RESUMEN

Glucose metabolic disorders, prevalent in numerous metabolic diseases, have become a pressing global public health concern. Artemisinin (ART) and its derivatives, including artesunate (ARTs) and artemether (ARTe), have shown potential as metabolic regulators. However, the specific effects of ART and its derivatives on glucose metabolism under varying nutritional conditions and the associated molecular mechanisms remain largely unexplored. In this study, we examined the impact of ART, ARTs, and ARTe on glucose homeostasis using a mouse model subjected to different dietary regimens. Our findings revealed that ART, ARTs, and ARTe increased blood glucose levels in mice on a normal-chow diet (ND) while mitigating glucose imbalances in high-fat diet (HFD) mice. Notably, treatment with ART, ARTs, and ARTe had contrasting effects on in vivo insulin signaling, impairing it in ND mice and enhancing it in HFD mice. Moreover, the composition of gut microbiota underwent significant alterations following administration of ART and its derivatives. In ND mice, these treatments reduced the populations of bacteria beneficial for improving glucose homeostasis, including Parasutterella, Alloprevotella, Bifidobacterium, Ileibacterium, and Alistipes. In HFD mice, there was an increase in the abundance of beneficial bacteria (Alistipes, Akkermanisia) and a decrease in bacteria known to negatively impact glucose metabolism (Coprobacillus, Helicobacter, Mucispirillum, Enterorhabdus). Altogether, ART, ARTs, and ARTe exhibited distinct effects on the regulation of glucose metabolism, depending on the nutritional context, and these effects were closely associated with modifications in gut microbiota composition.

The COVID-19 pandemic has severely affected healthcare worldwide and has led to the excessive use of disinfectants and antimicrobial agents. However, the impact of excessive disinfection measures and specific medication prescriptions on the development and dissemination of bacterial drug resistance during the pandemic remains unclear. This study investigated the influence of the pandemic on the composition of antibiotics, antibiotic resistance genes (ARGs), and pathogenic communities in hospital wastewater using ultra-performance liquid chromatography-tandem mass spectrometry and metagenome sequencing. The overall level of antibiotics decreased after the COVID-19 outbreak, whereas the abundance of various ARGs increased in hospital wastewater. After COVID-19 outbreak, blaOXA, sul2, tetX, and qnrS had higher concentrations in winter than in summer. Seasonal factors and the COVID-19 pandemic have affected the microbial structure in wastewater, especially of Klebsiella, Escherichia, Aeromonas, and Acinetobacter. Further analysis revealed the co-existence of qnrS, blaNDM, and blaKPC during the pandemic. Various ARGs significantly correlated with mobile genetic elements, implying their potential mobility. A network analysis revealed that many pathogenic bacteria (Klebsiella, Escherichia, and Vibrio) were correlated with ARGs, indicating the existence of multi-drug resistant pathogens. Although the calculated resistome risk score did not change significantly, our results suggest that the COVID-19 pandemic shifted the composition of residual antibiotics and ARGs in hospital wastewater and contributed to the dissemination of bacterial drug resistance.

Essential oils (EO) are promising feed additives for their antibacterial, antioxidant, and immune-enhancing abilities with low toxicity. Carvacrol, thymol, and cinnamaldehyde are commonly used to synthesize EO. However, few studies focus on combining these three EO in early-weaned piglets. In the present study, 24 piglets weaned at 21 d of age were randomly divided into 2 groups (6 replicate pens per group, 2 piglets per pen). The piglets were fed a basal diet (the control group) and a basal diet supplemented with 400 mg/kg EO (a blend consisting of carvacrol, thymol, and cinnamaldehyde, the EO group) for 28 days. At the end of the experiment, one piglet per pen was randomly chosen to be sacrificed. Growth performance, hematology, plasma biochemical indices, antioxidant capacity, intestinal epithelial development and immunity, colonic volatile fatty acids (VFA), and microbiota were determined. The results indicated that the diet supplemented with EO significantly improved average daily feed intake (ADFI, p < 0.01) and average daily gain (ADG, p < 0.05) in the day 0 to 28 period. EO supplementation led to a significant decrease in plasma lysozyme (p < 0.05) and cortisol levels (p < 0.01). Additionally, EO significantly promoted jejunal goblet cells in the villus, jejunal mucosa ZO-1 mRNA expression, ileal villus height, and ileal villus height/crypt depth ratio in piglets (p < 0.05). The ileal mucosal TLR4 and NFκB p-p65/p65 protein expression were significantly inhibited in the EO group (p < 0.05). Colonic digesta microbiota analysis revealed that bacteria involving the Erysipelotrichaceae family, Holdemanella genus, Phascolarctobacterium genus, and Vibrio genus were enriched in the EO group. In conclusion, these findings indicate that the EO blend improves ADG and ADFI in the day 0 to 28 period, as well as intestinal epithelial development and intestinal immunity in early-weaned piglets, which provides a theoretical basis for the combined use of EO in weaned piglets.

The occurrence of antibiotics in the feces of elderly individuals in Shenzhen, China, was investigated by monitoring 78 compounds to understand the adverse effects and its association with antibiotic residues in animal products collected from local markets. In total, 18 compounds belonging to 5 classes of antibiotics were identified in 74 of 140 fecal samples. Furthermore, 17.9% of the fecal samples contained at least two antibiotics, and 14.3% of the samples showed antibiotic concentrations higher than 100 µg/kg. Cephalothin exhibited the highest detection frequency (22.1%), followed by azithromycin (15.7%) and tilmicosin (12.9%). Oxytetracycline, norfloxacin, and azithromycin showed extremely high concentrations (> 1000 µg/kg). Eight antibiotics were detected in the animal products, with detection frequencies ranging from 4.8 to 40.0%. Five antibiotics exhibited similar detection frequencies and strong correlations between the human fecal and animal product samples. Health risk assessment based on hazard quotients showed that ciprofloxacin in animal products and human feces posed a medium and high risk, respectively. The hazard quotients of oxytetracycline, norfloxacin, and azithromycin in the feces were greater than 1, indicating a high health risk. These findings suggest that the elderly individuals were frequently exposed to antibiotics via the food chain and faced health risks posed by these antibiotics.

Bisphenol F (BPF) is a widely used bisphenol A (BPA) substitute plastic additive that has attracted increasing public concerns due to its potential toxic effects on animal and human health. Although previous studies have indicated that BPF might have harmful effects on metabolic homeostasis, the systematic effects of BPF on glucose disorders remain controversial. In this study, mice fed a normal chow diet (ND) and high-fat diet (HFD) were administered BPF at a dose of 100 µg/kg of body weight, and glucose metabolism was monitored after both short- and long-term treatment. Little change in glucose metabolism was observed in BPF-treated ND mice, but improved glucose metabolism was observed in BPF-treated HFD mice. Consistently, BPF treatment led to increased insulin signalling in the skeletal muscle of HFD mice. Additionally, liver metabolite levels also revealed increased carbohydrate digestion and improved TCA cycle progression in BPF-treated HFD mice. Our results demonstrate that sustained BPF exposure at an environmentally relevant dosage may substantially improve glucose metabolism and enhance insulin sensitivity in mice fed a high-fat diet.

Phorate is a systemic, broad-spectrum organophosphorus insecticide. Although it is commonly used worldwide, phorate, like other pesticides, not only causes environmental pollution but also poses serious threats to human and animal health. Herein, we measured the blood glucose concentrations of high-fat-diet-fed mice exposed to various concentrations of phorate (0, 0.005, 0.05, or 0.5 mg/kg); we also assessed the blood glucose concentrations of high-fat-diet-fed mice exposed to phorate; we also assessed the distribution characteristics of the resistance genes in the intestinal microbiota of these mice. We found that 0.005 and 0.5 mg/kg of phorate induced obvious hyperglycaemia in the high-fat-diet-fed mice. Exposure to phorate markedly reduced the abundance of Akkermansia muciniphila in the mouse intestine. The resistance genes vanRG, tetW/N/W, acrD, and evgS were significantly upregulated in the test group compared with the control group. Efflux pumping was the primary mechanism of drug resistance in the Firmicutes, Proteobacteria, Bacteroidetes, Verrucomicrobia, Synergistetes, Spirochaetes, and Actinobacteria found in the mouse intestine. Our findings indicate that changes in the abundance of the intestinal microbiota are closely related to the presence of antibiotic-resistant bacteria in the intestinal tract and the metabolic health of the host.

OBJECTIVES: China banned the use of colistin as animal growth promoter in April 2017. Herein, we report the prevalence of mcr-1 in the intestine of healthy humans and risk factors associated with mcr-1 carriage after the implementation of the ban. METHODS: We recruited 719 healthy volunteers from Shenzhen City from 1 March 2018 to 31 December 2019 to investigate the prevalence of mcr-1 in human intestine, and undertook a case-control study to ascertain the risk factors associated with the mcr-1-positive population. A further comparative study was conducted to identify differences between genetic characteristics of mcr-1-positive and mcr-1-negative Escherichia coli. RESULTS: Overall, 56 (7.8%, 95% CI 5.9%-10.0%, n = 719) individual faecal samples were positive for mcr-1, and prevalence of mcr-1 among individuals in 2019 (2.4%, 95% CI 8.7%-15.0%, 7/294) was significantly lower than that in 2018 (11.5%, 95% CI 1.0%-4.8%, 49/425) (p < 0.0001). After the colistin ban, animal-derived food (pork and chicken meat) was no longer a risk factor for mcr-1 carriage in human intestine, whereas a higher intake of fish and seafood (>75 g/day) and whole grains (>150 g/day) was associated with higher and lower risk of mcr-1 carriage, respectively (OR 2.175, 95% CI 1.047-4.517; OR 0.045, 95% CI 0.004-0.567). Compared with mcr-1-negative E. coli, the mcr-1-positive E. coli had different patterns of resistance genes and genetic heterogeneity. CONCLUSIONS: Our study implicates aquatic food as beeing associated with mcr-1 carriage in the healthy population, even after the ban on colistin. Dietary modification (e.g. whole grains) may help to combat mcr-1-positive bacterial colonization of the gut.

Although non-phthalate plasticizers are widely used in various consumer products as substitutes for phthalates, currently little is known about their environmental occurrence and the risks of human exposure. To characterize the pollution and human exposure risk in indoor environments, house dust samples collected from 83 households in Guangzhou were analyzed for twelve typical non-phthalate plasticizers. Results showed that dioctyl terephthalate (DEHT), trioctyl trimellitate (TOTM), acetyl lemon tributyl ester (ATBC), heptylnonyl adipate, di(2-ethylhexyl) adipate, and trioctyl trimellitate were detected in all samples. The total concentrations of non-phthalate plasticizers ranged from 22.4 to 615 µg ·g-1 with a median of 101 µg ·g-1. DEHT had the highest concentrations (9-487 µg ·g-1), followed by TOTM (0.3-87 µg ·g-1) and ATBC (1.7-50 µg ·g-1). Daily intakes via dust ingestion for adults and children were estimated based on the dust concentrations. The total daily intake of children was 367 ng ·(kg ·d)-1, which was 8-9 times higher than that of adults[43 ng ·(kg ·d)-1]. Further risk assessment through the hazard quotient approach showed that the exposure of adults and children to non-phthalic plasticizers via dust ingestion was within an acceptable risk range.

Bisphenol S (BPS), an industrial chemical that is a structural analogue of bisphenol A, has been widely reported to be involved in various biological processes. Epidemiological studies have demonstrated that exposure to BPS is associated with dysglycaemia-related health outcomes. The role of BPS in glucose metabolism, however, remains controversial. In this study, we aimed to investigate the effects of chronic exposure to environmentally relevant concentrations of BPS on glucose metabolism in different nutritionally conditioned mice. Our results revealed that 1-month exposure to a BPS dosage of 100 µg/kg bw slightly increased the insulin sensitivity of normal diet-fed mice, and that this effect was enhanced after 3-month exposure. It was also found that BPS exposure attenuated insulin resistance and reduced gluconeogenesis in high-fat diet-fed mice. Consequently, the concentrations of hepatic metabolites related to glucose metabolism were altered in both groups of mice. Moreover, thyroid hormone signalling was disrupted after BPS administration in both groups of mice. Taken together, our results demonstrated that chronic exposure to environmentally relevant concentrations of BPS exerted an unexpected hypoglycaemic effect in mice of different nutritional statuses, and that this was partly attributable to disrupted thyroid hormone signalling.

Despite phthalate monoesters (mono-PAEs) being commonly recognized as metabolic products of phthalate diesters (di-PAEs), investigations on their environmental occurrences, particularly in indoor environments, remain limited. The present study demonstrated the presence mono-PAEs, along with a variety of di-PAEs, in house dust collected from 83 South China families. Among 15 target mono-PAEs, monobutyl phthalate (median concentration, 21.54 µg/g) dominated over other mono-PAEs in indoor dust, followed by monoethylhexyl phthalate (9.44 µg/g), monoisobutyl phthalate (5.14 µg/g), monomethyl phthalate (MMP; 2.05 µg/g), and several others. The total concentrations of detectable mono-PAEs (median, 45.40 µg/g) constituted an average of 6.7 ± 3.7% of the total concentrations of their parent diesters in the same dust. Molar concentration ratios of mono-PAEs to their respective di-PAEs varied greatly among chemicals (median, 0.001-3.1), with the highest ratios determined for the MMP/dimethyl phthalate and mono-/diisopropyl phthalate pairs (i.e., 3.1 and 1.5, respectively). In addition, no significant associations were observed between dust-associated mono- or di-PAEs and urinary mono-PAEs detected in both children (n = 48) and adult participants (n = 41). We hypothesized that mono-PAEs in dust could originate from different sources (e.g., impurities in di-PAE formulas, degradation from di-PAEs, and direct application as commercial additives), while the relative importance of various origins could differ between chemicals. Our findings demonstrate broad occurrences of mono-PAEs in indoor environments, but future studies are needed to better elucidate their sources, fate in indoor and outdoor environments, and potential human health risks.

The present study investigated the occurrence of 20 organophosphate esters (OPEs) in house dust from 51 South China homes and the risks of human exposure to OPEs via two pathways: dust ingestion and hand-to-mouth contact. In addition to several traditional OPEs, five out of six novel OPEs, including bisphenol A bis(deiphenyl phosphate) (BPA-BDPP), t-butylphenyl diphenyl phosphate (BPDPP), cresyl diphenyl phosphate (CDP), isodecyl diphenyl phosphate (IDDPP), and resorcinol-bis(diphenyl)phosphate (RDP), were frequently detected in house dust (median concentration: 59.7-531 ng/g). Eight of the 20 target OPEs were frequently detected in hand wipes collected from adults and children ( n = 51 and 31, respectively), which in combination (referred to as Σ8OPEs) had a median mass of 76.9 and 58.9 ng, respectively. Increasing dust concentrations of Σ8OPEs or three individual substances among these eight OPEs, including tris(1-chloro-2-propyl) phosphate (TCIPP), tris(1,3-dichloro-2-propyl) phosphate (TDCIPP), and triphenyl phosphate (TPHP), were strongly associated with their levels in children's hand wipes ( p < 0.05 in all cases). By contrast, in adults' hand wipes only TPHP exhibited a marginally significant association with dust concentrations ( p = 0.04). Levels of Σ8OPEs in hand wipes from children, but not adults, were inversely influenced by hand washing frequency ( p = 0.002), while indoor temperature was inversely associated with hand wipe levels of Σ8OPEs from both children and adults ( p = 0.01 and 0.002, respectively). Exposure estimation suggests that hand-to-mouth contact represents another important pathway in addition to dust ingestion and that children are subjected to higher OPE exposure than adults.

To understand human exposure to dust-associated flame retardants in the biggest metropolitan area (city of Shanghai) of East China, our study determined a suite of legacy and emerging flame retardants in dust from dwellings, cars, and university computer labs. The results exhibited a consistent dominance of organophosphate flame retardants (OPFRs) over polybrominated diphenyl ethers (PBDEs) and other alternative flame retardants (AFRs) regardless of microenvironments. In addition to OPFRs, some alternative flame retardants, such as decabromodiphenyl ethane (DBDPE), 2-ethylhexyltetrabromobenzoate (EH-TBB), bis(2-ethylhexyl)-3,4,5,6-tetrabromobenzoate (BEH-TEBP), and 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), were also frequently detected. Among them, DBDPE exhibited concentrations comparable to those of PBDEs. Comparison with international studies indicated that concentrations of ∑PBDEs (0.2-12.3 µg/g dry weight or dw) and ∑OPFRs (3.8-165.5 µg/g dw) from Shanghai dwellings (bedroom and living room) were generally in the middle of concentration ranges reported worldwide, whereas elevated DBDPE concentrations (0.1-9.5 µg/g dw) was observed compared with most other countries or regions. OPFR compositions in house dust from this study also differed from those from many other countries. This suggested inter-regional differences in market demands on the quantities and types of flame retardants. Human intake estimation suggested elevated exposure for toddlers when compared with adults, although the daily intake estimations of individual flame retardants were generally 2-4 orders of magnitude lower than the reference doses. The findings from this preliminary study developed a baseline for future evaluation of the sources and fate of emerging flame retardants and related human exposure risks in East China.

House dust associated with organic pollutants is not only a potential source of pollutants to the outdoor environment, but also a source to human exposure. The present study investigated the occurrence and concentrations of organophosphate flame retardants (OPFRs) in house dust collected from South China dwellings (n = 20). The results revealed a universal presence of most target OPFRs in house dust, with concentrations of ΣOPFRs ranging from 2.06 to 19.95 µg/g. The median concentration of ΣOPFR (9.20 µg/g) was one order of magnitude greater than that of polybrominated diphenyl ethers (0.80 µg/g). The composition of OPFR chemicals in house dust was dominated by chlorinated OPFRs, such as tris(1,3-dichloro-2-propyl) phosphate (TDCPP) and tris(1-chloro-2-propyl) phosphate (TCPP). This compositional pattern was different from what has been reported in indoor dust from many other countries, where tris(2-butoxyethyl) phosphate (TBEP) was generally dominant. The daily intake of OPFRs by adults was estimated to be 1.6 and 4.2 ng/kg body weight/day under average and high exposure scenarios, respectively, and 31.7 and 127 ng/kg body weight/day for toddlers.
