RESUMEN

In our search for peroxisome proliferator-activated receptor (PPAR) agonists, five undescribed compounds, namely two acyclic diterpenes (1 and 2; cladopsol A and cladopsol B), two sesquiterpenes (3 and 4; cladopsol C and cladopsol D), and one C21-ecdysteroid (5; cladopsol E), and 15 known compounds were isolated from the jellyfish-derived fungus - Cladosporium oxysporum. The structures of the undescribed compounds were defined using UV, NMR, HR-ESI-MS, and electronic circular dichroism (ECD) spectroscopy and a modified Mosher's method. Luciferase reporter assay and docking analysis suggested that cladopsol B may function as a PPAR-γ partial agonist with a potential antidiabetic lead which may evade the side effects of full agonists. Moreover, cladopsol B stimulated glucose uptake in HepG2 cells with an efficacy comparable to that of rosiglitazone, but with less side effect induced by lipid accumulation in 3T3-L1 cells. Therefore, cladopsol B could serve as a molecular skeleton in a study of advanced antidiabetic lead with less side effect.

Melampsora medusae f. sp. deltoidae is causing serious foliar rust disease on Populus deltoides clones in India. In the present study, a novel fungal hyperparasite on M. medusae has been reported. The hyperparasitic fungus was isolated from the uredeniospores of the rust fungi and identified as Cladosporium oxysporum by morphological characterization and DNA barcode technique based on the Internal Transcribed Spacer (ITS) region of nrDNA and beta-tubulin (TUB) gene region. Hyperparasitism was further confirmed through leaf assay and cavity slide methods. Leaf assay method showed no adverse effect of C. oxysporum on poplar leaves. However, the mean germination percentage of urediniospores was significantly decreased (p < 0.05) in the cavity slide method when a conidial suspension (1.5 × 107 conidia per ml) of C. oxysporum was applied in different deposition sequences. Scanning and light microscopic observations were made to explore the mode of action of the hyperparasitism. The antagonistic fungus vividly showed three different types of antagonism mechanisms, including enzymatic, direct, and contact parasitism. Alternatively, by screening 25 high-yielding clones of P. deltoides, five clones (FRI-FS-83, FRI-FS-92, FRI-FS-140, FRI-AM-111, and D-121) were enlisted under highly resistant category. Present study revealed an antagonistic relationship between C. oxysporum and M. medusae, which could be an effective method of biocontrol in field plantations of poplar. Combining this biocontrol approach with the use of resistant host germplasm could be an environment friendly strategy for preventing foliar rust and increasing poplar productivity in northern India. RESUMEN

The Cladosporium fungi, one of the largest genera of dematiaceous hyphomycetes, could produce various bioactive secondary metabolites. From the AcOEt-soluble extract of Cladosporium oxysporum 170103, three new secopatulolides (1-3) and thirteen known compounds (4-16) were obtained. Their structures were established by detailed analysis of the NMR and HR-ESI-MS data. All sixteen compounds were tested for antibacterial activity against Vibrio parahemolyticus, ergosterol (10) presented moderate effect with the minimum inhibitory concentration (MIC) of 32 µM. It can destruct the membrane integrity of Vibrio parahemolyticus to change the cell shape.

Five undescribed 12-membered macrolides containing thioethers, thiocladospolides F-J, together with the known biogenetic related analogues, were isolated from a mangrove-derived endophytic fungus Cladosporium oxysporum. The absolute configuration of thiocladospolide F was deduced by single-crystal X-ray diffraction, and those of thiocladospolides G-J were determined based on specific optical rotation and electronic circular dichroism (ECD) data, as well as the biogenetic considerations. Thiocladospolide G display the best antimicrobial activity against the aquatic pathogen Edwardsiella tarda with minimal inhibit concentration (MIC) value of 4 µg/mL.

The genus Cladosporium is one of the largest genera of dematiaceous hyphomycetes, and is characterised by a coronate scar structure, conidia in acropetal chains and Davidiella teleomorphs. Based on morphology and DNA phylogeny, the species complexes of C. herbarum and C. sphaerospermum have been resolved, resulting in the elucidation of numerous new taxa. In the present study, more than 200 isolates belonging to the C. cladosporioides complex were examined and phylogenetically analysed on the basis of DNA sequences of the nuclear ribosomal RNA gene operon, including the internal transcribed spacer regions ITS1 and ITS2, the 5.8S nrDNA, as well as partial actin and translation elongation factor 1-α gene sequences. For the saprobic, widely distributed species Cladosporium cladosporioides, both a neotype and epitype are designated in order to specify a well established circumscription and concept of this species. Cladosporium tenuissimum and C. oxysporum, two saprobes abundant in the tropics, are epitypified and shown to be allied to, but distinct from C. cladosporioides. Twenty-two species are newly described on the basis of phylogenetic characters and cryptic morphological differences. The most important phenotypic characters for distinguishing species within the C. cladosporioides complex, which represents a monophyletic subclade within the genus, are shape, width, length, septation and surface ornamentation of conidia and conidiophores; length and branching patterns of conidial chains and hyphal shape, width and arrangement. Many of the treated species, e.g., C. acalyphae, C. angustisporum, C. australiense, C. basiinflatum, C. chalastosporoides, C. colocasiae, C. cucumerinum, C. exasperatum, C. exile, C. flabelliforme, C. gamsianum, and C. globisporum are currently known only from specific hosts, or have a restricted geographical distribution. A key to all species recognised within the C. cladosporioides complex is provided.
