BACKGROUND: Among the birds, chicken was the model used in the majority of studies on germ cells and other birds like quail, turkey and pheasant had some interest but ostrich has been lacking from these researches. AIMS: The present study was conducted to confirm the presence of ostrich primordial germ cells (PGCs) in the embryonic blood and to determine the appropriate time for having them in the blood. METHODS: Embryos were extracted in embryonic day (E) 6-12, their blood was obtained and the rest of the embryos were processed for histological examinations. Staining by periodic acid-Schiff (PAS) method and immunohistochemistry (IHC) using stage-specific embryonic antigen 1 (SSEA1) and stage-specific embryonic antigen 4 (SSEA4) antibodies (Abs) were used for identification of the PGCs. RESULTS: While the blood circulating ostrich PGCs were SSEA1-negative and SSEA4-positive, in the presumptive gonads PGCs showed negative immunoreactivity for both Abs. Although the ostrich PGCs were PAS-positive in both blood and presumptive gonad, their PAS-positive contents reduced during development from E 10 to E 12. CONCLUSION: E 9, when the hind limb buds appear, is the best time for detecting PGCs in the ostrich embryonic blood.

To distinguish between the direct effects of heat stress (HS) and reduced DMI on metabolic and physiological variables, morphometry, and gene expression of transporters involving in ruminal VFA absorption, 16 Holstein bull calves (178.1 ± 7.55 kg of BW) were used in a randomized complete design using 2 experimental periods. In period 1 (P1), all animals were housed in thermoneutral (TN) conditions (20.7 ± 3.0°C and a temperature-humidity index [THI] of 65.2 ± 3.1) and fed ad libitum for 8 d. In period 2 (P2; 9 d), half of the calves ( = 8) were subjected to HS condition (29.9 to 41.0°C and a THI ≥ 85 for 8 h/d) and the other half ( = 8) were maintained in TN conditions but were pair fed (pair-fed thermoneutral [PFTN]) to the HS calves. Heat stress decreased DMI (20%; < 0.01) and, by design, PFTN calves had similar reduced intakes. During P1, ADG was similar between groups (1.7 kg/d), but ADG was reduced similarly for both groups (0.14 kg/d) during P2 ( < 0.01). Compared with P1, HS calves had decreased plasma glucose levels (12.5%; < 0.01) in P2. Basal insulin levels increased in P2 for HS calves (71%; < 0.03), but there was no difference between periods for PFTN calves. Circulating NEFA concentrations did not differ between periods in the HS calves, but PFTN animals had increased basal NEFA levels (100%; < 0.01). Compared with P1, HS calves had increased blood urea nitrogen concentration in P2 ( < 0.01) but blood urea nitrogen did not differ between periods in the PFTN calves. Heat stress had no measurable effect on nutrient digestibility. Rumen papillae height increased (51%; < 0.04) in HS calves whereas papillae top width decreased (40%; < 0.03), but there was no difference between treatments for other morphometric parameters. Gene expression of VFA transporters in rumen epithelium was not affected by thermal treatments. These results confirmed that short-term HS in the current study had no effect on diet digestibility, rumen morphology, or VFA transporters genes. Therefore, HS-induced decrease in DMI is in large part responsible for gross changes in BW in growing calves (albeit tissue composition of growth is not unknown).

The aim of this study was to assess the effects of particle size (PS) of alfalfa hay on growth characteristics and rumen development in dairy calves at two levels of alfalfa supplementation. Fifty newborn dairy calves (42.7 ± 2.2 kg BW) were used in a 2 × 2 factorial arrangement with the factors supplementation level (low, 8%; or high, 16% on DM basis) and PS (medium, 2.92 mm; or long, 5.04 mm as geometrical means) of alfalfa hay. In addition, a control group without alfalfa hay was used. Hence, treatments were: control (C); low level with medium PS (LM); low level with long PS (LL); high level with medium PS (HM) or high level with long PS (HL). Growth performance of alfalfa-fed calves did not differ from control calves, but alfalfa supplementation decreased corneum thickness of the rumen wall. In alfalfa-fed calves, post-weaning starter intake was greater for LL calves than for LM calves. During the entire rearing period, starter intake was 26-32% higher for LL and HM calves than for LM calves. Pre-weaning average daily gain was higher for LL and HM calves than for HL calves, but this effect was not persistent over the entire rearing period. Final body weight decreased from 86 to 79 kg when the level of long PS alfalfa hay increased from 8 to 16%, but increased from 78 to 87 kg when the level of medium PS alfalfa increased from 8 to 16%. Regardless of PS and level, morphometric characteristics of rumen wall were generally similar among alfalfa feeding groups, but corneum thickness decreased from 8.7 to 6.1 µm with greater PS at the low level. These results indicate that adequate, but not excessive, physical stimulation is required for appropriate rumen development and growth performance of dairy calves.

The objective of this experiment was to investigate the effects of different levels of alfalfa hay (AH) and sodium propionate (Pro) added to starter diets of Holstein calves on growth performance, rumen fermentation characteristics, and rumen development. Forty-two male Holstein calves (40±2kg of birth weight) were used in a complete randomized design with a 3×2 factorial arrangement of treatments. Dietary treatments were as follows: (1) control = concentrate only; (2) Pro = concentrate with 5% sodium propionate [dry matter (DM) basis]; (3) 5% AH = concentrate + 5% alfalfa hay (DM basis); (4) 5% AH + Pro = concentrate + 5% alfalfa hay + 5% sodium propionate (DM basis); (5) 10% AH = concentrate + 10% alfalfa hay (DM basis); and (6) 10% AH + Pro = concentrate + 10% alfalfa hay + 5% sodium propionate (DM basis). All calves were housed in individual pens bedded with sawdust until 10wk of age. They were given ad libitum access to water and starter throughout the experiment and were fed 2L of milk twice daily. Dry matter intake was recorded daily and body weight weekly. Calves from the control, 10% AH, and 10% AH + Pro treatments were euthanized after wk 10, and rumen wall samples were collected. Feeding of forage was found to increase overall dry matter intake, average daily gain, and final weight; supplementing sodium propionate had no effect on these parameters. Calves consuming forage had lower feed efficiency than those on the Pro diet. Rumen fluid in calves consuming forage had higher pH and greater concentrations of total volatile fatty acids and molar acetate. Morphometric parameters of the rumen wall substantiated the effect of AH supplementation, as plaque formation decreased macroscopically. Overall, the interaction between forage and sodium propionate did not affect calf performance parameters measured at the end of the experiment. Furthermore, inclusion of AH in starter diets positively enhanced the growth performance of male Holstein calves and influenced both the macroscopic and microscopic appearances of the rumen wall. These benefits, however, were small when only sodium propionate was offered.

The effects of feeding clinoptilolite on hematology, performance, and health of newborn Balouchi lambs were evaluated in this experiment. In a completely randomized design, 30 newborn lambs were allocated to three groups and fed by basal diet (C0; without clinoptilolite) and C1 and C2 (the basal diet plus 1.5% and 3% clinoptilolite, respectively, for 6 weeks (3 weeks before and 3 weeks after weaning)). Blood samples were taken from all lambs, at the time when the animals were allocated to the experimental diet and at the end of each week of experiment, and analyzed for hematology, plasma fibrinogen, and total protein. Performance and health of all lambs were measured. Fecal consistency score and diarrhea severity were evaluated. There was no difference between lambs in case of hematological parameters. Lambs fecal consistency score and severity of diarrhea were lowest (P < 0.05) for lambs on C1 and C2 and highest for lambs on C0. Dry matter intake and feed conservation ratio were similar between the groups of lambs fed by different diets, but daily gain of lambs differed significantly (P < 0.05) and was higher in C2. It was concluded that addition of 3% clinoptilolite to starter diet of newborn lamb can reduce incidence and severity of diarrhea, although its effect on hematology and performance was negligible.

The objectives of the present research were to determine the effects of water stress on seed-quality traits and to map QTLs controlling the studied traits under two different water treatments in a population of sunflower recombinant inbred lines (RILs). Two experiments were conducted in greenhouse and field conditions, each with well-watered and water-stressed treatments. The experiments consisted of a split-plot design (water treatment and RIL) with three blocks. Analyses of variance showed significant variation among genotypes, and a water treatment x genotype interaction was also observed for most of the traits. Two to 15 QTLs were found, depending on trait and growth conditions, and the percentage of phenotypic variance explained by the QTLs ranged from 5% to 31%. Several QTLs for oil content overlapped with QTLs for palmitic and stearic acid contents in all four conditions. An overlapping region on linkage group 3 (QTLs 2.OC.3.1 and 4.SA.3.1) was linked to an SSR marker (ORS657). A principal component analysis was performed on four fatty acid traits. Two principal components, P1 and P2, were used for QTL analysis. This method improved the ability to identify chromosomal regions affecting the fatty acids. We also detected the principal-component QTLs that did not overlap with the fatty acid QTLs. The results highlight genomic regions of interest in marker-based breeding programmes for increasing oil content in sunflower.
