RESUMEN

Urinary tract infections (UTIs) are a significant cause of morbidity in healthcare systems and are prominently associated with applying urethral catheters, particularly in surgeries. Polyvinyl chloride (PVC) is extensively utilized in the fabrication of catheters. Biofilms, complex polymeric constructions, provide a protective milieu for cell multiplication and the enhancement of antibiotic resistance. Strategies to counteract biofilm development on medical apparatuses' surfaces incorporate antimicrobial agents such as N,N-dodecyl, and methyl polyethylenimine (DMPEI). This research endeavored to characterize the morphology of PVC and PVC-DMPEI surfaces utilizing Scanning Electron Microscopy (SEM) and Atomic Force Microscopy (AFM) and to gauge hydrophobicity through contact angle measurements. Employing Escherichia coli, Staphylococcus aureus, and Candida albicans in adhesion assays enabled the assessment of DMPEI's efficacy in preventing microbial adherence to PVC. Butanol successfully solubilized 2 mg.mL-1 DMPEI without altering the PVC structure. SEM results substantiated the formation of a DMPEI layer on the PVC surface, which led to decreased surface roughness, as validated by AFM, and increased hydrophilicity, as demonstrated by contact angle evaluations. E. coli, S. aureus, and C. albicans exhibited significant adhesion reduction, 89.3%, 94.3%, and 86.6% on PVC-DMPEI surfaces. SEM visualizations confirmed reduced cellular colonization on PVC-DMPEI and highlighted considerable morphological modifications in E. coli. Consequently, DMPEI films effectively minimize the adhesion of E. coli, S. aureus, and C. albicans on PVC surfaces. DMPEI, with its potential as a protective coating for innovative medical devices, promises to inhibit biofilm adherence effectively.

Gold nanoparticles (Au NPs) have been extensively used as artificial enzymes, but their performance is still limited. We address this challenge by focusing on multimetallic nanorattles comprising an Au core inside a bimetallic AgAu shell, separated by a void (Au@AgAu NRs). They were prepared by a galvanic replacement approach and contained an ultrathin and porous shell comprising an AgAu alloy. By investigating the peroxide-like activity using TMB oxidation as a model transformation, we have found an increase of 152 fold in activities for the NRs relative to conventional Au NPs. Based on the kinetics results, the NRs also showed the lowest Km, indicating better interaction with the substrate and faster product formation. We also observed a linear relationship between the concentration of the product and oxTMB as a function of H2O2 concentration, which could be further applied for H2O2 sensing applications (colorimetric detection). These data suggest that the NRs enable the combined effect of an increased surface area relative to solid counterparts, the possibility of exposing highly active surface sites, and the exploitation of nanoconfinement effects due to the void regions between the core and shell components. These results provide important insights into the optimization of peroxidase-like performances beyond what can be achieved in conventional NPs and may inspire the development of better-performing artificial enzymes.

BACKGROUND: Microorganisms can migrate from the external environment to the patient's organism through the insertion of catheters. Despite being indispensable medical device, the catheter surface can be colonized by microorganisms and become a starting point for biofilm formation. Therefore, new technologies are being developed in order to modify surfaces to prevent the adhesion and survival of microorganisms. Patents with the use of DMPEI have been filed. OBJECTIVE: In the present work, we coated latex catheter surfaces with 2 mg mL-1 DMPEI in different solvents, evaluated the wettability of the surface and the anti- biofilm activity of the coated catheter against Escherichia coli, Staphylococcus aureus, and Candida albicans. METHODS: We coated the inner and outer catheter surfaces with 2 mg mL-1 of DMPEI solubilized in butanol, dimethylformamide, and cyclohexanone and the surfaces were analyzed visually. Contact angle measurement allowed the analysis of the wettability of the surfaces. The CFU mL-1 count evaluated E. coli, S. aureus, and C. albicans adhesion onto the control and treated surfaces. RESULTS: The contact angle decreased from 50.48º to 46.93º on the inner surface and from 55.83º to 50.91º on the outer surface of latex catheters coated with DMPEI. The catheter coated with DMPEI showed anti-biofilm activity of 83%, 88%, and 93% on the inner surface and 100%, 92%, and 86% on the outer surface for E. coli, S. aureus, and C. albicans, respectively. CONCLUSION: Latex catheter coated with DMPEI efficiently impaired the biofilm formation both on the outer and inner surfaces, showing a potential antimicrobial activity along with a high anti-biofilm activity for medical devices.

O objetivo foi utilizar métodos complementares de diagnóstico (histopatológicos, bacteriológicos e moleculares), no julgamento de lesões suspeitas de tuberculose observadas durante a inspeção post mortem de rotina em abatedouros. Foi acompanhado o abate e a inspeção de 41.193 bovinos, sadios ao exame ante mortem, em sete abatedouros no estado de Mato Grosso. Carcaças de 198 (0,48%) animais apresentaram lesões, sendo 182 (92,0%) classificadas como granulomatosas ou piogranulomatosas na avaliação histopatológica. Entretanto, na baciloscopia, não foi evidenciada a presença de bacilo álcool-ácido resistente (BAAR). Mycobacterium bovis foi isolado em três (1,5%) lesões, provenientes de linfonodos retrofaringeanos de bovinos com até três anos de idade. Quando usado a PCR múltipla (m-PCR) diretamente nos fragmentos de tecido, detectou-se a presença de DNA de M. bovis em 14 (7,0%) lesões, incluindo as três amostras identificadas na análise bacteriológica. O julgamento das lesões pelo exame macroscópico concordou em 93,0% (184/198) com os resultados obtidos por meio da PCR. A fim de evitar equívocos durante a avaliação, principalmente das lesões paucibacilares, como as encontradas neste estudo, recomenda-se a utilização de testes complementares rápidos e confirmatórios. A m-PCR, associada à inspeção post mortem de rotina, demonstrou ser uma técnica promissora para a vigilância da tuberculose bovina em abatedouros, contribuindo para o sucesso do programa de erradicação da tuberculose bovina.


The aim of this study was used diagnostic methods (histopathological, bacteriological and molecular) in the trial of suspected tuberculosis lesions observed during routine post mortem inspection in abattoirs. A total of of 41,193 cattle, which appeared healthy in ante mortem examination, slaughtered in seven abattoirs in the state of Mato Grosso, Brazil were examined. The carcasses of 198 (0.48%) animals showed lesions, of which 182 (91.9%) were classified as granulomatous or pyogranulomatous by histopathological analysis. However, at bacilloscopy, the presence of acid-fast bacilli (AFB) was not detected. Mycobacterium bovis was recovered from 3 (1.5%) samples, all from retropharyngeal lymph nodes in cattle up to three years old. When multiplex PCR (m-PCR) was performed directly on fragments of injured tissue, M. bovis DNA was detected in 14 (7.0%) samples including the same 3 bacteriologically positive samples. Evaluation of lesions by macroscopic analysis agreed 93% (184/198) with bacteriological culturing and the molecular test. To avoid misinterpretation during the examination, mainly of paucibacillary lesions such as those found in the samples analyzed, the use of rapid and unequivocal complementary tests such as mPCR is recommended. Molecular diagnosis, combined with routine post mortem inspection, proved to be a promising technique to improve the surveillance of TB in abattoirs, contributing to the success of the bovine tuberculosis eradication program.

Introdução: A mandioca, Manihot esculenta Crantz, é um importante vegetal comestível na região amazônica que durante o processamento origina ácido cianídrico, podendo acarretar efeitos nocivos ao ser humano e animais. Objetivo: Avaliar a exposição humana ao ácido cianídrico oriundo dos glicosídeos cianogênicos presentes no vegetal, através da determinação dos teores urinários de tiocianato, principal produto de biotransformação do cianeto e seus derivado. Método: Análise espectrofotométrica dos teores urinários de tiocianato. Resultados: A concentração urinária média do metabólito nos indivíduos fumantes foi de 1.215 +/- 0,86mg/L, enquanto que em não fumantes de 0,93 +/- 0,87mg/L. Conclusão: Os resultados obtidos indicam que a exposição humana ao ácido cianídrico neste grupo populacional é semelhante à população geral, bem como, confirma a eficiência dos processos de remoção do ácido cianídrico presentes no vegetal utilizado na culinária do estado do Pará
