RESUMEN

OBJECTIVE: Magnolia biondii, a plant containing many magnolian-like compounds in its flowers or buds, exhibits anti-inflammatory and antiallergic effects; however, no study has addressed its effect on alleviating ultraviolet light (UV)-induced skin damage. We thus aimed at studying the effects of M. biondii flower extract (MB) on UVB-induced skin damage and determine the relationship between cell damage and damage-associated molecular patterns (DAMPs). METHODS: Reconstructed epidermal models and foreskin samples were selected to detect cellular reactions after UVB irradiation and MB treatment. MTT, haematoxylin-eosin and immunofluorescence staining were used to examine total viability, sunburned cells and expression and migration of DAMPs at 16 or 48 h. Prostaglandin E2 (PGE-2) and interleukin 8 (IL-8) levels were measured using enzyme-linked immunosorbent assays. A clinical UVB-damaged test was carried out on human arms subjected to MB pre- or post-treatment. Human skin probes were used to measure erythema, melanin, ITA° and transepidermal water loss (TEWL), while skin photos were captured using the VISIA system. RESULTS: MB is rich in lignans such as magnolin, pinoresinol dimethyl ether and fargesin, and shows weak UV absorption at 280-320 nm. Coculturing with MB for 16 or 48 h after UVB irradiation improved the tissue viability and structure of Skinovo-Epi, and reduced the expression and migration of high mobility group box protein B1 (HMGB1) as well as the expression of IL-8 and PGE-2. In the excised foreskin treated with MB after UVB irradiation, the generation of 8-hidroxy-2-deoxyguanosine and nuclear transfer of HMGB1 were reduced. When pre-treated with MB for 3 days, UVB-induced skin erythema and ITA° were significantly decreased. When post-treated with MB for 5 days, a decrease in skin erythema, melanin and TEWL values and an increase in skin ITA° were observed. CONCLUSIONS: Treatment with MB attenuated UVB-induced skin damage, such as erythema, pigmentation and skin barrier function, by improving the tissue viability and structure and reducing sunburned cells and skin inflammation. This effect may be related to DNA damage, which causes the migration of HMGB1 from the nucleus to the outside of the cell to induce skin inflammation.


OBJECTIF: Magnolia biondii, une plante dont les fleurs et les bourgeons contiennent de nombreux composés de type magnolien, possède des effets anti­inflammatoires et antiallergiques. Cependant, aucune étude n'a abordé son effet sur la réduction des lésions cutanées induites par la lumière ultraviolette (UV). Dès lors, nous avons cherché à étudier les effets de l'extrait de fleur de M. biondii sur les lésions cutanées induites par les UVB et à déterminer le lien entre les lésions cellulaires et les profils moléculaires associés aux lésions (PMAL). MÉTHODES: Des modèles épidermiques reconstruits et des échantillons de prépuce ont été sélectionnés pour détecter les réactions cellulaires après une irradiation aux UVB et un traitement par extrait de fleur de M. biondii. Le test MTT, l'hématoxyline­éosine (HE) et la coloration par immunofluorescence ont été utilisés pour examiner la viabilité totale, les cellules brûlées par le soleil, ainsi que l'expression et la migration des PMAL à 16 ou 48 h. Les taux de prostaglandine E2 (PGE­2) et d'interleukine 8 (IL­8) ont été mesurés par dosages immuno­enzymatiques (ELISA). Une analyse clinique des lésions dues aux UVB avant ou après traitement a été effectuée sur des bras humains traités par extrait de fleur de M. biondii. Des sondes cutanées humaines ont permis de mesurer l'érythème, le taux de mélanine, l'ITA° et la perte en eau transépidermique, tandis que la peau a été photographiée à l'aide du système VISIA. RÉSULTATS: L'extrait de fleur de M. biondii est riche en lignans, comme la magnoline, le pinorésinol diméthyléther et la fargésine, et montre une faible absorption des UV à une longueur d'onde de 280 à 320 nm. La mise en culture de l'extrait de fleur de M. biondii pendant 16 ou 48 h après irradiation aux UVB a amélioré la viabilité et la structure des tissus de Skinovo­Epi et réduit l'expression et la migration de la protéine B1 du groupe à haute mobilité (HMGB1), ainsi que l'expression de l'IL­8 et de la PGE­2. Dans le prépuce excisé traité par extrait de fleur de M. biondii après irradiation aux UVB, la génération de 8­hidroxy­2­désoxyguanosine et le transfert nucléaire de HMGB1 étaient réduits. Lors d'un prétraitement par extrait de fleur de M. biondii pendant 3 jours, l'érythème cutané induit par les UVB et l'ITA° avaient diminué significativement. Lors d'un post­traitement par extrait de fleur de M. biondii pendant 5 jours, une diminution des valeurs de l'érythème cutané, de la mélanine et de la perte en eau transépidermique et une augmentation de l'ITA° cutané ont été observées. CONCLUSIONS: Le traitement par extrait de fleur de M. biondii a atténué les lésions cutanées induites par les UVB, comme l'érythème, la pigmentation et la fonction de barrière cutanée, en améliorant la viabilité et la structure des tissus et en réduisant les cellules brûlées par le soleil et l'inflammation cutanée. Cet effet peut être lié à une altération de l'ADN, qui entraînent la migration du HMGB1 du noyau vers l'extérieur de la cellule, induisant ainsi une inflammation cutanée.

Abstract Magnolia biondii Pamp is an important ornamental tree species widely grown and used as a rootstock in the propagation of different Magnolia varieties. In the current studies, anatomical, physiological and endogenous hormones were studied to check the effect of IBA 750 mg/L on the adventitious rooting and to provide theoretical and technical support for the propagation of Magnolia biondii Pamp through stem cuttings. Two thousand stem cuttings were prepared and divided into two groups i.e., IBA treated cuttings and water control. For the evaluation of antioxidant enzyme activities, and endogenous hormones levels, samples were collected on the day of planting and each 5th day and further steps were carried out in the laboratory according to the protocols and proper precautions. For the anatomical observations, samples were collected on the 13th, 15th, and 17th day for IBA treated cuttings while 21st, 23rd, and 25th day for control. Collected samples were preserved in the FAA solution and further observations were carried out in the laboratory. Anatomical observations showed that it took 13 days for the differentiation of root primordia to the appearance of young adventitious roots in IBA treated cuttings, while it took 21 days to develop primordia in the control. Antioxidant enzyme activities involved in ROS were significantly higher in the IBA treated cuttings compared to control. POD showed a peak on the 13th day before the emergence of roots in IBA treated cuttings while it showed a peak on the 21st day in the control. PPO showed a peak on the 21st day in the IBA treated cuttings while it showed a peak on the 29th day in the control. SOD showed a peak on the 17th day in IBA treated cuttings, while it showed a peak on the 25th day in the control. Exogenous application of IBA enhanced the endogenous IAA and GA3 levels compared to CK, while it reduced the levels of ABA continuously at the time of rooting and then increased gradually. Inclusively, our study suggests that IBA 750 mg/L is efficient for the rooting of Magnolia biondii Pamp cuttings, as it enhanced the process of antioxidant enzyme activities, endogenous hormones levels and reduced the time of root formation which is evident from the anatomical observations.


Resumo Magnolia biondii Pamp é uma importante espécie de árvore ornamental muito cultivada e utilizada como porta-enxerto na propagação de diferentes variedades de Magnolia. Nos estudos atuais, hormônios anatômicos, fisiológicos e endógenos foram estudados para verificar o efeito do AIB na dose de 750 mg / L no enraizamento adventício e fornecer suporte teórico e técnico para a propagação de M. biondii Pamp por meio de estacas. Duas mil estacas foram preparadas e divididas em dois grupos, ou seja, tratadas com AIB e controle de água. Para a avaliação das atividades das enzimas antioxidantes e dos níveis de hormônios endógenos, as amostras foram coletadas no dia do plantio e a cada 5 dias, enquanto as demais etapas foram realizadas em laboratório de acordo com os protocolos e os devidos cuidados. Para as observações anatômicas, as amostras foram coletadas no 13º, 15º e 17º dias para estacas tratadas com AIB e no 21º, 23º e 25º dias para o controle. As amostras coletadas foram preservadas em solução FAA, e outras observações foram realizadas em laboratório. Observações anatômicas mostraram a necessidade de 13 dias para a diferenciação dos primórdios radiculares até o aparecimento de raízes adventícias jovens em estacas tratadas com AIB e de 21 dias para o desenvolvimento dos primórdios no controle. As atividades das enzimas antioxidantes envolvidas nas ROS foram significativamente maiores nas estacas tratadas com AIB em comparação com o controle. A POD apresentou pico no 13º dia antes da emergência das raízes nas estacas tratadas com AIB, enquanto no 21º dia apresentou pico no controle. A PPO teve pico no 21º dia nas estacas tratadas com AIB e no 29º dia no controle. A SOD apresentou pico no 17º dia nas estacas tratadas com AIB e no 25º dia no controle. A aplicação exógena de AIB aumentou os níveis endógenos de IAA e GA3 em relação ao controle, enquanto reduziu os níveis de ABA continuamente no momento do enraizamento e, em seguida, aumentou gradativamente. Inclusive, nosso estudo sugere que o AIB na dose de 750 mg / L é eficiente para o enraizamento de estacas de M. biondii Pamp, visto que potencializou o processo de atividades de enzimas antioxidantes e os níveis de hormônios endógenos, além de reduzir o tempo de formação de raízes, o que fica evidente nas observações anatômicas.

The dried flower buds of Magnolia biondii Pamp (herbal name, Xin-Yi) are a traditional Chinese medicine with a long history of clinical use in the treatment of allergic rhinitis and sinusitis. However, the constituents responsible for its antiallergic effects remain clearly unidentified. In the present study, totally 33 lignans were obtained from M. biondii. Among them, two novel furofuran lignans (1 and 2), two novel tetrahydrofuran lignans (3 and 4), and other 16 known lignans were isolated first time from M. biondii. The antiallergic effects of compounds 1-33 on mouse bone marrow-derived mast cells (BMMCs) degrunaliton were evaluated and results showed that compounds 7, 8, 13, 15 and 18 could significantly inhibited ß-hex release on BMMCs. The results proved that furofuran and tetrahydrofuran lignans were the main constituents in M. biondii and their antiallergic effects were related with suppressing mast cell activation.

Two new lignans, 3,4-(10-methoxy-phenylallyl)-9″-((10'-isopropanol-3',4'-furan)-phenylacetyl)-8″-dioxane-7″-O-ß-d-glucopyranoside (1), 3,4-benzolactone-9″-((12'-isopropanol-3',4'-furan)-phenylbutenone)-8″-dioxane-7″-O-ß-d-glucopyranoside (2), and nine known lignan derivatives (3-11) were isolated from the flower buds of Magnolia biondii Pamp. for the first time. Their structures were elucidated by 1D and 2D NMR, UV, IR, and MS data, as well as by comparison with those of the references. Compounds (1-11) were evaluated for their neuroprotective activities against 6-OHDA-induced cell death in SH-SY5Y cells. As a result, compounds 1, 2, and 5 exhibited significant neuroprotective activities with IC50 values in the range of 3.08-6.12 µM.

Two new phenylpropanoids, named (2'R*,3'R*)-2',3'-dihydroxy-4'-methoxy-caffeoyl butyrate (1), 9-acetoxy syringin (2), and a new dihydrostilbene, named (8'R)-4',5-dihydroxy-4,8'-dimethoxy-2-hydroxyethyl diphenylethane (3), together with five analogues (4-8), were isolated from the flower buds of Magnolia biondii Pamp. Their structures were elucidated by extensive spectroscopic analyses and comparison with literature data. The absolute configurations were deduced by comparison of experimental and calculated gauge-independent atomic orbital (GIAO) 1 D NMR data. Moreover, the isolated compounds (1-8) were evaluated in vitro for their acetylcholinesterase (AChE) inhibitory activities.

Mas-related G protein-coupled receptor X2 was a mast cell-specific receptor mediating anaphylactoid reactions by activating mast cells degranulation, and it was also identified as a target for modulating mast cell-mediated anaphylactoid and inflammatory diseases. The anti-anaphylactoid drugs used clinically disturb the partial effect of partial mediators released by mast cells. The small molecule of Mas-related G protein-coupled receptor X2 specific antagonists may provide therapeutic action for the anaphylactoid and inflammatory diseases in the early stage. In this study, the Mas-related G protein-coupled receptor X2 high expression cell membrane chromatography was coupled online with liquid chromatography and mass spectrometry and successfully used to screen anti-anaphylactoid components from Magnolia biondii Pamp. Fargesin and pinoresinol dimethyl ether were identified as potential anti-anaphylactoid components. Bioactivity of these two components were investigated by ß hexosaminidase and histamine release assays on mast cells, and it was found that these two components could inhibit ß hexosaminidase and histamine release in a concentration-dependent manner. This Mas-related G protein-coupled receptor X2 high expression cell membrane chromatography coupled online with liquid chromatography and mass spectrometry system could be applied for screening potential anti-anaphylactoid components from natural medicinal herbs. This study also provided a powerful system for drug discovery in natural medicinal herbs.

The quality of drug is vital to its curative effect, thus it is important to develop a comprehensive quality control method for commonly used drugs. In this study, we developed a Gas chromatography-mass spectrometry separation method for the qualitative and quantitative analysis of volatiles, together with a High-performance liquid chromatography-mass spectrometry separation method for lignans in Magnolia biondii Pamp.. 79 volatiles and 11 lignans were identified via comparing their chromatographic behavior and mass spectra data with those in the literature. The methods were then used to determine the contents of volatiles (1, 8-cineole, d-Limonene, α-terpineol, linalool, L-camphor brain and bornyl acetate) and lignans (epieudesmin, magnolin, epi-magnolin A and fargesin) in Magnolia biondii Pamp.. Subsequently, 13 qualitative models including volatiles (1, 8-cineole, d-Limonene, α-terpineol, linalool, L-camphor brain and bornyl acetate), water-soluble extractive, lignans (pinoresinol dimethyl ether, magnolin, epi-magnolin A and fargesin) and moisture were developed by Near-Infrared Spectroscopy based on partial least square regression herein. The reference values were obtained by High-performance liquid chromatography, Gas chromatography and etc., while the predicted values were attained from the NIR spectrum. Compared with the traditional detection methods, NIR technique methodology significantly improved the ability to evaluate the quality of Magnolia biondii Pamp., which had the advantages of convenience, celerity, highly efficiency, low cost, no harm to samples, no reagent consumption, and no pollution to the environment. Moreover, the systematic analysis method combined pharmaceutical analysis with pharmacochemistry was proposed to prepare volatiles, water-soluble extractive and lignans parts from the same sample. This way could extract more index components to be beneficial in the quality control of Magnolia biondii Pamp. roundly.

Over the past decades, periodontitis has become a rising health problem and caused various diseases. In the many studies shows that some extracts and compound to the prevention and treatment of periodontitis. This study focuses on the effects of inhibition of gingival damage and alveolar bone loss. The aim of this study was to evaluate the protective effects of Magnolia biondii extract (MBE) against ligature-induced periodontitis in rats. A ligature was placed around the molar teeth for 8 weeks, and MBE was administered for 8 weeks. Gingival tissue damage and alveolar bone loss were measured by microcomputed tomography (CT) analysis and histopathological examination. Serum Interluekin-1 ß (IL-1ß), tumor necrosis factor-α (TNF-α), cyclooxygenases-2 (COX-2), and receptor activator of nuclear factor-κB ligand (RANKL) levels were investigated using commercial kits to confirm the antiperiodontitis effects of MBE. We confirmed that ligature-induced periodontitis resulted in gingival tissue damage and alveolar bone loss. However, treatment for 8 weeks with MBE protected from periodontal tissue damage and downregulated serum inflammatory cytokine factors and RANKL levels. These results suggest that MBE exerts antiperiodontitis effects by inhibiting gingival tissue destruction and alveolar bone loss through regulation of anti-inflammatory cytokines in periodontitis-induced rats.

Dried flower buds of Magnolia biondii Pamp. are the main ingredient in "Xin-yi" in China, and the volatile oils of M. biondii flower buds are the principal medicinal component. Gas chromatographymass spectrometry (GC-MS) and microscopic techniques were employed to detect the volatile yields of M. biondii flowers at various growth stages. The volatile oil yields of M. biondii flowers differed significantly at different growth stages and were closely related to flower dry weight, oil cell density and degree of oil accumulation. In February 2016, flower buds had the highest dry weight, the maximum percentage of oil cells at the oil saturation stage and the highest density of oil cells, which coincided with the highest oil yield. In March 2016, flower buds had a lower dry weight, a higher percentage of oil cells at the oil-degrading stage and the lowest oil cell density, resulting in decreased oil yields. The total amounts of the major medicinal components in the M. biondii flower also showed regular changes at different growth stages. In January and February of 2016, M. biondii flowers had a higher dry weight, volatile oil yield and total content of medicinal ingredients, which was the best time for harvesting high-quality medicinal components. Our study reveals that volatile oil content and chemical composition are closely related to the growth stage of M. biondii flower buds. The results provide a scientific morphology and composition index for evaluating the medicinal value and harvesting of high-quality M. biondii medicinal herbs.

The present study aims to investigate the lignans from the flower buds of Magnolia biondii. The isolation and purification of the compounds were performed by column chromatographies on Diaion HP-20, silica gel, and Sephadex LH-20, combined with semi-preparative HPLC. Their structures were elucidated on the basis of spectral data and physiochemical properties. Four new lignans, a furofuran lignan medioresinol B (10) and three tetrahydrofuran lignans kobusinol C (16), 7'-methoxy magnostellin A (21), and mangnostellin D (23), along with 19 known lignans, were isolated from the flower buds of Magnolia biondii PAMP. The structures of the isolates were elucidated using spectroscopic analysis, mainly one- and two-dimensional NMR, high resolution-MS, and circular dichroism techniques as well as Mosher's esterification method. The anti-allergic effects of the isolated compounds were evaluated by analyzing the inhibition of interleukin-2 (IL-2) expression in Jurkat T-cells. Compounds 11-14 reduced IL-2 expression in a dose-dependent manner.

Chromatographic fingerprint analysis has been widely used in quality control of herbal medicines, and Similarity Analysis (SA) as a well-established method has been applied in the quality control practice as well as publications related to the study of herbal medicines and preparations. However, in some cases the results of SA do not fit well with those of other chemometric approaches and quantitative analysis, and the problem remains unsolved. In this study, a modified SA algorithm has been proposed, with its advantages discussed in theory. The extract of dried flower bud of Magnolia biondii Pamp. obtained by pressurized solvent extraction was then selected as a case to verify the modified algorithm. After identification of the components, fingerprint analysis was performed using different chemometric methods including Hierarchical Cluster Analysis (HCA), Principal Component Analysis (PCA) as well as original and modified SA methods, and the improved discriminative capacity of modified SA algorithm was illustrated. Characteristic chemical markers were then identified using the modified SA approach and then confirmed using PCA method. The quantitative results were then utilized to confirm the advantage of modified SA approach over the original one. The study made a modification to the widely applied SA algorithm, which was possibly a beneficial improvement in fingerprint analysis and quality control practice of herbal medicines.

RESUMEN

The isolation of the MeOH extract from the flower bud of Magnolia biondii Pamp. using various column chromatographies and HPLC led to eleven neoglignan derivatives (1 - 11). Their structures were mainly determined by 1D and 2D NMR spectral data analysis and physiological methods. The isolated compounds (1 - 11) were tested for anti-allergic effects using IL-2 inhibitory assay in Jurkat T cells.

RESUMEN

Four new monoterpenoid glycosides 1-4, named magnoliaterpenoid A-D, were isolated from a 50% aqueous acetone extract of flower buds of Magnolia biondii, along with one known compound, (1'R,3'S,5'R,8'S,2Z,4E)-dihydrophaseic acid 3-O-ß-d-glucopyranoside (5). Their structures and relative configuration were identified by extensive spectroscopic analysis (IR, UV, MS, 1D and 2D NMR). The aglycones of these four new compounds possess seven-membered rings systems, which are very rare. A plausible biosynthetic route for the four new compounds was proposed via the biogenetic isoprene rule. Compounds 1, 2, 3, and 4 showed no antimicrobial activity at the concentration range of 1.95-250 µg/mL.

OBJECTIVE: To study the chemical constituents of Magnolia biondii. Pamp. METHODS: The compounds were isolated and purified by Diaion HP-20, Toyopearl HW-40, silica gel column chromatography and semi-preparative HPLC and so on. The structures were elucidated on the basis of spectral data and physiochemical properties. RESULTS: Fourteen compounds were isolated and identified as 4-O-β-Z)-glucopyranosylvanillic acid(1), tachinoside(2), methyl 4-hydroxy-3-methoxybenzoate (3), caffeic acid(4), 3, 4, 5-trimethoxyphenyl-β-D-glucopyranoside(5), benzyl-O-β-D-glucopyranoside(6), benzyl-O-β-D-galactopyranoside(7), syringin(8), vanillic acid glucosyl ester(9), vanillic acid(10), 1'-(3, 4-dihydroxycinnamoyl)cyclopentane-2', 3'-diol(11), scopolin (12), 7-methoxycoumarin-6-O-β-D-glucopyranoside(13), and scopoletin(14). CONCLUSION: Compounds 1-9 and 11-13 are isolated from this plant for the first time.

OBJECTIVE: To investigate the anti-inflammatory effect and acute toxicity of standard Magnolia biondii Pamp oil (SMBO). METHODS: The anti-inflammatory effect of SMBO was evaluated in five experimental animal models, including xylene-in-duced mice ear edema, carrageenin-induced rat hind paw edema, histamine-induced rat vascular permeability, cotton pellet-induced rat granuloma formation, and guinea pig schneiderian membrane immuno-inflammation. In addition, the 7-day acute oral toxicity of SMBO was evaluated. RESULTS: Pretreatment with a single dose of SMBO produced significant dose-dependent anti-inflammatory effects on xylene-induced mice ear edema, carrageenin-induced rat hind paw edema, histamine-induced rat vascular permeability, cotton pellet-induced rat granuloma formation, and guinea pig schneiderian membrane immuno-inflammation. In addition, a single oral dose of SMBO at 2.5 g · kg-1 body weight produced no observable acute toxicity in mice within seven days. CONCLUSION: SMBO possesses potent anti-inflammatory effect and has no observable acute oral toxicity, and the mechanism may be related with the inhibition of multi-inflammatory mediators and inflammatory hyperplasia.

Objective To investigate the pharmacologic action of the Magnolia biondii Pamp volatile oil nanometer bangosome. MethodsThe pharmacologic action of the Magnolia biondii Pamp volatile oil nanometer bangosome was evaluated by observing its effect on Jimpy mice with swelling ear,capillary permeability and rats with allergic symptoms. Results The Magnolia biondii Pamp volatile oil nanometer bangosome could significantly relieve the degree of swelling in Jimpy mice extended by p-xylene,inhibit the increased capillary permeability in Jimpy mice extended by HAC,and combat the symptoms of rhinocnesmus,sneezing,nasal discharge with a better effect than the Magnolia biondii Pamp volatile oil. Conclusion The Magnolia biondii Pamp Volatile oil nanometer bangosome has a good anti-inflammation and anti-hypersensitiveness effect,which upgrades the effect of the Magnolia biondii Pamp volatile oil.

Objective To investigate the acute toxicity, subacute toxicity, nasal membrane local toxicity of the Magnolia biondii Pamp volatile oil nanometer bangosome. Methods Kunming mice and SD rats were selected as experimental material. Kunming mice were used in the acute and subacute toxicity tests by intragastric administration of Magnolia biondii Pamp volatile oil nanometer bangosome with different dosage and different time, and SD rats were used in the nasal membrane local toxicity test by nose dropping, while the control group was treated with intragastric administration of or nose dropping with normal saline for the same dosage. The treatment course lasted fifteen days. At the end of the tests, the general condition, routine blood test, function index of live and kidney, organ humid weight index, histological changes of liver and kidney and ultra microstructure change of rat nasal membrane were obtained and compared with the control group. Results In the acute toxicity test, the daily maximum tolerant dosage by intragastric administration was equal to 222.7 times of the clinical routine, with no marked toxic reaction. In the subacute toxicity test, the general condition, blood test, organ humid weight index and histological changes of live and kidney in different dosage groups were similar to the control group. While in the function index of live and kidney, uric acid was stepped down in the middle and large dose groups, and total bilirubin was decreased in the large dose group. The nasal membrane local toxicity test revealed that there was little change in the ultra microstructure of rat nasal membrane.Conclusion The Magnolia biondii Pamp volatile oil nanometer bangosome may have little toxicity by intragastric administration and nose dropping.
