RESUMEN

The fetal period is a critical stage in brain development, and understanding the characteristics of the fetal brain is crucial. Although some studies have explored aspects of fetal brain functional networks, few have specifically focused on sex differences in brain network characteristics. We adopted the graph theory method to calculate brain network functional connectivity and topology properties (including global and nodal properties), and further compared the differences in these parameters between male and female fetuses. We found that male fetuses showed an increased clustering coefficient and local efficiency than female fetuses, but no significant group differences concerning other graph parameters and the functional connectivity matrix. Our study suggests the existence of sex-related distinctions in the topological properties of the brain network at the fetal stage of development and demonstrates an increase in brain network separation in male fetuses compared with female fetuses.

China has a great wealth of lake resources over a great spatial extent and these lakes are highly sensitive to climate changes through their heat and water budgets. However, little is known about the changes in lake surface water temperature (LSWT) across China under the climate warming conditions over the past few decades. In this study, MODIS land surface temperature (LST) data were used to examine the spatial and temporal (diurnal, intra-annual, and inter-annual) variations in LSWT of China's lakes during 2001-2016. Our results indicated that 169 large lakes included in the study exhibited an overall increasing trend in LSWT, with an average rate of 0.26 °C/decade. The increasing rate of nighttime LSWT is 0.31 °C/decade, faster than that of daytime temperature (0.21 °C/decade). Overall, 121 (71.6%) lakes showed an increase in daytime temperature with a mean rate of 0.38 °C/decade, while the rest 48 (28.4%) lakes decreased in temperature with a mean rate of - 0.21 °C/decade. We also quantitatively analyzed the relationship of the lake surface temperature and diurnal temperature differences (DTDs) with geographical location, topography, and lake morphometry by utilizing multivariate regression analysis. Our analysis suggested that the geographical location (latitude and longitude) and topography (altitude) were primary driving factors in explaining the national lake water temperature variation (P < 0.001), which were also mediated by morphometric factors such as lake surface area and volume. Moreover, the diurnal lake temperature variations were significantly correlated with altitude, latitude, and lake surface area (R2 = 0.426, P < 0.001). Correlation analyses of LSWT trend and air temperature trend for each lake indicated that LSWT was positively correlated with air temperature in both daytime and nighttime for most lakes.

INTRODUCTION: Diabetic retinopathy (DR) is a damaging complication of the eye. Studies investigating molecular mechanisms of DR are lacking, leading to poor clinical outcomes. miR-20b-5p is up-regulated in DR. The present study aimed to confirm the involvement of miR-20b-5p in DR and the mechanism involved. METHODS: Microarray analysis was done to study the differentially expressed miRs. DR model was established using Sprague-Dawley rats, the expression of miR-20b-5p was altered using inhibitor or mimic as treatment. THBS1 was one of the potential genes identified by microarray bioinformatics analysis associated with DR. The expression of THBS1 was suppressed by siRNA to study the mechanism behind involvement of miR-20b-5p in DR. In addition, the levels of miR-20b-5p VEGF/PI3K/Akt pathway associated genes were studied. Correlation between THBS1 and miR-20b-5p was evaluated. Cell apoptosis, growth and tube formation assay was performed. RESULTS: The retinal tissues of DR rats showed over-expressed miR-20b-5p and decreased THBS1 via VEGF/PI3K/Akt cascade. THBS1 was confirmed as the target gene of miR-20b-5p by dual-luciferase reporter gene assay. Upregulation of miR-20b-5p or knockdown of THBS1 caused increased tube formation and cell proliferation, whereas it blocked the cell apoptosis of endothelial cells in rats. CONCLUSION: The outcomes suggested that silencing of miR-20b-5p resulted in inhibition of tube formation and cell growth in vascular endothelial cells of rats subjected to DR altering the VEGF/PI3K/Akt cascade by up-regulation of THBS1.

BACKGROUND: Long noncoding RNA (lncRNA) has been reported to play important roles in tumor initiation. However, how lncRNA ELFN1-AS1 affects retinoblastoma development remains unclear. Thus, we sought to elucidate its functions in retinoblastoma progression. METHODS: ELFN1-AS1 expression was measured in retinoblastoma tissues and normal tissues by qRT-PCR. CCK8, colony formation and Transwell assay were carried out to investigate the effects of ELFN1-AS1 knockdown on cell malignant behaviors. Bioinformatics analyses were performed to predict the relationship among ELFN1-AS1, miR-4270 and SBK1. RESULTS: ELFN1-AS1 was highly expressed in retinoblastoma tissues and cell lines. ELFN1-AS1 was positively correlated with retinoblastoma progression and prognosis. ELFN1-AS1 knockdown curtailed retinoblastoma proliferation, migration and invasion. ELFN1-AS1 was the competing endogenous RNA for miR-4270 and promoted SBK1expression. CONCLUSION: Altogether, our findings demonstrated that ELFN1-AS1 promotes retinoblastoma progression through mediating miR-4270/SBK1 axis and might be a promising therapeutic target.

Retinoblastoma (RB) is one of the most common cancer in children. However, the specific mechanism about RB tumorigenesis has not been fully understood. In this study, to comprehensively characterize the splicing alterations in the tumorigenesis of RB, we analyzed the differential alternative splicing events in RB. Specifically, the isoforms of RB1 were downregulated in the RB samples, and a large proportion of differentially expressed genes had multiple differentially expressed transcripts (64%). We identified 1453 genes with differential alternative splicing, among which, SE accounted for the majority, followed by MXE, RI, A3SS, and A5SS. Furthermore, the biological function related to the normal function of eyes, and E2F family TFs were significantly enriched by the genes with differential alternative splicing. Among the genes associated with visual sense, ABCA4 was found to have two mutually exclusive exons, resulting in two isoforms with different functionalities. Notably, DAZAP1 was identified as one of the critical splicing factors in RB, which was potentially involved in E2F and RB pathways. Functionally, differential binding sites in DAZAP1 protein were significantly observed between RB and normal samples. Based on the comprehensive analysis of the differential alternative splicing events and splicing factors, we identified some driver genes with differential alternative splicing and critical splicing factors involved in RB, which would greatly improve our understanding of the alternative splicing process in the tumorigenesis of RB.
