Interactive digital technologies have become relevant in modern education. The use of these technologies in the classroom contributes to the development of professional competencies and skills. The purpose of the study is to analyze the effectiveness of interactive learning environments and online platforms in learning history. The features of modern online learning platforms and the possibilities of their use in the study of history have been analyzed. The research was carried out at the Department of National History of Moscow City University (Moscow, the Russian Federation). The experiment involved 623 history students of different years of study of the following training profiles: "History" (124 students of 1-4 years of study) and "Pedagogical education" ("History and a foreign language", "History and social studies" - 399 students of 1-5 years of study). Also, the survey was addressed to 29 faculty members from the university under consideration. The results of the first survey show that the majority of students (62%) noted the use of traditional methods and 38% of respondents indicated the use of the modern ones. The majority of students (52%) also answered that digital tools are rarely used in the study of history while 71% of respondents reported that digital technology affects their motivation to learn. Therefore, the majority of students rated their level of motivation as average (43%) or low (47%). The most frequent professional competencies and skills are "technical competence (modern technologies)" (62%), "creativity" (65%), "critical thinking" (60%), "strategic thinking" (55%), and "practical skills" (78%). Based on the results obtained in the first survey, a program was developed to increase the motivation of students to study history and archeology, as well as to familiarize themselves with modern educational technologies that can be used in the learning process. The second survey showed an increase in the indicators of students' motivation to learn, which indicates the effectiveness of the developed program for the study of history. The majority of students (85%) answered that digital tools affect their motivation to learn. Thus, they rated it as medium (50%), high (20%) and low (30%). Accordingly, the indicators of professional competencies and skills increased by 10%: "technical competence (modern technologies)" (72%), "creativity" (75%), "critical thinking" (71%), "strategic thinking" (65%), and "practical skills" (88%). No less significant were the shifts in students' creative thinking development indicators - they changed from 18% (before the experiment) to 58% (after the experiment). The practical significance and prospects for further research are explained by the possibility of using the developed program for the study of history in other universities located in different countries. Also, the methodological basis of the proposed program can be used to study not only history but also other subjects as digital technologies are relevant in the modern education system.

A randomized, triple-blind, placebo-controlled trial involving 42 patients between 3 and 7 years of age with spastic cerebral palsy was performed. For the treatment test group (n=22), daily respiratory exercises with hypercapnic hypoxia were performed using a Carbonic training apparatus for 20 minutes per day; a total of 14 to 16 sessions were performed. Before the start of the study and the day after training was completed, the patients underwent neurological and neurophysiological examinations (electroencephalography, magnetic stimulation of the pyramidal tract, and cognitive potentials Р300).The evoked potentials showed a decrease in the peak latency of the P3 component of the test group after treatment (302 ms) that was more pronounced than that of the placebo group (305 ms; p<0.05). Magnetic stimulation showed that hypercapnic hypoxic training resulted in reductions in central motor conduction time by 2.2 to 2.5 ms (p<0.05) and in the excitation threshold of the motor cortex by 12% to 16% (р<0.01) depending on the lateralization; The strategy of adjusting to hypercapnic hypoxia, either unfavorable (hyperventilation and avoidance) or favorable (homeostatic with the achievement of preset values for hypercapnia and hypoxia), did not change during the process of training in the placebo group; however, it shifted considerably toward favorable (from 33% to 57%; р<0.05) in the test group.Respiratory training with hypercapnic hypoxia can have a positive impact on the functional state of the nervous system of children with cerebral palsy and can be considered a method of improving the efficiency of standard therapy.

A method is described for the direct detection of unstable cysteine peptidase activity in polyacrylamide gels after native electrophoresis using new selective fluorogenic peptide substrates, pyroglutamyl-phenylalanyl-alanyl-4-amino-7-methylcoumaride (Glp-Phe-Ala-AMC) and pyroglutamyl-phenylalanyl-alanyl-4-amino-7-trifluoromethyl-coumaride (Glp-Phe-Ala-AFC). The detection limit of the model enzyme papain was 17 pmol (0.29 µg) for Glp-Phe-Ala-AMC and 43 pmol (0.74 µg) for Glp-Phe-Ala-AFC, with increased sensitivity and selectivity compared to the traditional method of protein determination with Coomassie G-250 staining or detection of activity using chromogenic substrates. Using this method, we easily identified the target digestive peptidases of Tenebrio molitor larvae by matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) analysis. The method offers simplicity, high sensitivity, and selectivity compared to traditional methods for improved identification of unstable cysteine peptidases in multi-component biological samples.

This study describes the design, synthesis, and use of selective peptide substrates for cysteine peptidases of the C1 papain family, important in many biological processes. The structure of the newly synthesized substrates is Glp-Xaa-Ala-Y (where Glp=pyroglutamyl; Xaa=Phe or Val; and Y=pNA [p-nitroanilide], AMC [4-amino-7-methylcoumaride], or AFC [4-amino-7-trifluoromethyl-coumaride]). Substrates were synthesized enzymatically to guarantee selectivity of the reaction and optical purity of the target compounds, simplifying the scheme of synthesis and isolation of products. The hydrolysis of the synthesized substrates was evaluated by C1 cysteine peptidases from different organisms and with different functions, including plant enzymes papain, bromelain, ficin, and mammalian lysosomal cathepsins B and L. The new substrates were selective for C1 cysteine peptidases and were not hydrolyzed by serine, aspartic, or metallo peptidases. We demonstrated an application of the selectivity of the synthesized substrates during the chromatographic separation of a multicomponent set of digestive peptidases from a beetle, Tenebrio molitor. Used in combination with the cysteine peptidase inhibitor E-64, these substrates were able to differentiate cysteine peptidases from peptidases of other classes in midgut extracts from T. molitor larvae and larvae of the genus Tribolium; thus, they are useful in the analysis of complex mixtures containing peptidases from different classes.

Prolyl carboxypeptidase (PRCP) is a lysosomal proline specific serine peptidase that also plays a vital role in the regulation of physiological processes in mammals. In this report, we isolate and characterize the first PRCP in an insect. PRCP was purified from the anterior midgut of larvae of a stored product pest, Tenebrio molitor, using a three-step chromatography strategy, and it was determined that the purified enzyme was a dimer. The cDNA of PRCP was cloned and sequenced, and the predicted protein was identical to the proteomic sequences of the purified enzyme. The substrate specificity and kinetic parameters of the enzyme were determined. The T. molitor PRCP participates in the hydrolysis of the insect's major dietary proteins, gliadins, and is the first PRCP to be ascribed a digestive function. Our collective data suggest that the evolutionary enrichment of the digestive peptidase complex in insects with an area of acidic to neutral pH in the midgut is a result of the incorporation of lysosomal peptidases, including PRCP.
