RESUMEN

Nodular fasciitis (NF) is a self-limiting fibrous neoplasm that can be mistaken for a soft tissue sarcoma. It is characterised by rapid growth, slight pain and local tenderness. Although it is frequently found in the forearm, a lesion distal to the wrist is quite rare. We present two unusual cases of NF involving the palm, supported by detecting ubiquitin specific protease 6 gene rearrangement. The first patient had non-intraneural NF presenting as peripheral neuropathy affecting the digital nerve while the second patient suffered from painless, non-tender NF in the palm, which had not regressed spontaneously during the five months prior to surgery.

AIMS: The aims of this study were to analyse the long-term outcome of vascularised fibular graft (VFG) reconstruction after tumour resection and to evaluate the usefulness of the method. PATIENTS AND METHODS: We retrospectively reviewed 49 patients who had undergone resection of a sarcoma and reconstruction using a VFG between 1988 and 2015. Their mean follow-up was 98 months (5 to 317). Reconstruction was with an osteochondral graft (n = 13), intercalary graft (n = 12), inlay graft (n = 4), or resection arthrodesis (n = 20). We analysed the oncological and functional outcome, and the rate of bony union and complications. RESULTS: Five- and ten-year overall survival rates were 89% and 86%, respectively. Local recurrence occurred in two patients. Eight patients developed pulmonary metastases. Bone union was achieved in 44 patients (90%). Fracture occurred in six patients (12%), infection in three (6%), and nonunion in five (10%). The mean Musculoskeletal Tumor Society (MSTS) scores were as follows: osteochondral graft 70%; intercalary graft 73%; inlay graft 89%; and resection arthrodesis 83%. CONCLUSION: Although associated with a relatively high rate of complications, each reconstruction method is useful, with a high rate of successful limb salvage and a good long-term functional outcome. RESUMEN

To identify sequences on the human genome that are actually transcribed, we mapped expressed sequence tags (ESTs) of long cDNAs ranging from 4 kb to 7 kb along a 33.4-Mb sequence of human chromosome 22, the first human chromosome entirely sequenced. By the EST mapping of 30,683 long cDNAs in silico, 603 cDNA sequences were found to locate on chromosome 22 and classified into 169 clusters. Comparison of the genomic loci of these cDNA sequences with 679 genes already annotated on chromosome 22q revealed that 46 clusters represented newly identified transcribed sequences. To further characterize these sequences, we sequenced 12 cDNAs in their entirety out of 46 clusters. Of these 12 cDNAs, 6 were predicted to include a protein-coding region while the remaining 6 were unlikely to encode proteins. Interestingly, 3 out of the 12 cDNAs had the nucleotide sequences of the opposite strands of the genes previously annotated, which suggested that these genomic regions were transcribed bi-directionally. In addition to these newly identified 12 cDNAs, another 12 cDNAs were entirely sequenced since these cDNAs were likely to contain new information about the predicted protein-coding sequences previously annotated. In the cases of KIAA1670 and KIAA1672, these single cDNA sequences covered two separately annotated transcribed regions. For example, the sequence of a clone for KIAA1670 indicated that the CHKL and CPT1B genes were co-transcribed as a contiguous transcript without making both the protein-coding regions fused. In conclusion, the mapping of ESTs derived from long cDNAs followed by sequencing of the entire cDNAs provided indispensable information for the precise annotation of genes on the genome together with ESTs derived from short cDNAs.

RESUMEN

Churg-Strauss syndrome (CSS) is a disorder characterized by hypereosinophilia and systematic vasculitis occurring in individuals with bronchial asthma. We present a case of 24-year-old woman with CSS diagnosed by and followed with gastrointestinal fiberscopic examination and electroneuromyography. The duodenal biopsy showed granulomatous angiitis with eosinophilic infiltration, and the electroneuromyography showed peripheral nerve dysfunction. After steroid treatment, the duodenal erosion and ulcer were almost completely resolved. There was no improvement in electroneuromyography, but the patient was able to walk independently after intensive rehabilitative training.

This study was undertaken to observe osteoclast differentiation related to inflammatory progression in aggressive periodontitis induced in beagle dogs by ligature of the gingival sulcus. To monitor osteoclastic activity, we used histochemical methods (staining for tartrate-resistant acid phosphatase [TRAP]) to visualize osteoclasts and their TRAP-positive precursors and biochemical methods (ELISA assay of pyridinium crosslinks) to detect bone matrix degradation products in gingival crevicular fluid (GCF), serum, and urine. For histochemical study, tissue specimens were prepared from 3 adult female beagle dogs induced with experimental periodontitis by silk ligature placement below the gingival margin of mandibular molars ligated for 3, 7, and 21 days. For biochemical study for pyridinoline measurement, the 24 mandibular molars of 4 male beagle dogs were ligated. GCF, urine, and serum were collected at day 0 and at 3, 7, 14, and 21 days after ligation. In the early inflammatory phase of ligature-induced periodontitis (day 3), TRAP+ mononuclear and TRAP+ multinucleated cells were present in the gingival connective tissue, and active bone-resorbing cells were found in excavated lacunae at the alveolar crest, but osteoclasts were not infiltrating the periodontal ligament during this early phase. During later stages of the inflammatory process (7 and 21 days), osteoclasts appeared at both the gingival and ligament side of the alveolar bone. Osteoclastic bone resorption appeared to be more severe on the bone surface at the gingival side than on the bone surface of the periodontal ligament side. Measurement of pyridinoline significantly increased in GCF and urine 3 days after ligation. The results suggested that bone at the crest of the alveolar bone is rapidly resorbed within 3 days of inducing experimental periodontitis.

RESUMEN

The nucleotide sequence of pHT926, a cryptic plasmid found in Bacillus borstelensis HP926, was determined. pHT926 replicates by a rolling-circle mechanism and belongs to the pC194 plasmid family. The copy number of pHT926 was fourfold higher than that of pUB110 and very stably maintained in Bacillus choshinensis.

RESUMEN

We investigated the localization of chondroitin sulfate and dermatan sulfate proteoglycans in the dental pulp and dentin of rats, using a combination of an immunohistochemical technique coupled with specific enzymatic digestion. Chondroitin 4-sulfate and dermatan sulfate were found to be widespread in pulpal connective tissue, predentin and dentinal tubules. The response to predentin was found to be particularly strong. Chondroitin 6-sulfate was stretched in pulpal connective tissue and predentin, but showed only a weak response.

We investigated the immunohistochemical localization of the chondroitin sulfates (chondroitin-4 sulfate, -6 sulfate and dermatan sulfate) in demineralized rat periodontal tissue using monoclonal antibodies (2-B-6, 3-B-3). Also, fixative and demineralized methods were established using these monoclonal antibodies. The result showed that the most effective combination of fixative and demineralized methods was 2% glutaraldehyde 1% para-formaldehyde and 5% EDTA. Chondroitin-4 sulfate and dermatan sulfate were widespread in gingival connective tissue and periodontal membrane, with an especially strong response of dermatan sulfate shown along collagen fiber bundles. Chondroitin-6 sulfate was located in peripheral blood vessels. In alveolar bone, chondroitin-4 sulfate and dermatan sulfate were found inside Hareversion canals, Volkman's canals and lacunae. Chondroitin-6 sulfate was localized at peripheral blood in alveolar bone. In cementum, chondroitin-4 sulfate and dermatan sulfate were found at lacunae of cellular cementum and a part of embedding Sharpey's fiber.

RESUMEN

One of the goals of orthodontic treatment is to obtain the best balance and harmony of facial lines, especially it is important to retract the lip in bimaxillary case. A 12-year and 4-month-old boy was first seen with the chief complaint of excessive lips protrusion. We treated him by the edgewise method for 34 months, and evaluated the change of facial profile. Excessive lip protrusion was eliminated, and arch leveled. Overjet and overbite were reduced. Mandibular growth increased the prominence of the chin, and reduce the protrusion of the lower lip.

The patient is a 10-year-old girl (Hellman's developmental stage IIIB) with a deficiency of upper second premolar (right side) erupting space caused by on upper first molar (right side) mesial shift). The treatment was proceeded with the looped lingual arch in the first place, the sectional arch and the open coil spring with the soldered lingual arch for reinforced anchorage in the second place. Consequently, the upper first molar (right side) was moved bodily without curve of the root in a short period, and upper second premolar (right side) erupted in a normal position.

This study investigated the immunohistochemical localization of chondroitin sulfate (chondroitin, 4-sulfate and 6-sulfate) and dermatan sulfate proteoglycan (PG) in human gingival connective tissue, using monoclonal antibodies. Dermatan sulfate was found to be widespread in connective tissue, with an especially strong response shown in collagen fiber bundles under the epithelial basement membrane. Chondroitin 4-sulfate occurred widely in connective tissue but showed only a weak response. Chondroitin 6-sulfate was located in peripheral blood vessels. Chondroitin was not detected in gingival connective tissue.

We applied of granular hydroxyapatite HAP to periodontal bone defects, for the purpose of restoring the periodontal tissue destroyed by periodontal disease, the subjects were 6 patients (6 regions) with chronic marginal periodontitis who had passed more than 3 years since HAP implantation. After routine periodontal initial treatment, a flap operation was performed, and granular HAP, or a combination of granular and block-like HAP, was implanted into the periodontal bone defect. Clinical parameters were probing depth, clinical attachment level, clinical gingival margin, residual HAP volume and X-ray before operation, at 3, 6, 12 months, and 2, 3, and 4, 6 years after the operation. The results revealed that all measured values showed good results for both granular HAP and the combination of granular and block-like HAP after implantation, and that all cases showed stable results from 12 months on. No remarkable difference was noted between the implanted regions.

The purpose of this study was to investigate the changes in glycosaminoglycans (GAGs) of the gingiva during a period of experimental periodontitis induced by placing a silk ligature below the gingival margin of dog molars. Incorporation of 3H-glucosamine into the gingiva was determined autoradiographically. The gingiva was collected at 0, 7, 21, 60 and 90 days and cultured in vitro in the presence of 3H-glucosamine. The autoradiographs showed a predominantly epithelial location of the silver grains in all gingival epithelia. The location was intercellular in all epithelia. The results suggest greater 3H-glucosamine incorporation by the epithelium compared with the connective tissue and markedly more rapid metabolic turnover of epithelial GAGs.
