RESUMEN

Soybean and derived products are among the most important food for both humans and animals. China is the world's largest importer of soybeans, with more than 100 million tons of annual imports, mainly from the United States of America (US), Brazil, and Argentina. However, there have been limited studies on the microbiota associated with imported soybean grains. Here, we reveal the soybean microbiota using amplicon sequencing based on samples from four countries on three continents of North America (US), South America (Argentina, Brazil), and Asia (China). Our results showed that the soybean-associated microbiota from different continents significantly separated, presenting strong geographic variations. The core microbial taxa and geographically specified taxa were defined, with Alternaria, Enterobacter, Plectosphaerella, Stenotrophomanas, and Xeromyces defined as the core microbiota for soybean from Asia; Amanita, Aspergillus, Fusarium, Nigrospora, Herbiconiux, Pseudomonas, Saccharopolyspora, and Schumannella from North America; and Bradyrhizobium, Colletotrichum, Filobasidium, Phialosimplex, Mycosphaerella, Septoria, Sphingomonas, and Weissalla, from South America. In addition, we build the Random Forest (RF) model to predict the source of imported soybean grains. We could accurately predict the original countries of imported soybean grains within the RF prediction models, with accuracies greater than 95 %. We constructed a database of soybean-related quarantine pathogens using full-length sequences of fungal ITS region and bacterial 16S rDNA region. Two phytopathogenic fungi, Diaporthe caulivora and Cladosporium cucumerinum, listed in the Chinese quarantine catalog, were intercepted through metabarcoding sequencing. The former was further confirmed using an available national standard protocol of qPCR diagnosis. In summary, our NGS-based approach revealed the microbiota associated with soybeans. It could provide comprehensive information and valuable method on the trace the origin of soybean and detection of quarantine pathogens at Customs and departments of inspection and quarantine.

Although estrogen has crucial functions for endometrium growth, the specific dose and underlying molecular mechanism in intrauterine adhesion (IUA) remain unclear. In this study, we aimed to investigate the effects of estrogen on epithelial-mesenchymal transition (EMT) in normal and fibrotic endometrium, and the role of estrogen and Wnt/ß-catenin signaling in the formation of endometrial fibrosis. CCK-8 and immunofluorescence assay were performed to access the proliferation of different concentrations of estrogen on normal human endometrial epithelial cells (hEECs). qRT-PCR and western blot assay were utilized to explore the effect of estrogen on EMT in normal and fibrotic endometrium, and main components of Wnt/ß-catenin signaling pathway in vitro. Hematoxylin and eosin and Masson staining were used to evaluate the effect of estrogen on endometrial morphology and fibrosis in vivo. Our results indicated that the proliferation of normal hEECs was inhibited by estrogen at a concentration of 30 nM accompanied by upregulation of mesenchymal markers and downregulation of epithelial markers. Interestingly, in the model of transforming growth factor ß1 (TGF-ß1)-induced endometrial fibrosis, the same concentration of estrogen inhibited the process of EMT, which might be partially mediated by regulation of the Wnt/ß-catenin pathway. In addition, relatively high doses of estrogen efficiently increased the number of endometrial glands and reduced the area of fibrosis as determined by the reduction of EMT in IUA animal models. Taken together, our results demonstrated that an appropriate concentration of estrogen may prevent the occurrence and development of IUA by inhibiting the TGF-ß1-induced EMT and activating the Wnt/ß-catenin pathway.

The aim of this study was to elucidate the concise effects of a traditional herb pair, Curcumae rhizoma-Sparganii rhizoma (CRSR), on uterine leiomyoma (UL) by analyzing transcriptional profiling. The UL rat model was made by intramuscular injection of progesterone and gavage administration of diethylstilbestrol. From 11 weeks of the establishment of the model, rats of the UL+CRSR group were gavaged daily with CRSR (6.67 g/kg). The serum concentrations of progesterone (P) and estradiol (E2) were determined by radioimmunoassay, the uterine index was measured by caliper measurement, and the pathological status was observed by hematoxylin and eosin stain. Gene expression profiling was checked by NimbleGen Rat Gene Expression Microarrays. The results indicated that the uterine mass of UL+CRSR rats was significantly shrunk and serum P and E2 levels significantly reduced compared to UL animals and nearly to the level of normal rats. Results of microarrays displayed the extensive inhibition of CRSR upon the expression of proliferation and deposition of extracellular matrix (ECM)-related genes, and significantly regulated a wide range of metabolism disorders. Furthermore, CRSR extensively regulated key pathways of the UL process, such as MAPK, PPAR, Notch, and TGF-ß/Smad. Regulation of the crucial pathways for the UL process and ECM metabolism may be the underlying mechanisms of CRSR treatment. Further studies will provide clear clues for effectively treating UL with CRSR.

OBJECTIVE: To investigate whether in utero opioid exposure, which has been linked to adverse neurodevelopmental and social outcomes, is associated with altered DNA methylation of opioid-related genes at birth. STUDY DESIGN: Observational cohort study of 21 healthy methadone-maintained opioid-dependent mother-infant dyads consecutively delivered at >36 weeks of gestation, and 2 comparator groups: smoking, "deprived" opioid-naïve mother-infant dyads (n = 17) and nonsmoking, "affluent" opioid-naïve mother-infant dyads (n = 15). DNA methylation of ABCB1, CYP2D6, and OPRM1 genes for mothers and babies was determined from buccal swabs. Plasma methadone concentrations were additionally measured for methadone-maintained opioid-dependent mothers. RESULTS: DNA methylation for ABCB1 and CYP2D6 was similar in opioid-naïve infants compared with their mothers, but was less for OPRM1 (3 ± 1.6% vs 8 ± 1%, P < .0005). Opioid-exposed newborns had similar DNA methylation to their mothers for all genes studied and greater methylation of ABCB1 (18 ± 4.8% vs 3 ± 0.5%), CYP2D6 (92 ± 1.2% vs 89 ± 2.4%), and OPRM1 (8 ± 0.3% vs 3 ± 1.6%) compared with opioid-naïve newborns (P < .0005 for all 3 genes). Infant DNA methylation was not related to birth weight, length of hospital stay, maternal smoking, dose or plasma concentration of methadone at delivery, or postcode of residence. CONCLUSIONS: In utero exposure to opioids is associated with increased methylation of opioid-related genes in the newborn infant. It is not clear whether these findings are due to opioid exposure per se or other associated lifestyle factors.

The purpose of this work was to investigate the isolation, culture process of chicken gonadal primordial germ cells (PGCs) and study their biological characterization. PGCs were harvested from 5.5-day-old chicken embryonic genital ridges and explanted onto chicken embryonic fibroblasts (CEFs). The results showed that the primary cultivation of chicken PGCs on their own gonadal stroma cells were better than CEFs at first two days for reproduction. The conditioned media supported the growth and colony formation of PGCs for a prolonged time in vitro and maintained a normal diploid karyotype, which were positively stained by alkaline phosphatase (AKP), periodic acid Schiff (PAS) and reacted with anti-SSEA-1, SSEA-3, Oct4, Blimp1 and Sox2. Real-time PCR showed that they expressed the stage specific genes CVH, Blimp1 and Dazl, the stem cell specific genes Sox2, Pouv and Nanog. They also formed the embryoid bodies (EBs). These results suggested that the chicken PGCs cultured in vitro not only had strong self-renewal ability, but also had the potential capability of multi-lineage differentiation.

Literature searches of the Cochrane Library, PubMed, EMBASE, Web of Science, LILACS, China National Knowledge Infrastructure, and Wanfang Data databases were performed from 1966 to September 2014. Only randomized and quasi-randomized controlled clinical trials comparing operative and nonoperative treatments for displaced midshaft clavicle fractures were included. Data collection and extraction, quality assessment, and data analyses were performed according to the Cochrane standards. Thirteen studies were considered in the meta-analysis. Constant scores and the Disabilities of the Arm, Shoulder and Hand scores were improved in the operative fixation group at a follow up of one year or more. The nonunion and symptomatic malunion rates were significantly lower in the operative group. Additionally, the nonoperative group had a higher likelihood of neurological symptoms compared with the operative group. A significantly higher risk of complications was found in patients treated conservatively than in those who underwent operative fixation. However, when patients with nonunion and symptomatic malunion were excluded from the analysis, no significant differences in the complication rate were found. We concluded that based on the current clinical reports, operative treatment is superior to nonoperative treatment in the management of displaced midshaft clavicle fractures. However, we do not support the routine use of primary operative fixation for all displaced midshaft clavicle fractures in adults.

River-dominated continental shelf environments are active sites of air-sea CO2 exchange. We conducted 13 cruises in the northern Gulf of Mexico, a region strongly influenced by fresh water and nutrients delivered from the Mississippi and Atchafalaya River system. The sea surface partial pressure of carbon dioxide (pCO2) was measured, and the air-sea CO2 flux was calculated. Results show that CO2 exchange exhibited a distinct seasonality: the study area was a net sink of atmospheric CO2 during spring and early summer, and it was neutral or a weak source of CO2 to the atmosphere during midsummer, fall, and winter. Along the salinity gradient, across the shelf, the sea surface shifted from a source of CO2 in low-salinity zones (0≤S<17) to a strong CO2 sink in the middle-to-high-salinity zones (17≤S<33), and finally was a near-neutral state in the high-salinity areas (33≤S<35) and in the open gulf (S≥35). High pCO2 values were only observed in narrow regions near freshwater sources, and the distribution of undersaturated pCO2 generally reflected the influence of freshwater inputs along the shelf. Systematic analyses of pCO2 variation demonstrated the importance of riverine nitrogen export; that is, riverine nitrogen-enhanced biological removal, along with mixing processes, dominated pCO2 variation along the salinity gradient. In addition, extreme or unusual weather events were observed to alter the alongshore pCO2 distribution and to affect regional air-sea CO2 flux estimates. Overall, the study region acted as a net CO2 sink of 0.96 ± 3.7 mol m-2 yr-1 (1.15 ± 4.4 Tg C yr-1).

It is recognized that anthropogenic factors have had a major impact on carbon fluxes from land to the ocean during the past two centuries. However, little is known about how future changes in climate, atmospheric CO2, and land use may affect riverine carbon fluxes over the 21st century. Using a coupled hydrological-biogeochemical model, the Dynamic Land Ecosystem Model, this study examines potential changes in dissolved inorganic carbon (DIC) export from the Mississippi River basin to the Gulf of Mexico during 2010-2099 attributable to climate-related conditions (temperature and precipitation), atmospheric CO2, and land use change. Rates of annual DIC export are projected to increase by 65% under the high emission scenario (A2) and 35% under the low emission scenario (B1) between the 2000s and the 2090s. Climate-related changes along with rising atmospheric CO2 together would account for over 90% of the total increase in DIC export throughout the 21st century. The predicted increase in DIC export from the Mississippi River basin would alter chemistry of the coastal ocean unless appropriate climate mitigation actions are taken in the near future.

Apoptosis-inducing factor (AIF) is a mitochondrial flavoprotein that mediates both NADH-oxidizing and caspase-independent apoptosis. Further, the proapoptotic activity of AIF is located in the C-terminus of AIF, although the precise minimum sequence responsible for apoptosis induction remains to be investigated. In the present study, we generated two truncated AIFs, AIFDelta1-480-FLAG, which is a FLAG-tagged C-terminal peptide comprising amino acids from 481 to 613, and AIF360-480 containing amino acids from 360 to 480 of AIF. We used confocal microscopy to demonstrate that both the truncated proteins are expressed and located in the cytoplasm of transfected cells. AIFDelta1-480 but not AIF360-480 induces apoptosis in transfected cells. We also found that the expression of AIFDelta1-480 could initiate the release of cytochrome c from the mitochondria. The suppression of caspase-9 via siRNA blocked the proapoptotic activity of AIFDelta1-480. Therefore, AIFDelta1-480 is sufficient for inducing caspase-9-dependent apoptotic signaling, probably by promoting the release of cytochrome c. At last, we generated a chimeric immuno-AIFDelta1-480 protein, which comprised an HER2 antibody, a Pseudomonas exotoxin A translocation domain and AIFDelta1-480. Human Jurkat cells transfected with the immuno-AIFDeltal-480 gene could express and secrete the chimeric protein, which selectively recognize and kill HER2-overexpressing tumor cells. Our study demonstrates the feasibility of the immuno-AIFDeltal-480 gene as a novel approach to treating HER2-overexpressing cancers.

RESUMEN

The impacts of major tropical storms events on coastal waters include sediment resuspension, intense water column mixing, and increased delivery of terrestrial materials into coastal waters. We examined satellite imagery acquired by the Moderate Resolution Imaging Spectroradiometer (MODIS) ocean color sensor aboard the Aqua spacecraft following two major hurricane events: Hurricane Katrina, which made landfall on 29 August 2005, and Hurricane Rita, which made landfall on 24 September. MODIS Aqua true color imagery revealed high turbidity levels in shelf waters immediately following the storms indicative of intense resuspension. However, imagery following the landfall of Katrina showed relatively rapid return of shelf water mass properties to pre-storm conditions. Indeed, MODIS Aqua-derived estimates of diffuse attenuation at 490 nm (K_490) and chlorophyll (chlor_a) from mid-August prior to the landfall of Hurricane Katrina were comparable to those observed in mid-September following the storm. Regions of elevated K_490 and chlor_a were evident in offshore waters and appeared to be associated with cyclonic circulation (cold-core eddies) identified on the basis of sea surface height anomaly (SSHA). Imagery acquired shortly after Hurricane Rita made landfall showed increased water column turbidity extending over a large area of the shelf off Louisiana and Texas, consistent with intense resuspension and sediment disturbance. An interannual comparison of satellite-derived estimates of K_490 for late September and early October revealed relatively lower levels in 2005, compared to the mean for the prior three years, in the vicinity of the Mississippi River birdfoot delta. In contrast, levels above the previous three year mean were observed off Texas and Louisiana 7-10 d after the passage of Rita. The lower values of K_490 near the delta could be attributed to relatively low river discharge during the preceding months of the 2005 season. The elevated levels off Texas and Louisiana were speculated to be due to the presence of fine grain sediment or dissolved materials that remained in the water column following the storm, and may also have been associated with enhanced phytoplankton biomass stimulated by the intense vertical mixing and offshore delivery of shelf water and associated nutrients. This latter view was supported by observations of high chlor_a in association with regions of cyclonic circulation.
