Intrauterine environmental factors can be associated with perinatal complications and long-term health outcomes although the underlying mechanisms remain poorly defined. Meconium formed exclusively in utero and passed naturally by a neonate may contain proteins which characterise the intrauterine environment. The aim of the study was proteomic analysis of the composition of meconium proteins and their classification by biological function. Proteomic techniques combining isoelectrofocussing fractionation and LC-MS/MS analysis were used to study the protein composition of a meconium sample obtained by pooling 50 serial meconium portions from 10 healthy full-term neonates. The proteins were classified by function based on the literature search for each protein in the PubMed database. A total of 946 proteins were identified in the meconium, including 430 proteins represented by two or more peptides. When the proteins were classified by their biological function the following were identified: immunoglobulin fragments and enzymatic, neutrophil-derived, structural and fetal intestine-specific proteins. Meconium is a rich source of proteins deposited in the fetal intestine during its development in utero. A better understanding of their specific biological functions in the intrauterine environment may help to identify these proteins which may serve as biomarkers associated with specific clinical conditions/diseases with the possible impact on the fetal development and further health consequences in infants, older children and adults.

This study explores the relationship between Arterial Ketone Body Ratio (AKBR = acetoacetate/3-hydroxybutyrate) and the patients state after partially liver resections. Enzymatic methods of Mellanby and Williamson for the determination of ketone bodies (acetoacetate and 3-hydroxybutyrate) were used. Twelve surgical patients (8 women and 4 men) were examined. The control group consisted of 5 persons. Ketone bodies were measured in samples of arterial blood taken before operation and in the postoperative period. Then AKBR values were calculated. Patients were classified into three groups. Group A patients (8 subjects) had AKBR value higher than 0.7, group B patients had AKBR between 0.4 and 0.7 (2 cases), and group C patients had AKBR below 0.4 (2 cases). We observed that incidence of postoperative complications increased with the decrease of AKBR value. No postoperative complications and good general conditions of patients were observed in group A only. Group B patients had postoperative complications, whereas group C patients decreased in the postoperative period. These results indicate that Arterial Ketone Body Ratio is a good indicator for prognosis of postoperative survival of patients after partially liver resections.

We detected the presence of fructosamine in human and bovine semen. In seminal plasma of healthy normozoospermic men (N = 17) fructosamine was found in 53% of the cases (fru+). In fru+ semen samples the concentration of fructosamine was (mean +/- S.E.M., N = 9) 0.45 +/- 0.09 mmol/L and varied from 0.15 to 0.75 mmol/L. It was 3-12 times lower than in blood serum of healthy men. In semen of infertile men (N = 57) fructosamine was present only in 21% of the cases and its concentration was lower than in fertile men i.e. (mean +/- S.E.M., N = 12) 0.27 +/- 0.007 mmol/L. In bulls (N = 98) fructosamine was found in semen of 82% of animals. In fru+ semen samples the concentration of fructosamine was (mean +/- S.E.M., N = 80) 0.77 +/- 0.12 mmol/L and varied from 0.30 to 1.15 mmol/L. We did not find any correlation between the concentration of fructosamine on one hand, and that of fructose and glucose on the other hand, in either human or bull semen. The difference in the frequency of fructosamine appearance in semen of fertile and infertile men suggests that fructosamine may be in some way involved in the process of fertilisation.

RESUMEN

Ethanol, hexobarbital, chlorpromazine and haloperidol had no effect on the composition and total amount of gangliosides in rat and mouse brains. The gangliosides seemed to be resistant to ethanol and the above mentioned drugs. None of them either in single or repeated doses had a major effect on the sialic acid-containing glycosphingolipids. The results of this study are at variance with those reported by Klemm & Foster (Life Sci., 1986, 39, 897-902) who described a sharp decrease of the total level of brain gangliosides after injection of a single dose of ethanol.
