RESUMEN

A novel galactose-specific lectin, AJLec (18.5 kDa), was isolated from the sea anemone, Anthopleura japonica. AJLec was characterized using the hemagglutination assay, isothermal titration calorimetry (ITC), and glycoconjugate microarray analysis and we found that AJLec has a specificity for galactose monomers and ß-linked terminal galactose residues in complex carbohydrates, but not for N-acetylgalactosamine (GalNAc), which is commonly recognized by galactose-binding lectins. The primary structure of AJLec did not show homology with known lectins, and a crystal structural analysis also revealed a unique homodimeric structure. The crystal structure of AJLec complexed with lactose was solved by measuring the sulfur single-wavelength anomalous diffraction (S-SAD) phasing with an in-house Cu Kα source method. This analysis revealed that the galactose residue in lactose was recognized via its O2, O3, and O4 hydroxyl groups and ring oxygen by calcium coordination and two hydrogen bonds with residues in the carbohydrate-binding site, which demonstrated strict specificity for the ß-linked terminal galactose in this lectin.

RESUMEN

We propose a novel method to reconstruct event by event the full kinematics of the cascade decay process, h→τ^{+}τ^{-}→(π^{+}ν[over ¯])(π^{-}ν), which allows us to measure the τ^{+}τ^{-} spin correlation, a measure of the CP property of the Higgs boson. By noting that the τ^{±} momenta lie on the plane spanned by the accurately measured impact parameter and momentum vectors of charged pions, we can obtain the most likely momenta of the two missing neutrinos by using the probability distribution functions of the p[over →]_{T} vector and the location of the primary vertex. A simple detector level simulation shows an excellent agreement between the reconstructed and the true kinematics, both in the τ^{+}τ^{-} and the π^{+}π^{-} rest frames. The method can be tested in Z→τ^{+}τ^{-} events, which should exhibit no correlation.

The stomach cancer-associated protein tyrosine phosphatase 1 (SAP-1) is a receptor-type protein tyrosine phosphatase that is specifically expressed on the apical membrane of the intestinal epithelium. SAP-1 is known to maintain the balance of phosphorylation of proteins together with protein kinases; however, its biological function and impact on pharmacokinetics in the intestine remain unclear. The present study, therefore, aimed at clarifying the relationship between SAP-1 and the intestinal absorption behaviors of typical transporter substrates and macromolecules. The endogenous levels of glucose and total cholesterol in the blood were similar between wild-type and SAP-1-deficient mice (Sap1-/-), suggesting no contribution of SAP-1 to biogenic influx. Moreover, in vitro transport study with everted ileal sacs demonstrated that there was no difference in the absorption of breast cancer resistance protein, P-glycoprotein, and peptide transporter substrates between both mice. However, absorptive clearance of macromolecular model dextrans (FD-4 and FD-10) in Sap1-/- mice was significantly higher than that in wild-type mice, and this was confirmed by the trend of increased FD-4 absorption from colonic loops of Sap1-/- mice. Therefore, the results of this study suggest the partial contribution of SAP-1 to the regulated transport of hydrophilic macromolecules through paracellular tight junctions.

Abnormalities in tactile perception, such as sensory defensiveness, are common features in autism spectrum disorder (ASD). While not a diagnostic criterion for ASD, deficits in tactile perception contribute to the observed lack of social communication skills. However, the influence of tactile perception deficits on the development of social behaviors remains uncertain, as do the effects on neuronal circuits related to the emotional regulation of social interactions. In neonatal rodents, whiskers are the most important tactile apparatus, so bilateral whisker trimming is used as a model of early tactile deprivation. To address the influence of tactile deprivation on adult behavior, we performed bilateral whisker trimming in mice for 10 days after birth (BWT10 mice) and examined social behaviors, tactile discrimination, and c-Fos expression, a marker of neural activation, in adults after full whisker regrowth. Adult BWT10 mice exhibited significantly shorter crossable distances in the gap-crossing test than age-matched controls, indicating persistent deficits in whisker-dependent tactile perception. In contrast to controls, BWT10 mice exhibited no preference for the social compartment containing a conspecific in the three-chamber test. Furthermore, the development of amygdala circuitry was severely affected in BWT10 mice. Based on the c-Fos expression pattern, hyperactivity was found in BWT10 amygdala circuits for processing fear/anxiety-related responses to height stress but not in circuits for processing reward stimuli during whisker-dependent cued learning. These results demonstrate that neonatal whisker trimming and concomitant whisker-dependent tactile discrimination impairment severely disturbs the development of amygdala-dependent emotional regulation.

The influence of ultraviolet B (UVB) radiation on transglutaminase 1 (TGase 1), a major factor that regulates skin keratinization, has not been sufficiently characterized especially at the gene or protein level. Thus, we determined whether UVB affects the expression of TGase 1 in human keratinocytes and clarified the intracellular stress signaling mechanism(s) involved. Exposure of human keratinocytes to UVB significantly up-regulated the expression of TGase 1 at the gene and protein levels. Treatment with inhibitors of p38, MEK, JNK or NFκB significantly abolished the UVB-stimulated protein expression of TGase 1. Treatment with astaxanthin immediately after UVB irradiation did not attenuate the increased phosphorylation of Ser536/Ser468NFκBp65, c-Jun, ATK-2 and CK2, and did not abrogate the increased or diminished protein levels of c-Jun/c-Fos or I-κBα, respectively. However, the same treatment with astaxanthin significantly abolished the UVB-stimulated expression of TGase 1 protein, which was accompanied by the attenuated phosphorylation of Thr565/Ser376/Ser360MSK1, Ser276NFκBp65 and Ser133CREB. The MSK1 inhibitor H89 significantly down-regulated the increased protein expression of TGase 1 in UVB-exposed human keratinocytes, which was accompanied by an abrogating effect on the increased phosphorylation of Ser276NFκBp65 and Ser133CREB but not Thr565/Ser376/Ser360MSK1. Transfection of human keratinocytes with MSK1 siRNA suppressed the UVB-stimulated protein expression of TGase 1. These findings suggest that the UVB-stimulated expression of TGase 1 is mediated predominantly via the NFκB pathway and can be attenuated through a specific interruption of the p38/MSK1/NFκBp65Ser276 axis.

RESUMEN

Tris(2-chloroethyl) and tris(1,3-dichloro-2-propyl) phosphates are chlorinated persistent flame retardants that have recently emerged as environmental pollutants. Two bacterial strains that can degrade the compounds when they are the sole phosphorus sources have been isolated and identified as members of the sphingomonads. The strains can be useful for the bioremediation of environments contaminated with these compounds.

RESUMEN

We investigated an integrated optical modulator consisting of two Mach-Zehnder interferometers. The modulator can generate optical signals in various types of modulation formats, which have advantages for long-haul transmission, optical labeling, etc. By using a fabricated versatile optical modulator having traveling-wave electrodes designed for high-speed signals, we demonstrated generation of optical 40Gb/s frequency-shift-keying signals, which can be demodulated by an optical filter. 80Gb/s optical differential quadrature-shift-keying modulation was also demonstrated, where 40Gb/s in-phase and quadrature data were, simultaneously, fed to the modulator.

BACKGROUND: Expression of argyrophilic nucleolar organizer regions (AgNOR) was counted on mucosal epithelium to detect whether the AgNOR count (the number of AgNOR per nucleus) is, for 20 weeks, affected by streptozotocin-induced diabetes mellitus. METHODS: Diabetic rats were applied non-denture wearing, non-pressure contact, continuous pressure (0.5, 1.5 or 3.4 kPa) and intermittent pressure (10.5, 33.0 or 81.7 kPa), respectively. RESULTS: In the non-denture wearing or the non-pressure contact group, AgNOR count was almost constant (ranging 1.86-1.92) without significant difference. Under continuous pressure, AgNOR count significantly decreased at earlier stage and finally recovered to the non-pressure contact group. A similar effect of continuous pressure appeared to manifest itself in intermittent pressure, but to a greater degree. Consequently, AgNOR count under intermittent pressure showed no recovery at 20-week stage. CONCLUSION: : Diabetes mellitus exerted, by itself, no influence on the cellular proliferative activity under continuous pressure, whereas the activity deteriorated under intermittent pressure.
