RESUMEN

An 11-year-old spayed female mixed-breed dog was referred for evaluation of a mass on the right forelimb of 4 months' duration. Cytologic examination of a fine-needle aspirate of the mass resulted in a diagnosis of liposarcoma. The 3 x 3 x 1.5-cm firm mass was surgically removed. Histopathologic observations confirmed the diagnosis of liposarcoma. During processing, a 1 x 0.8-cm piece of glass was found within the mass. Association between a glass foreign body and liposarcoma is discussed.

OBJECTIVE: To evaluate accumulation of products of lipid peroxidation in free skin grafts in rats over 10 days after grafting. DESIGN: Prospective analysis. ANIMALS: 30 adult Sprague-Dawley rats. PROCEDURE: Free skin grafts were applied to 1 hemithorax of 30 rats (2 groups, n = 15/ group), the other hemithorax acting as a nongrafted control. Biopsy specimens were taken from the nongrafted side of all rats immediately before and from the nongrafted and grafted sides immediately after the procedure. Biopsy specimens of both sides of the thorax were performed on days 1, 3, and 7 after grafting in group-1 rats, and on days 2, 4, and 10 after grafting in group-2 rats. Thiobarbituric acid (TBA) analysis for malondialdehyde and other aldehydic products (TBA-reactive substances) was used to determine lipid peroxidation. Concentration of TBA-reactive substances was determined by absorbance spectrophotometry at a wavelength of 532 nm. RESULTS: Accumulation of products of lipid peroxidation, reflected by increase in absorbance, continued to increase over the 10 days of this study. Difference in absorption between the nongraft and graft biopsy specimens was significant (P = 0.0011). Absorbance on days 3 and 4 was significantly higher than control and day-0 values (P < or = 0.05). Absorbance on days 7 and 10 was significantly higher than control, day-0, and day-1 values (P < or = 0.05). Rate of increase in absorption was maximal at day 4 and rapidly decreased thereafter. CONCLUSION: Accumulation of lipid peroxidation products in skin grafts may be best explained by oxygen-derived free radical-induced injury attributable to postischemic reperfusion. Maximal rate of accumulation corresponded to the known time of graft recirculation and revascularization. CLINICAL RELEVANCE: Predictability of free radical damage may allow timely pharmacologic intervention to reduce radical formation and ameliorate effects of peroxidation. Such intervention may help increase free graft survival or mitigate the effects of vascular compromise to axial pattern flaps.

RESUMEN

Three dogs were presented for para-aural abscessation. Ear canal separations were suspected preoperatively, based on histories of trauma, physical examination findings of minimally inflamed external ear canals with abrupt endings, and radiographic evidence of discontinuity of the ear canals. Concurrent otitis media, based on radiographic findings, was diagnosed in all three dogs. Disruptions of the external ear canals were confirmed at surgery, and the dogs each responded to total ear canal ablation and lateral bulla osteotomy.

RESUMEN

It has been suggested that autoimmunity to the proteoglycan (PG) component of cartilage plays a major role in the etiology of adjuvant arthritis (AA), which occurs in rats, but not in mice, after injection of CFA. In order to more directly investigate this role, bovine and human cartilage PG were used to modulate AA, and immunity to PG was assessed. Immunization of rats or mice with PG by itself does not induce arthritis. However, in rats, a single i.v. injection of soluble PG, given 1 wk before injection of CFA, results in a significant increase in incidence and severity of the arthritis induced. Rats injected with CFA have both antibody and delayed type hypersensitivity (DTH) to PG. Upon pretreatment of rats with PG i.v., both DTH and antibody titers to PG are increased. Rats immunized with PG in IFA have high titers of anti-PG and strong DTH to PG, which are also enhanced by pretreatment with PG i.v., although none of these animals develops arthritis. In contrast to these findings in rats, when mice are pretreated with PG i.v., DTH to PG induced by injection of CFA is lower, whereas anti-PG titers are higher than in unpretreated controls. The results presented here show that, in rats, i.v. injection of PG synergizes with CFA in the induction of AA, and enhances both humoral and cellular immunity to PG. The findings support the hypothesis that immunity to PG is of importance in AA, although under the conditions of these experiments immunity induced by PG alone is clearly not sufficient for the induction of arthritis.

Six men handling barley were followed up over two working days and their flow volume curves were measured every two hours. All experienced falls in ventilatory capacity of up to 800 ml with changes in the shape of the flow volume curve in five men. Five volunteers not previously exposed to barley dust sat in a silo for two hours. Decreases in ventilatory capacity ranging from 200 ml to 800 ml were found, with recovery taking up to 72 hours. All subjects had decreases in flow at 50% vital capacity but little or no change in flow at 75% vital capacity. In three subjects there was a drop in specific conductance that lasted for less than 24 hours. Two subjects were similarly exposed on a second occasion when transfer factor for carbon monoxide was measured in addition to flow volume curves. There was a decrease in transfer factor but no change in transfer factor per unit volume (Kco).
