Due to their selective toxicity to insects, nicotinoid compounds have been widely used to control pests in crops and livestock around the world. However, despite the advantages presented, much has been discussed about their harmful effects on exposed organisms, either directly or indirectly, with regards to endocrine disruption. This study aimed to evaluate the lethal and sublethal effects of imidacloprid (IMD) and abamectin (ABA) formulations, separately and combined, on zebrafish (Danio rerio) embryos at different developmental stages. For this, Fish Embryo Toxicity (FET) tests were carried out, exposing two hours post-fertilization (hpf) zebrafish to 96 hours of treatments with five different concentrations of abamectin (0.5-11.7 mg L-1), imidacloprid (0.0001-1.0 mg L-1), and imidacloprid/abamectin mixtures (LC50/2 - LC50/1000). The results showed that IMD and ABA caused toxic effects in zebrafish embryos. Significant effects were observed regarding egg coagulation, pericardial edema, and lack of larvae hatching. However, unlike ABA, the IMD dose-response curve for mortality had a bell curve display, where medium doses caused more mortality than higher and lower doses. These data demonstrate the toxic influence of sublethal IMD and ABA concentrations on zebrafish, suggesting that these compounds should be listed for river and reservoir water-quality monitoring.

The indiscriminate use of pesticides is a worldwide concern due to the environment contamination since it can cause deleterious effects to non-target organisms including the fishes. The effects of abamectin, a pesticide from the avermectin family, were evaluated in adult zebrafish (Danio rerio) after exposure to a commercial formula commonly used in Brazil. The animals were submitted to acute (96 h) and to a short-term chronic exposure (15 days) of distinct concentrations of abamectin. LC50 was determined and a histological study followed by an immunohistochemistry analysis for P-gp and HSP70 identification were performed on livers of the animals submitted to the acute and chronic treatment, respectively. Moreover, behavior patterns were observed daily in both trials. A LC50 value of 105.68 µg/L was determined. The histological analysis revealed a morphological alteration of the hepatocytes, glycogen accumulation, degeneration, and disorganization of the cytoplasm, and a pyknotic, irregular, and laterally located nuclei. The immunohistochemistry for HSP70 and P-gp showed strong staining in the hepatocytes of the control groups and progressive decrease as the concentration of abamectin increased. Changes were observed in body posture, movement around the aquarium, opercular activity, body color and search for food in the groups treated with abamectin. The results presented suggest that abamectin can affect the behavioral pattern of the animals, promote morphological changes, and decrease the expression of HSP70 and P-gp in zebrafish liver.

In spite of killing the targeted pests, such insecticides also effect growth of predatory mites. Present study has been conducted to evaluate the impact of two acaricides, abamectin and hexythiazox, on seven species of predatory mites. Standard solutions of abamectin (8.4% w/v) and hexythiazox (10% w/w) were purchased and prepared for direct spray. Acaricides were tested at three serial concentrations of recommended dose (RD), half of the recommended dose (HRD) and double the recommended dose (DRD). Trials were made on eggs, nymphs and adults of the seven species of predatory mites under laboratory conditions. Effect of the applied acaricides was determined one week after the application. The results of the study revealed that all doses of hexythiazox and abamectin are toxic to the nymphs and adults of all seven species of predatory mites but to the variable extent. Furthermore, it was observed that hexythiazox and abamectin reduced the percentage of egg hatching for all seven species of predatory mites. Hexythiazox was found to be more toxic than abamectin to all seven predatory mites. Higher mortality was recorded at DRD and RD, while minimum mortality was recorded at HRD. Acaricidal effect is less severe on hatching eggs than on immatures and adults of the predatory mites. Species vary in susceptibility to acaricides and in some populations resistance has been observed. Based upon this study, it is recommended that the frequent use of acaricides against phytophagous mites should be avoided and feasibility of biological control programs should be promoted to protect the environment, health of living individuals and the non-target organisms.

RESUMEN
Several different tick species are known to infest horses. Aside from causing serious health and welfare issues, including anaemia, ill thrift, and immunosuppression, ticks can transmit a variety of important, sometimes zoonotic, pathogens. The successful prevention and treatment of tick infestations have been described, but the information is scarce and, in many instances, anecdotal. Here we describe a practical and affordable prevention of tick infestation by using abamectin-impregnated cattle ear tags affixed to a safety collar. We have assessed the radial distribution of abamectin by analyzing hair samples, as well as its efficacy against tick infestations. The study results show that abamectin distributes across horse skin from the site of application and its associated effectiveness in reducing the tick burden.

Resumen El impacto productivo de los nematodos gastrointestinales (NGI) sobre los de pequeños rumiantes es conocido a nivel mundial. Las infestaciones por NGI constituyen una limitante de alto impacto en la producción de rumiantes y las perdidas comprendidas por este problema sanitario varían desde reducciones subclínicas en la ganancia de peso hasta la muerte de animales con altas cargas parasitarias. El objetivo de este trabajo es presentar los resultados de un test de eficacia a campo utilizando una combinación comercial de Abamectina + Derquantel y del Moxidectin en cabras lecheras naturalmente infestadas con NGI en el Valle de Lerma, provincia de Salta. El ensayo se llevó a cabo en la Unidad Tambo Caprino de la Estación Experimental Agropecuaria Salta del INTA. Para la evaluación de eficacia de la droga en estudio, se realizó el test de reducción de conteos de huevos. En los coprocultivos pre-tratamientos se pudo observar una gran prevalencia de Trichostrongylus spp. (71%). seguido de Teladorsagia spp. (21%) y Haemonchus spp. (8%). En cuanto a los resultados del TRCH, en el GDQ se observaron valores del 100% de PRCH, mientras que en el GMX se observó un PRCH de 95,5 %, donde la eficacia para Haemonchus spp. y Teladorsagiaspp. fue del 100% y para Trichostrongylus spp. del 81,1%.Los resultados observados en este trabajo permiten concluir que en las condiciones de campo en donde fue llevado a cabo el TRCH, la combinación Derquantel-Abamectina posee una alta eficacia contra los géneros presentes.
Abstract The productive impact of gastrointestinal nematodes (NGI) of small ruminants are known worldwide. Infestations by NGI constitute a limiting factor in the production of ruminant and the losses included by this sanitary problem vary from subclinical reductions in weight gain to the death of animals with high parasite loads. The objective of this work is to present the results of an efficacy trial using a commercial combination of Abamectin + Derquantel and Moxidectin in dairy goats naturally infested with NGI in the Lerma Valley, Salta province. The test was carried out in the Dairy Goat Unit of the Salta Agricultural Experiment Station of INTA. In the pretreatment stool cultures, a high prevalence of Trichostrongylusspp. (71%). followed by Teladorsagiaspp. (21%) and Haemonchus spp. (8%) was observed. Regarding the results of the TRCH, in the GDQ values of 100% of PRCH were observed, while in the GMX a PRCH of 95.5% is shown, where the efficacy for Haemonchus spp. and Teladorsagia spp. was 100% and for Trichostrongylus spp. 81.1%. The results observed in this work allow us to conclude that under the field conditions where TRCH was carried out, the Derquantel-Abamectin combination has high efficacy against the genera present.

As a therapy, we have opted for a treatment based on natural antioxidants ginger extract (Zingiber officinale) mixed with olive oil to see the restorative effects during the experiment. The study was conducted on female Wistar rats. The latter was given an inhalation exposure of abamectin (ABM) at a dose CL50 > 5.04mg/L/1hr each morning for one week. The effects of this exposure showed a drop in body weight gain (g), a significant decrease in the relative and absolute weights liver (gm/100 gm body weight) accompanied by hepatic impairment indicating a significant disturbance of transaminases (AST/ALT), alkaline phosphatase (ALP) activity and significant behavioral changes. Administration of ginger (Zingiber officinale) extract at a dose of 3 ml mixed with olive oil at a dose of 1 ml/kg for 7 days orally (gavages) after inhalation indicates that ginger has a more remarkable and rapid effect on physical and mental health, it acts as a major means of defense and hepato-protective against toxicity.

An approach for the diagnosis of an abamectin outbreak in calves in the field is described and discussed. In a Midwestern Brazilian property, nine out of a 52 newborn calves were affected and died, making up for morbidity, mortality, and lethality ratios of 17.3%, 17.3%, and 100%, respectively. Major clinical signs included tremors in various muscle groups, inability to stand, and difficult, wheezing breathing. Each affected calf had been treated subcutaneously with abamectin (0.4mg/kg/body weight). No lesions were found at necropsy or at histological examination. Major diseases of newborn calves were included in the differential diagnosis.(AU)
Uma abordagem para o diagnóstico de um surto de abamectina em bezerros a campo é descrita e discutida. Numa propriedade do Centro-Oeste brasileiro, nove de um lote de 52 bezerros de 3 dias de idade foram afetados e morreram, perfazendo quocientes de morbidade, mortalidade e letalidade, respectivamente, de 17,3%, 17,3% e 100%. Os principais sinais clínicos incluíam tremores em vários grupos musculares, incapacidade em se manter em pé, e respiração difícil e estertorosa. An approach for the diagnosis of an abamectin outbreak in calves in the field is described and discussed. In a Midwestern Brazilian property, nine out of a 52 newborn calves were affected and died, making up for morbidity, mortality, and lethality ratios of 17.3%, 17.3%, and 100%, respectively. Major clinical signs included tremors in various muscle groups, inability to stand, and difficult, wheezing breathing. Each affected calf had been treated subcutaneously with abamectin (0.4mg/kg/body weight). No lesions were found at necropsy or at histological examination. Major diseases of newborn calves were included in the differential diagnosis.(AU)
Uma abordagem para o diagnóstico de um surto de abamectina em bezerros a campo é descrita e discutida. Numa propriedade do Centro-Oeste brasileiro, nove de um lote de 52 bezerros de 3 dias de idade foram afetados e morreram, perfazendo quocientes de morbidade, mortalidade e letalidade, respectivamente, de 17,3%, 17,3% e 100%. Os principais sinais clínicos incluíam tremores em vários grupos musculares, incapacidade em se manter em pé, e respiração difícil e estertorosa. The combination of synthetic anthelmintics and bioactive phytochemicals may be a pharmacological tool for improving nematode control in livestock. Carvone (R-CNE) has shown in vitro activity against gastrointestinal nematodes; however, the anthelmintic effect of bioactive phytochemicals either alone or combined with synthetic drugs has been little explored in vivo. Here, the pharmacological interaction of abamectin (ABM) and R-CNE was assessed in vitro and in vivo. The efficacy of this combination was evaluated in lambs naturally infected with resistant gastrointestinal nematodes. Additionally, the ligand and molecular docking of both molecules to P-glycoprotein (P-gp) was studied in silico. The presence of R-CNE produced a significant (p < 0.05) increase of Rho123 and ABM accumulation in the intestinal explants. After 60 min of incubation, Rho123 incubated with R-CNE had a 67 ± 21% higher concentration (p < 0.01) than when it was incubated alone. In the case of ABM, a significant increase in the intestinal concentrations was observed at 15 and 30 min after incubation with R-CNE. In the in vivo assay, no undesirable effects were observed after the oral administration of R-CNE. The coadministration of the natural compound prolonged ABM absorption in lambs. ABM T ½ absorption was 1.57-fold longer (p < 0.05) in the coadministered group. Concentrations of R-CNE between 420 and 2,593 ng/mL were detected in the bloodstream between 1 and 48 h posttreatment. The in vivo efficacy of ABM against gastrointestinal nematodes increased from 94.9 to 99.8% in the presence of R-CNE, with the lower confidence interval limit being >90%. In vitro/in vivo pharmacoparasitological studies are relevant for the knowledge of the interactions and the efficacy of bioactive natural products combined with synthetic anthelmintics. While ADMET (absorption, distribution, metabolism, excretion, and toxicity) predictions and the molecular docking study showed a good interaction between ABM and P-gp, R-CNE does not appear to modulate this efflux protein. Therefore, the pharmacokinetic-pharmacodynamic effect of R-CNE on ABM should be attributed to its effect on membrane permeability. The development of pharmacology-based information is critical for the design of successful strategies for the parasite control.

Abstract The biological control used for the control of Tetranychus urticae (Koch) is the predator mite Phytoseiulus persimilis (Athias-Henriot). It is important to the know the effects of acaricides on the biological behavior the Abamectin on the functional response of P. persimilis. The functional response of the predator was of type II exposed to concentration of Abamectin, the functional response parameters: successful attack rate (a'), handling time (Th), search efficiency and the maximum predation theory (T/Th) were affected by the acaricide. The predator spends more time in persecute, dominate, consume and prepair it self to the next searching comparing with the proof subject an the predation ability was affected.
Resumo O controle biológico utilizado para o controle de Tetranychus urticae (Koch) é o acaro predador Phytoseiulus persimilis (Athias-Henriot). É importante conhecer os efeitos dos acaricidas sobre o comportamento biológico do predador. Foi avaliado o efeito tóxico de a Abamectina na resposta funcional de P. persimilis. A resposta funcional do predador foi tipo II exposta a concentrações subletais de Abacmetina, os parâmetros da resposta funcional: taxa de ataque (a'), tempo de manipulação (Th), a eficiência na procura e predação teórica máxima (T/Th) foram afetados pelo acaricida. O predador passa mais tempo na procura, dominar, consumir e se preparar para a próxima procura em comparação com a testemunha e sua capacidade de predação foi afetada.

The aim of the current study was to evaluate the in vivo pharmacokinetic of ivermectin (IVM) after the administration of a long-acting (LA) formulation to sheep and its impact on potential drug-drug interactions. The work included the evaluation of the comparative plasma profiles of IVM administered at a single therapeutic dose (200 µg/kg) and as LA formulation at 630 µg/kg. Additionally, IVM was measured in different gastrointestinal tissues at 15 days posttreatment with both IVM formulations. The impact of the long-lasting and enhanced IVM exposure on the disposition kinetics of abamectin (ABM) was also assessed. Plasma (IVM and ABM) and gastrointestinal (IVM) concentrations were analyzed by HPLC with fluorescent detection. In plasma, the calculated Cmax and AUC0-t values of the IVM-LA formulation were 1.47- and 3.35-fold higher compared with IVM 1% formulation, respectively. The T1/2ab and Tmax collected after administration of the LA formulation were 2- and 3.5-fold longer than those observed after administration of IVM 1% formulation, respectively. Significantly higher IVM concentrations were measured in the intestine mucosal tissues and luminal contents with the LA formulation, and in the liver, the increase was 7-fold higher than conventional formulation. There was no drug interaction between IVM and ABM after the single administration of ABM at 15 days post-administration of the IVM LA formulation. The characterization of the kinetic behavior of the LA formulation to sheep and its potential influence on drug-drug interactions is a further contribution to the field.

In recent years, the need for the development of alternative test methods for the conventional acute fish toxicity test (AFT) with adult fish has often been discussed. In addition, concerns have been raised on the potential risks related with environmentally realistic pesticide mixtures since risk evaluations have traditionally been based on individual pesticides. The insecticide/acaricide abamectin and the fungicide difenoconazole are the main pesticides that are intensively used in Brazilian strawberry crop and are hence likely to occur simultaneously in edge-of-field waterbodies. The aim of the present study was therefore to evaluate the lethal and sublethal toxicity of single and mixture exposures of these pesticides to zebrafish early life stages (embryos and juveniles). By comparing the derived toxicity data of the individual compounds with that previously determined for zebrafish adults, the order of life stage sensitivity was juvenileâ¯>â¯adultâ¯>â¯embryo. The pesticide mixture revealed a dose-level dependent deviation of the independent action model, with antagonism at low dose levels and synergism at high dose levels. Sublethal parameters (especially those related with locomotion) were considerably more sensitive than lethality. Subsequently, the inclusion of sublethal parameters may greatly improve the sensitivity of FET tests and hence its suitability as a substitution of adult fish testing in risk assessment evaluations.

The present study was designed to evaluate possible adverse effects of different dosages of avermectins (abamectin and a combination of ivermectin + abamectin) administered subcutaneously in calves less than one month of age. Clinical as well as biochemical parameters of blood and cerebrospinal fluid (CSF) were evaluated for animals subjected to various treatments. Thirty-five calves were divided into seven groups of five animals each: T01 (control), T02 (abamectin 200⯵g/kg), T03 (abamectin 400⯵g/kg), T04 (abamectin 600⯵g/kg), T05 (ivermectin 450⯵g/kgâ¯+â¯abamectin 250⯵g/kg), T06 (ivermectin 900⯵g/kgâ¯+â¯abamectin 500⯵g/kg) and T07 (ivermectin 1350⯵g/kgâ¯+â¯abamectin 750⯵g/kg). Cerebrospinal fluid and blood samples were collected on day zero (treatment day) and on days 1, 7 and 14 following treatment. The biochemical parameters measured in blood samples were total protein and the AST, GGT and alkaline phosphatase enzymes. In CSF samples, color, aspects, pH, density, cellularity, total proteins, glucose and the CK, ALT and LDH enzymes were evaluated. No neurological toxicity or biochemical changes in the CSF or blood that could be correlated to administration of tested products were observed in this study. Though it is well known that abamectin may lead to intoxication in bovines less than four months of age, based on data from the present study, no adverse clinical effects to the CSF or blood were observed in calves <30â¯days old that had received up to 600⯵g/kg abamectin formulations and up to 1350⯵g/kg ivermectin +750⯵g/kg abamectin associations. It is possible that the level and activity of the multiple drug resistance gene (MDR1 - ABCB1) present in the calves may have influenced the results obtained in this study. Future studies should be carried out.
