RESUMEN

Three extremophile bacterial strains (BBCOL-009, BBCOL-014 and BBCOL-015), capable of degrading high concentrations of perchlorate at a range of pH (6.5 to 10.0), were isolated from Colombian Caribbean Coast sediments. Morphological features included Gram negative strain bacilli with sizes averaged of 1.75 × 0.95, 2.32 × 0.65 and 3.08 × 0.70 µm, respectively. The reported strains tolerate a wide range of pH (6.5 to 10.0); concentrations of NaCl (3.5 to 7.5% w/v) and KClO4- (250 to 10000 mg/L), reduction of KClO4- from 10 to 25%. LB broth with NaCl (3.5-30% w/v) and KClO4- (250-10000 mg/L) were used in independent trials to evaluate susceptibility to salinity and perchlorate, respectively. Isolates increased their biomass at 7.5 % (w/v) NaCl with optimal development at 3.5 % NaCl. Subsequently, ClO4- reduction was assessed using LB medium with 3.5% NaCl and 10000 mg/L ClO4-. BBCOL-009, BBCOL-014 and BBCOL-015 achieved 10%, 17%, and 25% reduction of ClO4-, respectively. The 16 S rRNA gene sequence grouped them as Bacillus flexus T6186-2, Bacillus marisflavi TF-11 (T), and Bacillus vietnamensis 15 - 1 (T) respectively, with < 97.5% homology. In addition, antimicrobial resistance to ertapenem, vancomycine, amoxicillin clavulanate, penicillin, and erythromycin was present in all the isolates, indicating their high adaptability to stressful environments. The isolated strains from marine sediments in Cartagena Bay, Colombia are suitable candidates to reduce perchlorate contamination in different environments. Although the primary focus of the study of perchlorate-reducing and resistant bacteria is in the ecological and agricultural realms, from an astrobiological perspective, perchlorate-resistant bacteria serve as models for astrobiological investigations.

Among the main sources of atmospheric mercury emissions are thermoelectric plants that use coal to generate electricity. This heavy metal is transported by air from its origin to neighboring coastal areas, where it is deposited in the sediment record. This research evaluated the content and temporal distribution of Hg in the coastal sediments of Central Chile, Coronel Bay, industrialized zone with two operative thermoelectric plants, and Coliumo Bay, reference zone free of industry. To this end, sediment cores from the centers of the two bays were obtained. Laboratory results show that the greatest Hg contents were found in the surface strata of the Coronel Bay core, with concentrations up to an order of magnitude greater than those obtained in the same strata in Coliumo Bay. This increase in Hg concentration coincides with the beginning of industrial activity in Coronel, with greater concentrations observed from 2012 on, the year in which the industrial operation of two coal-fired thermoelectric plants on the bay began. Based on 210Pb activity and the relationships between Hg content and organic matter, it was established that the main pathway of Hg to sediment is atmospheric deposition, while Coliumo presented inputs that reflect local non anthropogenic sources.

Over half of the organic carbon on Earth's surface is trapped in marine sediment as methane hydrates. Ocean warming causes hydrate dissociation and methane leakage to the water column, rendering the characterization of microbes from hydrate depositions a pressing matter. Through genomic, phylogenetic, and biochemical assays, we characterize the first microorganisms isolated from the Rio Grande Cone (Brazil), reservoir responsible for massive methane releases to the water column. From sediment harboring rich benthic communities, we obtained 43 strains of Brevibacillus sp., Paenibacillus sp. and groups of Bacillus sp. Methane-enriched samples yielded strains of the Pseudomonas fluorescens complex, exhibiting fluorescent siderophore production and broad multi-carbon catabolism. Genomic characterization of a novel Pseudomonas sp. strain indicated 32 genes not identified in the closest related type-species, including proteins involved with mercury resistance. Our results provide phylogenetic and genomic insights on the first bacterial isolates retrieved from a poorly explored region of the South Atlantic Ocean.

Antibiotics are extensively used for growth promotion purposes in intensive aquaculture. In Chile, the use of antibiotics in salmon farming is excessive, approximately 62 times more than is used in Norway. In the salmon industry, antibiotics such as oxytetracycline (OTC) are administered in the diet, both in the juvenile stage in freshwater and in the fattening process of salmon in marine sectors. We have investigated the fjords of Chile, where many salmon farms are located, searching for fungi able to degrade this tetracycline antibiotic. We have evaluated the OTC degradation ability of the following; Penicillium commune, Epicoccum nigrum, Trichoderma harzianum, Aspergillus terreus and Beauveria bassiana, isolated from sediments in salmon farms from southern Chile. In all these fungal strains, the amount of OTC decreased in the culture medium, as adsorbed in the mycelia, after the third day of exposure. These strains were capable of degrading OTC at remarkable rates up to 78%, by the 15th day. This is the first study showing that the mycelium of these fungal strains has the ability to degrade OTC. We believe the knowledge produced by these results has the potential to serve as a basis for implementing a bioremediation process in the near future.

This work aims to propose new standards to assess the degree of sediment contamination in saline and brackish environments, since the legislation currently used in Brazil is based on Canadian/American regulations, which do not comply with the conditions in Brazil. This study is based on geochemical analyses of 340 surface sediment samples collected in the Green Coast region (Rio de Janeiro, Brazil), including Mangaratiba, Angra dos Reis and Ribeira coves and Ilha Grande and Sepetiba bays. This region is influenced by industrial, harbor, urban and tourist activities and was affected by a dam rupture episode that released contaminated material. The results show heterogeneity in the distribution and range of metal concentrations in the study area depending on the supply of metals from natural and anthropogenic sources. Environmental characteristics such as coastal and tidal currents, water temperature and salinity, local depth, sediment grain size, sedimentary dynamics and biogeochemical processes influence the dispersion or retention of metals. The pollution load index (PLI) suggests that Sepetiba Bay is the region with the most environmental degradation due to anthropogenic contamination. In this context, we propose the establishment of new levels of contamination according to the Cd, Cr, Cu, Mn, Ni, Pb, V and Zn concentrations in sediments of salt and brackish waters, considering I) background level; II) level 1 - with anthropogenic influence; and III) level 2 - contaminated. The results of this work also suggest that, except for zinc, the range of metal concentrations admitted by Brazilian legislation are quite permissive and not adequate for Brazilian coastal environments.

Surficial marine sediments were sampled along the south-east coast and in Discover Bay Jamaica. Total elemental composition was determined for the sediments using three techniques. Total mass fractions of Al, As, Br, Ce, Co, Cr, Cs, Eu, Fe, La, Mn, Sb, Sc, Th, U, V, and Zn were determined using instrumental neutron activation analysis; Cu, Ni, Pb, Sr and Zr using energy dispersive X-ray fluorescence; and Hg using a direct mercury analyser. Potential anthropogenic hotspots were assessed using indices including enrichment factor (EF), geoaccumulation index (Igeo), ecological risk (Er) and potential ecological risk index (Ri). The quality of the sediments was also assessed using the sediment quality guidelines (SQGs). Temperature, pH, salinity, dissolved oxygen and organic carbon content (OC) were determined to assess variation across sample sites. Results indicated moderate to severe ecological risk at sites in Port Royal and Discovery Bay with ecological risk values as high as 381.9.

A total of 114 moderately halophilic bacteria were isolated from marine sediment environments. The isolates are belonged to 23 species based on the 16S rRNA sequence analysis. 63, 52, 47, 57, 74, 15 and 4 isolates are able to produce protease, amylase, lipase, pectinase, pulluanase, xylanase, cellulase, respectively. Combined hydrolytic enzyme activity analysis show that 15 strains present 1 hydrolytic activity, 32 strains present 2 hydrolytic activities, 21 strains present 3 hydrolytic activities, 26 strains present 4 hydrolytic activities, 11 strains present 5 hydrolytic activities and 2 strains present 6 hydrolytic activities. Hydrolase activities are widely distributed in a variety of species. The highest rates for production of protease, amylase, lipase, pectinase, pullanase, xylanase and cellulase were observed in species of B. baekryungensis, Hallobacillus sp., B. pumilus, B. megaterium or P. chungwhensis, B. amyloliquefaciens, B. pumilus, B. baekryungensis, respectively. However, the higher activities of protease, pectinase and pulluanase are frequently produced by the species of Halomonas sp. B. amyloliquefaciens or P. chungwhensis, and Vibrio sp. respectively. This investigation show that the diversity of halophilic bacteria from marine sediments could serve as a potential source of hydrolytic enzymes for industrial applications. (AU)


Um total de 114 bactérias moderadamente halofílicas foram isoladas de ambientes de sedimentos marinhos. Os isolados pertencem a 23 espécies com base na análise da sequência 16S rRNA. 63, 52, 47, 57, 74, 15 e 4 isolados são capazes de produzir protease, amilase, lipase, pectinase, pululanase, xilanase, celulase, respectivamente. A análise da atividade enzimática hidrolítica combinada mostra que 15 cepas apresentam 1 atividade hidrolítica, 32 cepas apresentam 2 atividades hidrolíticas, 21 cepas apresentam 3 atividades hidrolíticas, 26 cepas apresentam 4 atividades hidrolíticas, 11 cepas apresentam 5 atividades hidrolíticas e 2 cepas apresentam 6 atividades hidrolíticas. Atividades de hidrolase são amplamente distribuídas em uma variedade de espécies. As maiores taxas de produção de protease, amilase, lipase, pectinase, pululanase, xilanase e celulase foram observadas em espécies de B. baekryungensis, Hallobacillus sp., B. pumilus, B. megaterium ou P. chungwhensis, B. amyloliquefaciens, B. pumilus, B. baekryungensis, respectivamente. No entanto, as atividades mais elevadas de protease, pectinase e pululanase são freqüentemente produzidas pelas espécies de Halomonas sp. B. amyloliquefaciens ou P. chungwhensis e Vibrio sp. respectivamente. Esta investigação mostra que a diversidade de bactérias halofílicas de sedimentos marinhos pode servir como uma fonte potencial de enzimas hidrolíticas para aplicações industriais. (AU)

Rhodopsins are broadly distributed. In this work, we analyzed 23 metagenomes corresponding to marine sediment samples from four regions that share cold climate conditions (Norway; Sweden; Argentina and Antarctica). In order to investigate the genes evolution of viral rhodopsins, an initial set of 6224 bacterial rhodopsin sequences according to COG5524 were retrieved from the 23 metagenomes. After selection by the presence of transmembrane domains and alignment, 123 viral (51) and non-viral (72) sequences (>50 amino acids) were finally included in further analysis. Viral rhodopsin genes were homologs of Phaeocystis globosa virus and Organic lake Phycodnavirus. Non-viral microbial rhodopsin genes were ascribed to Bacteroidetes, Planctomycetes, Firmicutes, Actinobacteria, Cyanobacteria, Proteobacteria, Deinococcus-Thermus and Cryptophyta and Fungi. A rescreening using Blastp, using as queries the viral sequences previously described, retrieved 30 sequences (>100 amino acids). Phylogeographic analysis revealed a geographical clustering of the sequences affiliated to the viral group. This clustering was not observed for the microbial non-viral sequences. The phylogenetic reconstruction allowed us to propose the existence of a putative ancestor of viral rhodopsin genes related to Actinobacteria and Chloroflexi. This is the first report about the existence of a phylogeographic association of the viral rhodopsin sequences from marine sediments.

The electrochemical generation of antimony volatile species (stibine) using Au and Ag mercury amalgamated cathodes is described. Compared with some other cathode materials commonly used for electrochemical hydride generation, performance of the amalgamated cathodes is substantially better in the following aspects: higher interference tolerance, higher erosion resistance and longer useful working time. Using the amalgamated cathodes, it could be shown that interferences from major constituents at high concentrations, especially from transition metals, affecting stibine generation are not as significant as they are using other cathode types in regards to sensitivity and useful working time. Results obtained using the Ag/Hg amalgamated cathode showed a slightly higher sensitivity than the corresponding results obtained using the Au/Hg cathode. The Au/Hg cathode, which to our knowledge has not previously been used to generate stibine, showed considerably longer useful working time than the Ag/Hg one. The optimum catholytes for electrolytic generation of stibine (SbH3) from Sb(III) and Sb(V) using the Au/Hg electrode were aqueous solutions containing 0.5mol L(-1) H2SO4 and 0.5mol L(-1)HCl, respectively. Under optimized conditions, using the Au/Hg cathode and comparing to aqueous standards calibration curves, detection limits (3σ) of 0.027µg L(-1) for Sb(III) and 0.056µg L(-1) for Sb(V), were obtained. To check accuracy a marine sediment reference material (PACS-2, NRC) was analyzed using a method purportedly developed for this task. Good agreement, 95% confidence, was found between the certified and the experimental values for Sb. The proposed method was also applied to the determination of Sb in aqueous solutions of marine sediments samples from Comuna de Bajo Alto Provincia de El Oro-Ecuador. Recoveries of five replicate determinations of these samples were in the range of 98-103% thus showing acceptable accuracy in the analysis of real samples.

A simple and rapid method for the detection and extraction of oxolinic acid, flumequine, florfenicol and oxytetracycline from marine sediments was developed and validated. The analytes were extracted from the marine sediment using a solution of oxalic acid diluted in methanol with sonication before detection by HPLC using a diode-array detector (florfenicol and oxytetracycline) and fluorescence (oxolinic acid and flumequine). The quantification limits (QL) were 100 ng/g for oxytetracycline and florfenicol and 5 ng/g for oxolinic acid and flumequine. The coefficients of variation of the repeatability and intermediate precision were less than 10% in all of the analytes. The calibration curves were linear between 50 and 500 ng/ml for oxytetracycline and florfenicol and 1 and 20 ng/ml for oxolinic acid and flumequine. The recuperation rate for the analytes was above 86%.

El presente trabajo tuvo por objeto determinar el período de supervivencia en sedimentos marinos de bacterias con riesgo biológico o indicadoras. Se tomaron muestras de sedimento marino intermareal en dos playas de la ciudad de Comodoro Rivadavia, Argentina; una sometida a contaminación por efluentes domiciliarios, y otra sin pruebas de contaminación evidente. Sobre éstos se realizó una caracterización inicial por recuento de bacterias en diferentes medios de cultivo (TSA, VRB, VRBG, TCBS, agar bilis esculina y agar cetrimide) y por su identificación. También se realizó un estudio de supervivencia de los microorganismos de interés en microcosmos. Estos fueron monitoreados por conteo de bacterias y por identificación de los integrantes de interés. Los principales resultados indicaron que existe una marcada diferencia en la composición de la comunidad bacteriana entre ambas muestras. Los géneros bacterianos en la muestra contaminada fueron Aeromonas, Salmonella, Pseudomonas, Escherichia, Ochrobactum y Klebsiella, y en la playa no contaminada Bacillus, Paenibacillus, Micrococcus, Photobacterium y Vibrio. El ensayo en microcosmos demostró que, luego de 65 días, la comunidad bacteriana de la playa contaminada fue similar a la del sedimento sin contaminación, indicando que al retirar la fuente de contaminación, es probable la recuperación de la zona afectada.


The present study had the purpose of determining the survival period of bacteria which represent a biological risk and are indicators of contamination. Inter-tide marine sediment samples were taken at two beaches of the Comodoro Rivadavia city in Argentine; one the beaches was submitted to contamination due to household effluents, and the other without proof of evident contamination. These samples were initially characterized by bacterial counts in different culture media (TSA, VRB, TCBS, bile esculin agar and cetrimide agar) and by their identification. A study of the survival of the target bacteria in microcosm was also done, monitored by bacterial counts and identification of these bacteria. The main results indicated that there was a marked difference in the composition of the bacterial community between the two samples. In the contaminated sample the bacterial genus were Aeromonas, Salmonella, Pseudomonas, Escherichia, Ochrobactum and Klebsiella, and in the non-contaminated beach Bacillus, Paenibacillus, Micrococcus, Photobacterium and Vibrio. The microcosm assay showed that after 65 days the bacterial community of the contaminated beach was similar to that of the non-contaminated beach sediment, indicating that by withdrawing the contamination source, the affected zone can probably be recovered.
