RESUMEN

The harmful effects of pesticides can be extended beyond the exposure time scale. Appraisals combining exposure and long-term post-exposure periods appear as an unavoidable approach in pesticide risk assessment, thus allowing a better understanding of the real impact of agrochemicals in non-target organisms. This study aimed to evaluate the progression of genetic damage in somatic and germ tissues of the crayfish Procambarus clarkii, also seeking for gender-specificities, following exposure (7 days) to penoxsulam (23 µg L-1 ) and a post-exposure (70 days) period. The same approach was applied to the model genotoxicant ethyl methanesulfonate (EMS; 5 mg L-1 ) as a complementary mean to improve knowledge on genotoxicity dynamics (induction vs. recovery). Penoxsulam induced DNA damage in all tested tissues, disclosing tissue- and gender-specificities, where females showed to be more vulnerable than males in the gills, while males demonstrated higher susceptibility in what concerns internal organs, that is, hepatopancreas and gonad. Crayfish were unable to recover from the DNA damage induced by EMS in gills and hepatopancreas (both genders) as well as in spermatozoa. The genotoxicity in the hepatopancreas was only perceptible in the post-exposure period. Oxidative DNA lesions were identified in hepatopancreas and spermatozoa of EMS-exposed crayfish. The spermatozoa proved to be the most vulnerable cell type. It became clear that the characterization of the genotoxic hazard of a given agent must integrate a complete set of information, addressing different types of DNA damage, tissue- and gender-specificities, as well as a long-term appraisal of temporal progression of damage.

The knowledge on parental experiences is critical to predict how organisms react to environmental challenges. So, the DNA integrity of Procambarus clarkii spermatozoa exposed ex vivo to the herbicide penoxsulam (Px) or ethyl methanesulfonate (EMS; model genotoxicant) was assessed with and without the influence of in vivo parental exposure to the same agents. The parental exposure alone did not affect the DNA of unexposed spermatozoa. However, the history of Px exposure increased the vulnerability to oxidative lesions in Px-exposed offspring. Otherwise, parental exposure to EMS allowed the development of protection mechanisms expressed when F1 was also exposed to EMS, unveiling life history as a shield. The parental exposure to a different agent adverse and decisively affected Px spermiotoxic potential, pointing out life history as a shadow to progeny. Given the complexity of the aquatic contamination scenarios, involving mixtures, the spermiotoxicity of Px to wild P. clarkii populations emerged as probable.

The spermiotoxic properties of aquatic contaminants might be the cause of low fertilization rate and decreased prolificacy, affecting the success of the impacted populations. The genotoxic potential of pesticides in spermatozoa as an undesirable effect on non-target organisms, namely aquatic invertebrates with external fertilization, emerges as a key question in ecogenotoxicological research. Thus, this study aimed to clarify if DNA integrity of red swamp crayfish (Procambarus clarkii) spermatozoa is affected by waterborne pesticides at environmentally relevant concentrations. By adopting an ex vivo approach, six pesticides were addressed in a short-term assay: herbicides glyphosate (9 and 90 µg L-1) and penoxsulam (2.3 and 23 µg L-1); insecticides dimethoate (2.4 and 24 µg L-1) and imidacloprid (13.1 and 131 µg L-1); fungicides pyrimethanil (2.2 and 22 µg L-1) and imazalil (16 and 160 µg L-1). Genotoxicity was observed in higher concentrations of glyphosate, penoxsulam, dimethoate, pyrimethanil, and imazalil. Imidacloprid was the only pesticide that did not cause non-specific DNA damage, although displaying pro-oxidant properties. Overall, the present study demonstrated the suitability of the ex vivo approach on spermiotoxicity screening, highlighting the potential ecological impact of pesticides on non-target species, such as P. clarkii, compromising sperm DNA integrity and, subsequently, the population success.

In this study, few different evaluation concepts were used for the assessment of genotoxic potential at the stretch of the Danube River identified as a significant hotspot of pollution originated through the untreated wastewaters. Three sites were chosen: one site upstream of the wastewater outlet in Novi Sad (Serbia), one at the outlet of wastewaters, and one site few kilometer downstream. Ex situ approach comprised prokaryotic SOS/umuC test on Salmonella typhimurium TA1535/pSK1005 and comet assay on human hepatoma cell line (HepG2). In situ approach was based on the active monitoring (cage approach) using freshwater mussels Sinanodonta woodiana and fish Cyprinus carpio. The comet and micronucleus assays were selected for evaluation of DNA damage in mussel haemocytes and fish blood cells. Within the ex situ part of the study, our results indicated that the eukaryotic model system is more sensitive compared to the prokaryotic one. In situ bioassays are recommended for obtaining a better insight into ecosystem status and in the case of our study the complete insight of genotoxic pressure. However, the choice of animals as bioindicators also has a significant impact on the quality of the obtained information. Differential response between fish and mussels was observed at the highly polluted site suggesting possible involvement of additional protective mechanism such as valve closure in mussels.

The aim of this study is to provide an analysis of the impact of ecogenotoxicity on native flora abortivity in various urban areas. In which, there was an analysis of 5 groupings of locations with a differing environmental load intensity within the city of Bratislava (Slovakia) over a 2-year period. Our results show varying data depending on the proximity of each site relating to a direct source of pollution and the potential impact of localized wind currents on the distribution of pollutants in the urban environment. The highest value of pollen abortivity in the city was observed in a group of locations exposed to heavy traffic pollution loads. Abortivity of native flora near heavy traffic road areas correlated with the imissions data measured in the same area. Wind-exposed uncovered sites also experienced higher values of native flora abortivity. These results confirmed the varying intensity of genotoxic impact in differing localities and also suggest that xenobiotic effects on flora can occur remotely from the original source of pollution.

The Velika Morava River is the greatest national Serbian river and the significant tributary of the Danube River. The major problems in the Velika Morava River Basin (VMRB) represent untreated industrial and municipal wastewaters. In this study, the level of genotoxic potential at the sites along the VMRB was evaluated by parallel in vitro and in situ approach. Within in vitro testing, genotoxicity of native water samples collected from the sites in VMRB was evaluated by SOS/umuC test on Salmonella typhimurium TA1535/pSK1002 and by the comet assay on HepG2 cells. DNA damage in situ was assessed in bleak (Alburnus alburnus) erythrocytes by the comet (alkaline and Fpg-modified comet) and micronucleus assays. Additionally, the concentration of heavy metals in fish tissue was measured and this data, compiled with the data of the physico-chemical parameters measured in water, was used as a measure of the pollution pressure at the sites. Results showed that applied in vitro tests with native water samples are less sensitive in comparison with in situ tests and should be taken with precaution when making predictions on the status of the ecosystem. Within applied battery of in situ assays differential sensitivity of assays was observed where alkaline comet assay showed the highest potential in differentiation of the sites based on genotoxic potential. Integrated biomarker response showed that usage of the battery of bioassays provides better insight in a genotoxic effects in animals, and consequently, that the holistic approach is more suitable for this type of study.

The aim of the study was to analyze, update, and complete the results of research in the field of in situ phytoindication of environmental genotoxicity near the aluminum plant in Ziar nad Hronom in central Slovakia. The authors focused on two methodologies: pollen abortivity assay of native flora and Trad-MCN assay. Comparison of changes in responses of living systems to changes in precipitation was conducted, and it suggests that there is an existence of an impact from a dilution effect to the plants. Also, the gradual increase of pollen abortivity in the 1990s and its decrease from 2009 were observed in a majority of species of wild flora. On an annual basis, abortivity has declined, although it has risen up slightly within each season. Despite a gradual decrease in the micronucleus frequency, the study area is still influenced by the ecogenotoxic factors. Pollen analysis of native flora and introduced Tradescantia plants indicates the long-time presence of ecogenotoxicity in this region due to the presence of aluminum plant.

Specimens of the mussel Mytilus galloprovincialis were collected from five sites in the Boka Kotorska Bay (Adriatic Sea, Montenegro) during the period summer 2011-autumn 2012. Three types of tissue, haemolymph, digestive gland were used for assessment of DNA damage. Images of randomly selected cells were analyzed with a fluorescence microscope and image analysis by the Comet Assay IV Image-analysis system. Three parameters, viz. tail length, tail intensity and Olive tail moment were analyzed on 4200 nuclei per cell type. We observed variations in the level of DNA damage in mussels collected at different sites, as well as seasonal variations in response. Sum of ranking differences (SRD) was implemented to compare use of different types of cell and different measure of comet tail per nucleus. Numerical scales were transferred into ranks, range scaling between 0 and 1; standardization and normalization were carried out. SRD selected the best (and worst) combinations: tail moment is the best for all data treatment and for all organs; second best is tail length, and intensity ranks third (except for digestive gland). The differences were significant at the 5% level. Whereas gills and haemolymph cells do not differ significantly, cells of the digestive gland are much more suitable to estimate genotoxicity. Variance analysis decomposed the effect of different factors on the SRD values. This unique combination has provided not only the relative importance of factors, but also an overall evaluation: the best evaluation method, the best data pre-treatment, etc., were chosen even for partially contradictory data. The rank transformation is superior to any other way of scaling, which is proven by ordering the SRD values by SRD again, and by cross validation.
