BACKGROUND:The discovery of novel HLA aleles is accelerated by development of molecular biology and applications of numerous new technologies. These new findings not only rich HLA family, but also find a breakthrough point for the study of genetic superiority or disappearance of national gene. OBJECTIVE:To identify two novel HLA-A aleles and to analysis their nucleotide sequences. METHODS:The two samples from two volunteers of Chinese Marrow Donor Program were detected using PCR-SBT and GSSP methods. The HLA-A locus in the two samples were both abnormal genes, and the nucleotide sequence differences were analyzed. RESULTS AND CONCLUSION: The sequences of the samples were different from al aleles in the HLA databases. Sample 1 was found to be different from the closet matching alele A*24:02:01 in one nucleotide substitutions, 360 G > C in exon 3, resulting in amino acid changed from glutamine to histidine at codon 96; and sample 2 differed from A*26:01:01 in one nucleotide substitution, 97 T > C in exon 2, resulting in amino acid changed from tyrosine to histidine at codon 9. The novel aleles were identified and assigned the name HLA-A*24:233 and HLA-A*26:89 officialy by the WHO Nomenclature Committee.

BACKGROUND: A lot of work has been done to study the association between human leukocyte antigen (HLA) and papillary thyroid carcinoma (PTC) in various populations. But the results of the currently available studies are not consistent. The aim of this study was to evaluate the association of HLA-A, -B, -C, -DRB1, and -DQB1 with PTC in the Chinese Han population of the coastal areas of Shandong Province with respect to age and sex. METHODS: A total of 154 patients diagnosed with PTC were analyzed for HLA-A, -B, -C, -DRB1, and -DQB1 alleles by using a polymerase chain reaction sequence-based typing (PCR-SBT) method. Two hundred unrelated healthy individuals were typed as controls. RESULTS: Compared with the controls, the HLA-B*51:01 (8.8% vs. 4.5%, p=0.029, OR 2.039 [CI 1.101-3.775]) and HLA-C*07:06 (2.6% vs. 0.5%, p=0.024, OR 5.307 [CI 1.119-25.171]) allele frequencies were higher in the PTC patients, while the HLA-C*07:01 (1.3% vs. 6.0%, p=0.001, OR 0.206 [CI 0.071-0.601]) allele frequency was lower in the PTC patients that did not persist after Bonferroni correction for multiple tests. This showed no statistically significant correlation of the HLA-A, -DRB1, and -DQB1 alleles and PTC. The incidence of PTC was more frequent in females between 30 and 60 years old. There were no significant differences in the age and sex distributions between the total and the HLA-B*51:01 positive PTC patients. CONCLUSIONS: The HLA associations in this Chinese Han population differ markedly from studies done in Europeans and Caucasians. The results reveal that HLA-B*51:01 is more likely to be a susceptible allele for PTC in addition to age and sex in the coastal areas of Shandong Province.

AIM: To investigate the expression kinetics of PD-L1 and PD-L2 on the surface of the resting and activated B/T cells as well as monocytes from healthy human peripheral blood.METHODS: Fluorescent antibody staining together with flow cytometry were used to detect the percentages of the resting as well as the activated B cells and T cells that expressed PD-L1 and PD-L2.Meanwhile the percentages of the resting and activated monocytes that expressed PD-L2 were determined.RESULTS: Both resting B cells and T cells did not express PD-L1 on their surface,however PD-L1 expression was significantly up-regulated on the surface of the activated B cells after 6 h stimulation with LPS or pokeweed mitogen(PWM),and the percentages of B cells that expressed PD-L1 reached a plateau at 24 h,which were(46.26?10.71)% with LPS and(43.67?6.14)% with PWM stimulation,respectively.No markedly change of PD-L1 expression on the surface of the activated T cells after stimulation with LPS was observed,but upregulation of PD-L1 expression was observed when stimulation with PWM.The percentages of T cells that expressed PD-L1 reached a plateau at 24 h,which was(25.42?9.23)%.PD-L2 expression was not found on the resting as well as the activated B cells and T cells.In addition,the resting monocytes did not express PD-L2.Combination of INF-? plus LPS markedly induced the PD-L2 expression,and the percentages of monocytes that expressed PD-L2 reached a peak at 48 h,which was(28.70?14.22)%.CONCLUSION: The activated lymphocytes only express PD-L1,reaching a plateau at 24 h.PD-L2 is expressed on the surface of the activated monocytes,reaching a peak at 48 h.

Objective To study the polymorphism of human platelet antigen HPA-1 to HPA-5,and HPA-15 system in Qingdao Han population.Methods A total of 918 samples from regular voluntary platelet donors in Qingdao were genotyped for HPA-1 to-5 and HPA-15 by PCR-SSP.Results The gene frequencies of HPA-1a,-1b;HPA-2a,-2b;HPA-3a,-3b;HPA-4a,-4b;HPA-5a,-5b;HPA-15a,-15b were 0.9940,0.0060;0.9319,0.0681;0.5822,0.4178;0.9897,0.0104;0.9804,0.0196;0.4913,0.5087,respectively.Both a and b alleles were found in each of the 6 HPA systems,and a/a homozygosity was more common in HPA-1,-2,-4 and-5 systems.The HPA genotype frequencies followed Hardy-Weinberg principle.HPA-1 frequency of Qingdao people was significantly different from that of North China(P
