RESUMEN

A previous study on ovarian and hypothalami transcriptome analysis in white Muscovy duck revealed that MAP3K8 gene participated in MAPK signaling pathway that influence egg production. Additionally, MAP3K8 was predicted as a target gene of miRNA-509-3p that promotes the secretion of oestradiol which is an important hormone in egg ovulation. This suggested that MAP3K8 might have a functional role in the reproductive performance "egg production" of white Muscovy ducks. Herein, we focused on expression level of MAP3K8 in reproductive and non-reproductive tissues of highest (HP) and lowest (LP) egg producing white Muscovy ducks and identified the polymorphism in MAP3K8 and its association with three egg production traits; Age at first egg (AFE), number of eggs at 300 days (N300D) and 59 weeks (N59W). The results of expression level indicated that mRNA of MAP3K8 was significantly (p < 0.01) expressed in the oviduct than in the ovary and hypothalamus. Seven synonymous SNPs were detected, and association analysis showed that g.148303340 G>A and g.148290065 A>G were significantly (p < 0.05) associated with N300D and N59W. The results of this study might serve as molecular marker for marker-assisted selection of white Muscovy ducks for egg production.

RESUMEN

The pathological mechanisms of cataract remain largely unknown due to the lack of appropriate in vitro cellular models. We developed a stable in vitro system, namely, a "fried egg" differentiation method to generate functional lentoid bodies (LBs) from induced pluripotent stem cells (iPSCs). The iPSCs-derived LBs exhibited crystalline lens-like morphology and a transparent structure, and expressed lens-specific markers. TEM examination and optical analysis further demonstrated that it has the same cell arrangement structure and magnifying ability as lens.

Squamate reptiles are central for studying phenotypic correlates of evolutionary transitions from oviparity to viviparity because these transitions are numerous, with many of them being recent. Several models of life-history theory predict that viviparity is associated with increased female size, and thus more female-biased sexual size dimorphism (SSD). Yet, the corresponding empirical evidence is overall weak and inconsistent. The lizard Zootoca vivipara, which occupies a major part of Northern Eurasia and includes four viviparous and two non-sister oviparous lineages, represents an excellent model for testing these predictions. We analysed how sex-specific body size and SSD is associated with parity mode, using body length data for nearly 14,000 adult individuals from 97 geographically distinct populations, which cover almost the entire species' range and represent all six lineages. Our analyses controlled for lineage identity, climatic seasonality (the strongest predictor of geographic body size variation in previous studies of this species) and several aspects of data heterogeneity. Parity mode, lineage and seasonality are significantly associated with female size and SSD; the first two predictors accounted for 14%-26% of the total variation each, while seasonality explained 5%-7%. Viviparous populations exhibited a larger female size than oviparous populations, with no concomitant differences in male size. The variation of male size was overall low and poorly explained by our predictors. Albeit fully expected from theory, the strong female bias of the body size differences between oviparous and viviparous populations found in Z. vivipara is not evident from available data on three other lizard systems of closely related lineages differing in parity mode. We confront this pattern with the data on female reproductive traits in the considered systems and the frequencies of evolutionary changes of parity mode in the corresponding lizard families and speculate why the life-history correlates of live-bearing in Z. vivipara are distinct. Comparing conspecific populations, our study provides the most direct evidence for the predicted effect of parity mode on adult body size but also demonstrates that the revealed pattern may not be general. This might explain why across squamates, viviparity is only weakly associated with larger size.

Due to consumer demands and institutional pressure, the egg production sector, is looking for alternative protein sources for laying hen feed to support more sustainable, circular production. black soldier fly (BSF) larvae could be used as a protein source. In addition to protein the larvae contain large quantities of fat and can either be fed to laying hens unprocessed (alive) or processed (meal and oil). The current study was performed with 560 Brown Nick laying hens from 20 to 27 wk of age. The laying hens were divided over 5 treatments, each replicated 8 times. Treatments consisted of standard laying hen feed (control) and standard feed in which soybean meal was partly exchanged with live BSF larvae or BSF larvae meal and oil combined, at 2 inclusion levels. During the experiment production parameters, egg-quality, and length and weight of various organs were measured. Laying hens fed BSF larvae products consumed less feed compared to those of the control group. Most egg production parameters were similar, however laying hens fed diets with BSF larvae meal plus oil produced eggs with lower egg weight during the last 2 wk of the experiment, compared to the control group. All egg-quality characteristics remained the same across treatments, except for darker yolk colors when feeding BSF meal and oil and high inclusion of live BSF larvae. This is a favorable characteristic for European consumers. The weight of intestinal organs was largely unaffected by the treatments. The jejunum and ileum weight of laying hens fed live larvae was lower compared to the control group. As FCRs were similar or improved compared to the control group, we assume that nutrient utilization was not impaired. For most detected differences the type of BSF larvae product (live larvae or meal plus oil) rather than inclusion level was of significance.

Despite the economic incentives evidenced in the recruitment strategies of the Iranian fertility industry for egg donors, the official discourse put forward by policymakers conveys egg donation as an altruistic act. Drawing on ethnographic fieldwork in two fertility clinics in Tehran, I center the narratives of paid egg donors to investigate how multiple meanings are attributed to egg donation as a form of labor, demonstrating how reproductive inequalities are perpetuated in this context. Following feminist theorists of reproductive bioeconomies, I argue that Iranian donors experience and articulate their participation in local egg market through the prism of their economic marginality, gendered responsibilities, and religiously informed beliefs, including divine reward.

Egg-laying is an important trait in chickens, and it is affected by many factors, such as hormones regulated by the hypothalamic-pituitary axis and precursors synthesized by the liver. Recent studies showed that gut microbiota was associated with egg-laying, however, its underlying mechanism remains unclear. We comprehensively analyzed the host transcriptome, gut microbiota and metabolome in broiler breeder hens during the pre-laying, peak-laying and late-laying periods. The transcriptome analysis of the tissues related to the hypothalamic-pituitary-liver (HPL) axis revealed dynamic gene expression during egg-laying periods. Differentially expressed genes (DEGs) (i.e., PENK, NPY, AVP, PRL, RLN3, and FST) from the hypothalamus and pituitary gland were involved in female gonadal development, hormone secretion, response to endogenous stimulus, liver development, and amide metabolism. In liver, DEGs (i.e., FABP3, VTG1, LPL, APOA5, APOV1, and RBP5) were enriched in efferocytosis, sphingolipid metabolism, amide and peptide biosynthesis. Alpha and beta diversity changed significantly in cecum microbiota during different laying periods. The abundance of Firmicutes was decreased and the abundance of Bacteroidota was increased during the peak-laying period. Functional analysis showed that the biosynthesis of secondary metabolites, amino acids, purine, and steroid hormones were altered during laying. The metabolome analysis from cecal contents showed that amino acid metabolism and steroid hormone biosynthesis changed during laying. Integrated analysis of the cecal microbiota and metabolites showed the genus Megasphaera was involved in amino acid metabolism, which included 3-phenyllatic acid, quinic acid, caffeic acid, and folic acid, and the genus Hungatella participated in steroid hormone biosynthesis through its strong correlation with estradiol. These results explored the dynamic changes in tissues related to the HPL axis and cecal microbiota, and provided new insights into the interaction between the host and microbiota during egg-laying in chickens.

The genotoxic and clastogenic/aneugeneic potentials of four α,ß-unsaturated aldehydes, 2-phenyl-2-butenal, nona-2-trans-6-cis-dienal, 2-methyl-2-pentenal, and p-methoxy cinnamaldehyde, which are used as fragrance materials, were assessed using the Chicken Egg Genotoxicity Assay (CEGA) and the Hen's egg micronucleus (HET-MN) assay, respectively. Selection of materials was based on their chemical structures and the results of their previous assessment in the regulatory in vitro and/or in vivo genotoxicity test battery. Three tested materials, 2-phenyl-2-butenal, nona-2-trans-6-cis-dienal, and 2-methyl-2-pentenal, were negative in both, CEGA and HET-MN assays. These findings were congruent with the results of regulatory in vivo genotoxicity assays. In contrast, p-methoxy cinnamaldehyde, which was also negative in the in vivo genotoxicity assays, produced evidence of DNA damage, including DNA strand breaks and DNA adducts in CEGA. However, no increase in the micronucleus formation in blood was reported in the HET-MN study. Such variation in responses between the CEGA and HET-MN assay can be attributed to differences in the dosing protocols. Pretreatment with a glutathione precursor, N-acetyl cysteine, negated positive outcomes produced by p-methoxy cinnamaldehyde in CEGA, indicating that difference in response observed in the chicken egg and rodent models can be attributed to rapid glutathione depletion. Overall, our findings support the conclusion that CEGA and/or HET-MN can be considered as a potential alternative to animal testing as follow-up strategies for assessment of genotoxic potential of fragrance materials with evidence of genotoxicity in vitro.

Egg-laying is one of the key aspects of female reproductive behavior in insects. Egg-laying has been studied since the dawn of Drosophila melanogaster as a model organism. The female's internal state, hormones, and external factors, such as nutrition, light, and social environment, affect egg-laying output. However, only recently, neurobiological features of egg-laying behavior have been studied in detail. fruitless and doublesex, two key players in the sex determination pathway, have become focal points in identifying neurons of reproductive significance in both central and peripheral nervous systems. The reproductive tract and external terminalia house sensory neurons that carry the sensory information of egg maturation, mating and egg-laying. These sensory signals include the presence of male accessory gland products and mechanical stimuli. The abdominal neuromere houses neurons that receive information from the reproductive tract, including sex peptide abdominal ganglion neurons (SAGs), and send their information to the brain. In the brain, neuronal groups like aDNs and pC1 clusters modulate egg-laying decision-making, and other neurons like oviINs and oviDNs are necessary for egg-laying itself. Lastly, motor neurons involved in egg-laying, which are mostly octopaminergic, reside in the abdominal neuromere and orchestrate the muscle movements required for laying the egg. Egg-laying neuronal control is important in various evolutionary processes like cryptic female choice, and using different Drosophila species can provide intriguing avenues for the future of the field.

The result of a collaboration between an anthropologist and a photographer, in this photo essay we aim to visualize the medical process of egg donation and the quotidian lives of egg donors in Spain. By extending their biographies beyond the moment of extraction, we shed light on the intertwined messiness of medical procedures and everyday life and the precarious circumstances in which egg donation takes place in Spain today. Our aim is to highlight the participants who, although they matter most in the egg donation economy, are concealed: the egg donors.

Gnetum gnemon L. is an evergreen tree that belongs to the Gnetaceae family and is commonly used as a vegetable and medicinal plant among indigenous people. The key goal of this study was to assess the antibacterial efficacy of ethanolic G. gnemon leaf extract (EGLE) against six food-borne pathogens. The antimicrobial activity of EGLE was evaluated using multiple methods, including the well diffusion assay (WDA), minimum inhibitory concentration (MIC), minimum bactericidal concentration (MBC), and time-kill assay. Gas Chromatography-Mass Spectrometry (GC-MS) analysis was used to identify active volatile compounds responsible for EGLE's antibacterial activities. Total plate count (TPC) was conducted to measure microbial populations and evaluate the efficacy of EGLE as a natural preservative in raw quail eggs. 100 g of dried and powdered sample yielded an average of 11.58 ± 0.38 % post-extraction. The inhibition zone in WDA ranged from 11.00 ± 0.57-13.50 ± 0.58 mm, MIC ranged from 6.25 to 50.00 mg/mL, and MBC values were between 12.5 and >50 mg/mL. Results from the time-kill study showed that at 4 × MIC Bacillus pumilus and B. megaterium were completely killed in 1 h incubation time and other bacteria were killed within 2-4 h. Findings from TPC demonstrated that at the highest tested concentration of EGLE, there was no significant bacterial growth for a 30-day observation period. Thereby, suggesting that it had the potential to function as a natural preservative for raw quail eggs. EGLE may be a viable alternative to synthetic preservatives in combating food-borne pathogens.

As an essential food ingredient with good nutritional and functional properties and health benefits, eggs are widely utilized in food formulations. In particular, egg proteins have good emulsification properties and can be commonly used in various food products, such as mayonnaise and baked goods. Egg protein particles can act as stabilizers for Pickering emulsions because they can effectively adsorb at the oil-water interface, reduce interfacial tension, and form a stable physical barrier. Due to their emulsifying properties, biocompatibility, controlled release capabilities, and ability to protect bioactive substances, egg proteins have become ideal carriers for encapsulating and delivering functional substances. The focus of this review is to summarize current advances in using egg proteins as emulsifiers. The effects of influencing factors (temperature, pH, and ionic strength) and various modification methods (physical, chemical, and biological modification) on the emulsifying properties of egg proteins are discussed. In addition, the application of egg proteins as emulsifiers in food products is presented. Through in-depth research on the emulsifying properties of egg proteins, the optimization of their applications in food, biomedical, and other fields can be achieved.

Given the significant environmental consequences of current poultry feed practices and the heavy dependence of the European Union on imported soybeans, studying alternatives is crucial. This study evaluated the potential benefits of using locally sourced alternative plant-based ingredients and whole dry black soldier fly larvae in the diet of laying hens. The experiment involved 120 Isazul hens at 23 weeks old, which were divided into three groups with five replicates each (eight hens per replicate): a control diet (CON) based on soybean meal and cereals, an alternative diet (ALT) replacing the soybean meal with locally sourced plant-based resources (peas, distillers' dried grains with solubles, and sunflower meal), and the ALT diet supplemented with 5% whole dry black soldier fly larvae (ALT+DBSFL). Over 15 weeks, the hens were fed ad libitum, and the production performance, egg quality, and plasma biochemical parameters were assessed across three experimental sub-periods. The results showed no significant differences in body weight, feed intake, egg production, egg weight, egg mass, or feed conversion ratio across the diets (p > 0.05). The egg quality remained consistent across all the groups; however, the hens fed the ALT+DBSFL diet exhibited higher Haugh units in the first experimental sub-period (p < 0.05) and lower plasma cholesterol and triglycerides at 32 weeks of age (p < 0.05). The findings of this study indicate that incorporating these alternative ingredients and whole DBSFL into layers' diets does not compromise production performance, egg quality, or biochemical parameters related to health status, supporting their potential as sustainable feed alternatives.

Chlorella vulgaris (CV) is a microalga with considerable nutritional value, containing high levels of protein, carotenoids, and polyunsaturated fatty acids, which have the potential to positively influence the productive performance and egg quality of laying hens. CV emerges as a more sustainable ingredient than soybean meal (SBM) as it can be produced locally and with fewer inputs. In this regard, a study was conducted with 48 H&N Brown Nick strain laying hens, at 19 weeks of age, over a period of 16 weeks. The hens were divided into four treatments, with 12 replicates each. The treatments consisted of providing four different diets: a control diet based on corn and SBM without the inclusion of CV, and three other diets with partial substitution of SBM by 2.5, 5, and 10% of CV. The results showed that the inclusion of CV in the diets did not significantly affect feed intake, feed conversion ratio, or egg production (p > 0.05). In addition, moderate CV levels increased egg weight, while higher levels reduced it. Haugh units, yolk index, albumen index, egg surface area, specific density, and translucency were not affected (p > 0.05), while shell index and shell thickness were lower in the groups that received the CV (p < 0.0001). Yolk color improved significantly with increasing CV levels (p < 0.0001), with darker, more intensely colored yolks at higher CV concentrations. The results of this study suggest that the incorporation of CV in the diet of laying hens had a limited impact on performance parameters. In addition, CV supplementation can positively influence egg quality and yolk color, although careful consideration of optimal levels is necessary to avoid negative effects on other parameters.

Applications of millet bran dietary fiber (MBDF) in the food industry are limited by its poor hydration properties. Herein, MBDF was modified by heating, xylanase and cellulase treatment separately combined with carboxymethylation, acetylation, and phosphate crosslinking, and the effects of the modified MBDFs on heat-induced egg white protein gel (H-EWG) were studied. The results showed that three composite modifications, especially heating and dual enzymolysis combined with carboxymethylation, increased the surface area, soluble fiber content, and hydration properties of MBDF (p < 0.05). MBDF and the modified MBDFs all made the microstructure of H-EWG denser and decreased its α-helix content. Three composite modifications, especially heating and dual enzymolysis combined with carboxymethylation, enhanced the improving effect of MBDF on the WRA (from 24.89 to 35.53 g/g), pH, hardness (from 139.93 to 323.20 g), chewiness, and gumminess of H-EWPG, and enhanced the gastric stability at 3-5 g/100 g. MBDFs modified with heating and dual enzymolysis combined with acetylation or crosslinking were more effective in increasing the antioxidant activity of the gastrointestinal hydrolysates of H-EWG than MBDF (p < 0.05). Overall, heating, xylanase and cellulase treatment separately combined with carboxymethylation, acetylation and crosslinking can enhance the hydration properties and the improving effect of millet bran fibers on H-EWG properties.

The Taihe Black-Bone Silky Fowl (Gallus gallus domesticus Brisson) possesses significant value in terms of consumption, medicinal applications, and ornamental appeal, representing a precious genetic resource and traditional Chinese medicinal material. However, considerable variation exists within populations regarding egg-laying performance. This study integrates a whole-genome selection signal analysis (SSA) with a transcriptome analysis to identify genes associated with egg-laying traits in Taihe Black-Bone Silky Fowls. We identified 31 candidate genes under selection from the high-yield chicken (HC) and low-yield chicken (LC) groups. Additionally, through RNA-seq analysis, 257 common differentially expressed genes (DEGs) were identified from four comparative groups. Two overlapping genes-LPL and SETBP1-were found in both the selected gene and DEG lists. These selected genes and DEGs were enriched in pathways related to ovarian development, including the lysosome pathway, the ECM-receptor interaction pathway, the TGF-beta signaling pathway, the Wnt signaling pathway, the PPAR signaling pathway, and the glycerolipid metabolism pathway. These research findings contribute to the breeding of Taihe Black-Bone Silky Fowls with high egg production traits and provide a theoretical foundation for exploring the regulatory mechanisms of avian reproduction.

As the second most widely consumed eggs, duck eggs are made into preserved eggs, salted duck eggs, and roasted duck eggs to extend their shelf-life. To investigate the differences in potent odorants (POs) between salted duck egg yolk (SDEY) and roasted duck egg yolk (RDEY), the volatiles in SDEY and RDEY were extracted through solvent extraction coupled with solvent-assisted flavor evaporation and were assayed with gas chromatography-mass spectrometry-olfactometry. A total of 45 volatiles were identified in two samples, 24 odor-active compounds (OACs) were screened, and more OACs were in RDEY. The flavor-dilution (FD) factors of OACs were obtained by aroma extract dilution analysis and ranged from 3 to 6561. Twenty-two OACs with FD factors ≥ 9 were quantitated, and the results indicated the concentrations of OACs in yolk increased greatly after salted duck eggs were roasted. Based on the concentrations and thresholds, odor activity values (OAVs) were determined; 17 odorants with OAVs ≥ 1 were determined as POs. Acetoin was the most PO in SDEY; there were more POs in RDEY, including 2-ethyl-3,6-dimethylpyrazine, acetoin, 2-acetyl-3-methylthiophene, dihydro-4-hydroxy-2(3H)-furanone, etc. The outcomes obtained have reference values for making better use of duck eggs in the food industry.

Targeted metabolomics and flavouromics combined with relative odor activity value were performed to explore the effect of degradation and oxidation of matrix mediated by pH on the formation of characteristic volatiles in preserved egg yolk (PEY) during pickling. It was found that the oxidation of proteins and lipids in PEY induced by pH sequentially occurred in early and later periods, and degradation both mainly occurred in early stage. Moreover, 1-octen-3-one, heptanal, trimethylamine, etc., compounds and 5-HETrE, proline, etc., components were confirmed as up-regulated characteristic volatiles and differential metabolites in PEY during pickling. The formation of octanal-M/D and benzeneacetaldehyde-M was attributed to ß-oxidation of hydroxyeicosapentaenoic acid and L-isoleucine catalyzed by strong alkali at early period based on correlation network between them, respectively. Meanwhile, the generation of 1-octen-3-one-M/D mainly depended on L-serine and could be promoted by phosphatidylcholines oxidation. At later stage, the formation of heptanal-M/D was primarily attributed to phosphatidylethanolamines oxidation induced by alkali, and the enrichment of heptanal-M/D and nonanal were both enhanced by oxidized lipids. Lastly, trimethylamine was derived from L-lysine under alkaline conditions and promoted by protein oxidation during the whole process. This manuscript provided insight into the differential contribution of oxidation and degradation from matrix regulated by exogenous factors on the formation pathway for characteristic volatiles in foods.

The thermal sterilization process of protein beverages inevitably leads to the formation of insoluble thermal aggregates, greatly reducing the texture and taste of protein beverages. In this study, homogenized egg white (HEW) was obtained by ultrahigh-high-pressure (UHP) homogenization pretreatment of egg white (EW), and then a special egg white fluid gel (EWFG) was prepared by water bath heating. The results showed that the optimal conditions for preparing EWFG were three cycles at 20 MPa homogenizing pressure and heating in a water bath at 72℃ for 10 min. Under these conditions, the EWFG was a milky-white homogeneous liquid with an average particle size of about 560 nm. Measurements of the physicochemical properties of HEW and EWFG showed that the UHP homogenization treatment reduced the viscosity of HEW, decreased the particle size of protein aggregates, and increased the zeta potential, which altered the interactions of proteins during the subsequent heating process and facilitated the formation of homogeneous and dispersed EWFG. EWFG showed good stability at pH 6-10 and in low-concentration saline and medium-concentration sucrose solutions. The EWFG obtained by the present treatment is more suitable for factory-scale production and has great potential for protein beverage applications.

The continuous increasing demand for egg quality and quantity, and the expanding market share have enabled the egg industry to achieve significant benefits through genetic improvement. This study aims to estimate the genetic parameters and explore selectable breeding traits in the purebred Rhode Island Red (RIR) and White Leghorn (WL), which are 2 high-yielding layer breeds, and better understand their underlying genetic basis and accelerate genetic progress. The DMU software was utilized to analyze 12 egg quality traits, including egg length (EL), egg width (EW), egg shape index (ESI), egg weight (EWT), albumen height (AH), yolk color (YC), Haugh unit (HU), yolk weight (YW), albumen weight (AW), albumen-to-egg weight ratio (AWR), yolk-to-albumen ratio (YAR), and yolk-to-egg weight ratio (YWR). In RIR, the heritability of egg quality traits ranged from 0.196 to 0.427, while the repeatability ranged from 0.395 to 0.668. In WL, the heritability of egg quality traits ranged from 0.203 to 0.347, and the repeatability ranged from 0.424 to 0.656. In both RIR and WL, highly strong genetic correlations were observed between AW and EW, as well as between AW and EWT. The genetic correlations for AW and EW were 0.902 in RIR and 0.864 in WL, while the genetic correlations for AW and EWT were 0.981 in RIR and 0.960 in WL. The egg quality traits in both breeds showed moderate heritability, indicating great genetic potential for improvement through selective breeding. This can help breeders meet the increasingly diverse egg preferences of consumers through genetic selection. Additionally, there is a highly strong correlation between egg width/egg weight, and albumen weight in both breeds. In practical production, it is feasible to estimate albumen weight by measuring egg width and egg weight, which can simplify the method for measuring albumen weight. In conclusions, our finding provided valuable insights into the genetic architecture of egg quality traits in RIR and WL chickens. They help our understanding of the potential for genetic improvement of these traits through selective breeding programs.
