RESUMEN

Proteins of the trefoil factor family (TFF) participate in mucosal repair and are formed by single or tandemly repeated trefoil domains. TFFs have been extensively studied in mammals and amphibians, but they have not been functionally characterized in other animals. Here we report the identification of two genes expressed in the hydroid Hydractinia symbiolongicarpus, predicted to encode trefoil domain-containing peptides, one with four trefoil domains in tandem and the other one with a trefoil domain flanked by two ShKT domains. Differential expression analyses by qPCR after an immune challenge and an induced mechanical damage, reveal that the former gene (hysyTFF) had no significant changes in expression after the inductions. However, the latter (hysyTFF-like) was overexpressed after three hours post immune challenge and was downregulated after the first hour post epithelial damage. Immunoblot analyses using specific IgY antibodies revealed that hysyTFF is secreted as a high molecular weight complex. Finally, whole mount immunofluorescence assays showed that hysyTFF was predominantly expressed in the endoderm of stolons and polyps, and sparsely in the ectoderm of both polyps and larvae. Thus, the tissue distribution and expression dynamics of trefoil factor genes in H. symbiolongicarpus suggest that hysyTFF is part of an ancient mechanism of epithelial restitution, and the newly reported hysyTFF-like might act as an immune effector gene, perhaps encoding an antibacterial peptide.

The present study describes a new species of Henneguya infecting the ornamental fish Caquetaia spectabilis from the Brazilian Amazon. Fish specimens were collected where the Tapajós and Amazon rivers merge, municipality of Santarém in the State of Pará, Brazil. Infections were intense, with several plasmodia spread on the opercula, fins and eye. Phylogenetic characterization and host-parasite relationship studies of the new Henneguya species used a combination of small subunit ribosomal DNA (ssrDNA) and morphological (photonic and transmission electron microscopy) analyses. Plasmodia were white round to ellipsoidal measuring up to 1.8 mm. The myxospores body measured 20.5 ± 3.9 (15-27) in length, 7.9 µm (6.2-10.8) in width, 6.7 µm (6.0-7.6) in thickness, 20.5 µm (14.4-32.3) in caudal appendages length, and 40.6 µm (34.2-54.6) in total length. The two polar capsules were elongated and equal in size, measuring 4.3 µm (3.3-5.4) in length and 2.1 µm (1.3-2.8) in width. Histological analysis revealed the parasite development in connective tissues of the fins, eyes and opercula. The skin of the fins and opercula presented detachment of the epidermis, however, no inflamatory infiltrate was observed. In the eye were observed inflammatory infiltratate in the epithelium and stroma of the cornea. Ultrastructure analysis showed the connective tissue capsule composed by an inner cellular layer with fibroblasts and outer layer where collagen fibers arranged transversely yet interspersed by layers of fibers arranged longitudinally. Numerous invaginations and extensive pinocytotic channels were observed in the plasmodial membrane. A layer of microfilament-like microfilament-like material was observed in the ectoplasm area and along to the internal surface of the plasmodial membrane. Generative cells and early stages of sporogenesis were seen more internally. The ssrDNA based phylogeny showed the South American species grouped in two lineages and the new species arises in a well-sustained subclade as sister branch of the clade composed by Henneguya spp. parasites of cichlids fish.

Myxozoans are a diverse group of parasitic cnidarians of wide distribution. A new species, Myxobolus matogrossoensis n. sp., is herein described infecting wild specimens of tetra mato-grosso Hyphessobrycon eques, caught in the Pantanal biome, the world's largest tropical wetland area. Cysts were found in 3 of the 30 examined fishes. Mature myxospores were ovoid in shape in frontal and measured 6.6 ± 0.4 µm (6.2-7.0 µm) in length and 3.5 ± 0.2 µm (3.3-3.7 µm) in width. The two polar capsules were elongated in shape, equal in size and occupying almost half of the myxospore body. They measured 3.3 ± 0.2 µm (3.1-3.5 µm) in length and 1.8 ± 0.1 µm (1.7-1.9 µm) in width. The polar tubules presented three to four turns. Phylogenetic analysis placed the new species within a clade containing myxobolid species from South American characiforms fish and appears as a close species of Myxobolus piraputangae and Myxobolus umidus. Nevertheless, the sequences of the new species and P. umidus and P. piraputangae have a large genetic divergence of 12 and 12.2% in their 18S rDNA gene, respectively. To the best of our knowledge, this is the first report of a Myxobolus species parasitizing the tetra fish mato-grosso, thus increasing our knowledge of cnidarian myxosporean diversity from South America.

Neuroinflammation is a condition associated with several types of dementia, such as Alzheimer's disease (AD), mainly caused by an inflammatory response to amyloid peptides that induce microglial activation, with subsequent cytokine release. Neuronal caspase-1 from inflammasome and cathepsin B are key enzymes mediating neuroinflammation in AD, therefore, revealing new molecules to modulate these enzymes may be an interesting approach to treat neurodegenerative diseases. In this study, we searched for new caspase-1 and cathepsin B inhibitors from five species of Brazilian marine invertebrates (four cnidarians and one echinoderm). The results show that the extract of the box jellyfish Chiropsalmus quadrumanus inhibits caspase-1. This extract was fractionated, and the products monitored for their inhibitory activity, until the obtention of a pure molecule, which was identified as trigonelline by mass spectrometry. Moreover, four extracts inhibit cathepsin B, and Exaiptasia diaphana was selected for subsequent fractionation and characterization, resulting in the identification of betaine as being responsible for the inhibitory action. Both molecules are already found in marine organisms, however, this is the first study showing a potent inhibitory effect on caspase-1 and cathepsin B activities. Therefore, these new prototypes can be considered for the enzyme inhibition and subsequent control of the neuroinflammation.

PURPOSE: Henneguya Thélohan, 1892 is one of the most species-rich genera of myxosporean parasites and infects fish around the world. The present study describes a new species infecting the gill filaments, fins, and kidneys of Plagioscion squamosissimus (Heckel, 1840), an economically important freshwater fish distributed in watersheds in the north of South America. METHODS: A total of 108 P. squamosissimus specimens were examined from three geographic localities in the Amazon basin: the Lago Grande do Curuai, a marginal lake of the Amazon River; the Tapajós River, in the state of Pará; and the Solimões River, in the state of Amazonas, Brazil. The analyses were based on the myxospore morphology, ribosomal DNA sequencing, phylogeny, prevalence, and geographic distribution of the host and its parasite. RESULTS: Parasite prevalences were 50% in both the Tapajós and Solimões rivers, and 35.4% in the Lago Grande do Curuai. In terms of the site of infection, the prevalence total was 23.1% in the gill filament, 29.6% in the fins, and 1.8% in the kidney. Regarding gender, the prevalence was 59.5% for males, 32.5% for females, and 21.7% for undetermined sex. The specimens found here were both morphologically and molecularly identical regardless of the infected organ and geographic locality, but distinct from all other Henneguya species, revealing that the parasite reported represents a novel species named Henneguya longisporoplasma n. sp. Despite the sampling being carried out in three different geographic localities of the Amazon basin, no population-level genetic variation was observed, even in the typically more variable ITS-1 region, revealing a panmictic population of H. longisporoplasma n. sp. in this large watershed. Maximum likelihood and Bayesian analyses showed the novel Henneguya clustered as a sister branch of the subclade formed of Henneguya that infect fish belonging to the family Cichlidae. CONCLUSIONS: A novel Henneguya species was identified parasitizing P. squamosissimus. The parasite presented wide geographic distribution in the Amazon basin and genetic analyses showed it as revealing a panmictic population.

About forty-two species of Myxobolus have been previously described to parasitize characiform fishes in South America. Here, we described a new myxozoan species, Myxobolus parodontidis n. sp., in the gills of Apareiodon piracicabae (Characiformes, Parodontidae) from the streams of the Middle Paranapanema River basin, Brazil. The proposed new species is supported by a combination of morphological, ultrastructural, and molecular characterization (small subunit ribosomal DNA). Thirteen specimens of A. piracicabae were analyzed and 30.8% was infected by Myxobolus parodontidis n. sp. The myxospores was classified as intralamellar asymmetric type. A few aberrant myxospores with three polar capsules were observed: the spore length and width were the same of normal myxospores, but the polar capsules had smaller sizes. Ultrastructural analysis showed that the plasmodial membrane of Myxobolus parodontidis n. sp. was in direct contact with the host tissue and a connective capsule surrounding the plasmodium was not observed. The phylogenetic analysis showed that the main influence in the clustering of species of myxobolids seems to be related to the phylogenetic relationships established among fish hosts, mainly at the level of family and order. This is the first record of a myxozoan species parasitizing parodontid fish, contributing to the knowledge of the biodiversity of myxozoans from Brazil.

Metazoans with worm-like morphologies across diverse and disparate groups typically demonstrate motility generated by hydrostatic skeletons involving tissue layers (muscles and epithelia). Here we present representative morphological, behavioural and molecular data for parasitic cnidarians (myxozoans) that demonstrate unprecedented variation in form and function, developing as cellular hydrostats. Motile elongate plasmodia characterise a remarkable radiation of species in the genus Ceratomyxa. The vermiform plasmodia inhabit gall bladders of a range of South American freshwater fish and exhibit undulatory motility reminiscent of nematodes but achieved at the cellular level. Collective insights from ultrastructure, confocal and light microscopy along with videos depicting movements highlight key features that we propose explain the unique motility of the plasmodia. These features include cytoskeletal elements (net forming microfilaments and microtubules), a large internal vacuole, a relatively rigid outer glycocalyx and peripherally arranged mitochondria. These constituents provide collective evidence for repurposing of the cnidarian epitheliomuscular cell to support worm-like motility at the cellular level. The apparent restriction of vermiform ceratomyxids to South American freshwaters suggests an origination via Cretaceous or Miocene marine transgressions and subsequent radiation.

The present study describes the morphological, histopathological and SSU rDNA data of a new myxosporean species infecting farmed Astyanax lacustris fish from the state of São Paulo, Brazil. Henneguya lambariensis sp. nov. was found infecting the gills, and the plasmodial development resulted in displacement, blood congestion, compression, deformation and lamellar fusion, stretching of the epithelia, hyperplasia of the epithelial cells, edema, and mild infiltration of the mast cells and lymphocytes. The SSU rDNA sequencing resulted in the sequencing of 1804 nucleotides that did not correspond to any myxosporean sequences deposited in GenBank. The closest phylogenetic affinity of the new species was to the South American Henneguya loreotoensis and Henneguya guanduensis, which also parasite the gills. The present study suggests this new myxosporean species has considerable pathogenic potential, and health monitoring should be implemented in A. lacustris fish farms to ensure production.

Three new sea fan species of Leptogorgia were discovered during multiple scuba diving expeditions along the Gulf of California coast and islands. Leptogorgia iridis sp. nov. is distributed in the southern region of the gulf (Mexican Province), inhabiting tropical rocky reefs of the Islas Marías Archipelago (Nayarit) and Bahía Banderas (Jalisco). This species has small colonies (< 7 cm height) with at least five clearly distinct chromotypes. Leptogorgia martirensis sp. nov. was found exclusively on the rocky reefs of San Pedro Mártir and San Esteban Islands located in the northern region of the Gulf of California (northern region of Cortez Province). Leptogorgia enrici sp. nov. is distributed from the south to the northern region of the Gulf of California (Cortez Province), inhabiting substrates of rocky reefs, sandy and pebbly sea floors. Comprehensive ecological diving expeditions to identify and classify octocorals in the Mexican Pacific (1995-2019) indicate that L. iridis sp. nov. and L. martirensis sp. nov. are likely to be micro-endemics and L. enrici sp. nov. is endemic to the Gulf of California, which defines their currently known biogeographic distribution ranges.

The farming of Colossoma macropomum has intensified in recent years, leading to an increased need for research into the health of the fish. We therefore investigated the diversity of myxosporeans (Cnidaria: Myxozoa) infecting C. macropomum in a breeding system in the municipality of Rio Branco, in the state of Acre, Brazil. Twenty-four fish specimens were examined from June to August 2018. Our results revealed a high prevalence of infection, with 23 specimens (95.8%) exhibiting myxosporean plasmodia. Morphological analysis, based on light and electron microscopies, and molecular analysis (small subunit ribosomal DNA [SSU rDNA] sequencing) revealed the occurrence of three novel species of the genus Myxobolus. Plasmodia of Myxobolus guttae n. sp. were found in the fins of 75% of the specimens, and the myxospores were pear-shaped, measuring 12.3 ± 0.6 (10.3-13.5) µm in length, 8.1 ± 0.3 (7.1-8.6) µm in width, and 5.1 ± 0.6 (4.5-6.5) µm in thickness. The polar capsules were elongated and equal in size, measuring 6.8 ± 0.5 (5.8-7.6) µm in length and 2.5 ± 0.3 (1.8-3.1) µm in width, exhibiting polar tubules with 9-10 coils. The plasmodia of Myxobolus longus n. sp. were found in the gills of 45.8% of the C. macropomum specimens, and the myxospores were fusiform, measuring 16.4 ± 0.6 (14.7-17.3) µm in length, 7.1 ± 0.2 (6.8-7.7) µm in width and 5.5 ± 0.6 (4.4-6.6) µm in thickness. The polar capsules occupied more than half of the myxospore, exhibiting different sizes, the largest measuring 9.2 ± 0.5 (7.3-10.1) µm in length and 2.5 ± 0.3 (2.0-3.1) µm in width, while the smallest measured 8.5 ± 0.4 (7.1-9.1) µm in length and 2.4 ± 0.2 (1.9-3.0) µm in width. Both polar capsules contained polar tubules with 10-12 coils. For Myxobolus tambaquiensis n. sp., plasmodia were found in the opercular cavity of 41.7% of the fish specimens, and the myxospores had an oval shape, measuring 10.0 ± 0.4 (9.5-11.3) µm in length, 6.5 ± 0.2 (6.1-7.1) µm in width, and 4.6 ± 0.4 (3.9-5.6) µm in thickness. The polar capsules were elongated and equal in size, measuring 4.9 ± 0.2 (4.4-5.3) µm in length and 1.9 ± 0.2 (1.5-2.2) µm in width, closing with 8-9 coils of the polar tubule. The morphological and sequencing data of the SSU rDNA showed that the three species studied herein remain unknown to science, increasing the diversity of myxosporeans infecting C. macropomum, an iconic fish in South American freshwater fish farming. The SSU rDNA based phylogenetic analysis revealed that Myxobolus spp. parasites of C. macropomum did not have a monophyletic origin, identifying different times and pathways of the acquisition of parasites by this host species.

A new cnidarian myxosporean infecting the spleen of an economic and ecological important bryconid fish (Salminus franciscanus) is described based on integrative taxonomic approach including morphological, ultrastructural, biological traits, geography, molecular data and phylogenetic analysis. In a total of thirty specimens examined, nineteen (63.3%) were infected by an undescribed parasite species belonging to the genus Myxobolus. Plasmodial development was asynchronous, with young development in the periphery and mature myxospores in the central area and without projections and microvilli in the plasmodial wall. Mature myxospores were ovoid in shape and measured 7.9 ± 0.2 µm (7.6-8.1 µm) in length and 5.4 ± 0.1 µm (5.0-5.6 µm) in width. The two polar capsules were equal in size, occupying a little more than half of the myxospore body, measuring 4.0 ± 0.2 µm (3.9-4.1 µm) in length and 1.7 ± 0.1 µm (1.5-1.8 µm) in width. The polar tubules coiled in six turns, perpendicular to the long axis of polar capsule. Phylogenetic analysis placed the new species within a clade containing nine myxobolid species from South American characiforms fish and appears as a close species of Myxobolus pantanalis. Nevertheless, the sequences of the new species and M. pantanalis have a large genetic divergence of 13.5% in their SSU rDNA. In light of the differences observed from the integrative taxonomy, we confidently considered that this isolate is a new species of cnidarian myxosporean, M. douradae n. sp., increasing the knowledge of diversity of this enigmatic group of cnidarians.

A total of 30 specimens of the Amazonian electric knifefish, Brachyhypopomus beebei Schultz, 1944 (Gymnotiformes: Hypopomidae), were collected from the Peixe-Boi River in the state of Pará, Brazil (1°0659 S; 47°1826 W). Fragments of the brain tissue were extracted for analysis via optical microscopy, and 18 specimens (60%) presented microparasites of the genus Myxobolus, with unequal capsules. The spores were 18.6 µm (17.7-19.8 µm) long and 8.6 µm (8.4-9.0 µm) wide; the largest polar capsule was 13.0 µm (12.4-13.4 µm) long and 5.6 µm (5.3-6.0 µm) wide, and the smallest capsule was 5.0 µm (4.5-5.3 µm) long and 2.5 µm (2.3-2.6 µm) wide. Infected brain fragments were extracted for histological processing and staining with hematoxylin-eosin and Ziehl-Neelsen. Some fragments were conserved in ethanol for molecular genetics analysis. A partial sequence of the 18S DNA gene was obtained from the spores, which did not correspond to any other sequences deposited in GenBank, although it did form a clade with other Myxobolus parasites of the nervous system. The morphological data, together with molecular phylogeny, supported the designation of a new species Myxobolus freitasi n. sp.(AU)


Um total de 30 espécimes do peixe-faca elétrico da Amazônia, Brachyhypopomus beebei Schultz, 1944 (Gymnotiformes: Hypopomidae), foram coletados no rio Peixe-Mani, no estado do Pará, Brasil (1 ° 06'59 S; 47 ° 18 ' 26 W). Fragmentos de tecido cerebral foram extraídos para análise em microscopia óptica, sendo que 18 espécimes (60%) apresentavam microparasitos do gênero Myxobolus, com cápsulas desiguais. Os esporos apresentavam 18,6 µm (17,7-19,8 µm) de comprimento e 8,6 µm (8,4-9,0 µm) de largura; a maior cápsula polar tinha 13,0 µm (12,4-13,4 µm) de comprimento e 5,6 µm (5,3-6,0 µm) de largura, e a menor cápsula tinha 5,0 µm (4,5-5,3 µm) de comprimento e 2,5 µm (2,3-2,6 µm) de largura. Fragmentos cerebrais infectados foram extraídos para processamento histológico e coloração com hematoxilina-eosina e Ziehl-Neelsen. Alguns fragmentos foram conservados em etanol para análise genética molecular. Dos esporos, foi obtida uma sequência parcial do gene 18S do DNA, que não correspondeu a nenhuma outra sequência depositada no GenBank, embora tenha formado um clado com outros parasitas do gênero Myxobolus do sistema nervoso. Abstract A total of 30 specimens of the Amazonian electric knifefish, Brachyhypopomus beebei Schultz, 1944 (Gymnotiformes: Hypopomidae), were collected from the Peixe-Boi River in the state of Pará, Brazil (1°06'59" S; 47°18'26" W). RESUMEN

Morphological analysis and microscopic observations of the Porites baueri holotype and specimens of other Porites species from different sites of the Mexican Pacific, allowed confirmation that Porites baueri is subjective synonym of Porites lobata. The holotype of Porites baueri was collected in the Marías Islands in 1957, and described as a new species by Squires in 1959. Since then, this species was considered as endemic, which is due to the lack of other specimens or a review of the taxonomic characters until now. The Mexican Pacific marine fauna loses a valid reef coral species, currently at 34, but gains in taxonomic clarity by ending almost 60 years of controversy.

RESUMEN

The family Hormathiidae Carlgren, 1932 is one of the largest families of sea anemones with ca. 130 species around the world, mostly in the deep sea. In Brazilian waters, only six species have been reported so far. Herein we record four hormathiids from deep-sea sites at the Potiguar Basin continental slope at Northeast Brazil. Monactis vestita (Gravier, 1918) and Phelliactis robusta Carlgren, 1928 represent the first records of both genera for the Brazilian coast. The new species Paraphelliactis labiata n. sp. is described. We also found another species of the genus, Phelliactis sp. The new species possesses fourth and fifth cycles of incomplete mesenteries, unlike all other Paraphelliactis species. These results increase the total number of hormathiid sea anemones in Brazil to nine and contribute to the knowledge of the Brazilian deep sea, still little explored.

RESUMEN

The diversity of Ceriantharia is known from studies formally describing species from the late 18th Century onwards. However, no nomenclators including a list and discussion of all valid species have been produced since a list discussed by Carlgren in 1912. The present nomenclator presents a complete list of adult species of Ceriantharia of the World, including a discussion on each species. It includes the three families (Arachnactidae, Botrucnidiferidae, Cerianthidae) and the currently accepted 54 species based on their adult form. This study serves as a presentation of the "state-of-the-art" list of species of Ceriantharia, and includes a species identification key to support taxonomic identification. Additional in-depth species-by-species investigations for almost all cerianthid species is still needed, as the information available for most of these species is quite superficial.
