Hylaseptin-4 (HSP-4, GIGDILKNLAKAAGKAALHAVGESL-NH2) is an antimicrobial peptide originally isolated from Hypsiboas punctatus tree frog. The peptide has been chemically synthetized for structural investigations by CD and NMR spectroscopies. CD experiments reveal the high helical content of HSP-4 in biomimetic media. Interestingly, the aggregation process seems to occur at high peptide concentrations either in aqueous solution or in presence of biomimetic membranes, indicating an increase in the propensity of the peptide for adopting a helical conformation. High-resolution NMR structures determined in presence of DPC-d38 micelles show a highly ordered α-helix from amino acid residues I2 to S24 and a smooth bend near G14. A large separation between hydrophobic and hydrophilic residues occurs up to the A16 residue, from which a shift in the amphipathicity is noticed. Oriented solid-state NMR spectroscopy show a roughly parallel orientation of the helical structure along the POPC lipid bilayer surface, with an insertion of the hydrophobic N-terminus into the bilayer core. Moreover, a noticeable pH dependence of the aggregation process in both aqueous and in biomimetic membrane environments is attributed to a single histidine residue (H19). The protonation degree of the imidazole side-chain might help in modulating the peptide-peptide or peptide-lipid interactions. Finally, molecular dynamics simulations confirm the orientation and preferential helical conformation and in addition, show that HSP-4 tends to self-aggregate in order to stabilize its active conformation in aqueous or phospholipid bilayer environments.

Abstract It is a fact that the regions that cultivate the most maize crop do not have fully adequate technologies to measure productivity losses caused by irregularities in water availability. The objective of this study was to evaluate the physiological characteristics of maize hybrids tolerant (DKB 390) and sensitive (BRS 1030) to drought, at V5 growth stage and under water restriction, in order to understand the mechanisms involved in the induction of tolerance to drought by chitosan in contrasting maize genotypes. Plants were cultivated in pots at a greenhouse, and chitosan 100 ppm was applied by leaf spraying. The water restriction was imposed for 10 days and then leaf gaseous exchange and chlorophyll fluorescence were evaluated. The tolerant hybrid (DKB 390) showed higher photosynthesis, stomatal conductance, carboxylation efficiency, electron transport rate, and non-photochemical quenching when chitosan was used. Plants from tolerant genotype treated with chitosan were more tolerant to water stress because there were more responsive to the biopolymer.
Resumo As regiões que cultivam milho como cultura principal ainda não possuem tecnologias adequadas para mensurar as perdas na produtividade decorrentes na disponibilidade irregular de água. O objetivo desse estudo foi avaliar as características fisiológicas de híbridos de milho tolerante (DKB 390) e sensível (BRS1030) à seca, no estádio de crescimento V5 e sob restrição hídrica, para compreender os mecanismos envolvidos na indução de tolerância à seca pela quitosana em genótipos contrastantes. As plantas foram cultivadas vasos na casa de vegetação e a quitosana 100 ppm foi aplicada por pulverização foliar. A restrição hídrica durou 10 dias e foram avaliadas as trocas gasosas e a fluorescência da clorofila. O híbrido tolerante (DKB 390) apresentou maior fotossíntese, condutância estomática, eficiência de carboxilação, taxa de transporte de elétrons e quenching não fotoquímico quando aplicada a quitosana. As plantas do genótipo tolerante tratadas com quitosana foram mais tolerantes ao déficit hídrico porque foram mais responsivas ao biopolímero.

Glycerol can be used as a substitute for corn for feeding poultry, but there are concerns about how it may affect growth performance and health of the birds. We evaluated the expression of mRNA of growth hormone (GH) and insulin-like growth factor I (IGF-I) in 35-day-old Japanese quails fed different glycerol levels (0, 4, and 8% dietary glycerol instead of corn). Total RNA was extracted from the breast muscle and cDNA was amplified with the use of specific primers for these genes using real-time PCR. Quails fed the diet with 8% glycerol supplementation had significantly lower GH mRNA and IGF-I mRNA expression than those fed no glycerol or 4% glycerol. No significant effect of the treatments was found on quail weight gain or feed intake. Feed conversion ratio was influenced by dietary glycerol levels: the group fed 8% glycerol displayed the worst feed conversion ratio (2.54) compared with that of quail fed the control diet (2.35) or 4% glycerol (2.36). Considering quail performance and the expression of the genes GH and IGF-I, a level of 4% glycerol can be used in quail feeding without any harmful effects.

An effective strategy for managing protein databases is to provide mechanisms to transform raw data into consistent, accurate and reliable information. Such mechanisms will greatly reduce operational inefficiencies and improve one's ability to better handle scientific objectives and interpret the research results. To achieve this challenging goal for the STING project, we introduce Sting_RDB, a relational database of structural parameters for protein analysis with support for data warehousing and data mining. In this article, we highlight the main features of Sting_RDB and show how a user can explore it for efficient and biologically relevant queries. Considering its importance for molecular biologists, effort has been made to advance Sting_RDB toward data quality assessment. To the best of our knowledge, Sting_RDB is one of the most comprehensive data repositories for protein analysis, now also capable of providing its users with a data quality indicator. This paper differs from our previous study in many aspects. First, we introduce Sting_RDB, a relational database with mechanisms for efficient and relevant queries using SQL. Sting_rdb evolved from the earlier, text (flat file)-based database, in which data consistency and integrity was not guaranteed. Second, we provide support for data warehousing and mining. Third, the data quality indicator was introduced. Finally and probably most importantly, complex queries that could not be posed on a text-based database, are now easily implemented. The aim of this work was to evaluate the correlation between in vitro and in vivo determinations of sun protection factors (SPF) of sunscreen lotions containing the synthetic sunscreen octyl methoxycinnamate. Three sunscreen lotions containing either 2, 4.5 or 7.5% octyl methoxycinnamate were prepared. The in vitro SPFs of these sunscreen lotions were determined according to the spectrophotometric method of Mansur et al. The in vivo SPF values were determined according to the Food and Drug Administration (FDA) method. The results indicate that there was a good correlation between the in vitro and in vivo determinations for the sunscreen lotions examined.

Amitraz, a formamidine insecticide and acaricide used in veterinary practice, presents side effects related to its pharmacological activity on alpha 2-adrenergic receptors. The present study was undertaken to investigate the antinociceptive effect of amitraz in rats and mice. The tail-flick test was used to determine the duration of the antinociceptive effect of the intraperitoneal (ip) administration of amitraz (1 and 2 mg/kg, 10 animals per group) in male Wistar rats weighing 180-220 g. The writhing test (using 10 ml/kg of a 0.6% acetic acid solution as a painful stimulus) was performed in 140 male Swiss mice weighing 20-30 g, divided into 14 groups that received ip injections of saline, amitraz (0.5, 1.0, 1.5, 2.0 and 4.0 mg/kg), xylazine or detomidine (1.0, 1.5, 2.0 and 4.0 mg/kg), in order to compare the effect of amitraz to that caused by xylazine and detomidine, and to investigate the participation of alpha 2-adrenergic receptors which were blocked by idazoxan (1 mg/kg). Amitraz induced a significant antinociceptive effect in both rats and mice. This effect is blocked in mice by idazoxan.
