RESUMEN

Applying biotechnological tools to the selection of higher-yielding bioeconomic crops is a promising and remarkable means of reducing the burden on production on a global scale. In the present study, 25 germplasms of Indian ginseng (Withania somnifera (L.) Dunal) were examined for their genetic diversity by using morphological, biochemical, and molecular markers for twenty plant growth traits. The properties of plant growth differed significantly in the maximum genotypes of Indian ginseng, the markers of randomly amplified polymorphic DNA (RAPD), and inter simple sequence repeat (ISSR) showed considerable diversity between the genotypes. The combined unweighted pair group technique with arithmetic mean (UPGMA) dendrogram of morphological, biochemical, and molecular markers grouped all 25 genotypes into two main clusters at 0.61 coefficient value. In addition to this, secondary metabolite profiling by high-performance liquid chromatography (HPLC), there were high variations for withanolide B (WL-B), withanoside-V (WS-V), wedelolactone (WDL), withanoside-IV (WS-IV), and withaferin A (WF-A) content between different genotypes. For the total alkaloid and withanolide concentration in the roots and leaves, high heritability with an increased genetic gain was observed, indicating that selection based on these traits could be an effective method in breeding programs. Furthermore, the path coefficient analysis showed a direct positive impact of the total root fiber, WL-B (leaves), WF-A (leaves), WS-IV (roots), WDL (roots), and the total alkaloid content on the dry root yield. High content of WDL, a high-quality bioactive withanolide, was also described for the first time in the genotype UWS23. These properties can further be exploited to improve the dry root yield in W. somnifera genotypes. The outcomes of the present study also provide an essential foundation for the selection of high-yielding bioeconomic varieties that could be utilized to improve Ashwagandha breeding programs.

RESUMEN

Adipocyte differentiation-associated long noncoding RNA (ADNCR) is a newly discovered lncRNA. It plays function by targeting miR-204 to significantly regulates the expression of the target SIRT1 gene in preadipocytes both at the level of mRNA and protein, thereby inhibiting adipogenesis. The tetra-primer amplification refractory mutation system PCR (T-ARMS-PCR) strategy is fast and accuracy at a negligible cost for SNP genotyping in large samples. In the study, a novel SNP g.1263T>A in intron 1 of bovine ADNCR gene was found. Herein, the T-ARMS-PCR assay was applied to detect the genotypes of the novel SNP of bovine ADNCR gene in 1017 individuals from seven cattle breeds and validated the accuracy by DNA sequencing assay of ninety animals representing three different genotypes. The concordance between two different methods was 100%. The association analysis indicated that this locus was significantly associated with the body weight (P = 0.010), chest girth (P = 0.014) and rump length (P = 0.038) in Jinnan cattle, hucklebone width (P = 0.032) in Qinchuan cattle, the cannon circumference (P = 0.019) in Jinjiang cattle, respectively. These novel findings may be used for marker-assisted selection (MAS) and contribute to the performance of beef cattle in the future.

Lysine-specific demethylase 1B (KDM1B) which plays a crucial role in regulating methylation status at lysine 4 of histone 3 is important for male fertility. The aim of this study was to explore the KDM1B mRNA expression profiles and to identify novel genetic variants of the pig KDM1B gene, as well as to determine the association between these variants and testis measurement traits in male piglets. The KDM1B mRNA expression profiles indicated that this gene widely expressed in all tested organs. In addition, a novel 17-bp deletion (NC_010449.4:g.31142_31159delCATGGATAGTAGTTGCT) within KDM1B gene was found. Notably, this deletion sequence was inconsistent with the prediction by NCBI. Association analysis revealed that the 17-bp indel locus was significantly associated with the testis weight in 40-day-old Large White pigs (P < 0.05). Furthermore, through bioinformatics analysis, transcriptional factor heat shock factor-1 could combine the 17-bp sequence. These results not only extend the genetic variations of the pig KDM1B gene but also contribute to implementing marker-assisted selection in pig breeding.

Pearl millet is a climate resilient crop and one of the most widely grown millets worldwide. Heterotic hybrid development is one of the principal breeding objectives in pearl millet. In a maiden attempt to identify heterotic groups for grain yield, a total of 343 hybrid parental [maintainer (B-) and restorer (R-)] lines were genotyped with 88 polymorphic SSR markers. The SSRs generated a total of 532 alleles with a mean value of 6.05 alleles per locus, mean gene diversity of 0.55, and an average PIC of 0.50. Out of 532 alleles, 443 (83.27%) alleles were contributed by B-lines with a mean of 5.03 alleles per locus. R-lines contributed 476 alleles (89.47%) with a mean of 5.41, while 441 (82.89%) alleles were shared commonly between B- and R-lines. The gene diversity was higher among R-lines (0.55) compared to B-lines (0.49). The unweighted neighbor-joining tree based on simple matching dissimilarity matrix obtained from SSR data clearly differentiated B- lines into 10 sub-clusters (B1 through B10), and R- lines into 11 sub-clusters (R1 through R11). A total of 99 hybrids (generated by crossing representative 9 B- and 11 R- lines) along with checks were evaluated in the hybrid trial. The 20 parents were evaluated in the line trial. Both the trials were evaluated in three environments. Based on per se performance, high sca effects and standard heterosis, F1s generated from crosses between representatives of groups B10R5, B3R5, B3R6, B4UD, B5R11, B2R4, and B9R9 had high specific combining ability for grain yield compared to rest of the crosses. These groups may represent putative heterotic gene pools in pearl millet.
