RESUMEN

This paper presents comparative hemagglutination inhibition (HI) assay data obtained using ferret or rat antisera to analyze influenza viruses. The results indicate that rat antisera can be successfully applied both for identification and for antigenic analysis of human influenza A and B viruses. Data gained with rat antisera were comparable to those obtained with ferret antisera. In-depth statistical analysis, based on Confusion Matrix analysis and Receiver Operating Characteristic (ROC) analysis, confirmed good coincidence between ferret antisera-based and rat antisera-based results. Two-dimensional antigenic mapping, based on HI assays using rat and ferret antisera, supported these findings. Both antisera types yielded identical antigenic attributions for the viruses analyzed, and both permitted visualization of contemporary human influenza virus evolutionary trends.

RESUMEN

We performed screening of nootropic properties of 10 new derivatives of quinolizidine alkaloid (-)-cytisine. Compounds with ß-endo stereochemistry were more active than α-endo-isomers. Under stress conditions (3aR,4S,8S,12R,12aS,12bR)-10-methyl-2-phenyloctahydro-1H-4,12a-etheno-8,12-methanopyrrolo[3',4':3,4]pyrido[1,2-a] [1,5]diazocine-1,3,5(4H)-trione enhanced memory and had a positive effect on cognitive functions of rats. According to molecular docking data, the nootropic activity of the compound can be associated with its affinity for the glutamate-binding subunits GluK1 and GluR2 of the kainate and AMPA receptor, respectively.

Human A (H3N2) influenza viruses are distinguished by a high rate of evolution and regularly cause epidemics around the world. Their ability to adapt and to escape from the host's immune response and to change their receptor specificity is very high. Over the past 20 years, these viruses have lost the ability to agglutinate red blood cells of chickens and turkeys and have practically ceased to propagate in chicken embryos - the main source of influenza vaccines. Isolation of viruses in the MDCK cell culture led to the selection of strains that lose one of the potential glycosylation sites. Many of the A (H3N2) strains have acquired mutations in neuraminidase, which distort the results of antigenic analysis in the hemagglutination inhibition test - the cornerstone method for the analysis of the match between viral isolates circulating in human population to strains selected for the influenza vaccines. In this regard, the characteristics of the antigenic properties of influenza A (H3N2) viruses by traditional methods become poorly informative, and the selection of vaccine strains of this subtype is erroneous, which is reflected in the discrepancy between vaccine and circulating A (H3N2) viruses in recent years (2013-2014, 2014 -2015, 2015-2016). The search, development and implementation of new algorithms for the isolation and antigen analysis of influenza A (H3N2) viruses are extremely urgent.

Glycyrrhizic acid (GA) conjugates with methyl and ethyl esters of D-amino acids (D-Trp, D-Phe, D-Tyr, D-Val, D-Leu) have been synthesized by the activated esters method using mixtures of N-hydroxybenzotriazole or N-hydroxysuccinimide with N,N'-dicyclohexylcarbodiimide. GA conjugate with D-Trp ethyl ester exhibited antiviral activity against influenza viruses A/H3N2, A/H1N1/pdm09, A/H5N1, B (SI > 10-29), and HRSV (SI > 25). GA conjugate with D-Trp methyl ester inhibited influenza virus A/H1N1/pdm09 (SI > 30).

Balanced chromosomal translocations do not generally have phenotypic manifestation, but lead to increased risk of miscarriage and live-birth of chromosomally unbalanced offspring in carriers. Frequently prenatal diagnosis of an unbalanced translocation may incidentally detect a balanced translocation in the family. Here, we report a unique case of trisomy 3q (karyotype 46,XYder(3)t(3;21)(q11;p11)), detected prenatally due to abnormal findings of the fetus ascertained through ultrasound assessment like growth retardation, vermal agenesis, micrognathia, cystic hygroma of the neck, dextra position of arcus aortae, shorter for the gestational week long bones In order to determine the paternity of this chromosomal aberration, the cytogenetic analyses of the parents was performed. A balanced paternal translocation 46,XY,t(3;21)(q11;p11) wase identified. During the next pregnancy the same balanced translocation of paternal origin wase also identified. This case demonstrate the significance of prenatal ultrasound screening of the fetus; the necessity of cytogenetic analysis of a fetus with prenatal ultrasound abnormalities; genetic counseling of such families with aim of prenatal care and prevention during next pregnancies.

INTRODUCTION: The researches of organism's adaptive possibilities of indigenous population of the Arctic region are extremely important in modern conditions of the development of northern territories. The functional reserve of basic physiological systems and their interaction define "the health's resources", its potential level. AIM: This research has substantiated the adaptive reactions of organism of the North inhabitants on the basis of medical-physiological results. MATERIALS AND METHODS: Physical development was estimated by the standard anthopometrical method: the height and body mass were measured, index of body mass (BM), Quetelet's index, Rohrer's index were defined, area of body surface was calculated. Carbon dioxide in the air was defined by portable infra-red gas analyzer of firm "Fudji" for measurements indoors and outdoors. The parametres of blood acid-base state (ABS) were analyzed by standard procedures with microevaluator application ABL-330 (Denmark). Computer spirometry was done on the hardware-software diagnostic complex (HSDC) for analyses of external respiration function (NSRIMT - Russia) and "Pneumoscrin-2" of Erih Eger company (Germany). Arterial pressure was measured by Korotkov's method. RESULTS: The inhabitants of the North irrespective of ethnicity have conditions for more effective lung ventilation to satisfaction of the high metabolic requirements of the organism. CONCLUSIONS: Stability of arterial pressure among indigenous population, microcirculation optimisation and transcapillary tissues exchange, conserves steady level of metabolism that testifies a high level of adaptation of the person to severe climate- geographic North conditions.

To establish the anthropo-ecological portrait of the current population of Yakutia, it is necessary to study the regularities of the variability of physical status in different gender groups and develop regional standards of the physical development of different age-sex groups of the population with regard to the peculiarities of the environment. The aim of this study is to do research on the gender features of anthropometric, cephalometric and bioimpedance parameters in the students of Yakut ethnicity. The measures of 228 Yakut girls and 168 Yakut youths born and permanently resident in Yakutia were analyzed. Anthropometric measurements were performed using the method of V.V. Bunak (1941), somatotyping--using the Rees-Eysenck index (1945). Head type was determined by the cephalic index, face type--by the upper facial index. Bioelectrical impedance analysis with the use of ABC--01 Medass analyzer was performed to estimate body composition. Adipose tissue mass (AM), skeletal muscle mass (SMM), lean mass (LM), active cell mass (ACM) and phase angle (PA) were determined. Obtained material was processed by the method of variation statistics with the use of SPSS 17.0 software package. Overall measures of the soma of the examined groups have gender features of body organization that are expressed in the significantly higher values of all measured parameters in the youths of Yakutia. In our study, the sexual dimorphism in the distribution of somatotypes by the Rees-Eysenck index has not been identified. The analysis of cephalometric measures has revealed reliably higher parameters of absolute head sizes in the youths. Gender differences in the distribution of head and face types have not been found in the examined ethno-age group. Significantly larger absolute amount of AM has been established in the girls. The measures of SMM, ACM and LM are reliably higher in the youths. The values of active and reactive tissue resistance are reliably higher in the girls. Gender features of the anthropometric, cephalometric and bioimpedance parameters of the students have been established. The conducted study presents the anthropo-ecological image of this age-sex group of the population of Yakutia.

The objective of the present study was to evaluate sperm quality and reproductive hormonal level in an unselected population of men who were permanent residents of the Yakutsk city and compare the semen and hormonal parameters with the World Health Organization (WHO, 2001) recommended normal values and the results from other countries. A total of 145 men (mean age 24 years) agreed to be included in the study. All the volunteers completed a questionnaire, received a general and reproductive health examination by an andrologist, gave semen and blood samples. There are no differences between Yakut and Slavs in anthropometric, hormonal or sperm measures, but height and ejaculate volume were higher in Slavs than Yakut men. The data obtained indicated high frequency of suboptimal quality of sperm in males of both nations from this Siberian region in comparison with other Russian regions or European countries.

The analysis was implemented concerning diagnostic parameters of commercial quick tests (immune chromatographic tests BinaxNOW Influenza A&B and BinaxNow RSV Alere, Scarborough Inc., USA) under detection of antigens of influenza virus A and respiratory syncytial virus in clinical materials. The polymerase chain reaction in real-time and isolation ofviruses in cell cultures. The analysis of naso-pharyngeal smears from 116 children demonstrated that sensitivity and specifcity of detection of influenza virus A using device mariPOC in comparison with polymerase chain reaction made up to 93.8% and 99.0% correspondingly at total concordance of results of both techniques as 98.3%. At diagnosing of respiratory syncytial virus using device mariPOC parameters made up to 77.3%, 98.9% and 862% as compared with polymerase chain reaction. The sensitivity, specificity and total concordance of results of immune chromatographic tests BinaxNOW in comparison ofpolymerase chain reaction made up to 86.7%, 100% and 96.2% correspondingly at detection of influenza virus A and 80.9%, 97.4% and 91.6% correspondingly at detection of respiratory syncytial virus. In comparison with isolation technique in cell cultures sensitivity of system mariPOC and immune chromatographic tests proved to be in 1.3-1.4 times higher at detection of influenza virus A and in 1.7-2 times higher in case of isolation of respiratory syncytial virus. There is no statistically significant differences between diagnostic parameters received for mariPOC and immune chromatographic tests at diagnosing influenza virus A and respiratory syncytial viral infection.

All the volunteers completed a questionnaire, received a general and reproductive health examination by an andrologist, gave semen and blood samples. We found that the means were 34,69 - 106 spermatozoa/mL for sperm concentration and 39,6% for total motile spermatozoa. Among all volunteers studied 4,3% had azoospermia, 27,0% reported oligozoospermia, 68,1% - asthenozoospermia. There are no differences between Yakut and Slavs in anthropometric, hormonal or sperm measures, but height and ejaculate volume were higher in Slavs than Yakut men. The data obtained indicated high frequency of suboptimal quality of sperm in males of both nations from this Siberian region in comparison with other Russian regions or European countries.

The aim of the study was to analyze differences in the serum levels of 8 selected biological parameters between monoclonal gammopathy of undetermined significance (MGUS) and different stages of multiple myeloma (MM), potentially beneficial for distinguishing between the two conditions. The analyzed group of 131 subjects comprised 62 individuals with MGUS and 69 MM patients examined at the time of diagnosis. The serum levels were determined by a quantitative immunoradiometric assay (insulin-like growth factor 1, IGF-1) and quantitative sandwich enzyme immunoassay (osteopontin, OPN; endostatin, ES; macrophage inflammatory proteins 1α/ß, MIP-1α/ß; angiogenin, ANG; and interleukin 17, IL-17). The analysis showed a statistically significant difference in serum concentrations between MGUS and the symptomatic form of MM using the Durie-Salmon (D-S) staging system only in the cases of OPN and stages II and III (0.001 and MM. More benefit may be expected from analyses using multiparametric immunophenotyping of plasma cells and molecular biology methods including gene expression analysis and proteomics.

INTRODUCTION: This study aimed to measure serum levels of 12 selected parameters in patients with monoclonal gammopathy of undetermined significance (MGUS) and initial, asymptomatic phase of multiple myeloma (MM) to assess their potential benefit in differentiating both conditions. PATIENT SAMPLE AND METHODOLOGY: The analysed sample of 131 patients consisted of 62 patients with MGUS and 69 patients with MM fulfilling the criteria of the International Myeloma Working Group (IMWG). The following techniques were used to assess serum levels: quantitative immunoradiometry (bone-type alkaline phosphatase - bALP and insulin-like growth factor-1 - IGF-1), chemiluminescent enzyme immunochemical reaction (parathormone--PTH), quantitative sandwich enzyme immunoassay (osteopontin--OPN, endostatin (ES), macrophage inflammatory protein-1alpha/beta--MIP-1alpha/beta, angiogenin--ANG a IL-17). Pearson chi2 test and Mann-Whitney U-test were applied during the statistical analysis. RESULTS: The analysis showed statistically significant differences in serum concentrations between MGUS and symptomatic form of MM (Durie-Salmon (D-S) stage 2-3) for: albumin, beta2-microglobulin (beta2-M) and OPN (p = 0.0001 and < 0.0001); osteocalcin for stage 2 (p = 0.006) and MIP-1alpha for stage 3 patients (p = 0.0002). Using the International Staging System (ISS), statistically significant differences between MGUS and all stages of MM (1-3) were identified for albumin and OPN (p = 0.003 and 0.001 and 0.00009, respectively), stages 2 and 3 for beta2-M and ES (p = 0.015 and 0.0001, respectively), stage 2 for ANG (p = 0.014) and stage 3 for MIP-1alpha (p = 0.00001). Statistically significant differences between MGUS and initial, asymptomatic phase of MM (D-S stage 1) was found only for bALP (p = 0.01) and for albumin (p = 0.004) and OPN (p = 0.003) when ISS was applied. Renal function impairment (D-S substage B) showed in comparison to MGUS significant elevation of serum levels of beta2-M (p < 0.0001), OC (p = 0.011), IGF-1 (p = 0.014), OPN (p = 0.003), ES (p = 0.0001), MIP-1alpha (p = 0.0004) and ANG (p = 0.005) and for albumin (p < 0.0001), beta2-M (p < 0.0001) and OPN (p < 0.0001) only when compared to substage A. CONCLUSION: Only albumin and OPN showed useful even though non-specific potential to differentiate MGUS from all ISS stages of MM, beta2-M and ES for ISS stages 2 and 3, the other parameters differed for stage 2 (ANG), stage 3 (MIP-1alpha) only and stage 1 (bALP), stage 2 (OC) and stage 3 (MIP-1alpha) when D-S stratification was applied.

BACKGROUNDS: The aim of the study was to evaluate the serum levels of 18 selected parameters in monoclonal gammopathy of undetermined significance, and the initial, asymptomatic phase of multiple myeloma, also from the point of view of the potential contribution to the differentiation of these two units. MATERIALS AND METHODS: The analyzed 119-patient group consisted of 59 individuals with monoclonal gammopathy of undetermined significance and 60 patients with multiple myeloma assessed at the time of diagnosis before the start of the treatment. For the evaluation of serum levels we used radioenzyme assay (thymidine kinase), immunoradiometry (IGF-1), enzyme immunoassay (osteocalcin, osteoprotegerin, ICTP), electrochemiluminiscence (PINP), quantitative sandwich enzyme immunoassay (MIP-1 alpha and MIP-1 beta, IL-17, osteopontin, HGF, VEGF, angiogenin, endostatin, syndecan-1/CD138), and for the assessment of serum levels of free light chains kappa and lambda, the Freelite system. Statistical evaluation was done using the Pearson chi-quadrat test and the U-test according to Mann-Whitney (p < 0.05). RESULTS: Statistically significant differences between monoclonal gammopathy of undetermined significance and multiple myeloma were found in the case of serum levels of thymidine kinase (0.0002), ICTP (0.001), MIP-1 alpha (0.002), osteopontin (<0.0001), HGF (< 0.0001), syndecan-1 (<0.0001), and the kappa/lambda ratio (0.0002), while lower significance was found in the case of angiogenin (0.031) and endostatin (0.011). Statistically non-significant differences between multiple myeloma and monoclonal gammopathy of undetermined significance were within the serum levels of IGF-1, osteocalcin, bALP, PINP, OPG, MIP-1 beta, IL-17, parathormon and VEGF. CONCLUSION: Statistical analysis revealed significant differences between monoclonal gammopathy of undetermined significance and multiple myeloma in 9 of the 18 evaluated parameters. However, due to the significant overlapping of the measured values, none of the parameters is unambiguously able to distinguish between the units. A certain contribution in the discrimination of multiple myeloma from monoclonal gammopathy of undetermined significance was found in markedly increased serum levels of thymidine kinase, MIP-1 alpha, osteopontin, HGF and significant pathology of the kappa/lambda index.

RESUMEN

Recent literature review is suggesting the need to renew the traditional concept of SHBG as only a transport protein. The new role of SHBG relates to the evidence of its specific receptor on the cell membrane and an alternative signal pathway for sexual steroids. This review points to the importance of simultaneous measurements of SHBG along with determinations of the total sexual steroid concentration and informs of some new areas where SHBG can be used in laboratory diagnostics. In obstetrics the low levels of SHBG in the first half of pregnancy were reported as a promising early risk marker of the later development of preeclampsia.

A relationship between heliogeophysical disturbances and exacerbations of cardiovascular disease was studied. The index of exacerbations of cardiovascular disease was a number of calls for ambulance in Yakutsk city to patients with hypertension, hypertension crisis, myocardial infarction, cerebral stroke. Statistical processing of the experimental data, using superposed epoch technique, has allowed defection of "pre-storm" (2-4 days before the maximum of geophysical disturbances) and "post-storm" (2-4 days after it) maxima in the dynamics of referring of patients with cardiovascular pathology for emergency care.

RESUMEN

The endometrium acquires the ability to implant a hatched blastocyst only within a specific time termed the receptive phase. Ovarian steroid hormones are essential for structural and functional changes that prepare the endometrium to be receptive. Pinopodes have been suggested to be markers of uterine receptivity. The aim of this study was to compare the pinopode expression and serum levels of ovarian steroid hormones in the mid-luteal phase of the natural cycle and in a "mock" cycle in the same subject. Sequentional endometrial biopsies within 48 hours were obtained from women in the mid-luteal phase (ovulation +5, ovulation +7) of the natural cycle and in the "mock" cycle (progesterone supplementation +5 and +7). Biopsies were examined under a scanning electron microscope for pinopode detection. The expression of pinopodes was similar in both cycles, where pinopodes covered about 5 % of the endometrial surface. The developmental stages were also similar with a slight increase of fully developed pinopodes in both samples in the "mock" cycles. Our findings suggest that hormonal preparation of the endometrium do not change the timing of pinopode expression.

UNLABELLED: The aim of the study was to establish the distribution of the lymphocyte populations in peripheral blood in pregnant women with antiphospholipid antibodies (aPL) and a history of recurrent pregnancy loss as well as to make a study on the influence of the intravenous IgG-treatment (IVIg) on the levels of antibodies and peripheral lymphocytes. MATERIALS AND METHODS: 14 pregnant women were investigated. The latter had two or more recurrent abortions and positive aPL--anticardiolipin (aCL) and/or anti-beta 2-Glycoprotein I (anti-beta 2-GPI). The lymphocyte populations were examined, using a flowcytometric method in peripheral blood before 6-7th gestational week and on the 10-14 day after the second IVIg infusion. 10 pregnant women wer used as controls who had no aPL antibodies and no pregnancy loss. RESULTS: 13 out of 14 pregnancies were successful Lymphocyte populations of the examined patients before the first infusion compared to the controls showed statistically significant differences for: T-lymphocyted Th(reduced) and CD3-CD56 + NK (increased) Significant increase after therapy was noticed for the levels of T-lymphocytes, and Th while the NK-cell populations were significantly reduced. CONCLUSIONS: aPL-positive pregnancies most probably were associated with elevated and activated peripheral NK-cell subpopulations. The low doses of IVIg successfully reduce the levels of aPL, as well as peripheral CD3-CD56 + NK cells.

OBJECTIVE: To verify if and to which extent the interaction with grapefruit juice can increase bioavailability of orally administered sexual steroids. DESIGN: Pilot pharmacokinetics study. SETTING: Department of Obstetrics and Gynecology and Institute of Pharmacology, Medical Faculty, Palacký University, Olomouc; Department of Nuclear Medicine, University Hospital, Olomouc. METHODS: 2 mg of estradiol valerate and 100 mg of micronized progesterone were given to eight healthy postmenopausal volunteers. Blood samples were collected at time 0, 2, 3, 5 and 24 hours after tablets application. The same trial was repeated a week later but tablets were swallowed with 200 ml of grapefruit juice. Serum levels of estradiol and progesterone were measured by RIA. Results were statistically evaluated using the Wilcoxon's nonparametric paired test. RESULTS: Though grapefruit juice on average slightly increased serum levels of estradiol (E2) and progesterone, this increase reached statistical significance only for the E2 level 24 hours after application of tablets. The mean area under curve (AUC) of estradiol rose significantly to 117%. The even greater increase in the mean AUC of progesterone (to 125%) was not statistically significant because of marked individual variability of response. CONCLUSIONS: Our results suggest that grapefruit juice may increase bioavailability of orally administered estradiol and progesterone. The response varies markedly between individuals. This observation may be of some importance also for users of OC and HRT.
