RESUMEN

Leishmania mexicana is one of the causal agents of cutaneous leishmaniasis. Current antileishmanial chemotherapeutics have demonstrated adverse side effects; thus, alternative treatments are needed. In this study, we performed in silico and in vitro analyses of the leishmanicidal potential of the most abundant phenolic compounds identified in black sesame sprouts biostimulated with Bacillus clausii. The molecular docking analysis showed strong interactions (binding free energies between -6.5 and -9.5 kcal/mol) of sesaminol 2-O-triglucoside, pinoresinol dihexoside, isoverbascoside, and apigenin with the arginase, leishmanolysin, cysteine peptidase B, and pyruvate kinase leishmanial enzymes. Furthermore, almost all phenolic compounds interacted with the active site residues of L. mexicana enzymes. In vitro, the B. clausii-biostimulated sprout phenolic extracts and apigenin inhibited the growth of promastigotes with IC50 values of 0.08 mg gallic acid equivalent/mL and 6.42 µM (0.0017 mg/mL), respectively. Additionally, in the macrophage infection model, cells treated with B. clausii-biostimulated sprout phenolic extracts and infected with L. mexicana exhibited significantly (P < 0.05) reduced nitric oxide production and decreased parasite burden. Altogether, our study provides important data related to high efficacy and less toxic natural antileishmanial candidates against promastigotes of L. mexicana.

RESUMEN La estrongiloidiasis comúnmente produce problemas gastrointestinales. Presentamos el caso de un varón, cadete en la marina de guerra del Perú, de 30 años, procedente de lima; que desarrolló síndrome de hiperinfección por Strongyloides Stercoralis, teniendo como antecedente el diagnóstico presuntivo de polimiositis, por lo cual recibió un ciclo corto de corticoesteroides. No portador del virus htlv 1/2. Presentó al ingreso hiporexia, debilidad generalizada, caquexia, diarrea intermitente autolimitada, intolerancia oral y leve distensión abdominal. El paciente llegó a la etapa de diseminación, lo que resultó en un daño severo a nivel intestinal. La baja excreción de larvas en las heces dificultó el diagnóstico. Se brindó tratamiento con ivermectina parenteral a dosis de 1.2ml vía subcutánea cada 48 horas por tres dosis, con buena respuesta clínica y posteriormente con buena tolerancia oral. La importancia de presentar el caso es comentar sobre el abordaje diagnóstico y terapéutico de esta geohelmintiasis endémica del Perú.


Abstract Strongyloidiasis commonly causes gastrointestinal problems. We present the case of a male, a 30-year-old cadet in the peruvian navy from lima, who developed a hyperinfection syndrome due to strongyloides stercoralis, having a presumptive diagnosis of polymyositis for which he received a short cycle of corticosteroids. He was not a carrier of the htlv 1/2 virus. Upon admission, he presented with hyporexia, generalized weakness, cachexia, intermittent self-limited diarrhea, oral intolerance, and mild abdominal distension. The patient reached the dissemination stage, resulting in severe intestinal damage. The low excretion of larvae in the feces made the diagnosis difficult. Treatment was provided with parenteral ivermectin at a dose of 1.2ml subcutaneously every 48 hours for three doses, with a good clinical response and subsequently good oral tolerance. The importance of presenting the case is to comment on the diagnostic and therapeutic approach to this endemic geohelminthiasis of peru.

INTRODUCCIÓN: El nuevo índice de obesidad basado en el volumen corporal (ABSI) ha sido asociado con enfermedades crónicas no transmisibles y mortalidad, independientemente de los valores del índice de masa corporal (IMC); sin embargo, se desconoce su asociación con diabetes mellitus tipo 2 (DMT2) en población chilena. OBJETIVO: determinar la asociación entre ABSI con glicemia, hemoglobina glicosilada (HbAc1) y DMT2 en población adulta chilena. MATERIALES Y MÉTODOS: Estudio transversal, incluyó a 4.874 participantes (edad media 43,3 años, 50,9% mujeres) de la Encuesta Nacional de Salud 2016-2017. ABSI fue calculado según la fórmula propuesta (basado en circunferencia de cintura, IMC y estatura. La Regresión de Poisson fue utilizada para investigar la asociación entre ABSI con DMT2 y regresión lineal para glicemia y HbAc1. Los análisis se ajustaron según factores sociodemográficos, de estilos de vida e IMC. RESULTADOS: ABSI fue positivamente asociado con glicemia (p < 0,001), HbA1c (p < 0,001) y DMT2 (p < 0,001). En el modelo más ajustado, por cada 0.025 unidad de aumento de ABSI, la glicemia aumentó en 1,78 mg/dL (95% IC: 1,21; 2,35) y la HbAc1en un 0,92% (95% IC: 0,49; 1,35). En cuanto a DMT2, la razón de prevalencia fue de 1,14 (95% IC: 1,09; 1,20), independiente de factores sociodemográficos, estilos de vida e IMC. CONCLUSIONES: ABSI se asoció de forma lineal con una mayor probabilidad de padecer DMT2, mayores niveles de glicemia y HbA1c en adultos chilenos. En este contexto, ABSI podría ser un índice complementario, independiente del IMC, para evaluar el riesgo de alteraciones metabólicas asociadas a obesidad.

Regarding T2DM, the prevalence ratio was 1.14 (95% CI: 1.09, 1.20), independent of sociodemographic factors, lifestyles, and BMI. CONCLUSIONS: ABSI was linearly associated with a higher probability of suffering from T2DM and higher levels of glycemia and HbA1c in Chilean adults. In this context, ABSI could be a complementary index, independent of BMI, to assess the risk of metabolic disorders associated with obesity.

This research aimed to evaluate the antimicrobial activity of essential oils (EOs) against clinically and environmentally isolated Salmonella serotypes. Oregano, thyme, and grapefruit EO compounds were identified, and the antimicrobial activity was evaluated against the S. Saintpaul, Oranienburg, and Infantis serotypes. In addition, molecular docking was performed to explore the possible mechanisms between compounds of EOs with microbial enzymes. Thymol was the main compound identified in oregano (44.0%) and thyme (31%) EOs, while d-limonene was present in a greater proportion in grapefruit EO. Oregano EO had the highest antimicrobial activity, followed by thyme and grapefruit EOs. Oregano and thyme EOs illustrated a greater inhibitory capacity to all serotypes, particularly with the environmental S. Saintpaul. Oregano EO presented values of minimum inhibitory concentration (MIC) and minimum bactericidal concentration of 0.1 µL/mL for all serotypes, while thyme and grapefruit EOs presented MIC values of 0.1 µL/mL for the clinical serotypes S. Infantis and S. Oranienburg, respectively. Molecular docking analysis showed the optimal binding free energies for thymol and carvacrol with glucokinase, ATP-dependent-6-fructokinase, outer membrane porin C, and topoisomerase IV. Our results indicate that these EOs can inhibit clinically and environmentally isolated Salmonella serotypes and can be used as alternatives for developing natural food preservatives.

Salmonella enterica serotype Oranienburg is a multi-host, ubiquitous, and prevalent Non-typhoidal Salmonella (NTS) in subtropical rivers, particularly in sediments; little studied so far possible the adaptation and establishment of this microorganism based on its genetic content. This study was focused on the first five genomes of S. Oranienburg in sediments through whole-genome sequencing (WGS) and 61 river water genomes isolated in previous studies. Results showed an open pangenome with 5,594 gene clusters (GCs), and the division of their categories showed; 3,303 core genes, 741 persistent genes, 1,282 accessory genes, and 268 unique genes. Additionally, it showed three main subclades within the same serotype and showed a conserved genetic content, suggesting the display of different adaptation strategies to its establishment. Nine genes for antimicrobial resistance were detected: aac (6') - Iy, H-NS, golS, marA, mdsABC, mdtK, and sdiA, and a mutation in the parC gene p. T57S generating a resistance. In addition, virulence genes and pathogenicity islands (SPI's) were analyzed, finding 92 genes and an identity above 80 % in the SPI's 1 to 5, and the centisomes 54 and 63. The environmental strains of S. Oranienburg do not represent a concern as multidrug resistance (MDR) bacterium; however, virulence genes remain a potential health risk. This study contributes to understanding its adaptation to aquatic environments in Mexico.

OBJETIVO: Identificar las características generales por género y grado de sobrepeso y obesidad y su afectación sobre la calidad de vida de pacientes con leve y moderada depresión en Sinaloa México. MATERIAL Y MÉTODOS. Se realizó un estudio de corte transversal con 182 pacientes con diagnósticos depresivos de leve a moderado. La muestra se caracterizó por género y grado de sobrepeso y obesidad, y se determinó su impacto y asociación sobre la calidad de vida. La calidad de vida se determinó con la encuesta SF-36. RESULTADOS. Se encontraron diferencias significativas entre los pacientes con depresión leve y moderada (p=0.000). El rol físico se vio afectado significativamente por el género (p=0.047), mientas que la función física (p=0.016) y dolor (p=0.040) por el grado de sobrepeso y obesidad en pacientes con depresión leve. Además, se encontraron correlaciones significativas entre la función física y el dolor con el rol físico, salud general y vitalidad. CONCLUSIONES: La depresión leve afectó las dimensiones de la calidad de vida de función física, dolor y rol físico. La depresión y las dimensiones de función física y dolor se ven afectadas por el grado de sobrepeso y obesidad en los pacientes con leve depresión.


OBJECTIVE: To identify the general characteristics by gender and degree of overweight and/or obesity and its effect on the quality of life of patients with mild and moderate depression in Sinaloa, Mexico. MATERIAL AND METHODS. A cross-sectional study was conducted with 182 patients with mild to moderate depressive diagnoses. The sample was characterized by gender and degree of overweight and/or obesity, and its impact and association on quality of life were determined. Quality of life was determined with the SF-36 survey. RESULTS: Significant differences were found between patients with mild and moderate depression (p = 0.000). Physical role was significantly affected by gender (p = 0.047), while physical function (p = 0.016) and pain (p = 0.040) by the adegree of overweight and/or obesity in patients with mild depression. Furthermore, significant correlations were found between physical function and pain with physical role, general health and vitality. CONCLUSIONS: Mild depression affected the quality of life dimensions of physical function, pain, and physical role. Depression and the dimensions of physical function and pain are affected by the degree of overweight and/or obesity in patients with mild depression

El objetivo de la presente investigación fue evaluar el valor nutrimental, actividad antioxidante e inhibición de la enzima α-glucosidasa de brownies libres de gluten enriquecidos principalmente con leguminosas. Los brownies fueron elaborados principalmente con leguminosas cocidas de frijol pinto (Phaseolus vulgaris L.), alubia chica (Phaseolus vulgaris L.), garbanzo (Cicer arietinum L.) o lenteja (Lens culinaris L.) (300 g de producto). Cada producto fue dividido en porciones de 100 g cada uno. Posteriormente, se determinó su composición nutrimental, y de los extractos etanólicos libres y ligados obtenidos, se evaluó el contenido de fenólicos totales, flavonoides, actividad antioxidante y el potencial inhibitorio de la enzima α-glucosidasa. Los brownies a base de leguminosas cocidas fueron significativamente mejores que los comerciales, y presentaron un contenido de proteínas entre 6.2 y 6.9%, 1.6-4.2% de lípidos, 1.2-5.8% de fibra dietaria soluble y 11.7-21.5% de fibra dietaria insoluble. Los productos presentaron en mayor proporción a los ácidos grasos linolénico y linoleico, además fueron ricos en minerales. El contenido de fenólicos totales, y flavonoides varió dependiendo de la leguminosa con la cual se desarrolló el producto, así como de la fracción que se evaluó. La actividad antioxidante total fue mayor en los brownies de alubia. La mayor inhibición de α-glucosidasa la presentaron los extractos de brownies elaborados a base de lenteja (fracción libre) y garbanzo (fracción ligada). Los brownies elaborados con lenteja fueron sensorialmente los más aceptados. La introducción de este tipo de productos promoverá un consumo nutritivo de alimentos por personas de diferentes edades y con efectos benéficos en la salud(AU)


The aim of the present study was to evaluate the nutritional value, antioxidant activity and inhibition of α-glucosidase enzyme of gluten-free brownies enriched mainly with legumes. The brownies were elaborated mainly with cooked legumes of pinto (Phaseolus vulgaris L.), haricot bean (Phaseolus vulgaris L.), chickpea (Cicer arietinum L.) or lentils (Lens culinaris L.) (300 g of product). Each product was divided into portions of 100 g and the chemical and nutritive composition were determined. From the free and bound ethanolic extracts obtained, the content of total phenolics, flavonoids, antioxidant activity and the potential inhibitor of the α-glucosidase enzyme were evaluated. Brownies based on cooked legumes were significantly better than commercial brownies, with a protein content between 6.2 and 6.8%, 1.6-4.2% lipids, 1.2-5.8% soluble dietary fiber and 11.7-21.5% insoluble dietary fiber. The products had a higher proportion of linolenic and linoleic fatty acids, and they were rich in minerals. The content of total phenolics and flavonoids varied depending on the legume as well as the fraction that was evaluated. Total antioxidant activity was higher in haricot bean-brownies. The highest inhibition of α-glucosidase was detected in extracts obtained from lentil (free fraction) and chickpea (bound fraction) brownies. Brownies based on lentil were the most accepted in the sensory test. The introduction of this type of products promotes a nutritious consumption of foods by people of different ages with beneficial effects on health(AU)

Escherichia coli O157:H7 has become a global public health and a food safety problem. Despite the implementation of control strategies that guarantee the safety in various products, outbreaks persist and new alternatives are necessary to reduce this pathogen along the food chain. Recently, our group isolated and characterised lytic bacteriophages against E. coli O157:H7 with potential to be used as biocontrol agents in food. To this end, phages need certain requirements to allow their manufacture and application. The aim of this study was to determine the physical stability and allergenic potential of free and microencapsulated (ME) bacteriophage cocktails against E. coli O157:H7. In vitro and in vivo studies were performed to determine phage survival under different pH, gastrointestinal conditions, temperature and UV light intensities. Results showed that the stability of ME phages was significantly (P<0.05) higher than free phages after ultraviolet irradiation, pH conditions between 3 to 7, and exposure to temperatures between at -80°C and 70°C. Both formulations were highly sensitive to very low pH in simulated gastric fluid, but stable in bile salts. In vivo studies in mice confirmed these phages passed through the gastrointestinal tract and were excreted in faeces. In silico, full-length alignment analysis showed that all phage proteins were negative for allergenic potential, but different predicting criteria classified seven phage proteins with a very low probability to be an allergen. In conclusion, these data demonstrated that microencapsulation provided a greater stability to phage formulation under stress conditions and assure a more suitable commercial formulation for the biological control of E. coli O157:H7.

This study was performed to evaluate in vitro the adherence and invasiveness capacity of Salmonella Oranienburg and Saintpaul (isolated from river water) exposed to laboratory and river water growth conditions and inoculated into epithelial HEp-2 cell. Results showed that Salmonella Oranienburg and Salmonella Saintpaul showed lower ability to adhere and invade epithelial HEp-2 cells under both growth conditions as compared to Salmonella Typhimurium reference strain. S. Oranienburg adhesion capacity was not affected by the growth conditions, while S. Saintpaul exposed to river water significantly (p < 0.05) decreased its adhesion capacity by 75.7 %. On the contrary, S. Oranienburg exposed to river water reduced its invasion efficiency by 80 %, whereas S. Saintpaul showed no differences between growth conditions. In conclusion, this study suggests that the exposure to non-host conditions, such as river water, adversely affects the adhesion and invasiveness of Salmonella serotypes differently, impacting on their ability to re-enter a new host.

Blastocystis sp. es un parásito de distribución mundial, que se transmite por vía fecal-oral, colonizando el tracto gastrointestinal, tanto de animales como de humanos. Existen múltiples factores que favorecen su transmisión tales como el pobre saneamiento, el consumo de alimentos contaminados y la propagación de vectores. Este parásito ha despertado gran interés científico y clínico en los últimos años, por su potencial asociación con enfermedad; sin embargo, no ha sido posible establecer con exactitud su rol patógeno. También ha sido considerado agente oportunista, por su alta prevalencia en pacientes inmunosuprimidos, específicamente en aquellos con VIH/SIDA y cáncer. Actualmente, en el GenBank se han descrito secuencias de 33 subtipos (STs) diferentes, si bien en la literatura solo se encuentran reportados 17 subtipos hasta el momento, de los cuales 9 han sido aislados en el ser humano. Aunque buena parte de los estudios reportan a ST3 como el de mayor prevalencia, unos cuantos autores sugieren predominio de ST1 y ST4 en ciertas zonas, indicando que algunos subtipos varían dependiendo de la distribución geográfica. Varios de estos STs se han relacionado directamente con sintomatología gastrointestinal aguda y crónica e incluso con manifestaciones clínicas extraintestinales, encontrándose con mayor frecuencia el ST3 en pacientes sintomáticos, generalmente seguido de ST1 y en algunos casos de ST2. Además, se han reportado infecciones mixtas, de las cuales la más común se ha presentado entre los ST1 - ST3. En Latinoamérica, y particularmente en Colombia, se han desarrollado pocas investigaciones orientadas a identificar y definir la prevalencia, coinfección y potencial patógeno de las diferentes formas y subtipos conocidos de Blastocystis sp., por lo cual el conocimiento que se tiene hasta el momento es escaso e inespecífico, requiriéndose más estudios que permitan obtener mayor información al respecto.


Blastocystis sp. is a globally distributed parasite which is transmitted by the fecal-oral route by colonizing the gastrointestinal tract of both animals and humans. There are many factors which allow its transmiBion such as poor sanitation, the consumption of contaminated food and vectors propagation. This parasite has awakened scientific and clinical interest in recent years because of its aBociation with disease. However, its pathogen role has not been accurately established. Also, it has been considered as an opportunistic agent due to its high prevalence in immunosuppreBed patients, specifically in those with HIV / AIDS and cancer. Currently, in GenBank database the sequences of 33 different subtypes (STs) have been described even though only 17 subtypes are reported in the literature so far, from which nine have been isolated in humans. Although many studies indicate the subtype 3 (ST3) as the most prevalent, some authors suggest the predominance of ST1 and ST4 in particular areas, suggesting that some subtypes vary depending on the geographical distribution. Several of these STs have been directly related to acute and chronic gastrointestinal symptoms and even with clinical extraintestinal manifestations, being ST3 found with more frequency in symptomatic patients followed by ST1 and ST2 in some cases. Furthermore, mixed infections have been reported of which the most common has been present between ST1 - ST3. In Latin America, and particularly in Colombia, little research has been developed that aims to identify and define the prevalence, coinfection, and pathogenic potential of different forms and known subtypes of Blastocystis sp., therefore the knowledge at hand so far is scarce and unspecific, requiring more studies which allow obtaining more information about it.

Carbon nanostructures have played an important role in creating a new field of materials based on carbon. Chemical modification of carbon nanostructures through grafting has been a successful step to improve dispersion and compatibility in solvents, with biomolecules and polymers to form nanocomposites. In this sense carbohydrates such as chitosan are extremely valuable because their functional groups play an important role in diversifying the applications of carbon nanomaterials. This paper reports the covalent attachment of chitosan onto graphene oxide, taking advantage of this carbohydrate at the nanometric level. Grafting is an innovative route to modify properties of graphene, a two-dimensional nanometric arrangement, which is one of the most novel and promising nanostructures. Chitosan grafting was achieved by redox reaction using different temperature conditions that impact on the morphology and features of graphene oxide sheets. Transmission Electron Microscopy, Fourier Transform Infrared, Raman and Energy Dispersive spectroscopies were used to study the surface of chitosan-grafted-graphene oxide. Results show a successful modification indicated by the functional groups found in the grafted material. Dispersions of chitosan-grafted-graphene oxide samples in water and hexane revealed different behavior due to the chemical groups attached to the graphene oxide sheet.

In one Los Angeles study, 20 % of day laborers reported excessive drinking. Older adults are more sensitive to alcohol's effects, yet heavy drinking persists among Latinos until they are in their 60s. No interventions to reduce heavy drinking exist for aging day laborers. We recruited 14 day laborers aged 50 and older in Los Angeles. We identified their unhealthy alcohol use behaviors and comorbidities and conducted semi-structured interviews to understand their perceptions of unhealthy alcohol use. We found social disadvantages and conditions exacerbated by alcohol use, like depression. Participants were concerned with dying and premature aging, and reported that family could influence behavior change. An intervention should consider (1) integrating family values and (2) increasing knowledge about alcohol use and comorbidities. Further studies are needed to explore family influence on aging Latino day laborers.

RESUMEN

El tumor de Frantz-Gruber es una masa sólida pseudopapilar de páncreas, poco común y con bajo potencial de malignidad, descrita por primera vez por Franz en 1959. Predomina en mujeres entre la 3era y 4ta década de la vida. La sintomatología está dada principalmente por efecto de masa y se caracteriza por dolor vago y sensación de plenitud. El diagnóstico radiológico incluye estudios como tomografía axial computalizada (TAC) y resonancia magnética, y los estudios histológicos confirman la sospecha clínica. El abordaje es quirúrgico en la mayoría de los casos e incluye una resección completa de tumor. La presencia de metástasis hepática no es infrecuente. La sobrevida a 5 años es por arriba del 90 por ciento. Se reporta el caso de un paciente de 15 años, sin antecedentes patológicos o quirúrgicos, visto en el Servicio de Emergencias por epigastralgia y vómito. Un ultrasonido (US) abdominal demostró una lesión en la cola pancreática, luego por TAC y US endoscópico se logró documentar una masa de aspecto quístico y contenido denso que abarca también el cuerpo del páncreas. Se recomendó abordaje quirúrgico; durante la laparotomía exploratoria se halla tumoración mixta sólidoquística. Se practicó pancreatectomía distal y esplenectomía. La evolución y manejo postoperatorio fue sin complicaciones.

OBJECTIVE: Nosocomial transmission of hepatitis C virus (HCV) infection had been related with anesthesia procedures. The study aim was to measure the association between anesthesia procedures in cases with previous surgery and HCV infection. MATERIAL AND METHODS: In a case-control study were included subjects that attended to the Central Blood Bank of the West Medical National Center, Mexican Institute of the Social Security in Guadalajara, Jalisco between july 2005 and september 2007. Cases were patients with positive hepatitis C antibody (anti-HCV) confirmed by recombinant immunoblot assay (RIBA) and/or nucleic acid test (HCV RNA); the control group was blood donors with negative antibody. An exhaustive questionnaire about risk factors for hepatitis C, was applied. The risk of HCV infection was determined with the Odds Ratio (OR) and multivariate analysis was made by logistic regression. RESULTS: We included 362 subjects, 211 cases and 151 controls; in 70 (33.2%) cases were found significant association between the anesthesia procedures and HCV infection in patients with previous surgery (OR adjusted 2.44, CI 95% 1.44 - 4.11) CONCLUSION: This is the first study in México that demonstrate association between history of anesthesia procedures and HCV infection in cases with previous surgery.

OBJETIVO: Medir la asociación entre el antecedente de procedimientos anestésicos y el riesgo de infección por el virus de la hepatitis C (VHC) en pacientes con cirugía previa. MATERIAL Y MÉTODOS: Diseño de casos y controles; los casos fueron pacientes con anticuerpo (anti-VHC) positivo confirmado por RIBA y/o RNA VHC y los controles fueron sujetos con el anti-VHC negativo. El riesgo de infección se estimó por razón de momios (RM) y análisis multivariado con regresión logística. RESULTADOS: Se incluyeron 362 sujetos, 211 casos y 151 controles; en 70 casos (33.2 por ciento), con cirugía previa, los procedimientos anestésicos se identificaron como único factor de riesgo significativo para la infección por el VHC (RM ajustada 2.44, IC 95 por ciento 1.44 - 4.11). CONCLUSIONES: Este es el primer estudio en México que demuestra asociación de riesgo significativa con el antecedente de procedimientos anestésicos por cirugía previa, en uno de cada tres enfermos con hepatitis C.

Enteropathogenic Escherichia coli (EPEC) produces attaching and effacing (A/E) lesions and watery diarrhea, both of which are intimin and EspA dependent. In this work, we explored the mucosal immune response by detecting cytokine induction in rabbits with diarrhea caused by rabbit EPEC (REPEC). Orally inoculated rabbits exhibited weight loss and mucosal inflammation, developed watery diarrhea, and died (day 7). At day 6 postinoculation, animals were analyzed for the induction of proinflammatory cytokines in enterocytes. The role of lymphocyte-dependent immunity was determined through the expression of proinflammatory cytokines by lymphocytes from Peyer's patches (PP) and the spleen. EspA and intimin mutants were used to explore the role of A/E lesions in the expression of these cytokines. REPEC-infected rabbit enterocytes showed increased interleukin 1beta (IL-1beta), IL-6, IL-8, and tumor necrosis factor alpha (TNF-alpha) mRNA expression, but that of anti-inflammatory IL-10 was increased only slightly. In contrast, intimin mutant-infected rabbits were unable to produce this proinflammatory cytokine profile but did produce a remarkable increase in IL-10 expression. Bacteria lacking EspA increased the expression of IL-8 and TNF-alpha, but that of IL-10 was increased only slightly. PP lymphocytes also produced proinflammatory cytokines, which were dependent on EspA (except for TNF-alpha) and intimin, while IL-10 was induced by EspA and intimin mutants. In contrast, spleen lymphocytes (systemic compartment) were unable to produce IL-1beta and TNF-alpha. These data show the importance of the proinflammatory cytokines secreted by enterocytes and those expressed locally by PP lymphocytes, which can activate effector mechanisms at the epithelium. Furthermore, this cytokine profile, including IL-6 and IL-1beta, which may be involved in the diarrhea produced by EPEC, depends on intimin.
