RESUMEN

Glioblastomas (GBs) are one of the most aggressive and invasive intracranial cancers. Recently, it has been postulated that, among other factors, the hedgehog (HH) pathway may be a key factor in this phenomenon. Moreover, it has been reported that small-size silver nanoparticles (AgNPs) are characterized by a high cytotoxic effect towards GBs. However, their effect on the sonic hedgehog (SHH) pathway has never been demonstrated in any cancer cells. Therefore, the aim of the present study was to evaluate the impact of the anti-proliferative properties of 5-nm AgNPs on the SHH pathway in the GB cell line (U-87MG) in vitro. The results showed a time- and dose-dependent decrease in the metabolic activity in the U-87MG cells treated with AgNPs, with IC50 reaching 30.41 and 21.16 µg/mL after 24 h and 48 h, respectively, followed by an increase in the intracellular reactive oxygen species (ROS) level. The co-treatment of the cells with AgNPs and Robotnikinin (SHH inhibitor) abolished and/or strengthened the effect of AgNPs, especially on the SHH mRNA levels and on the PCNA, PTCH1, Gli1, and SUFU protein levels. Interestingly, no changes in the level of ERK1/2, Akt, and SRC kinase protein expression were detected, suggesting a direct impact of AgNPs and/or ROS on the inhibition of the canonical SHH pathway. However, more studies are needed due to the increase in the mTOR protein expression after the treatment of the cells with AgNPs, as in the Robotnikinin treatment. In conclusion, small-size AgNPs are able to inhibit the proliferation of GB cells in vitro by suppressing the canonical SHH pathway.

The prevalence of oral mucosa lesions (OMLs) among children varies from 4.1% up to 69.5%. There is a lack of sufficient epidemiological data and adequate knowledge about OMLs in relation to minors' gender and age. The aim of the study was to evaluate the prevalence of OMLs in children, patients of the oral pathology clinic in south-western parts of Poland, and to investigate the potential correlation between the occurrence of particular types of OMLs and the gender and age. A retrospective study was performed using a total of 2474 clinical charts from 2015 to 2019. Data collected included age, gender, and OMLs' type. The prevalence of OMLs in minors was 5.21%. Aphthae was the most frequent diagnosis. Boys were more commonly affected, and traumatic erosion and ulcers were significantly more often detected in males. The mean age of children was 8 y/o, preschoolers were significantly more often diagnosed with geographic tongue, while Morsicatio buccarum was significantly more common in school children and adolescents. Clinicians should be familiar with OMLs prevalence and with its specific frequency in connection to age and gender of children. Furthermore, they should be aware of the diversity of OMLs found in the oral cavity of children and that their frequency in the pediatric population is different from that in adults.

Currently, the development of the use of biomaterials and their application in medicine is causing rapid changes in the fields of regenerative dentistry. Each year, new research studies allow for the discovery of additional possibilities of dental tissue restoration. The structure and functions of teeth are complex. They consist of several diverse tissues that need to act together to ensure the tooth's function and durability. The integrity of a tooth's enamel, dentin, cementum, and pulp tissue allows for successful mastication. Biomaterials that are needed in dentistry must withstand excessive loading forces, be biocompatible with the hosts' tissues, and stable in the oral cavity environment. Moreover, each tooth's tissue, as well as aesthetic qualities in most cases, should closely resemble the natural dental tissues. This is why tissue regeneration in dentistry is such a challenge. This scientific research focuses on paediatric dentistry, its classification of caries, and the use of biomaterials in rebuilding hard dental tissues. There are several methods described in the study, including classical conservative methods such as caries infiltration or stainless-steel crowns. Several clinical cases are present, allowing a reader to better understand the described methods. Although the biomaterials mentioned in this work are artificial, there is currently ongoing research regarding clinical stem cell applications, which have a high potential for becoming one of the most common techniques of lost dental-tissue regeneration in the near future. The current state of stem cell development is mentioned, as well as the various methods of its possible application in dentistry.

FT-NIR spectroscopy and color measurements were applied to evaluate the influence of packaging on cosmetic cream stored at room and refrigerated conditions. Commercial cosmetic cream was stored for 2 and 4 mo in five containers intended for cosmetics: aluminum jar with polypropylene thermos (Al/PP), acrylic jar with polypropylene thermos (Ac/PP), glass (G) jar, polypropylene (PP) jar, and styrene acrylonitrile (SAN) jars. Principal component analysis (PCA) of the FT-NIR spectra showed the effect of time on tested samples stored in all applied packagings; separate groups were formed by fresh samples and samples stored for 2 and 4 mo. The changes in samples stored in the cold for 2 mo were similar for all applied packagings as compared with fresh samples, although samples stored in SAN jars formed a separate group. After 4 mo, a separate group was formed by samples stored in G jars. For samples stored at room temperature, the influence of packaging material on cosmetic emulsion was clearly visible; four separate groups (Al/PP with PP, Ac/PP, G, and SAN) were created by samples stored for 4 mo. Using partial least squares (PLS1) regression, it was found that the FT-NIR spectra of tested samples correlated with their lightness L*, a* parameter, and total color difference ΔE * ab . FT-NIR spectroscopy is a rapid technique which could be useful to make the best choice of packaging for cosmetics protecting the original quality of products during long-term storage.

RESUMEN

BACKGROUND: The right aortic arch with mirror-image of branching arteries without coexisting congenital heart disease is a very rare anomaly. CASE REPORT: We report a case of the right-sided aortic arch with aplasia of the left brachiocephalic trunk in a 64-year-old women, presenting difference in systolic blood pressure between upper extremities. The history of the patient and angio-CT findings were described and visualized with images. CONCLUSIONS: The knowledge of vascular variations is important for the clinical and therapeutic aspects.

BACKGROUND: Twin to twin transfusion syndrome occurs in 15% of monochorionic twin pregnancies. Untreated, TTTS has been reported to have a mortality of nearly 100%. Two main therapies include serial amnioreduction and fetoscopic laser coagulation for the vascular anastomoses. OBJECTIVES: The aim of the project was to investigate the optimal diagnostic and therapeutic procedure in pregnancies complicated by TTTS. Additionally the study was supposed to compare non-invasive and invasive methods of treatment and to show antenatal and postnatal follow - up to 4 months of age. METHODS: 42 pregnant women with twin-to-twin transfusion syndrome were assigned to laser therapy using diode laser and 33 pregnant women underwent only several amnioreductions. Selected parameters characterizing the pregnancy were compared in both groups. RESULTS: In the amnioreduction group, the perinatal survival rate seven days after the delivery was 31.8%. The survival rate of at least one twin was 39.4%. As compared to the amnioreduction group, in the laser group the survival rate of at least one twin was observed in 31 cases (31/42) and it was equal to 74%. Neurological complications in the amninoreduction group were observed in 19% (4/21) of cases, in the laser group and in 5% (2/40) of neonates at 4 months of age. CONCLUSIONS: Currently the preferred and only method that addresses the cause of the disease is the endoscopic laser coagulation of anastomoses. Comparison of the two treatments shows better outcomes with higher survival rates and minor neurological defects in cases treated with laser coagulation.
