RESUMEN

Human pluripotent stem cell-derived ß cells (hPSC-ß cells) show the potential to restore euglycemia. However, the immature functionality of hPSC-ß cells has limited their efficacy in application. Here, by deciphering the continuous maturation process of hPSC-ß cells post transplantation via single-cell RNA sequencing (scRNA-seq) and single-cell assay for transposase-accessible chromatin sequencing (scATAC-seq), we show that functional maturation of hPSC-ß cells is an orderly multistep process during which cells sequentially undergo metabolic adaption, removal of negative regulators of cell function, and establishment of a more specialized transcriptome and epigenome. Importantly, remodeling lipid metabolism, especially downregulating the metabolic activity of ceramides, the central hub of sphingolipid metabolism, is critical for ß cell maturation. Limiting intracellular accumulation of ceramides in hPSC-ß cells remarkably enhanced their function, as indicated by improvements in insulin processing and glucose-stimulated insulin secretion. In summary, our findings provide insights into the maturation of human pancreatic ß cells and highlight the importance of ceramide homeostasis in function acquisition.

RESUMEN

The preparation of water-based adhesives with high bonding strength for various substrates is challenging. Moreover, to construct a sustainable society, it is highly desirable to develop a cost-effective way to achieve the reuse of plastic wastes. Herein, using polymethylmethacrylate (PMMA) chemicals or wastes as raw materials, water-based adhesives with high bonding strength for various substrates are prepared through a simple one-step hydrolysis strategy. The adhesives possess the maximum bonding strength of 7.1 MPa to iron, 4.2 MPa to wood, and ∼1.5 MPa to plastics. The adhesives have a world-record bonding strength to metal when compared with that of current reported water-based adhesives. Our method is low cost, simple, environmentally friendly, and suitable for large-scale industrial production. More importantly, using plastic wastes as raw materials opens up a new and low-cost way to turn wastes into valuables, which will greatly contribute to construct a sustainable society.

Vulvar lichen sclerosus (VLS) is a chronic inflammatory skin disease that brings life-long and psychological distress to patients. It remains unclear whether this condition is related to changes in the skin microbial community. The aim of this study was to evaluate the compositional characteristics of the vulvar skin microbiota between VLS patients and asymptomatic postmenopausal women. We included 60 cases of postmenopausal patients in the outpatient vulvar clinic of Peking University First Hospital from August 2020 to October 2020. Thirty-one patients were diagnosed with VLS by vulvar skin biopsy (VLS group), while 29 women were asymptomatic volunteers (control group). DNA was extracted from vulvar skin swabs of the VLS and control groups. The V3-V4 fragments of 16S rRNA were targeted for high-throughput sequencing and gene sequence analysis. The sequencing results were analysed by α diversity, ß diversity, species composition, LEfSe analysis to compare the compositional differences of the vulvar skin microbiota between the two groups. Our study revealed that at the phylum level, patients with VLS had a lower relative abundance of Firmicutes (p < 0.0001) and a higher relative abundance of Proteobacteria than the control group (p < 0.0001). At the genus level, Lactobacillus spp. accounted for the largest proportion of the microflora in the asymptomatic controls, while the proportion of Prevotella spp. in the VLS group was the highest. In the VLS group, the relative abundance of Finegoldia spp., Ralstonia spp., Peptoniphilus spp., Anaerococcus spp., Campylobacter spp., Providencia spp. Kelbsiella spp., Ezakiella spp., and Escherichia-Shigella spp. was significantly increased compared with the control group. Although there was no significant difference in the α diversity of the vulvar skin microbiota, the ß diversity differed significantly between the two groups.

RESUMEN

A reusable magnetic-quantum dot material (MNP-SiO2-QD) with good magnetic properties and high fluorescence retention was successfully fabricated from linked magnetic nanoparticles and quantum dots. The resulting material can qualitatively and quantitatively detect four kinds of antibiotics and maintain high recovery rates.
