RESUMEN

We study the multifractal behavior of coherent states projected in the energy eigenbasis of the spin-boson Dicke Hamiltonian, a paradigmatic model describing the collective interaction between a single bosonic mode and a set of two-level systems. By examining the linear approximation and parabolic correction to the mass exponents, we find ergodic and multifractal coherent states and show that they reflect details of the structure of the classical phase space, including chaos, regularity, and features of localization. The analysis of multifractality stands as a sensitive tool to detect changes and structures in phase space, complementary to classical tools to investigate it. We also address the difficulties involved in the multifractal analyses of systems with unbounded Hilbert spaces.

Abstract In order to better understand the ossification processes in anurans our study was carried out on tadpoles and adults of Lithobates catesbeianus. In this sense, we characterized the kinetic properties of alkaline phosphatase with p-nitrophenylphosphatase (pNPP) and pyrophosphate (PPi) and evaluated the activities of tartrate-resistant acid phosphatase and acid phosphatase. The enzyme extracts were obtained from tadpoles and adult femurs, which were divided into epiphysis and diaphysis. After homogenization, the samples were submitted to differential centrifugation to obtain cell membranes and, further, to phospholipase C (PIPLC) treatment, to remove membrane-bound proteins anchored by phosphatidylinositol. The average of specific activity for pNPP hydrolysis (at pH 10.5) by alkaline phosphatase released by phosphatidylinositol-specific phospholipase C (PIPLC) from Bacillus cereus among different bone regions at different animal ages was 1,142.57 U.mg-1, while for PPi hydrolysis (at pH 8.0), it was 1,433.82 U.mg-1. Among the compounds tested for enzymatic activity, the one that influenced the most was EDTA, with approximately 67% of inhibition for pNPPase activity and 77% for PPase activity. In the case of kinetic parameters, the enzyme showed a "Michaelian" behavior for pNPP and PPi hydrolysis. The Km value was around 0.6mM for pNPPase activity and ranged from 0.01 to 0.11mM for PPase activity, indicating that the enzyme has a higher affinity for this substrate. The study of pNPP and PPi hydrolysis by the enzyme revealed that the optimum pH of actuation for pNPP was 10.5, while for PPi, which is considered the true substrate of alkaline phosphatase, was 8.0, close to the physiological value. The results show that regardless of the ossification type that occurs, the same enzyme or isoenzymes act on the different bone regions and different life stages of anurans. The similarity of the results of studies with other vertebrates shows that anurans can be considered excellent animal models for the study of biological calcification.


Resumo Para melhor compreender o processo de ossificação em anuros, nosso estudo foi conduzido em girinos e adultos de Lithobates catesbeianus. Nesse sentido, as propriedades cinéticas da fosfatase alcalina com p-nitrofenilfosfato (pNPP) e pirofosfato (PPi) foram caracterizadas, e as atividades enzimáticas das fosfatases ácida e ácida tartarato resistente foram avaliadas. Os extratos enzimáticos foram obtidos de fêmur de girinos e adultos, divididos em epífise e diáfise. Após a homogeneização as amostras foram submetidas à centrifugação diferencial para obter membrana celular e, em seguida, ao tratamento com fosfolipase C (PIPLC), para remover as proteínas de membrana ancoradas por fosfatidilinositol. A média da atividade específica da fosfatase alcalina, liberada pela PIPLC de Bacillus cereus, para a hidrólise de pNPP (pH 10,5) nas diferentes regiões do fêmur e idades dos animais foi de 1.142,57 U.mg-1, enquanto para a hidrólise do PPi (pH 8,0) foi de 1.433,82 U.mg-1. Entre os compostos testados para a atividade enzimática, o de maior influência foi o EDTA, inibindo aproximadamente 67% e 77% das atividades de pNPPase e PPase, respectivamente. Quanto aos parâmetros cinéticos, a enzima apresentou comportamento Michaeliano para a hidrólise dos dois substratos. O valor de Km foi de 0,6 mM para a atividade de pNPPase e variou de 0,01 a 0,11 para a atividade de PPase, indicando uma maior afinidade por esse substrato. O estudo da hidrólise de pNPP e PPi revelou que o pH ótimo aparente de atuação foi de 10,5 para o pNPP e 8,0 para o PPi, próximo ao fisiológico, sendo que esse é considerado o substrato natural da fosfatase alcalina. Os resultados demonstram que, apesar do tipo de ossificação que ocorre, a mesma enzima ou isoenzimas, atuam nos diferentes locais do osso e estágios de vida dos anuros. A similaridade dos estudos com os realizados com outros vertebrados apontam que os anuros podem ser considerados excelentes modelos animais para o estudo da calcificação biológica.

ABSTRACT The objective was to evaluate the inoculation with Aspergillus terreus and/or Trichoderma longibrachiatum on fermentation, chemical and microbiological composition of elephant grass 'Cameroon' silage (Cenchrus purpureus). Treatments were A. terreus at 105 colony forming units (CFU)/g (AT15), T. longibrachiatum at 105 CFU/g (TL20), a mixture of both at 105 CFU/g (MIX), and a control group without inoculation (CONTR). The design was completely randomized with seven replicates. The MIX silage was most stable, while CONTR, AT15, and TL20, had lower dry matter losses. There was no effect of inoculation in the chemical composition of silages. Only MIX silage (4.40) had pH above the minimum of 4.2 for humid grass silage and above the control (4.05). Bacteria from Diplococcus genus was identified at the opening of TL20 and CONTR silages. After air exposure, the population of rods, Lactobacillus, and total lactic acid bacteria was higher in theTL20 and MIX. The inclusion of a T. longibrachiatum and A. terreus mixture increases dry mater loss and silage pH. T. longibrachiatum was more efficient in maintaining populations of total lactic acid bacteria after opening; therefore, this strain has potential as an additive for elephant grass 'Cameroon' silage.


RESUMO O objetivo foi testar a inoculação com Aspergillus terreus e Trichoderma longibrachiatum sobre a fermentação, a composição bromatológica e microbiológica de silagem de capim-elefante cultivar 'Cameroon' (Cenchrus purpureus). Os tratamentos foram A. terreus a 105 unidades formadores de colônias (UFC)/g (AT15), T. longibrachiatum a 105 UFC/g (TL20), a mistura de ambos a 105 UFC/g (MIX), cada, e um controle não inoculado (CONTR). O delineamento foi inteiramente ao acaso, com sete repetições. A silagem MIX foi mais estável após abertura, enquanto CONTR, AT15 e TL20 apresentaram menor perda de massa seca. Não houve efeito de inoculação sobre a composição bromatológica das silagens. Apenas a silagem MIX (4,40) apresentou pH acima do mínimo de 4,2 para silagem de capim úmido e superior ao controle (4,05). Bactérias do gênero Diplococcus foram identificadas na abertura das silagens TL20 e CONTR. Após exposição ao ar, a população de bastonetes, Lactobacillus e bactérias láticas totais foram maiores em TL20 e MIX. A mistura de T. longibrachiatum e A. terreus aumenta a perda de matéria seca e o pH da silagem. T. longibrachiatum é mais eficiente em manter as populações de bactérias láticas totais após a abertura. Portanto, essa cepa tem potencial como aditivo para silagem de capim-elefante 'Cameroon'.

The knowledge of biological trace minerals and phytase requirements for modern broiler genotypes is not established and the pressure to reduce their usage in animal feeding due to environmental issues is increasing. Here, the alkaline phosphatase (ALP) and tartarate-resistant acid phosphatase (TRAP) of the tibia and serum of broilers fed with diets containing various levels of phytase and reduced levels of zinc, manganese, and copper was evaluated. The experiment was performed using 1,200 male Cobb broilers raised according to standard commercial husbandry techniques. Data were analyzed as a 4×3 factorial arrangement with four concentrations of zinc (0.34, 0.49, 0.64, and 0.79 ppm), manganese (0.18, 0.43, 0.68, and 0.93 ppm), or copper (0.09, 0.12, 0.15, and 0.18 ppm) and three concentrations of phytase (0, 500, and 1,000 FTU/kg) for age periods of 1-21 and 36-42 days. While the dietary supplementation with copper did not induce a significant effect in bone tissue biochemical markers, serum TRAP activity of 42-day old broilers increased with higher copper levels. Increasing dietary zinc levels linearly increased ALP activity in tibia growth, suggesting that zinc is essential for longitudinal bone growth. Phytase significantly promoted the increase of TRAP and ALP activities, suggesting that manganese increased growth plate activity, accelerated calcification, and remodeled the newly formed tissue into trabecular bone. Although not every enzymatic activity was affected by the treatments, the phytase use, along with trace minerals, improved the animal response to the rapid growth required nowadays and provided the nutrients for adequate bone metabolism.(AU)

This study evaluated the viability of using soy gum (residue from soy processing to obtain degummed oil) as an emulsifier in starter diets for broilers (1 to 21 days). For this, 600 1-day-old male broilers (Cobb® 500) were randomly assigned in a factorial arrangement (3 x 4), with three levels of gum inclusion (0, 1.25, and 2.5%) and four levels of soybean oil (0,1.3, 2.6, and 3.9%), with 5 replicates of 10 birds each. At 7, 14, and 21 days of age, we analyzed the performance parameters, pancreatic lipase activity and digestibility coefficients. Inclusion of soy gum improved (p<0.05) the performance and the digestibility coefficient of the ether extract, increased (p<0.05) the levels of AME and AMEn. The higher inclusion of gum (2.5%) as an emulsifier resulted in improved performance, showing the best values of feed conversion, with increased ether extract digestibility, increased AME content of the diets, and a lower requirement for pancreatic lipase in micelle formation.(AU)

Quantum systems whose classical counterparts are chaotic typically have highly correlated eigenvalues and level statistics that coincide with those from ensembles of full random matrices. A dynamical manifestation of these correlations comes in the form of the so-called correlation hole, which is a dip below the saturation point of the survival probability's time evolution. In this work, we study the correlation hole in the spin-boson (Dicke) model, which presents a chaotic regime and can be realized in experiments with ultracold atoms and ion traps. We derive an analytical expression that describes the entire evolution of the survival probability and allows us to determine the time scales of its relaxation to equilibrium. This expression shows remarkable agreement with our numerical results. While the initial decay and the time to reach the minimum of the correlation hole depend on the initial state, the dynamics beyond the hole up to equilibration is universal. We find that the relaxation time of the survival probability for the Dicke model increases linearly with system size.

Diarrhoeagenic Escherichia coli (DEC) is a leading cause of infectious diarrhoea worldwide. In recent years, Escherichia albertii has also been implicated as a cause of human enteric diseases. This study describes the occurrence of E. coli pathotypes and serotypes associated with enteric illness and haemolytic uremic syndrome (HUS) isolated in Brazil from 2011 to 2016. Pathotypes isolated included enteropathogenic E. coli (EPEC), enteroaggregative E. coli (EAEC), enterotoxigenic E. coli (ETEC), enteroinvasive E. coli (EIEC) and Shiga toxin-producing E. coli (STEC). PCR of stool enrichments for DEC pathotypes was employed, and E. albertii was also sought. O:H serotyping was performed on all DEC isolates. A total of 683 DEC and 10 E. albertii strains were isolated from 5047 clinical samples. The frequencies of DEC pathotypes were 52.6% (359/683) for EPEC, 32.5% for EAEC, 6.3% for ETEC, 4.4% for EIEC and 4.2% for STEC. DEC strains occurred in patients from 3 months to 96 years old, but EPEC, EAEC and STEC were most prevalent among children. Both typical and atypical isolates of EPEC and EAEC were recovered and presented great serotype heterogeneity. HUS cases were only associated with STEC serotype O157:H7. Two E. albertii isolates belonged to serogroup O113 and one had the stx2f gene. The higher prevalence of atypical EPEC in relation to EAEC in community-acquired diarrhoea in Brazil suggests a shift in the trend of DEC pathotypes circulation as previously EAEC predominated. This is the first report of E. albertii isolation from active surveillance. These results highlight the need of continuing DEC and E. albertii surveillance, as a mean to detect changes in the pattern of pathotypes and serotypes circulation and provide useful information for intervention and control strategies.

In order to assess the impact of nanoplastics on marine species, polystyrene nanoparticles (PS NPs) have been largely used as model particles. Here we studied the effects of 50 nm amino-modified PS-NH2 on Mediterranean sea urchin Paracentrotus lividus immune system cells (coelomocytes) in the presence of celomic fluid (CF) and at different NP concentrations (1, 5, 10, and 25 µg mL-1) and experimental conditions (absence or presence of EDTA). PS-NH2 acquired a protein corona once incubated with CF, dominated by the toposome precursor protein (TPP). In short-term cultures, a significant concentration- and time-dependent decrease in lysosomal membrane stability and apoptotic-like nuclear alterations were observed in phagocytes upon exposure to PS-NH2 (10 and 25 µg mL-1) in CF but they resulted abolished in the presence of EDTA confirming the role of TPP in triggering PS-NH2-coelomocytes interaction and toxicity. PS-NH2 did not alter MXR phenotype but the observed dose-dependent decrease in calcein accumulation suggests the ability of PS-NH2 to affect pump's efflux activity. Overall results encourage additional studies on positively charged nanoplastics, since the observed effects on sea urchin coelomocytes as well as the TPP corona formation might represent a first step for addressing their impact on sensitive marine species.

Background This study was performed to assess adhesion molecules in systemic lupus erythematosus (SLE). Methods This case-control study examined 126 SLE patients and 48 healthy individuals. Blood levels of six adhesion molecules, cortisol, nuclear autoantibody (ANA) and anti-double stranded DNA (anti-dsDNA) titers were measured, while disease activity was assessed using the SLE Disease Activity Index (SLEDAI) score. Results Platelet endothelial cell adhesion molecule 1 (PECAM-1), vascular cell adhesion molecule 1 (VCAM-1), E-selectin, P-selectin, and plasminogen activator inhibitor type-1 (PAI-1) were significantly higher in SLE patients than in controls. Binary logistic regression analysis showed that PECAM-1 and PAI-1 predicted SLE with a sensitivity of 86.5% and a specificity of 81.3%. ANA titers were significantly and positively associated with PECAM-1, VCAM-1, E-selectin, and PAI-1, whereas there were no associations between anti-dsDNA titers and adhesion molecules. Cortisol was negatively associated with PCAM-1 and ICAM-1. There were significant associations between metabolic syndrome (MetS) and E-selectin and PAI-1. 14.8% of the variance in the SLEDAI score was explained by the regression on PECAM-1 and MetS. Conclusions Our data show that adhesion molecules, especially PECAM-1, are significantly associated with SLE and disease activity, suggesting that they play a role in SLE pathophysiology. While MetS, ANA titers and cortisol levels modulate adhesion molecule levels, these associations do not explain the increased levels of adhesion molecules in SLE. Increased levels of adhesion molecules are new drug targets in SLE.

The objective of this study was to evaluate the chemical composition of tomato bagasse silage containing increasing inclusion levels of semi-purified glycerin. The experiment was conducted as a completely randomized design consisting of five treatments and four replicates, with exclusive tomatog bagasse silage and four inclusion levels of semi-purified glycerin (7.5%, 15%, 22.5% and 30% added based on natural matter). The inclusion of glycerin during ensiling increased dry matter content and linearly reduced silage pH, but did not alter ammoniacal nitrogen. Crude protein and neutral/acid detergent fiber decreased linearly, while in situ dry matter digestibility increased in a linear manner with the inclusion of glycerin in tomato bagasse silage. Objetivou-se com este estudo avaliar a composição química das silagens de bagaço de tomate com níveis crescentes de inclusão da glicerina semipurificada. O experimento foi conduzido em delineamento inteiramente casualizado, com cinco tratamentos e três repetições, sendo silagem exclusiva de bagaço de tomate e quatro níveis de inclusão da glicerina semipurificada à silagem (7,5%, 15%, 22,5% e 30%), sendo adicionada com base na matéria natural. A inclusão de glicerina na ensilagem elevou os teores de matéria seca e reduziu linearmente o pH da silagem, mas não alterou o nitrogênio amoniacal. Os teores de proteína bruta, fibra em detergente neutro e fibra em detergente ácido das silagens reduziram linearmente, já a digestibilidade in situ da matéria seca aumentou de forma linear com a inclusão da glicerina à ensilagem do bagaço de tomate. A inclusão de até 30% de glicerina semipurificada viabiliza a ensilagem do bagaço de tomate por melhorar a qualidade fermentativa e o valor energético da silagem.

The objective of this study was to evaluate the chemical composition of tomato bagasse silage containing increasing inclusion levels of semi-purified glycerin. The experiment was conducted as a completely randomized design consisting of five treatments and four replicates, with exclusive tomatog bagasse silage and four inclusion levels of semi-purified glycerin (7.5%, 15%, 22.5% and 30% added based on natural matter). The inclusion of glycerin during ensiling increased dry matter content and linearly reduced silage pH, but did not alter ammoniacal nitrogen. Crude protein and neutral/acid detergent fiber decreased linearly, while in situ dry matter digestibility increased in a linear manner with the inclusion of glycerin in tomato bagasse silage. The fungus Xylaria arbuscula was isolated as an endophyte from Cupressus lusitanica and has shown to be a prominent producer of cytochalasins, mainly cytochalasins C, D and Q. Cytochalasins comprise an important class of fungal secondary metabolites that have aroused attention due to their uncommon molecular structures and pronounced biological activities. Due to the few published studies on the ESI-MS/MS fragmentation of this important class of secondary metabolites, in the first part of our work, we studied the cytochalasin D fragmentation pathways by using an ESI-Q-ToF mass spectrometer coupled with liquid chromatography. We verified that the main fragmentation routes were generated by hydrogen and McLafferty rearrangements which provided more ions than just the ones related to the losses of H2 O and CO as reported in previous studies. We also confirmed the diagnostic ions at m/z 146 and 120 as direct precursor derived from phenylalanine. The present work also aimed the production of structurally diverse cytochalasins by varying the culture conditions used to grow the fungus X. arbuscula and further insights into the biosynthesis of cytochalasins. HPLC-MS analysis revealed no significant changes in the metabolic profile of the microorganism with the supplementation of different nitrogen sources but indicated the ability of X. arbuscula to have access to inorganic and organic nitrogen, such as nitrate, ammonium and amino acids as a primary source of nitrogen. The administration of 2-13 C-glycine showed the direct correlation of this amino acid catabolism and the biosynthesis of cytochalasin D by X. arbuscula, due to the incorporation of three labeled carbons in cytochalasin chemical structure. AIMS: Although Shiga toxins (Stx) are well-established virulence traits of O113:H21 Shigatoxigenic Escherichia coli (STEC) strains, a shortage in the knowledge of other virulence properties that may contribute to pathogenesis may exist in this serotype. This study investigated biofilm, invasiveness and colicinogeny capabilities in O113:H21 STEC isolated in Brazil, mostly from animal reservoirs. A search for genes that were reported to participate in the process of biofilm formation was also performed. METHODS AND RESULTS: The 34 O113:H21 STEC isolates analysed were assayed for biofilm production in polystyrene microplates. Genes for biofilm were investigated by PCR. Invasion of cell lineages was assessed in gentamicin protection assays and colicinogeny was investigated by phenotypic tests. Fifty per cent of the strains were biofilm formers, and 35% exhibited an invasive behaviour. The pattern of distribution of biofilm-related genes did not correlate with biofilm phenotypes observed, and a high percentage of the investigated strains were able to secrete colicins. CONCLUSION: Ability to form biofilm, invasiveness and colicinogeny is demonstrated for the first time in a collection of O113:H21 STEC. SIGNIFICANCE AND IMPACT OF THE STUDY: The ability to express three additional phenotypes besides Stx production may be a factor influencing the pathogenicity and persistence potential of O113:H21 STEC.

Diversidade genética de Mycoplasma hyopneumoniae tem sido relatada em análise múltipla de repetições em tandem em número variável (MLVA). O objetivo deste estudo foi descrever a distribuição espacial e a heterogeneidade genética de tipos de M. hyopneumoniae no Brasil, bem como investigar a correlação entre regiões de repetição 1 (RR1) e 3 (RR3) de duas adesinas importantes (P97 e P146). Foram identificados 39 tipos de MLVA baseados no número de repetições em tandem em P97 RR1 e RR3 P146. A correlação negativa significativa (Spearman's rho = -0,26; P = 0,022) entre P97 RR1 e RR3 P146 foi observada, o que sugere um possível mecanismo compensatório que permitiria a bactéria manter a sua capacidade de adesão. Os resultados contribuem para compreender a epidemiologia das M. hyopneumoniae no quarto maior país produtor de suínos do mundo.(AU)

Nitric oxide (NO) plays an important role as a leishmanicidal agent in murine macrophages. NO resistant Escherichia coli and Mycobacterium tuberculosis have been associated with poor outcomes of their resulting diseases. NO resistant Leishmania braziliensis has also been identified and exacerbates the clinical course of human leishmaniasis. We report, for the first time, natural resistance of Leishmania chagasi promastigotes to NO. These parasites were isolated from humans and dogs with visceral leishmaniasis. We also demonstrate that this resistance profile was associated with a greater survival capacity and a greater parasite burden in murine macrophages, independent of activation and after activation by IFN-γ and LPS.

RESUMEN

A total of 76 samples of Streptococcus suis isolated from meningitis cases in pigs from 2005 to 2009 were evaluated. The samples were collected from 3 to 21-week-old pigs raised in 30 farms located in Paraná state, Brazil. The samples of S. suis were obtained as part of routine procedures and were serotyped by coagglutination test using rabbit hyperimmune sera for the serotypes 1 to 9 and ½. S. suis type 2 was the most frequent, followed by serotypes 1, ½ and 3.
