Introduction: Cattle health and welfare are monitored via the analysis of the haematological profile, and it shows cattle's ability to adapt to changing environmental conditions, pregnancy and lactation; profile changes also indicate reproductive disorders. The literature lacks reports of the examination of the haematological profile in cows up to the 50th day of pregnancy (dop). Therefore, this research examined that in cows up to this pregnancy stage. Material and Methods: A total of 101 Polish Holstein-Friesian black-and-white cows were divided into groups. The control groups consisted of non-pregnant heifers (group C00) and non-pregnant cows (group C0), and the experimental groups were pregnant heifers (group T1 at dop ≤ 28 and group T2 at dop ≥ 29-dop < 45) and pregnant cows (group T3 at dop ≥ 29-dop ≤ 50). In addition, the T3 group was divided into cows pregnant for up 45 dop and cows between 45 and 50 dop. Blood samples were collected in March and April 2021 from each animal and analysed. A transrectal ultrasound examination was performed to detect and confirm pregnancy. Results: Statistically significant differences (P ≤ 0.01) between the group of cows at dop < 45 dop and those at dop ≥ 45-dop ≤ 50 dop were noted in granulocyte percentage (GRA%), white and red blood cell counts (WBC/RBC), platelets (PLT), platelet distribution width (PDW), haematocrit (HCT) and lymphocyte percentage (LYM%). No statistically significant differences were found in the mean corpuscular haemoglobin, monocytes (MON), monocyte percentage (MON%), mean platelet volume (MPV), thrombocrit or red blood cell distribution width (RDW). Similar statistically significant differences (P ≤ 0.01) emerged between the groups of heifers in PLT, GRA, RBC, lymphocytes, LYM% and HCT, and no significant differences were found between MPV, MON, MON% or RDW. Conclusion: Examining the haematological profile in high-yielding cattle is vital in maintaining herd profitability and high reproduction, which depend on the quick diagnosis of disorders facilitated by haematology. This study analysed the haematology profile of dairy cattle at dop ≤ 50 for the first time, indicating changes in lymphocyte levels, which suggests that the animals experienced direct stress during the study.

This study aimed to assess the effect of milk source and bear garlic addition on the selected properties of soft rennet cheese. Cheeses were produced from cow milk derived from two sources: Polish Red cows (PR) and Polish Holstein-Friesian cows (PHF) with a 0.5% (w/w) addition of bear garlic (Allium ursinum L.) dried leaves. Chemical composition and fatty acid profiles (GC) were determined in fresh cheeses. Fresh and stored for two weeks cheeses were subjected to microbiological studies, i.e., total aerobic bacteria count (TABC); count of Lactococcus sp., yeast and molds; coliforms; analysis of the proteolysis extension by means of o-phthaldialdehyde (OPA) assay and free amino acids content (HPLC); antioxidant capacity as 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and ferric reducing antioxidant power (FRAP); as well as pH and water activity. Cheeses with bear garlic herbs were more prone to proteolysis but this was not accompanied by any effect on the microbial counts, water activity or pH. Cheeses produced from PR milk contained less monounsaturated fatty acids (MUFA) but were richer in n-3 PUFA and had a lower n-6/n-3 FA ratio than cheeses from PHF milk. Bear garlic addition increased DPPH anti-radical power but had less of an effect on the FRAP values.

The assay aimed to evaluate the effect of feeding a recently developed phytogenic feed additives mixture in diets of lactating Friesian cows (n = 30; 514 ± 10.1 kg body weight) for 3 months. Cows were stratified into three groups of 10 cows each and fed a control diet alone or the control diet supplemented with the additives mixture at 3 g (PHY3) or 6 g (PHY6)/cow daily. Menthol, levomenthol, ß-linaloolm, anethole, hexadecanoic acid and p-menthane were the principle compounds identified in the additives mixture. The PHY3 increased (p < 0.01) intake and nutrient digestibility. PHY3 and PHY6 increased (p < 0.01) ruminal pH, total volatile fatty acids, propionate and acetate. PHY3 and PHY6 improved serum total protein and antioxidant capacity and decreased the concentrations of serum urea-N, triglycerides, total lipids, cholesterol and malondialdehyde (p < 0.05). PHY3 increased milk production and milk content of total solids, protein, lactose and fat. Both PHY3 and PHY6 did not affect mineral concentrations in blood or milk. It is concluded that the inclusion of 3 g/cow/d of feed additives mixture in the lactating Friesian cows diet enhanced milk production and feed utilization, with negative effects observed with increasing the dose of additives mixture to 6 g/cow daily.

Milk lipids are known for a variety of biological functions, however; little is known about compositional variation across breeds, especially for Jaffarabadi buffalo, an indigenous Indian breed. Systematic profiling of extracted milk lipids was performed by mass spectrometry across summer and winter in Holstein Friesian cow and Jaffarabadi buffalo. Extensive MS/MS spectral analysis for the identification (ID) of probable lipid species using software followed by manual verification and grading of each assigned lipid species enabled ID based on (a) parent ion, (b) head group, and (c) partial/full acyl characteristic ions for comparative profiling of triacylglycerols between the breeds. Additionally, new triacylglycerol species with short-chain fatty acids were reported by manual interpretation of MS/MS spectra and comparison with curated repositories. Collectively, 1093 triacylglycerol species belonging to 141 unique sum compositions between the replicates of both the animal groups were identified. Relative quantitation at sum composition level followed by statistical analyses revealed changes in relative abundances of triacylglycerol species due to breed, season, and interaction effect of the two. Significant changes in triacylglycerols were observed between breeds (81%) and seasons (59%). When the interaction effect is statistically significant, a higher number of triacylglycerols species in Jaffarabadi has lesser seasonal variation than Holstein Friesian.

OBJECTIVE: The aim of the present study was to investigate the effect of dietary zinc supplementation of Friesian cows on chemical-nutritional and aromatic properties of Caciocavallo cheese after 7 days (C7) and 120 days (C120) of ripening. METHODS: Twenty eight Friesian cows, balanced for parity, milk production and days in milk, were randomly assigned to 2 groups. The control group (CG) was fed with a conventional complete diet, while the experimental group (zinc group, ZG) received a daily zinc supplementation of 60 mg for kg of dry complete feed. During the experimental period, the milk yield was monitored and samples of milk and caciocavallo cheese were collected and analyzed for chemical-nutritional composition and aromatic profile. RESULTS: The enrichment of dairy cows diet with zinc, did not influence milk yield and composition, however a marked reduction of somatic cell count was evidenced. Both in milk and cheese the ZG samples were characterized by a lower concentration of satured fatty acids and an increase in oleic, vaccenic and rumenic acids. The aromatic profile of dairy products was also positively affected by dietary zinc intake, with an increase in concentration of carboxylic acids, esters and lactones. CONCLUSION: The present results suggest a positive role of dietary zinc intake in improving the quality of bovine milk and related cheese, in particular for the increase in concentration of bioactive fatty acids such as rumenic acid. The changes evidenced in cheese through the analysis of the volatile profile, would be consistent with the development of interesting organoleptic properties, although further evaluations should be performed to confirm the consumer acceptability of these changes.

Bovine milk contains different components with nutritional and immunological benefits. It is easily accessible and a rich source of potential markers reflective of pathophysiological conditions; however, little is known about the changes in protein abundance associated with variation across breeds and seasons. In this study, we performed a comprehensive proteomic profiling of whey proteins from Holstein Friesian cow and Murrah buffalo across summer and winter seasons. Collectively, 490 proteins were identified with 113 and 144 differentially expressed proteins across seasons in cow and buffalo, respectively. Breed specific proteins like secretoglobin, e-cadherin and cathepsin-L were detected exclusively in HF, while basigin, conglutinin, and thrombomodulin were identified exclusively in Mu. Acute phase proteins (e.g. haptoglobin and α1AG) were more abundant in summer while antimicrobial proteins (e.g. conglutinin and osteopontin) were upregulated in winter. Similarly, proteins involved in lipid homeostasis (e.g. perilipin 2 and acyl CoA binding protein) showed breed specific variations. Selected representative mass spectrometric proteins (e.g. gelsolin and osteopontin) were validated by Western blot analysis. Results of this study indicate the dynamic nature of milk protein and provide a foundation for future studies of whey proteins which may be linked to diseases specific across breeds and seasons.

Amniotic fluid is a rich source of multipotent mesenchymal stem cells (MSCs). Amniotic fluid stem cells (AFSCs) have become a new source of stem cells; they have low immunogenicity and are easily harvested. For this reason, they may be useful in clinical tissue engineering. Moreover, AFSCs have anti-inflammatory properties and can repair tissues. This study evaluated the utility of AFSC injection to treat bilateral ovarian dystrophy in Holstein-Friesian cows. Bovine AFSCs (BAFSCs) were collected at slaughter from Holstein-Friesian cows during the third or fourth month of pregnancy and cultured in vitro. The BAFSCs began to show a fibroblast-like morphology. They were positive for ß-integrin, CD44, CD73, CD106 and Oct4 and negative for CD34 and CD45. After induction, the cells differentiated into mesodermal lineages. Bilateral ovarian dystrophy was confirmed by ultrasonography in 16 lactating cows. The subsequent experiment lasted 15 weeks. Serum was collected weekly to analyse progesterone concentrations, and weekly ultrasonography recorded ovarian changes. Each cow was equipped with an automatic heat detection system to facilitate oestrus observation and breeding records. The progesterone concentration of two cows in the treatment group (25%) significantly increased during weeks 10-15. On ultrasonography, the treatment group demonstrated mature follicles after BAFSCs injection, and foetuses were visualized approximately 40 days after artificial insemination (AI). Oestrus rates in the control and treatment groups were 0% (0/8) and 50% (4/8), respectively; pregnancy rates were 0% (0/8) and 25% (2/8), respectively. Calves were successfully delivered in both cases of pregnancy. These results show that BAFSCs can alleviate bovine ovarian dystrophy and restore fertility.

The present study was undertaken to examine the effect of cow genetic merit on enteric methane (CH4) emission rate. The study used a data set from 32 respiration calorimeter studies undertaken at this Institute between 1992 and 2010, with all studies involving lactating Holstein-Friesian dairy cows. Cow genetic merit was defined as either profit index (PIN) or profitable lifetime index (PLI), with these two United Kingdom genetic indexes expressing the expected improvement in profit associated with an individual cow, compared with the population average. While PIN is based solely on milk production, PLI includes milk production and a number of other functional traits including health, fertility and longevity. The data set had a large range in PIN (n=736 records, -£30 to +£63) and PLI (n=548 records, -£131 to +£184), days in milk (18 to 354), energy corrected milk yield (16.0 to 45.6 kg/day) and CH(4) emission (138 to 598 g/day). The effect of cow genetic merit (PIN or PLI) was evaluated using ANOVA and linear mixed modelling techniques after removing the effects of a number of animal and diet factors. The ANOVA was undertaken by dividing each data set of PIN and PLI into three sub-groups (PIN:£15, PLI:£67) with these being categorised as low, medium and high genetic merit. Within the PIN and PLI data sets there was no significant differences among the three sub-groups in terms of CH(4) emission per kg feed intake or per kg energy corrected milk yield, or CH(4) energy (CH(4)-E) output as a proportion of energy intake. Linear regression using the whole PIN and PLI data sets also demonstrated that there was no significant relationship between either PIN or PLI, and CH(4) emission per kg of feed intake or CH(4)-E output as a proportion of energy intake. These results indicate that cow genetic merit (PIN or PLI) has little effect on enteric CH(4) emissions as a proportion of feed intake. Instead enteric CH(4) production may mainly relate to total feed intake and dietary nutrient composition.

Milk is a complex fluid whose proteome displays a diverse set of proteins of high abundance such as caseins and medium to low abundance whey proteins such as ß-lactoglobulin, lactoferrin, immunoglobulins, glycoproteins, peptide hormones, and enzymes. A sample preparation method that enables high reproducibility and throughput is key in reliably identifying proteins present or proteins responding to conditions such as a diet, health or genetics. Using skim milk samples from Jersey and Holstein-Friesian cows, we compared three extraction procedures which have not previously been applied to samples of cows' milk. Method A (urea) involved a simple dilution of the milk in a urea-based buffer, method B (TCA/acetone) involved a trichloroacetic acid (TCA)/acetone precipitation, and method C (methanol/chloroform) involved a tri-phasic partition method in chloroform/methanol solution. Protein assays, SDS-PAGE profiling, and trypsin digestion followed by nanoHPLC-electrospray ionization-tandem mass spectrometry (nLC-ESI-MS/MS) analyses were performed to assess their efficiency. Replicates were used at each analytical step (extraction, digestion, injection) to assess reproducibility. Mass spectrometry (MS) data are available via ProteomeXchange with identifier PXD002529. Overall 186 unique accessions, major and minor proteins, were identified with a combination of methods. Method C (methanol/chloroform) yielded the best resolved SDS-patterns and highest protein recovery rates, method A (urea) yielded the greatest number of accessions, and, of the three procedures, method B (TCA/acetone) was the least compatible of all with a wide range of downstream analytical procedures. Our results also highlighted breed differences between the proteins in milk of Jersey and Holstein-Friesian cows.

The aim of this study was to detect new polymorphisms in the bovine ß-casein (ß-CN) gene and to evaluate association of (new) ß-CN protein variants with milk production traits and milk protein composition. Screening of the ß-CN gene in genomic DNA from 72 Holstein Friesian (HF) bulls resulted in detection of 19 polymorphisms and revealed the presence of ß-CN protein variant I in the Dutch HF population. Studies of association of ß-CN protein variants with milk composition usually do not discriminate protein variant I from variant A2. Association of ß-CN protein variants with milk composition was studied in 1857 first-lactation HF cows and showed that associations of protein variants A2 and I were quite different for several traits. ß-CN protein variant I was significantly associated with protein percentage and protein yield, and with αs1 -casein (αs1 -CN), αs2 -casein (αs2 -CN), κ-casein (κ-CN), α-lactalbumin (α-LA), ß-lactoglobulin (ß-LG), casein index and casein yield. Inferring ß-κ-CN haplotypes showed that ß-CN protein variant I occurred only with κ-CN variant B. Consequently, associations of ß-κ-CN haplotype IB with protein percentage, κ-CN, α-LA, ß-LG and casein index are likely resulting from associations of κ-CN protein variant B, while associations of ß-κ-CN haplotype IB with αs1 -CN and αs2 -CN seem to be resulting from associations of ß-CN variant I.
