Surface-active compounds (SACs) of microbial origin are an active group of biomolecules with potential use in the formulation of emulsions. In this sense, the present study aimed to isolate and select yeasts from fruits that could produce SACs for essential oil emulsions. The Candida krusei M4CK was isolated from the Byrsonima crassifolia fruit to make SACs. This emulsification activity (E24) was equal to or greater 50% in all carbon sources, such as olive oil, sunflower oil, kerosene, hexane, and hexadecane. E24 followed exponential growth according to the growth phase. The stability of emulsions was maintained over a wide range of temperatures, pH, and salinity. The OMBE4CK (melaleuca essential oil emulsion) had better and more significant inhibitory potential for biofilm reduction formation. In addition, bioemulsifier BE4CK alone on Escherichia coli and Pseudomonas aeruginosa biofilm showed few effective results, while there was a significant eradication for Staphylococcus aureus biofilms. The biofilms formed by S. aureus were eradicated in all concentrations of OMBE4CK. At the same time, the preformed biofilm by E. coli and P. aeruginosa were removed entirely at concentrations of 25 mg/mL, 12.5 mg/mL, and 6.25 mg/mL. The results show that the bioemulsifier BE4CK may represent a new potential for antibiofilm application.

A Staphylococcus aureus isolate (SA01) obtained from bloodstream infection exhibited a remarkable drug resistance profile. In this study, we report the draft genome sequence of S. aureus ST 5 SA01, a multidrug-resistant isolate, and analyzed the genes associated with drug resistance and virulence. The genome sketch of S. aureus ST5 SA01 was sequenced with Illumina and annotated using the Prokka software. Rapid Annotation Subsystem Technology (RAST) was used to verify the gene functions in the genome subsystems. The Comprehensive Antibiotic Resistance Database (CARD) and Virulence Factor Database (VFDB) were used in the analysis. The RAST indicated a contribution of 25 proteins to host adenine, fibronectin-binding protein A (FnbA), and biofilm formation as an intercellular polysaccharide adhesive system (PIA). The MLST indicated that S. aureus ST 5 SA01 belongs to ST5 (CC5). In silico analyses also showed an extensive repertoire of genes associated with toxins, such as LukGH leukocidin, enterotoxins, and superantigen staphylococcal classes (SSL). The 11 genes for antimicrobial resistance in S. aureus ST 5 SA01 showed similarity and identity above ≥ 99% with nucleotide sequences deposited in GenBank. Although studies on ST5 clones in Brazil are scarce, monitoring the clone of S. aureus ST 5 SA01 is essential, as it has become a problem in pediatrics in several countries.

Vernonanthura brasiliana (L.) H. Rob is a medicinal plant used for the treatment of several infections. This study aimed to evaluate the antileishmanial activity of V. brasiliana leaves using in vitro and in silico approaches. The chemical composition of V. brasiliana leaf extract was determined through liquid chromatography-mass spectrometry (LC-MS). The inhibitory activity against Leishmania amazonensis promastigote was evaluated by the MTT method. In silico analysis was performed using Lanosterol 14alpha-demethylase (CYP51) as the target. The toxicity analysis was performed in RAW 264.7 cells and Tenebrio molitor larvae. LC-MS revealed the presence of 14 compounds in V. brasiliana crude extract, including flavonoids, flavones, sesquiterpene lactones, and quinic acids. Eriodictol (ΔGbind = -9.0), luteolin (ΔGbind = -8.7), and apigenin (ΔGbind = -8.6) obtained greater strength of molecular interaction with lanosterol demethylase in the molecular docking study. The hexane fraction of V. brasiliana showed the best leishmanicidal activity against L. amazonensis in vitro (IC50 12.44 ± 0.875 µg·mL-1) and low cytotoxicity in RAW 264.7 cells (CC50 314.89 µg·mL-1, SI = 25.30) and T. molitor larvae. However, the hexane fraction and Amphotericin-B had antagonistic interaction (FICI index ≥ 4.0). This study revealed that V. brasiliana and its metabolites are potential sources of lead compounds for drugs for leishmaniasis treatment.

Endotoxemia is a condition caused by increasing levels of lipopolysaccharide (LPS) characterized by an impaired systemic response that causes multiple organ dysfunction. Lacticaseibacillus rhamnosus ATCC 9595 is a strain with probiotic potential which shows immunomodulatory properties. The incorporation of this bacterium in food rich in bioactive compounds, such as cupuaçu juice (Theobroma grandiflorum), could result in a product with interesting health properties. This work evaluated the effects of the oral administration of cupuaçu juice fermented with L. rhamnosus on the outcome of LPS-induced endotoxemia in mice. C57BL/6 mice (12/group) received oral doses (100 µL) of saline solution and unfermented or fermented cupuaçu juice (108 CFU/mL). After 5 days, the endotoxemia was induced by an intraperitoneal injection of LPS (10 mg/kg). The endotoxemia severity was evaluated daily using a score based on grooming behavior, mobility, presence of piloerection, and weeping eyes. After 6 h and 120 h, the mice (6/group) were euthanized for analysis of cell counts (in peritoneal lavage and serum) and organ weight. L. rhamnosus grew in cupuaçu juice and produced organic acids without the need for supplementation. The bacteria counts were stable in the juice during storage at 4 °C for 28 days. The fermentation with L. rhamnosus ATCC 9595 changed the metabolites profile of cupuaçu juice due to the biotransformation and enhancement of some compounds. In general, the administration of L. rhamnosus-fermented juice allowed a significant improvement in several characteristics of endotoxemic status (weight loss, hypothermia, severity index, cell migration). In addition, treatment with fermented juice significantly reduced the weight of the spleen, liver, intestine, and kidneys compared to the saline-treated endotoxemic group. Taken together, our data show that short-term intake therapy of cupuaçu juice fermented with L. rhamnosus ATCC 9595 can reduce systemic inflammation in an experimental model of LPS-induced endotoxemia in mice.

O Brasil é um país que possui um litoral onde se deparam grandes cidades e com grande fluxo de atividades humanas, o que pode levar a contaminação por microrganismos e parasitos. Diante desse contexto, o objetivo desse trabalho foi avaliar a ocorrência parasitária e microbiológica em areia de praias de São Luís, Maranhão. Para tal, foram coletadas quatro amostras de três praias, totalizando assim 12 amostras, cada área foi dividida em quatro faixas distintas, sendo duas secas e duas úmidas, e para cada amostra foram introduzidos coletores estéreis na areia com profundidade de 20 centímetros. As amostras foram identificadas e colocadas em caixa de isopor e foram conduzidas ao laboratório para análises, a parasitológica foi através do método de sedimentação espontânea e centrifugação e para a microbiológica utilizou-se o kit COLItest® para determinação de coliformes totais e termotolerantes (Escherichia coli). As areias, 100% (12/12) encontravam-se positivas para helmintos e/ou protozoários, sendo que o helminto e protozoário de maior ocorrência foram: 100% (12/12) Trichuris trichiura e 83,3% (10/12) Giardia lamblia, respectivamente. As análises microbiológicas, apresentaram 66,6% (8/12) com contaminação por coliformes totais, e 58,3% (7/12) para Escherichia coli, o ponto de coleta 3 da areia seca da praia de São Marcos (Sm3S) apresentou a maior contaminação por Escherichia coli (4,33x102 UFC/mL). As praias da orla marítima chamada Litorânea em São Luís estão contaminadas por parasitas como também por bactérias oriundas de fontes que entraram em contato com fezes seja de animais ou de humanos. Assim, aumenta-se a probabilidade de indivíduos que frequentem esses ambientes de adquirirem infecções.
Brazil has a coastline with large cities and a great flow of human activities, which can lead to contamination by microorganisms and parasites. Given this context, the objective of this study was to evaluate the parasitic and microbiological events in the sand of beaches in São Luís, Maranhão. Therefore, four samples were collected from three beaches, thus totaling 12 samples. Each area was divided into four distinct segments, two dry and two wet, and for each sample, sterile collectors were inserted into the sand at a depth of 20 centimeters. The samples were identified, placed in a Styrofoam box, and taken to the laboratory for analysis. The parasitological analysis was performed through spontaneous sedimentation and centrifugation, while the microbiological analysis used the COLItest® kit to determine total and thermotolerant coliforms (Escherichia coli). All the sand samples were positive (100%) for helminths and/or protozoa, and the most frequent helminth and protozoan were Trichuris trichiura (100%; n=12 ) and Giardia lamblia (83.3%; n=10), respectively. Microbiological analyses showed that 66.6% (8/12) were contaminated by total coliforms and 58.3% (7/12) by Escherichia coli. Collection point 3 of dry sand from São Marcos beach (Sm3S) showed the highest contamination by Escherichia coli (4.33x102 CFU/mL). The beaches on the seafront called Litorânea in São Luís are contaminated by parasites and fecal bacteria from animals or human sources on the beach. Thus, the probability of acquiring infections increases among individuals visiting these environments.

This is a cross-sectional study conducted with pregnant women who underwent prenatal care at basic health units in São Luís City, Maranhão State, Brazil. The authors used a semistructured questionnaire to assess the socioeconomic, demographic, and clinical characteristics of pregnant women as well as the Edinburgh Scale to investigate depressive symptoms. In order to assess the association between the explanatory variable and the outcome variable, Poisson logistic regression was performed with statistical significance at p < 0.05. A total of 205 women were interviewed, most aged between 18 and 29 years (66.83%). Of this total, 74.63% had not planned their pregnancy and 26.67% had depressive symptoms. The variables unplanned pregnancy (PR = 1.41; CI = 0.99−2.00; p = 0.05) and not undergoing psychological counseling (PR = 1.42; CI = 0.51−0.83; p ≤ 0.01) correlated with depressive symptoms during pregnancy. It is thus possible to link the variables unplanned pregnancy (p > 0.05) and not undergoing psychological counseling (p = 0.001) to depression. Therefore, it is important to monitor the mental health of pregnant women, especially in situations of vulnerability.

Fruit juices have been emerging as excellent vehicles for development of probiotic products due to their nutritional properties and presence of bioactive compounds. This work evaluated the growth and viability of Limosilactobacillus fermentum ATCC 23271 and Lacticaseibacillus rhamnosus ATCC 9595 in bacuri juice (Platonia insignis Mart., Clusiaceae). Both strains were able to grow in bacuri juice, without any supplementation. Viability was kept after 28 days of storage; however, growth was significantly higher for L. rhamnosus ATCC 9595 (7.40 ± 0.04 Log CFU/mL). Following this, the effects of bacterial inoculum and pulp concentration on growth and lactic acid production by L. rhamnosus ATCC 9595 were investigated using a central composite rotational design. The inoculum concentration was the main factor for obtaining the most favorable relation between growth and organic acid production (G/pH ratio). Among the tested conditions, those used in assay 6 allowed the best G/pH ratio (2.13) and higher lactic acid production (4.14 g/L). In these conditions, L. rhamnosus ATCC 9595 grown in bacuri juice showed the same resistance towards acidification or addition of lysozyme than when cultivated in MRS. Finally, the anti-infective effects of fermented and non-fermented juices were analyzed using Tenebrio molitor larvae infected by enteroaggregative Escherichia coli 042. The pre-treatment with supernatants of both fermented and non-fermented juices significantly increased the survival of E. coli-infected larvae. However, only the L. rhamnosus-fermented juice had protective effects when inoculated 2 h after infection. Collectively, the results obtained in this research allowed the basis for the development of a non-dairy probiotic product from bacuri juice.

The objective of this study was to evaluate the antibacterial action of filamentous bacteria isolated from the Byrsonima crassifolia leaf. An endophytic bacterium has been identified by classical and molecular techniques as Streptomyces ansochromogene. Screening for antibacterial action against pathogens with medical relevance (Klebsiella pneumoniae ATCC 700603, Pseudomonas aeruginosa ATCC 15692, Staphylococcus aureus ATCC 6538, Corynebacterium diphtheriae ATCC 27012, Mycobacterium abscessus, Cryptococcus gattii ATCC 24065, and Cryptococcus neoformans ATCC 24067) demonstrated activity against the bacterium P. aeruginosa ATCC 0030 with inhibition diameter zones (IDZ) of 17.6 ± 0.25 mm in the preliminary screening in solid medium. After fermentation in liquid medium, an IDZ of 19.6 ± 0.46 mm and a minimum inhibitory concentration (MIC) of 0.5 mg/mL were detected. The antibiofilm action was observed with 100% inhibition of biofilm formation at a concentration of 0.250 mg/mL. When the infection curve was prepared, it was observed that the metabolite was effective in protecting the larvae of Tenebrio molitor. The metabolite does not show toxicity for eukaryotic cells. The leishmanicidal activity demonstrated that the metabolite presented a dose-dependent effect on the promastigotes forms of Leishmania amazonensis growth and the estimated IC50/72 h was 71.65 ± 7.4 µg/mL. Therefore, it can be concluded that the metabolite produced by the endophytic bacterium Streptomyces sp. is promising for future use as an alternative strategy against bacterial resistance.

The occurrence of damage on bacterial DNA (mediated by antibiotics, for example) is intimately associated with the activation of the SOS system. This pathway is related to the development of mutations that might result in the acquisition and spread of resistance and virulence factors. The inhibition of the SOS response has been highlighted as an emerging resource, in order to reduce the emergence of drug resistance and tolerance. Herein, we evaluated the ability of betulinic acid (BA), a plant-derived triterpenoid, to reduce the activation of the SOS response and its associated phenotypic alterations, induced by ciprofloxacin in Staphylococcus aureus. BA did not show antimicrobial activity against S. aureus (MIC > 5000 µg/mL), however, it (at 100 and 200 µg/mL) was able to reduce the expression of recA induced by ciprofloxacin. This effect was accompanied by an enhancement of the ciprofloxacin antimicrobial action and reduction of S. aureus cell volume (as seen by flow cytometry and fluorescence microscopy). BA could also increase the hyperpolarization of the S. aureus membrane, related to the ciprofloxacin action. Furthermore, BA inhibited the progress of tolerance and the mutagenesis induced by this drug. Taken together, these findings indicate that the betulinic acid is a promising lead molecule in the development helper drugs. These compounds may be able to reduce the S. aureus mutagenicity associated with antibiotic therapies.

Este trabalho teve como principal objetivo analisar os sedimentos do rio Moji-Guaçu, numa tentativa de avaliação do impacto dos metais sobre o meio ambiente. A Bacia Hidrográfica do Moji-Guaçu, localizada entre os estados de São Paulo e Minas Gerais, é um importante sistema de sustento de diversas atividades econômicas regionalizadas. Em decorrência de atividades antropogênicas, tais como uso e ocupação inadequada do solo; utilização de pesticidas e fertilizantes nas lavouras; lançamento de esgoto sanitários in natura e de efluentes industriais; desmatamento e extração de areia e argila, entre outras, verificou-se que o rio Moji-Guaçu sofre diferentes graus de degradação ao longo do seu curso. Para avaliar os efeitos desses impactos, foram realizadas coletas de sedimentos, em quatro épocas diferentes, duas no período seco (agosto de 2000 e junho de 2001) e duas no chuvoso (novembro de 2000 e março de 2001),15 pontos de coleta distribuídas desde a nascente até a foz. Foram analisadas as porcentagens de matéria orgânica e inorgânica, a granulometria e os metais presentes. A análise qualitativa dos metais foi feita pela técnica espectroscópica de fluorescência de raios-X e a quantitativa foi realizada por absorção atômica. Os resultados mostraram que as porcentagens de matéria orgânica foram consideradas elevada para rios, e ocorreram na quase totalidade dos pontos e em todos os períodos de amostragem. A análise granulométrica indicou a presença de uma elevada fração de argila e silte na maioria dos pontos de amostragem, sugerindo que os sedimentos dos trechos paulista e mineiro têm importante papel na redistribuição dos metais na coluna d'água. A espectroscopia de fluorescência de raios-X mostrou a presença dos metais Zr, Cu, Fe, Cr, Ti, Ca, K, P, S, Si e Al. De um modo geral, as concentrações dos metais totais de Cd, Pb apresentaram-se elevadas para todos os pontos e Fe, Cu, Cr e Mn em alguns pontos, o que pode caracterizar um ambiente impactado e um sistema com alto grau de contaminação e interferência antrópica.
