RESUMEN

High-grade gliomas (grades III and IV) are highly malignant and aggressive brain tumors that present significant treatment challenges. Despite advances in surgery, chemotherapy, and radiation therapy, the prognosis for patients with glioma remains poor, with a median overall survival (mOS) range of 9-12 months. Therefore, exploring new and effective therapeutic strategies to improve glioma prognosis is of utmost importance and ozone therapy is a viable option. Ozone therapy has been used in various cancers, such as colon, breast, and lung, yielding significant results in preclinical studies and clinical trials. Only a few studies have been conducted on gliomas. Furthermore, since the metabolism of brain cells involves aerobic glycolysis, ozone therapy may improve the oxygen condition and enhance glioma radiation treatment. However, understanding the correct ozone dosage and optimal time of administration remains challenging. Herein, we hypothesize that ozone therapy should be more effective in gliomas compared with other tumors. This study provides an overview of the use of ozone therapy in high-grade glioma, including mechanisms of action, preclinical data, and clinical evidence.

RESUMEN

Breast cancer is the most common malignant tumor in women and remains a major global challenge, with ~1.4 million cases per year, worldwide. Numerous studies have shown that changes in cell metabolism are associated with the regulation of tumor progression. In the present study, the anti-cancer properties of glyoxal (GO), which is the smallest dialdehyde formed in the oxidation-reduction reaction and involved in electron transfer and energy metabolism, in breast cancer was investigated. The biological functions and molecular mechanisms of GO were investigated in breast cancer cell lines using MTT and crystal violet assays, flow cytometry, western blot analysis, 3D laser scanning confocal microscopy and transmission electron microscopy. The results showed that GO strongly inhibited cell proliferation, promoted cell apoptosis and cell cycle G2/M arrest, induced the disappearance of cellular microvilli, and enlarged mitochondria. In addition, the protein expression level of AKT, mTOR and p70-S6K decreased in the AKT-mTOR pathway, accompanied by an increase in p-ERK and p-MEK in the MAPK pathway. The results from the present study indicate that GO suppressed breast cancer progression via the MAPK and AKT-mTOR pathways. Taken together, these results provide the basis for a potential therapeutic strategy for breast cancer.

RESUMEN

Plasmalogens and Platelet-Activating Factor (PAF) are both bioactive ether phospholipids. Whereas plasmalogens are recognized for their important antioxidant function and modulatory role in cell membrane structure and dynamics, PAF is a potent pro-inflammatory lipid mediator known to have messenger functions in cell signaling and inflammatory response. The relationship between these two types of lipids has been rarely studied in terms of their metabolic interconversion and reciprocal modulation of the pro-inflammation/anti-inflammation balance. The vinyl-ether bonded plasmalogen lipid can be the lipid sources for the precursor of the biosynthesis of ether-bonded PAF. In this opinion paper, we suggest a potential role of plasmalogenic analogs of PAF as modulators and PAF antagonists (anti-PAF). We discuss that the metabolic interconversion of these two lipid kinds may be explored towards the development of efficient preventive and relief strategies against PAF-mediated pro-inflammation. We propose that plasmalogen analogs, acting as anti-PAF, may be considered as a new class of bioactive anti-inflammatory drugs. Despite of the scarcity of available experimental data, the competition between PAF and its natural plasmalogenic analogs for binding to the PAF receptor (PAF-R) can be proposed as a mechanistic model and potential therapeutic perspective against multiple inflammatory diseases (e.g., cardiovascular and neurodegenerative disorders, diabetes, cancers, and various manifestations in coronavirus infections such as COVID-19).

RESUMEN

Neonatal respiratory distress syndrome (NRDS) is a type of newborn disorder caused by the deficiency or late appearance of lung surfactant, a mixture of lipids and proteins. Studies have shown that lung surfactant replacement therapy could effectively reduce the morbidity and mortality of NRDS, and the therapeutic effect of animal-derived surfactant preparation, although with its limitations, performs much better than that of protein-free synthetic ones. Plasmalogens are a type of ether phospholipids present in multiple human tissues, including lung and lung surfactant. Plasmalogens are known to promote and stabilize non-lamellar hexagonal phase structure in addition to their significant antioxidant property. Nevertheless, they are nearly ignored and underappreciated in the lung surfactant-related research. This report will focus on plasmalogens, a minor yet potentially vital component of lung surfactant, and also discuss their biophysical properties and functions as anti-oxidation, structural modification, and surface tension reduction at the alveolar surface. At the end, we boldly propose a novel synthetic protein-free lung surfactant preparation with plasmalogen modification as an alternative strategy for surfactant replacement therapy.

RESUMEN

OBJECTIVE: To investigate the effects of Buyang huanwu decoction on the expression of MMPs and TIMPs in cardiac tissue of viral myocarditis (VMC) model mice. METHODS: Male BALb/c mice were randomly divided into control group, model group, positive control group [captopril, 100 mg/(kg·d)], Buyang huanwu decoction low-dose, medium-dose and high-dose groups [6, 18, 36 g/(kg·d)], with 24 mice in each group. Except for control group, other groups were given Coxsackie virus B3 once intraperitoneally to induce VMC model. After modeling, control group and model group were given constant volume of normal saline intragastrically; administration groups were given relevant medicine intragastrically; once a day, for consecutive 30 days. The general situation of mice in each group was observed. The day of inoculation was set at 0 d, heart mass to body mass ratio (HW/BW) was measured at 4, 10, 20, 30 d after inoculation. The morphological characteristics of myocardium were observed by HE staining, and the myocardial histopathological scores of myocardium were evaluated. The distribution of type Ⅰ and Ⅲ collagen in myocardium was observed by Abcam picrosirius red staining, and the ratio of type Ⅰ to Ⅲ collagen was calculated. At 30 d, relative expressions of MMP-1, MMP-3, MMP-9 and TIMP-1 in cardiac tissue were detected by Western blotting assay, and the ratio of MMPs to TIMPs was calculated. RESULTS: Compared with control group, mice in model group suffered from irritability, arch back, alleviation of stimulation response, reduction of body mass and even mental depression. Typical inflammatory changes and local interstitial hyperemia were observed in the myocardium, accompanied by a large number of lymphocyte infiltration and distribution of type Ⅰ and Ⅲ collagen. HW/BW (at different time points of 10-30 d), myocardial histopathological score (at different time points of 4-30 d), ratio of type Ⅰ and Ⅲ collagen (at different time points of 4-30 d), the expression of MMP-1 and MMP-9, ratio of MMPs to TIMPs were increased significantly, while the expression of MMP-3 and TIMP-1 were decreased significantly (P<0.05). Compared with model group, above symptoms of mice in administration groups were improved to different extents. HW/BW [at different time points of 10-30 d in administration groups (except for 10 d in Buyang huanwu decoction low-dose group)], myocardial histopathological score (at different time points of 10-30 d in administration groups), ratio of type Ⅰand Ⅲ collagen (at different time points of 4-10 d in positive control group and Buyang huanwu decoction high-dose group, at different time points of 20-30 d in Buyang huanwu decoction low-dose and medium-dose groups), the expression of MMP-1 (positive control group and Buyang huanwu decoction high-dose group) and MMP-9 (administration groups), ratio of MMPs to TIMPs (administration groups) were decreased significantly, while the expression of MMP-3 (positive control group, Buyang huanwu decoction low-dose and high-dose groups) and TIMP-1 (administration groups) were increased significantly (P<0.05). CONCLUSIONS: Buyang huanwu decoction can inhibit myocardial fibrosis of VMC model mice by inhibiting myocardial collagen hyperplasia, regulating the expression of MMPs and TIMPs, improving MMPs/TIMPs imbalance.

RESUMEN

OBJECTIVE: To observe the effect of acupuncture intervention on depressive-like behavior and nuclear factor-κB (NF-κB), inducible nitric oxide synthase (iNOS) and nitric oxide (NO) levels in the prefrontal cortex in chronic stress induced depression rats, so as to explore its mechanism underlying antidepressant effect. METHODS: SD rats were randomly divided into control, model, acupuncture and medication (Fluoxetine) groups (8 rats/group). The depression model was established by chro-nic unpredictable stress stimulation (fasting, water deprivation, cold water swimming, tail clamping, constraining, etc., and solitary raising) for 28 days. Acupuncture treatment was applied to "Baihui" (GV 20) and unilateral "Neiguan"(PC 6) once every other day for 14 days. Fluoxetine (10 mg/kg) was given to rats of the medication group by gavage (p.o.) before every stress stimulation, once every day for 28 days. The animals' behavior was tested by using sucrose intake and open field tests (number of crossings and rearings in 5 min). The expression of NF-κB in the prefrontal cortex was detected by Western blot, and the contents of prefrontal iNOS and NO were detected by ELISA and nitric reductases, respectively. RESULTS: After modeling, the sucrose intake and the numbers of crossing and rearing times were significantly decreased (P0.05). CONCLUSION: Acupuncture treatment can improve the depressive-like behavior of depression rats, which is closely related to its effects in down-regulating the levels of NF-κB protein, iNOS and NO in the prefrontal cortex to reduce brain inflammatory damage.

RESUMEN

Three new receptors (1a-c) bearing a p-dimethylaminobenzamide fluorophore have been synthesized and evaluated in terms of their fluoroionophoric properties towards various metal ions. Notably, receptors 1a and 1c exhibited dramatic fluorescent enhancement towards Cu2+ in acetonitrile. Subsequent investigations revealed that the highly selective behavior of these receptors towards Cu2+ could be attributed to the Cu2+-mediated oxidative cyclization of these compounds to the corresponding 1,3,4-oxadiazoles. Solvent effects and quantum calculations indicated that 1a and 1c both possessed an intramolecular charge transfer channel, which could be obstructed by the oxidative cyclization of these receptors. Receptor 1a was successfully applied to the determination of the Cu2+ in drug sample with a low detection limit of 2.2×10-8molL-1.

RESUMEN

OBJECTIVETo observe the effect of SSd on reversing the malignant phenotype of HepG2 cells and to investigate its mechanism in order to prove that SSd is a new choice to prevent and treat HCC.

METHODSHepG2 cells were cultured and treated by different concentration (0 mg/L, 2.5 mg/L, 5.0 mg/L, 10.0 mg/L and 20.0 mg/L) of SSd for 24 h, and treated by 10 mg/L of SSd for 0 h, 6 h, 12 h, 24 h, 48 h and 72h respectively. The cell inhibition rates were measured by MTT assay. Then cells were treated by 10 mg/L SSd for 48 hr in experimental group and treated by no SSd as a control, their morphological changes were observed by contrast phase microscope. The concentrations of ALB and AFP in clear supernatant liquid of cells were detected by radio-immunity and chemiluminescence. The cell migration rates were observed by transwell method, the relative expression levels of p27 mRNA were measured by RT-PCR.

RESULTSThe inhibitive effect of 10 mg/L SSd was the most significant among different concentrations ( F = 265.06, P less than 0.01). The shape of HepG2 from experimental group turned into small and round, and their volume ratios of nucleus to plasma decreased. ALB in supernatant liquid of HepG2 was higher ( t = 7.83, P less than 0.05, and its AFP was lower ( t = -10.72, P less than 0.01) as compared to control group. Cells migrated were fewer and p27 mRNA expression of HepG2 was higher in experimental group than that in control group (t = 22.00, P less than 0.05).

CONCLUSIONSSd could reverse the malignant phenotype of HepG2 cells. It was suggested that the up-regulation of p27 mRNA expression play an important role in the differentiation of HepG2 cells treated by SSd.

RESUMEN

OBJECTIVE: To study the frequency distribution in polymorphism of the apoprotein B 3' variable number tandem repeat (ApoB3'VNTR) and influence factors on hyperlipemia in civil aircrew. METHODS: ApoB genotypes were determined by PCR technology and agarose gel electrophoresis. The blood lipids were measured by routine kits. Personal information of flight personnel was collected by questionnaire. RESULTS: Prevalence of the total dyslipidemia (49.5%) and overweight (55.6%) of flight personnel were much higher than that of domestic general population (29.2% and 49.1%) respectively (P < 0.05). There were 16 alleles and 54 kinds of genotypes of ApoB3'VNTR in the 682 flight personnel. The frequency distribution of alleles and genotypes of aircrew in the two air companies had same trend, which was different from the general population. The frequency of the homozygote was 76.54%, which was much higher than that of the other peoples home and abroad (21.50%). The frequency of the big allele (VNTR > or =39) in hyperlipemia groups were higher than that of normal groups. By analysis of co-variance, the body mass index (BMI), low density lipoprotein (LDL) and the total cholesterol(TC) increased with the cumulate flight hours (P < 0.05). In multivariate logistic regression analysis, the BMI was the only factor influencing blood lipids, and the cumulate flight hours was only factors affecting the BMI. Taking the cumulate flight hours logarithm as the independent variable(X), and the BMI as dependent variable(Y), the linearity equations was: Y = 2.730X + 13.584 (R2 = 0.159, P < 0.01). CONCLUSION: There are perhaps special genetic characteristics in the polymorphism of the ApoB3'VNTR in the aircrew. The big allele is correlated with the hyperlipemia. The flight burden not only directly affects the BMI and blood lipids levels, but also it can indirectly affect the lipids levels by BMI.

RESUMEN

AIM:To study the expression of proapoptotic gene Bax in human gastric carcinoma and its significance.METHODS:Using immunohistochemistry methods, the Bax protein expression in 57 specimens of gastric carcinoma and its relationship with clinical status and pathomorphological parameters were observed.RESULTS:Thirty- three (57.9%) cases were positive for Bax protein staining which was mainly located in the cytoplasm of tumor cells. The rate of Bax protein expression was not correlated with the tumor size, lymph node metastasis, depth of invasion, clinical stages of tumors and age and sex of patients (P >0.05), but strongly associated with the morphological type and differentiation degree of tumors. It was significantly higher in intestinal type and well or moderately differentiated gastric carcinoma than in diffuse type and poorly differentiated gastric carcinoma (P < 0.05 and P <0.01).CONCLUSION:The proapoptotic gene Bax is differently expressed in most of gastric carcinoma and may take part in the modulation of apoptosis in gastric carcinoma. The expression of Bax might be associated with the occurrence of intestinal type gastric carcinoma and the differentiation of gastric carcinoma.

RESUMEN

AIM:To further study the role of bcl-2 protein expression in gastric carcinogenesis and tumor progression.METHODS:Using immunohistochemical staining, the bcl-2 protein expression in 50 cases of gastric carcinoma and its relation to clinical status and pathomorphological parameters were observed.RESULTS:Forty-one (82%) cases were positive for bcl-2 protein staining which was located in the cytoplasm and nuclear membrane of tumor cells. The rate of bcl-2 protein expression was not correlated with the patient, sex, tumor size, lymph node status or clinical stages (P > 0.05). It was strongly associated with intestinal-type tumors and poorly differentiated tumors (P < 0.05 andP <0.01).CONCLUSION:Aberrant bcl-2 protein expression appears to be specifically associated with development of intestinal-type gastric carcinoma, bcl-2 protein expression might play an important role in the early development/promotion and phenotypic differentiation of gastric carcinomas, but not in tumor progression.
