitacional Programado 2 foi associado à presença de clusters. Embora não significativo, é relevante o fato de que 26 por cento das cepas em cluster apresentaram resistência primária. Estes achados, embora possivelmente subestimados devido à impossibilidade de analisar todas as cepas, fornecem subsídios para a melhoria do programa local de controle da tuberculose.
