RESUMEN

A water-soluble polysaccharide BSP was extracted from the basal part of bamboo shoot, a main by-product of bamboo shoot processing. BSP is composed of glucose (72.8%), xylose (19.43%) and a small amount of galactose, arabinose, glucuronic acid and mannose. The effects of BSP on mice with antibiotic-associated diarrhea (AAD) were investigated. The mice fed with BSP exhibited significant higher bodyweight gain, lower pH value and higher concentrations of SCFAs in the feces compared with those fed with saline. BSP administration reduced the inflammatory cells in the small intestine and colon in the AAD mice, and Firmicutes/Bacteroidetes ratio in the gut was decreased from 0.56 to 0.19. Moreover, BSP administration affected the composition and diversity of the gut microbiota in the AAD mice, particularly on the improvement of beneficial bacteria such as Bacteroides, Lactobacillus and Lachnospiraceae_NK4A136_group. Our results suggest that the polysaccharides from bamboo shoot by-products could be an attractive natural component for gut health and AAD treatment.

RESUMEN

Synchronous front-face fluorescence spectroscopy has been developed for the discrimination of used frying oil (UFO) from edible vegetable oil (EVO), the estimation of the using time of UFO, and the determination of the adulteration of EVO with UFO. Both the heating time of laboratory prepared UFO and the adulteration of EVO with UFO could be determined by partial least squares regression (PLSR). To simulate the EVO adulteration with UFO, for each kind of oil, fifty adulterated samples at the adulterant amounts range of 1-50% were prepared. PLSR was then adopted to build the model and both full (leave-one-out) cross-validation and external validation were performed to evaluate the predictive ability. Under the optimum condition, the plots of observed versus predicted values exhibited high linearity (R(2)>0.96). The root mean square error of cross-validation (RMSECV) and root mean square error of prediction (RMSEP) were both lower than 3%.

RESUMEN

Cronobacter spp. is an emerging pathogen that causes meningitis, sepsis, bacteremia, and necrotizing enterocolitis in neonates and children. The present study developed an assay integrating real-time PCR and high resolution melting (HRM) analysis targeting the OmpA gene for the specific detection and rapid identification of Cronobacter spp. (formerly Enterobacter sakazakii) in powdered infant formula. Eleven Cronobacter field isolates and 25 reference strains were examined using one pair of primers, having the accuracy of 100% in reference to conventional methods. The assay was proved to be highly sensitive with a detection limit of 10(2) CFU/ml without pre-enrichment, and highly concordant (100%) when compared with ISO-IDF 22964 in 89 actual samples. The method performed for Cronobacter spp. detection was less than 24 h, drastically shortened, compared to several days using standard culturing method, it is probe-free and reduces a risk of PCR carryover. Moreover, all Cronobacter strains examined in this study were genotyped into two species according to their HRM profiles. The established method should provide a molecular tool for direct detection and simultaneous genotyping of Cronobacter spp. in powdered infant formula.
