RESUMEN

Visual processing depends on sensitive and balanced synaptic neurotransmission. Extracellular matrix proteins in the environment of cells are key modulators in synaptogenesis and synaptic plasticity. In the present study, we provide evidence that the combined loss of the four extracellular matrix components, brevican, neurocan, tenascin-C, and tenascin-R, in quadruple knockout mice leads to severe retinal dysfunction and diminished visual motion processing in vivo. Remarkably, impaired visual motion processing was accompanied by a developmental loss of cholinergic direction-selective starburst amacrine cells. Additionally, we noted imbalance of inhibitory and excitatory synaptic signaling in the quadruple knockout retina. Collectively, the study offers insights into the functional importance of four key extracellular matrix proteins for retinal function, visual motion processing, and synaptic signaling.

Bionic vision systems are currently limited by indiscriminate activation of all retinal ganglion cells (RGCs)- despite the dozens of known RGC types which each encode a different visual message. Here, we use spike-triggered averaging to explore how electrical responsiveness varies across RGC types toward the goal of using this variation to create type-selective electrical stimuli. A battery of visual stimuli and a randomly distributed sequence of electrical pulses were delivered to healthy and degenerating (4-week-old rd10) mouse retinas. Ganglion cell spike trains were recorded during stimulation using a 60-channel microelectrode array. Hierarchical clustering divided the recorded RGC populations according to their visual and electrical response patterns. Novel electrical stimuli were presented to assess type-specific selectivity. In healthy retinas, responses fell into 35 visual patterns and 14 electrical patterns. In degenerating retinas, responses fell into 12 visual and 23 electrical patterns. Few correspondences between electrical and visual response patterns were found except for the known correspondence of ON visual type with upward deflecting electrical type and OFF cells with downward electrical profiles. Further refinement of the approach presented here may yet yield the elusive nuances necessary for type-selective stimulation. This study greatly deepens our understanding of electrical input filters in the context of detailed visual response characterization and includes the most complete examination yet of degenerating electrical input filters.

Optimal stimulus parameters for epiretinal prostheses have been investigated by analyzing retinal ganglion cell (RGC) spiking responses to white-noise electrical stimulation, through a spike-triggered average (STA) analysis technique. However, it is currently unknown as to activation of which retinal cells contribute to features of the STA. We conducted whole-cell patch clamping recordings in ON and OFF RGCs in response to white-noise epiretinal electrical stimulation by using different inhibitors of synaptic transmission in a healthy retina. An mGluR6 agonist, L-AP4, was firstly used to selectively block the output of photoreceptors (PRs) to ON bipolar cells (BCs). We subsequently fully blocked all synaptic inputs to RGCs using a combination of pharmacological agents. Our data shows that PRs dominate the ability of ON RGCs to integrate electrical pulses and form a unique STA shape, while BCs do not contribute in any way. In addition, our results demonstrate that the ability of OFF RGCs to integrate pulses is consistently impaired after blocking the PR to ON BC pathway. We hypothesise that the mechanisms underlying this co-effect are related to the narrow field AII amacrine cells connecting ON and OFF pathways.Clinical Relevance-Recent retinal studies recorded mirror-inverted STAs in ON and OFF retinal pathways, thus raising the possibility of designing a stimulation approach that can differentially activate ON and OFF pathways with electrical stimulation. However, the detailed contribution of three major retinal cell layers in forming characteristic STAs is still unclear. It is of great clinical relevance to investigate the isolated contribution of PRs to the electrically driven STA since PRs progressively degenerate in the course of retinal disease.

Irradiated dose to the organs at risk surrounding the paranasal sinuses was compared in cone beam computed tomography (CBCT) and multi spiral computed tomography with respect to the organs' relative positions to the imaging field. A head and neck Alderson-Rando phantom equipped with thermoluminescence dosemeters pellets was irradiated according to three routine CBCT protocols and one protocol in multi spiral computed tomography. Dose value of organs outside the imaging field as well as those measured dose of organs inside the field were assessed. The highest measured doses were obtained from CT scan for most of the organs investigated in this study, whereas the lowest one was associated with the low-resolution mode of CBCT. Also, statistical analysis showed no significant differences between the dose values of out-of-field organs in all CBCT modes, whereas significant differences were observed between the radiation doses of CT and CBCT modes for all organs at risk inside and outside of the imaging field. CBCT is recommended on the basis of having a lower dose; however, the image qualities were the same in the two employed modalities, so the approach of lower dose can be made.

The evaluation of building facades is one of the most important elements in built environments for helping architects and professionals to develop future designs. The form or shape of windows in building facades has direct impacts on perceivers' affective state and emotions. To understand the impacts of geometric windows on the subject's feedback and cortical activity, psychophysics experiments and electroencephalogram (EEG) recordings were measured from the participants. Our behavioral results show a distinguished categorization of the window shapes as pleasant and unpleasant stimuli. The rectangular, square, circular and semi-circular arch were determined as the pleasant window shapes, while the triangular and triangular arch window shapes were distinguished as unpleasant. Furthermore, event-related potential (ERP) components (N1, P2 and P3) were investigated to determine the influence of window shapes on the local brain activity. To measure reliable cortical responses, a Butterworth notch filter (50 Hz), band pass filter (0.1-60 Hz) and ADJUST filter were employed to remove the artifacts. The electrophysiological results show increased activity for the unpleasant in comparison to the pleasant windows (p < 0.05, Rank-Sum test) in both frontal (for P2 component) and posterio-occipital (ERP amplitudes; the N1 through to the P3 peak) channels. The ERP amplitudes of the right hemisphere were significantly larger than in the left hemisphere, not only in response to the unpleasant (p < 0.001) but also to the pleasant window stimuli (p < 0.001, Signed-Rank test). However, the unpleasant stimuli evoked significantly larger ERP amplitude than the pleasant stimuli. Moreover, the significant ERPP2 amplitude was more distinguished for unpleasant (p = 0.01, Signed-Rank test) than pleasant windows (p = 0.01, Rank-Sum test) between frontal and central cortical lobes. Overall, our behavioral and electrophysiological studies demonstrate a distinguished categorization of pleasant and unpleasant window shapes and more significant ERP modulations in the right than left hemisphere for unpleasant windows compared to pleasant ones.

A new PID controller for resistant differential control against load disturbance is introduced that can be used for load frequency control (LFC) application. Parameters of the controller have been specified by using imperialist competitive algorithm (ICA). Load disturbance, which is due to continuous and rapid changes of small loads, is always a problem for load frequency control of power systems. This paper introduces a new method to overcome this problem that is based on filtering technique which eliminates the effect of this kind of disturbance. The object is frequency regulation in each area of the power system and decreasing of power transfer between control areas, so the parameters of the proposed controller have been specified in a wide range of load changes by means of ICA to achieve the best dynamic response of frequency. To evaluate the effectiveness of the proposed controller, a three-area power system is simulated in MATLAB/SIMULINK. Each area has different generation units, so utilizes controllers with different parameters. Finally a comparison between the proposed controller and two other prevalent PI controllers, optimized by GA and Neural Networks, has been done which represents advantages of this controller over others.
