RESUMEN

Diabetic wounds arise great attention as they are difficult to heal and easily suffer from serious bacterial infection. However, the overuse of antibiotics increases the resistance of bacteria and makes common drugs ineffective. Here, we developed a photothermal hydrogel (TFP/NP) composed of tremella fuciformis polysaccharides (TFPs) and cuttlefish ink-derived melanin nanoparticles (NPs). The NPs can produce reliable photothermal effects under near-infrared laser (NIR) irradiation and help to remove the bacteria in the wounds, while TFPs were able to form hydrogel frameworks which possessed anti-inflammatory effects and could be applied to promote wound healing. The TFP/NP hydrogels produced stable thermal effects under NIR irradiation and could continuously kill bacteria. The experiment on a full-layer skin wound sMRSA activity and could improve the healing efficiency. The wounds of the mice could be repaired within 14 days after reasonable treatment. In addition, the hydrogels play significant roles in promoting collagen deposition, anti-inflammation, angiogenesis, and cell proliferation during the therapeutic process. This research provides a simple and effective method for the therapy of bacterial infection wounds through the synergistic effect of TFPs and NPs.

Skin could protect our body and regenerate itself to against dysfunctional and disfiguring scars when faced with external injury. As wound dressings, hydrogels are biocompatible, hydrophilic and have a 3D structure similar to the extracellular matrix (ECM). In particular, hydrogels with drug-releasing capabilities are in acute wound healing. In this paper, photocrosslinked hydrogels served as wound dressing based on sodium carboxymethylcellulose (CMC) were prepared to promote wound healing. Photocrosslinked hydrogels were prepared by grafting lysine and allyl glycidyl ether (AGE) onto CMC and encapsulating curcumin (Cur). The synthesized hydrogels had the unique 3D porous structure with a swelling ratio up to 1300 % in aqueous solution. The drug release ratios of the hydrogels were 20.8 % in acid environment, and 14.4 % in alkaline environment. Notably, the hydrogels showed good biocompatibility and antibacterial properties and also exhibited the ability to accelerate the process of skin wound healing while prevent inflammation and scar formation when applied to a mouse skin wound model. As a result, the prepared hydrogels Gel-CLA@Cur showed great potential in wound healing.

The increasing prevalence of drug-resistant Escherichia coli (E. coli) resulting from the excessive utilization of antibiotics necessitates the immediate exploration of alternative approaches to counteract pathogenic E. coli. Phages, with their unique antibacterial mechanisms, are considered promising candidates for treating bacterial infections. Herein, we isolated a lytic Escherichia phage Tequatrovirus YZ2 (phage YZ2), which belongs to the genus Tequatrovirus. The genome of phage YZ2 consists of 168,356 base pairs with a G + C content of 35.34% and 269 putative open reading frames (ORFs). Of these, 146 ORFs have been annotated as functional proteins associated with nucleotide metabolism, structure, transcription, DNA replication, translation, and lysis. In the mouse model of a skin wound infected by E. coli, phage YZ2 therapy significantly promoted the wound healing. Furthermore, histopathological analysis revealed reductions in IL-1ß and TNF-α and increased VEGF levels, indicating the potential of phages as effective antimicrobial agents against E. coli infection.

Controlled and accurate drug release at the target site have been the focus of research. Especially in cancer therapy, economical, convenient and accurate delivery strategies could help to reduce the toxic effects of drugs on normal tissues and improve drug availability. In the study, glutathione (GSH)-responsive microcapsules (FA-RSMCs) were prepared by sonochemical method based on thiolated modified starch. 10-Hydroxycamptothecin (HCPT) was designed as a reactive oxygen species (ROS)-responsive polyprodrug (polyHCPT), which was loaded into the core of the microcapsules to obtain stepwise released drug delivery carriers. In the tumor microenvironment, FA-RSMCs first triggered GSH-responsive cleavage to release polyHCPT, followed by ROS-responsive cleavage of polyHCPT to release intact HCPT drug molecules. The results of experiments in simulated tumor microenvironment showed that FA-RSMCs exhibited good cascade-response release properties in vitro. It exhibited good anti-tumor ability and protection of normal cells in cytotoxicity in vitro. This strategy enhanced the accuracy and safety of targeted delivery of HCPT via microcapsules, which has potential for clinical application.

The hydrogel wound dressing with self-healing and adhesive property can provide better protection to the wound and prolong the service life of the material. Inspired by mussels, a high-adhesion, injectable, self-healing and antibacterial hydrogel was designed in this study. The lysine (Lys) and the catechol compound 3,4-dihydroxyphenylacetic acid (DOPAC) were grafted onto chitosan (CS). The presence of catechol group endows the hydrogel strong adhesion and antioxidation. In the experiment of wound healing in vitro, the hydrogel can adhere to the wound surface and promote wound heal. In addition, it has been proved the hydrogel has good antibacterial properties against S. aureus and E. coli. The treatment of CLD hydrogel, the degree of wound inflammation was significantly alleviated. The levels of TNF-α, IL-1ß, IL-6 and TGF-ß1 were reduced from 39.8379 %, 31.6768 %, 32.1015 % and 38.4911 % to 18.5931 %, 12.2275 %, 13.0524 % and 16.9959 %, respectively. And the levels of PDGFD and CD31 were increased from 35.6054 %, 21.7394 % to 51.8555 %, 43.9326 %, respectively. These results indicated that the CLD hydrogel has a good ability to promote angiogenesis, thickening of skin and epithelial structures.

Polysaccharide materials have attracted a widespread interest in the biomedical materials field due to their non-toxic, biocompatible and biodegradable properties. In this research, starch was modified with chloroacetic acid, folic acid (FA) and thioglycolic acid and then starch-based nanocapsules loaded with curcumin (FA-RSNCs@CUR) were prepared by the convenient oxidation method. The nanocapsules were prepared with stable particle size distribution of 100 nm. In the drug release test simulating the tumor microenvironment in vitro, the cumulative CUR release rate at 12 h was 85.18 %. Due to FA and FA receptor mediation, it only took 4 h for FA-RSNCs@CUR to achieve internalization by HeLa cells. In addition, cytotoxicity confirmed that starch-based nanocapsules have good biocompatibility as well as protection of normal cells in vitro. And FA-RSNCs@CUR showed certain antibacterial properties in vitro. Therefore, FA-RSNCs@CUR has good potential for future applications in food preservation and wound dressing, and so on.

Microcapsules have attracted attention due to their widespread applications. Among the numerous wall materials of microcapsules, chitosan is a promising candidate due to its various merits, such as natural, nontoxic, biocompatible and biodegradable. Chitosan-based microcapsules have broad application scope and application prospects. Recently, chitosan-based microcapsules are used in biomedical and food science increasingly, which show the great practical application potential of chitosan microcapsules. This review discussed and analyzed the preparation methods and design strategies of chitosan-based microcapsules for various applications. And summarized the design considerations of the chitosan-based microcapsule in different applications for biomedical and food science. It is expected to provide researchers with inspiration and design ideas about chitosan-based microcapsules.

Fluorescence sensing can not only identify a target substrate qualitatively but also achieve the purpose of quantitative detection through the change of the fluorescence signal. It has the advantages of immense sensitivity, rapid response, and excellent selectivity. The proposed aggregation-induced emission (AIE) concept solves the problem of the fluorescence of traditional fluorescent molecules becoming weak or quenched in high concentration or aggregated state conditions. Schiff base fluorescent probes have the advantages of simple synthesis, low toxicity, and easy design. They are often used for the detection of various substances. In this review we cover late developments in Schiff base compounds with AIE characteristics working as fluorescence sensors.

Biocompatible and biodegradable polysaccharides are abundant and renewable natural materials. Polysaccharides and their derivatives are developed into various carrier materials for biomedical applications. In particular, advanced polysaccharide-based nano/microcapsules have received extensive attention in biomedical applications due to their good encapsulation ability and tunability. In recent years, polysaccharide-based nano/microcapsules have been widely used in drug carriers, gene carriers, antigen carriers, wound dressings, bioimaging and biosensors. Numerous research results have confirmed the feasibility, safety, and effectiveness of polysaccharide-based nano/microcapsules in the above-mentioned biomedical applications. This review discussed and analyzed the latest research strategies and design considerations for these applications in detail. The preparation methods, application strategies, and design considerations of polysaccharide-based nano/microcapsules are summarized and analyzed, and their challenges and future research prospects in biomedicine are further discussed. It is expected to provide researchers with inspiration and design ideas.

Aggregation-induced emission (AIE) active compounds are fascinated due to their unique properties of limiting intramolecular rotation, and they have been developed in the biomedical fields. In this work, AIE material based on the Schiff base compound of chitosan (Cs) and salicylaldehyde (SA) was designed and synthesized. Cs-SA emits weak light in dilute aqueous solution, and emits bright light in concentrated solution and solid, showing obvious AIE performance. In addition, Cs-SA can also be used as a biosensor to detect Fe3+, and Cu2+, it has good bioimaging behavior. In addition, it can also be used as biosensor to quantitatively detect gram-positive bacteria and gram-negative bacteria, Moreover, Cs-SA shows excellent broad spectrum antibacterial performance in inhibiting E. coli and S. aureus.

Hydrogels are a class of polymer materials with three-dimensional networks structure, which can absorb a large number of water and biological fluids. Natural polysaccharides are ideal materials for the preparation of biomimetic hydrogels because of their good biocompatibility and biodegradability. The self-healing hydrogel can achieve healing without external force and prolong the service life of the hydrogel. Therefore, self-healing hydrogels are of great interest in tissue engineering, wound dressings, drug delivery, and tissue engineering. This review aims to introduce the preparation methods, self-healing properties of polysaccharide-based self-healing hydrogels and its potential applications in biomedicine and other fields.

The unique natural advantages of polysaccharide materials have attracted attention in biomedical applications. The abundant modifiable functional groups on the polysaccharide materials surface can facilitate the synthesis of various multifunctional drug delivery carriers. Especially in tumor therapy, the designs of polysaccharide-based drug delivery carriers are diverse. Therefore, this review summarized several latest types of polysaccharide-based drug carriers designs, and focused on the latest design strategies and considerations of drug carriers with polysaccharides as the main structure. It is expected to provide some design ideas and inspiration for subsequent polysaccharide-based drug delivery systems.

Traditional and single treatment strategies are difficult to achieve good results due to tumor resistance and complex mechanisms. Combination therapy through co-delivery systems is one of the methods to improve the effectiveness of cancer treatment. The polyprodrug platform has inherent advantages such as high drug loading and strong stability. Herein, a new reactive oxygen species (ROS)-responsive micelle composed of poly 10-hydroxycamptothecin (pHCPT) and PEG is reported, which loaded dexamethasone (DEX) as synergistic drugs. The micelles collapse in the complex microenvironment of tumor cells to release DEX. The first released DEX can increase the ROS level of tumor cells, thereby facilitating the cleavage of thioketal bonds to release intact HCPT molecules. Meanwhile, DEX can normalize tumor blood vessels, reduce adverse reactions, and further improve the efficacy of HCPT. This co-delivery system shows an ideal tumor suppressive effect in vivo and in vitro. Designing drugs into a modular multi-drug platform and selecting appropriate synergistic drugs according to the treatment plan provides a convenient strategy for future clinical treatment.

In this study, a biocompatible folate-decorated reductive-responsive carboxymethylcellulose-based nanocapsules (FA-RCNCs) were designed and prepared via sonochemical method for targeted delivery and controlled release of hydrophobic drugs. The shell of FA-RCNCs was cross-linked by disulfide bonds formed from hydrosulfuryl groups on the thiolated carboxymethylcellulose (TCMC) and encapsulated hydrophobic drug dispersed in the oil phase into nanocapsules. Moreover, the size and morphology of drug loaded FA-RCNCs were characterized by DLS, SEM and CLSM which indicated that the synthesized nanocapsules have suitable size range and excellent stability for circulating in the bloodstream. The drug release rate of FA-RCNCs could be controlled by adjusting their sizes and shell thickness, which could be dominated by the concentration of TCMC and sonochemical conditions. Furthermore, the obtained FA-RCNCs could be ingested into Hela cells via folate-receptor (FR)-mediated endocytosis and quickly release drugs under reductive environment, which demonstrated that FA-RCNCs could become potential hydrophobic drugs carries for cancer therapy.

Infectious diseases, such as the coronavirus disease-19, SARS virus, Ebola virus, and AIDS, threaten the health of human beings globally. New viruses, drug-resistant bacteria, and fungi continue to challenge the human efficacious drug bank. Researchers have developed a variety of new antiviral and antibacterial drugs in response to the infectious disease crisis. Meanwhile, the development of functional materials has also improved therapeutic outcomes. As a natural material, chitosan possesses good biocompatibility, bioactivity, and biosafety. It has been proven that the cooperation between chitosan and traditional medicine greatly improves the ability of anti-infection. This review summarized the application and design considerations of chitosan-composed systems for the treatment of infectious diseases, looking forward to providing the idea of infectious disease therapy.

Polymer gels can be classified as chemical and physical gels, depending on the type of cross-link. Physical gels usually form by physical cross-linking, such as hydrogen bonding, van der Waals and/or p-p interactions, which can avoid the use of additional cross-linking agents. Polyamidoamine (PAMAM) dendrimers possess abundant active groups on their surfaces. Modified dendrimers have been used as versatile linkers in some projects. In this work, polymer gels composed of PAMAM dendrimers without any covalent bonding cross-linking agents were prepared. The number of amino groups and ester groups on the surface of the dendrimers was precisely regulated to help form hydrogen bonds between adjacent dendrimers. The prepared dendrimer-based polymer gels retain the properties of PAMAM dendrimers such as antibacterial properties, and the unique structures make the gels exhibit high compressive strengths but relatively low tensile strengths. Interestingly, the prepared gels show good anti-inflammatory properties in acute inflammation models of mice with ear edema. The inflammatory inhibition rate and hematoxylin-eosin (H&E) staining method were used to confirm the anti-inflammatory effect. This present study demonstrates that the dendrimer-based polymer gels achieved through hydrogen bonding could be a versatile platform for tissue engineering.

Polymeric micelles (PMs) have been applied widely to transport hydrophobic drugs to tumor sites for cancer treatment. However, the low load efficiency of the drug in the PMs significantly reduces the therapeutic efficiency. We report here that disulfide-linked camptothecin (CPT) as a kind of dimeric drug can be effectively embedded in the core of poly(ε-caprolactone)-poly(ethylene glycol)-poly(ε-caprolactone) (PCL-PEG-PCL) PMs for improving drug-loading efficiency, and PEG can be used as a hydrophilic shell. Moreover, the dimeric CPT-loaded PCL-PEG-PCL PMs exhibited excellent solubility in phosphate-buffered saline (PBS) media and significant cytotoxicity to cancer cells.

Soil is the loose solum of land surface that can support plants. It consists of minerals, organics, atmosphere, moisture, microbes, et al. Among its complex compositions, soil moisture varies greatly. Therefore, the fast and accurate inversion of soil moisture by using remote sensing is very crucial. In order to reduce the influence of soil type on the retrieval of soil moisture, this paper proposed a normalized spectral slope and absorption index named NSSAI to estimate soil moisture. The modeling of the new index contains several key steps: Firstly, soil samples with different moisture level were artificially prepared, and soil reflectance spectra was consequently measured using spectroradiometer produced by ASD Company. Secondly, the moisture absorption spectral feature located at shortwave wavelengths and the spectral slope of visible wavelengths were calculated after analyzing the regular spectral feature change patterns of different soil at different moisture conditions. Then advantages of the two features at reducing soil types' effects was synthesized to build the NSSAI. Thirdly, a linear relationship between NSSAI and soil moisture was established. The result showed that NSSAI worked better (correlation coefficient is 0.93) than most of other traditional methods in soil moisture extraction. It can weaken the influences caused by soil types at different moisture levels and improve the bare soil moisture inversion accuracy.

The spatio-temporal distribution and variation of soil moisture content have a significant impact on soil temperature, heat balance between land and atmosphere and atmospheric circulation. Hence, it is of great significance to monitor the soil moisture content dynamically at a large scale and to acquire its continuous change during a certain period of time. The object of this paper is to explore the relationship between the mass moisture content of soil and soil spectrum. This was accomplished by building a spectral simulation model of soil with different mass moisture content using hyperspectral remote sensing data. The spectra of soil samples of 8 sampling sites in Beijing were obtained using ASD Field Spectrometer. Their mass moisture contents were measured using oven drying method. Spectra of two soil samples under different mass moisture content were used to construct soil spectral simulation model, and the model was validated using spectra of the other six soil samples. The results show that the accuracy of the model is higher when the mass water content of soil is below field capacity. At last, we used the spectra of three sampling points on campus of Peking University to test the model, and the minimum value of root mean square error between simulated and measured spectral reflectance was 0.0058. Therefore the model is expected to perform well in simulating the spectrum reflectance of different types of soil when mass water content below field capacity.

Farmland drought has the characteristics of wide range and seriously affecting on agricultural production, so real-time dynamic monitored has been a challenging problem. By using MODIS land products, and constructing the spectral space of LST and LAI, the temperature LAI drought index (TLDI) was put forward and validated using ground-measured 0-10 cm averaged soil moisture of Ningxia farmland. The results show that the coefficient of determination (R2) of both them varies from 0.43 to 0.86. Compared to TVDI, the TLDI has higher accuracy for farmland moisture monitoring, and solves the saturation of NDVI during the late development phases of the crop. Furthermore, directly using MODIS land products LST and LAI and avoiding the complicated process of using the original MODIS data provide a new technical process to the regular operation of farmland drought monitoring.
