RESUMEN

The results obtained at our laboratory during several last years suggest that a specific telomeric chromatin is formed at the terminal regions of Drosophila melanogaster DNA of 4-5 kb in length. In this work, we have studied how the telomeric chromatin influences the Polycomb-dependent repression. Many Polycomb proteins are involved in the formation of subtelomeric chromatin and inhibit the expression of a transgene inserted into it. However, the question on a positive or negative effect of telomeric chromatin on the assembly of subtelomeric protein complexes is yet open. Using a model system, in which a P element inserted upstream of the yellow gene promoter and located at the end of a terminally deleted chromosome can serve as a binding site for Pc-G proteins in the presence of php1 mutant allele, we have shown that a specific structure of the telomeric chromatin negatively influences the formation of Polycomb-dependent repression complex. These results suggest an antagonism between the telomeric and subtelomeric (Pc-G-dependent) chromatins.

Our recent study demonstrated that alteration of expression of the east gene affects the transcription of the yellow gene of Drosophila melanogaster. Furthermore, this influence was shown to be associated with the presence of retrotransposon MDG4 sequences in the pre-promoter gene region. In the present study, we analyzed the effects of different concentrations of EAST protein on the expression of different genes of Drosophila. In addition, it was analyzed whether this effect was associated with the presence of MDG4 in the allelic gene variant of interest. It was demonstrated that the effect of EAST protein on the expression of different genes was not always directly associated with the presence of MDG4 sequences. For instance, an increase in the EAST concentration had no effect on the expression of mutant alleles of the achaete-scute complex, some of which contained the MDG4 sequences. Moreover, enhancement of the east expression resulted in the alteration of the phenotypic manifestation of the mutant allele of the cut locus irrespective of the presence of MDG4. Finally, the alteration of the EAST concentrations had no substantial effect on transgene repression if they were inserted either in heterochromatin or in telomeric regions of chromosome 4, as well as on the repression induced by Polycomb group proteins. The data obtained suggested that EAST protein belonged to a new regulatory system, which was common to most of the genes, and repressed transcription of the genes located in the euchromatin regions of the chromosomes.

The article deals with the issues of conceptual and methodological motivation of stages related with working out and approving the draft projects of regional healthcare legislation through superpositioning the legislative activity of the subjects of the Russian Federation on the provisions of the basic law "On Healthcare". The common features of the basic law are described; the specificity of the recommended structure and item-by-item sequence of principles and norms of regional laws "On Healthcare" is defined.

Many-year complex expert and analytical studies of the valid and newly created laws in this sphere helped design a conceptual model of the development and improvement of the system of sanitary legislation.
