RESUMEN

Peripheral T-cell Lymphoma (PTCL) represents a heterogenous group of aggressive non-Hodgkin Lymphomas with poor prognostic outcomes and limited treatment options. The development and refinement of therapeutic strategies for PTCL are impeded by a paucity of reliable preclinical models that accurately mimic the disease's pathophysiology. There is a dire need for more physiologically relevant models for PTCL. Here we describe a spontaneousCD8+ peripheral T-cell lymphoma cell line (LM-23) derived from a 12-week-old female Balb/cJ mouse. Both intravenous and subcutaneous administration of this cell line to syngeneic Balb/cJ mice resulted in rapid establishment of tumor growth. CHOP and anti-PD1 treatment both displayed no benefit to mice in regulating tumor growth. Such results along with its phenotypic characteristics, rapid growth, and metastatic behavior in syngeneic mice highlight its value in studying the elusive disease and discovery of novel therapeutics.

The mutation I.3.2 was previously identified in a FLP/FRT screen of chromosome 2R for conditional growth regulators. Here we report the phenotypic characterization and genetic mapping of I.3.2 by undergraduate students participating in Fly-CURE, a pedagogical program that teaches the science of genetics through a classroom research experience. We find that creation of I.3.2 cell clones in the developing eye-antennal imaginal disc causes a headless adult phenotype, suggestive of both autonomous and non-autonomous effects on cell growth or viability. We also identify the I.3.2 mutation as a loss-of-function allele of the gene centromere identifier ( cid ), which encodes centromere-specific histone H3 variant critical for chromosomal segregation.

The invasive mussel Mytilus galloprovincialis is a heat-tolerant species relative to its competative congener M. trossulus, that dominates warm seawater environments but it is unknown how multiple stressors (MS) may affect its physiology. Our study determined the effects of MS on the metabolic rate (MR), superoxide dismutase (SOD) antioxidant enzyme activity, and clearance rate (CR) of M. galloprovincialis. Mussels were exposed for 7 d to hyposalinity (20, 28, 34 ppt) then to heat shock (17, 20, 25 °C) after which MR and SOD activity were determined. CR was quantified following a 30 min MS exposure. We found a significant influence of MS on MR, SOD, and CR. We identified synergistic effects on MR under the most extreme treatment. SOD activity was the greatest under 20 °C exposure while CR declined under heat shock. Thus, our study suggests that mussels experiencing MS may become energy limited as MR increases and feeding rates decrease.
