RESUMEN

Glucose concentration in the saliva is increased in type 1 diabetes mellitus. This parameter directly correlates with markers of the disease in the blood serum. Increased concentration of 8-oxo-2'-deoxyguanosine (8-OHdG) and diene conjugates, markers of oxidative stress, and reduced activities of superoxide dismutase and catalase were also observed in this pathology. Correlation analysis revealed a strong positive correlation between glucose concentration and the levels of oxidative stress markers and a negative correlation between activity of antioxidant enzymes and glucose concentration. The results indicate that the level of 8-OHdG, diene conjugates, and superoxide dismutase and catalase activities can serve as diagnostic markers of pathophysiological changes in the body in type 1 diabetes mellitus.

RESUMEN

Genetic diversity and colonization routes of noble salmons were studied using a partial nucleotide sequence of the mitochondrial COI gene. The brown trout S. trutta, which is the most ancient species of the genus, was concluded to originate from the modern southeastern Pontic-Caspian area, which is currently inhabited by members of the subspecies S. trutta oxianus. Migrating westward while the Paratethys was in existence (5-34 million years ago), species of the genus colonized ancient water bodies in the modern Mediterranean basin and formed many isolated populations that survived desiccation of the Mediterranean Sea (5-6 million years ago). The Strait of Gibraltar mediated brown trout migrations to Northern Europe; the subspecies S. trutta trutta belongs to a relatively young phylogenetic lineage of the species. A separate brown trout lineage, currently classified as the subspecies S. trutta labrax, formed most likely in the area of the modern Danube basin, which was a relatively separate part of the Paratethys and was sometimes isolated as the Pannonian Lake. A highly divergent phylogenetic lineage of Atlantic salmon (S. salar) haplotypes originates from a haplotype of the brown trout that inhabited the area of the modern Strait of Gibraltar.

AIM: Determination of the level of zinc and its fractions, as well as the enzyme neutrophilic elastase and albumin in persons suffering from chronic obstructive pulmonary disease (COPD), as well as smoking actively and passively. MATERIALS AND METHODS: The study involved 30 patients with a diagnosis of COPD and 90 healthy persons (60 of them smoking at the present time, 30 - no) who underwent spirometry and determination of zinc levels and its pools, albumin, and neutrophil elastase. All data are subject to statistical processing. RESULTS: It is determined that the studied parameters differ significantly in the groups of smokers with COPD, healthy smokers and non-smokers, and correlate with the volume of forced exhalation for 1 second as a percentage of the due. CONCLUSION: The revealed regularities make it possible to consider the indicator "share of bound zinc fraction" introduced in the study as a screening criterion in diagnosing COPD in smokers.

Population aging is a global social phenomenon that affects the most important health indicators of the population. The older generation is particularly vulnerable in the current conditions of urbanization and the associated environmental situation. The comprehension of indicators of disability and morbidity of the population, the study of the factors that determine the negative processes of health damage, taking into account regional features of these processes are necessary for the planning of preventive measures. In the age structure of the primary disability of the adult population in the Voronezh region elderly disabled people make up 53,3%, middle and young people with disabilities are less - 30,1 and 16,6%, respectively. These indicators are higher than the average Russian. The results of non-carcinogenic risk and primary morbidity analysis in population of Levoberezhniy district of Voronezh under permanent anthropotechnogenic load have presented. It has been established that non-carcinogenic risk coefficients were bigger that acceptable level (HQ>1) by for nitrogen dioxide, acrylonitrile, acrolein, 1,3-butadiene, copper oxide, chromium+6. Unacceptable level of non-carcinogenic risk is typical for respiratory organs pathology, pathologies of cardiovascular system and endocrine system. Analysis of the primary morbidity of these nosological forms shows their high distribution in adult population morbidity. Under rehabilitation measures in patients with respiratory organs diseases, circulatory system and endocrine system diseases it is necessary to consider anthropotechnogenic load and relationship between agents and disease development.

RESUMEN

DNA fragmentation, caspase-1 and caspase-3, aconitate hydratase (AH) activities, and citrate content have been investigated in the blood of patients with type 2 diabetes mellitus complicated by steatohepatitis. These indicators of apoptotic processes intensity and oxidative stress development were estimated after basic treatment and a combined therapy including epifamin. Before treatment DNA fragmentation blood leukocytes, decrease of AH activity and increase of caspases activities in the serum of patients were detected. Treatment with epifamin provided more pronounced changes in the investigated parameters towards control values as compared to basis therapy. Epifamin caused a positive effect on the citrate content in the serum of patients. Epifamin inclusion to the basic therapy was accompanied by a more pronounced changes towards the normal values of such biochemical parameters as ALT, AST, b-lipoproteins, cholesterol, fasting glucose and postprandial glucose levels. All these changes may be obviously attributed to epifamin-induced correction of the melatonin level and manifestation of adaptogenic properties and antioxidant effects of the hormone.

The purpose of the investigation was in the study of the influence of aerotechnogenic load on children and adolescents as a cause of the occurrence of diseases and exacerbation of the course of the chronic disease. There was performed an analysis of the daily air pollution by nitrogen dioxide, particulate matter, soot, sulfur dioxide, formaldehyde, phenol, nitrogen oxide, carbon monoxide, and appealability for medical aid of children and young people residing in the area with the high level of aerotechnogenic load. In the course of the work the correlation analysis was used in three ways: when the date of appealability for medical care coincided with daily indices of the level of air pollution; with delayed of 1 day (shift of data was carried out for 1 day); with delayed of 2 days (shift of data was carried out for 2 days). The number of examined children's population under the age of 14 years and adolescents up to 17 years accountedfor 7303 children, the study period was 1 year. In the course of the work there was established a causal relationship between the level of air pollution and the appealability of the children for the medical care: the pair correlation coefficients for certain classes of diseases amounted from 0.16 to 0.82. It is revealed that the highest number of requests for medical assistance was established to be accounted for the date following the date after exposure to elevated concentrations of pollutants in ambient air Therefore, the aerotechnogenic load, to which the human body is exposed to throughout life, from childhood can be considered as a stress factor that reduces the compensatory capacities.

RESUMEN

Type 2 diabetes mellitus complicated by steatohepatitis is accompanied by a decrease in aconitate hydratase activity, increase in the content of diene conjugates, decrease in the concentration of α-tocopherol, and change in the citrate level in the blood serum of patients, which is evidence of the development of oxidative stress as a result of the intensification of free radical oxidation of biosubstrates and decreasing degree of antioxidant defense of the organism. Combined therapy with melaxen provided a more significant change of the investigated parameters toward the norm (on the average by 36%, p 0.05) in comparison with basic treatment. This result was evidently associated with implementation of the antioxidant effect of melatonin, the level of which is corrected under the action of the drug employed.

A comparative study of the activity of caspase-1 and caspase-3, the glutathione antioxidant system and NADPH-generating enzymes (glucose-6-phosphate dehydrogenase and NADP-isocitrate dehydrogenase) and a study of DNA fragmentation in the blood serum of patients with chronic alcoholic hepatitis during basic treatment and combination therapy including melaxen have been carried out. It was found that the blood serum level of reduced glutathione, which decreases in pathology, increased more significantly in patients receiving melaxen as compared to the group of patients receiving the standard treatment. More significant changes in the activity of caspase-1 and caspase-3, glutathione reductase, glutathione peroxidase, glutathione-S-transferase, glucose-6-phosphate dehydrogenase and NADP-isocitrate dehydrogenase toward the control values were observed during the combination therapy. The correction in the melatonin level under the influence of melaxen apparently had a positive effect on the free-radical homeostasis in patients, which resulted in more pronounced changes in the investigated parameters towards the normal values as compared to the basic treatment. RESUMEN

Combined treatment including melatonin resulted in the improvement of general conditions in 81.7% of the patients with drug-induced hepatitis compared with 66.5% after standard therapy including hepatoprotectors. Normalization of biochemical characteristics was more pronounced after treatment with melatonin that caused no side effects as shown by laboratory and instrumental studies.

RESUMEN

A combined therapy with melaxen led to a decrease in the activity of gamma-glutamyl transpeptidase and the level of diene conjugates in blood serum of patients with the drug-induced hepatitis developing on the background of administration of antituberculous preparations. These changes are indicative of pronounced antioxidant and hepatoprotective properties of the drug. In addition, there was a decrease in the content of citrate and a change in the activity of aconitate hydratase toward a normal level, which reflects a decrease in the degree of pathologic oxidative stress development and is evidence of the antiradical effect of melaxen.

Methods of light optical microscopy were used to explore histoarchitectonics, topography and tinctorial properties of the extracellular phase of fibers of jejunum wall intersticium in Mongolian gerbils following 12-day orbital flight aboard Foton-M3, ground-based simulation of the spaceflight factors in the KONTUR-L facility, and in the group of biological control Postflight destructive changes were found in reticulin fibers (type-III collagen) of villi stroma, intercrypt intersticium and submucosa. Local acidophilia and fiber homogenization formed in type I collagen present in the intestinal subserous layer, muscular layers endomysium and submucose against the background of progressing edema and arterial, venous plethora and lymphostasis. Elastic component of the intersticium was disarranged in the structures of internal elastic membrane of submucous vessels, fragmented and partly reduced. Simulation of the orbital factors, except for microgravity, in the KONTUR-L facility called forth similar, although less often and diffuse, changes in intersticium fibers. The results of examination of intestinal intersticium fibers in the vivarium control gerbils discovered expressed species characters that should be taken into account by investigators, especially when comparing with data obtained from other animal species.

RESUMEN

Activity of the glutathione antioxidant system has been studied in patients with drug-induced hepatitis treated with standart base therapy and combined with epifamin therapy. In blood serum of patients before treatment the decrease of reduced glutathione (GSH) level and the increase of glutathione peroxidase (GP) and glutathione reductase (GR) activities versus control were observed. Combined treatment with epifamin changed GSH level to the normal values. Treatment with epifamin was accompanied by pronounced increase in GP and GR activities compared with standard therapy. Activities of the NADPH-generated enzymes, glucose-6-phosphate dehydrogenase and NADP-isocitrate dehydrogenase, which decreased at this pathology, also demonstrated a more significant increase than standart treatment.

Methods of light-optical microscopy, morpho- and cytometry and histochemisty were used to study the epithelial lining of herbal's small gut after 12-day flight in biosat "Foton M3". Changes in mucous coat histoarchitectonics included branching of villi and cystic lumps lined with prismatic epithelium. Shortening of the mucous membrane villi was accompanied by reduction of prismatic epithelium height, increase in the number of goblet cells and change of their dislocation, stimulation of excretion of biosynthesis products on the brush border surface pointing to impairment of the interstitial barrier function. Nothing evidenced change in mitotic index of the crypts epithelium in animals of the vivarium control, in the Kontur facility simulating the flight condition, and in the space flown herbals. There was a minor individual variability of changes in the epithelium mucous membrane due to the simulated flight factors.

Analysis of the maturation and spawning times of the Black Sea brown trout bred at the fish-farming plants and inhabiting natural waterways of the Northwestern Caucasus has demonstrated a considerable variation depending on environmental conditions, first and foremost, temperature. This fact, as well as the analysis of literature data, suggests that the duration and timing of the spawning season cannot be used as self-sufficient criteria for identifying species of the genus Salmo.

RESUMEN

Intensity of free radical oxidation processes and superoxide dismutase activity at patients with drug-induced hepatitis at the combined therapy with melaxen or epifamin. Free radical homoeostasis state at patients with drug-induced hepatitis being on standard treatment, including hepatoprotectors, and the combined therapy with melaxen or epifamin has been investigated. Biochemiluminescence parameters in blood serum point out to intensification of free radical processes and inhibition of antioxidant defense of organism, at this time superoxide dismutase activity in blood serum increased. After leading of standard therapy a decline of oxidative stress expression was observed. Incorporation to base treatment of melaxen or epifamin accompanied by correction of antioxidant status of organism and provided more expressed antioxidant effect, that confirmed by changing of biochemiluminescence parameters and superoxide dismutase activity. Obviously, it is related to antiradical properties of melatonin the level of which is corrected under the action of these preparations.

The paper compares and contrasts the results of measuring the hepatocyte cytoplasm area and RNA content in 35 gerbils in three series of experiments, i.e. the vivarium control, modeled space flight (synchronous control) and exposure to the factors of 12-d Foton-M3 orbital flight. Central, intermediate and peripheral zones of hepatic lobes were subjected to histological and histochemical analyses to measure the hepatocyte cytoplasm area; the RNA content was determined from the level of cytoplasm basophilia after azure staining. Cytometric and cytophotometric investigations were performed using image analyzer Video-7-Test-Morpho. In the vivarium animals, hepatocytes with the largest cytoplasm localized predominantly in the intermediate and central zones of the lobes. Judging from the results of microdensitometry, the RNA content was particularly high in binucleate hepatocytes of the intermediate zone. In the synchronous control, hepatocytes tended to grow in size, in the peripheral zone specifically, whereas RNA content was largely equal no matter hepatocyte topography. After space flight, cytoplasm enlargement transcended this process in the vivarium animals. The cytoplasm RNA content along the entire liver parenchyma made a significant decrease equally as compared with the vivarium and synchronous control animals.

Morphofunctional state of hepatocytes nuclear apparatus was analyzed in the liver of Mongolian gerbils Meriones unguiculatus returned from 12-d space flight of Foton-M3 (SF) and their vivarium and ground synchronous controls. Volume, ploidy and number of hepatocyte nuclei, nucleolus dimensions and number as well as contacts with karyolemma were determined in the central, intermediate and peripheral areas of the liver classical lobe. Also, total number of mitoses and amitoses was determined in the liver parenchyma. The vivarium control animals displayed specifics of the nucleus apparatus structure that depended on intralobe topography. Based on the selected criteria, high functional activity was characteristic of cells in the intermediate area. According to the criteria, nuclear apparatus in the synchronous control tended to down the functional activity The adaptive adjustment of nuclei in SF seemed to have been initiated by changes in the hepatic blood flow: volumes of hepatocyte nuclei and nucleoli increased as did the number of nuclei in cell, whereas ploidy made a decrease, especially in the intermediate area. Under the SF conditions, particularly important compensatory mechanism for the liver function was intensification of amitosis and consequent increase of the population of dinuclear hepatocytes.

The work purpose was research of degree of free-radical processes intensity and catalase activity in blood serum of patients with a thyreotoxicosis syndrome at traditional treatment and the combined therapy with epifamin. Patients (n=25) have been divided into 2 groups: the first group of patients--12 persons who are on traditional treatment (antithyreoid drugs, beta-adrenoblockers); the second group--13 persons, are padding to basic therapy received epifamin on 1 tablet (10 mg) 3 times a day 10-15 minutes prior to meal within 7 days. Patients with thyreotoxicosis syndrome had an intensification of free-radical oxidation that was traced at measurement of biochemiluminescence parameters in blood serum. At the combined therapy with epifamin less expressed intensity of free-radical processes, and also increasing of the general antioxidant activity of an organism that did not descend at traditional treatment was observed. At carrying out of standard therapy of thyreotoxicosis syndrome, including application of antithyreoid drugs and beta-adrenoblockers, catalase activity decreased, that unfavorable affected antioxidant potential of an organism. Glycogen cytochemistry and distribution in hepatocytes of the classic liver lobules were studied in three groups of gerbils Meriones unguiculatus: vivarium, synchronous control and flown in the 12-d Foton-M3 mission. The control animals were shown to have the central glycogen distribution with a large pool of polysaccharides found in hepatocytes of the pericentral and intermediate lobules and a small pool in the periportal area. Glycogen in hepatocyte plasm was within the physiological norm in the alpha- and beta-granules, typically localized on the cell periphery. Exposure to the spaceflight conditions decreased significantly glycogen concentrations in each functional region of the hepatic lobules and reduced the gradient of polysaccharide distribution from the portal triads toward the central vein. In parallel, high glycogen heterogeneity formed in adjacent hepatocytes and loci. The presence of glycosomes evidenced disturbance of carbohydrates metabolism. In addition, intracellular topography of glycogen granules in cytoplasm was altered. Trends of glycogen in gerbils of the synchronous control were similar to the space flown animals but much less pronounced.

Experimental thyrotoxicosis in rats is accompanied by the increase of serum alanine aminotransferase (AlA), aspartate aminotransferase (AsA), creatine kinase-MB (CK-MB) activities and content of primary products of lipid peroxidation--conjugated dienes--in liver, heart and blood. This suggests impairments in these organs accompanying free radical processes intensification. Administration of melatonin decreased AlA, AsA and CK-MB activities and CD level decreased. Thyrotoxicosis increased catalase activity in liver, heart and blood. Exogenous melatonin decreased specific activity ofcatalase in blood and in heart in comparison with animals subjected to hyperthyroidism. However, some increase of catalase specific activity (approximately 15%) was observed in liver. alpha-Tocopherol content, raising in rat tissues in thyrotoxicosis development conditions, decreased after melatonin treatment. Thus, exogenous melatonin is capable to reduce lipid peroxidation intensity at thyrotoxicosis and to act as an adoptogen, regulating free radical homeostasis.
