RESUMEN

Objective To establish and evaluate a method for rapid and sensitive S.xylosus detection using qPCR(real-time quantitative PCR).Methods A gehM gene fragment was selected as the target for S.xylosus.A set of specific primers was synthesized and a qPCR method was established to detect S.xylosus.A S.xylosus standard strain and other non-target strains were chosen for analysis.DNA of S.xylosus was diluted 10-fold to determine its sensitivity.Clinical samples were tested,and positive products were sequenced.The result were compared with those of bacterial culture.Results S.xylosus had a specific amplification curve,whereas other non-S.xylosus species did not,indicating that the primers were specific for S.xylosus.Sensitivity was 100 fg/μL DNA.Repeatability within and between groups was less than 3%.A total of 60 clinical samples were analyzed,of which five samples had a typical S curve.qPCR products were sequenced and BLAST searched.The similarity of the gene sequences was 99.63%,indicating that the sample was positive for the S.xylosus gehM gene with a positivity rate of 8.3%.However,the positivity rate of bacterial culture was 6.7%.The positivity rate of qPCR was slightly higher than that of the culture.Conclusions The established qPCR method is rapid with high sensitivity and specificity,and can be used to detect S.xylosus.

RESUMEN

In recent years, with the rapid development of life sciences, the use of laboratory fish in toxicology, genetics, developmental biology and medicine has increased dramatically, and they have gradually become important new model organisms. At the same time, the welfare of laboratory fish has also received increasing attention. Although the research level of experimental fish welfare is still in a relatively early stage compared to terrestrial experimental animals, developed regions such as Europe and America have established corresponding legal frameworks to safeguard the welfare of laboratory fish in research. This article elucidates the current developmental status of laboratory fish welfare, discusses the rationale behind the imperative to prioritize and enhance their welfare, deeply investigates factors influencing their welfare from the feeding stage and experimental stage. Moreover, it explores strategies for augmenting welfare standards, with the overarching aim of propelling the continual improvement of laboratory fish welfare in our country.

RESUMEN

ObjectiveTo observe the fine structure of the trunk kidney in zebrafish, and to identify its secreted exosomes. MethodsThe microstructure and ultrastructure of the trunk kidney in zebrafish were observed by light microscopy and electron microscopy, and the particle size of exosomes was detected by nanoparticle tracking analysis (NTA). ResultsThe trunk kidney was close and parallel to the spine in adult zebrafish. The nephron consisted of renal tubules and renal corpuscles. The renal tubules could be further divided into three types: proximal convoluted tubules, distal convoluted tubules, and cervical segments. The renal corpuscles were composed of glomerulus and renal capsules. The periodic acid-Schiff (PAS) staining results revealed that there were abundant glycogen granules in the proximal convoluted tubules, with brush-like outline in the apical surface of epithelial cells. Under transmission electron microscopy (TEM), there were exosomes distributed in the lumen of renal tubules, with numerous late endosomes and few number of multivesicular bodies (MVBs) in the cytoplasm of the epithelial cells concentrating on the apical side. Meanwhile, MVBs were also distributed in the apical regions of the renal tubules and the podocytes of the renal glomeruli. Immunohistochemical staining results showed that CD9, CD63 and TSG101 were strongly expressed in the lumen surface of the renal tubules, but weakly expressed in the corpuscles and lumen. NTA and TEM results showed that the exosomes isolated from zebrafish trunk kidney were saucer-like outline, and the particle size mode was 144.4 nm, which was consistent with the characteristics of morphological futures of exosome. ConclusionThe zebrafish somatic kidney has the typical structure of the mammalian kidney and is the urinary organ in the body. The renal tubules have the ability to secrete exosomes, and their formation is a process of releasing poly-vesicles to the free surface of epithelial cells into the extracellular space. This study laid a morphological foundation for further study of exosomes in urinary function in aquatic experimental animals as well as the development and application of related models.

RESUMEN

ObjectiveTo establish a method for rapid and sensitive detection of Staphylococcus aureus. MethodsThe specific gene nuc of Staphylococcus aureus was selected as the target gene. A pair of specific primers and a TaqMan probe were designed and synthesized according to the published sequence of the nuc gene. Establish a nucleic acid detection method for nuc gene using fluorescence quantitative PCR technology, and apply it clinically in the detection of fecal samples from rats and mice. ResultsThe DNA extracted from Staphylococcus aureus and other non-Staphylococcus aureus strains was detected by qPCR. The results showed that Staphylococcus aureus had a specific amplification curve, while other non-Staphylococcus aureus did not, indicating that the designed primers and probes were specific for Staphylococcus aureus. The sensitivity of this method was determined by diluting the DNA of Staphylococcus aureus by 10 times. The results showed that the detection limit of this method was 10 fg DNA, which was 2 orders of magnitude higher than that of ordinary PCR method. A total of 91 clinical samples were detected in this study, of which 4 rat samples from the same facility had a typical S-curve. The PCR products were sequenced and BLAST compared. The gene sequence of this sample was 100% similar to that of Staphylococcus aureus, indicating that the sample was positive for the nucleic acid of Staphylococcus aureus nuc gene, with a positive rate of 4.40%. The result was consistent with that obtained by bacterial culture method. The nucleic acid extraction adopted a full-automatic nucleic acid purification instrument, and the time required from nucleic acid extraction to detection result determination was less than 1.5 h. ConclusionThe qPCR method established in this study to identify Staphylococcus aureus with nuc gene as the target gene has the advantages of fast, high sensitivity and specificity, and can be used for the detection of Staphylococcus aureus in feces of rats and mice.

PURPOSE OF THE STUDY: Effective screening questionnaires are essential for early detection of obstructive sleep apnea (OSA). The STOP-Bang questionnaire has high sensitivity but low specificity. Dry mouth is a typical clinical sign of OSA. We hypothesised that adding dry mouth in the STOP-Bang questionnaire would improve its specificity. STUDY DESIGN: A survey of the incidence of dry mouth was performed in a general population group and suspected sleep apnea clinical population group. Patients with suspected OSA were assessed by laboratory polysomnography and STOP-Bang questionnaire was performed. Adding the option of dry mouth to the OSA screening questionnaire resulted in a new quesionnaire, where cut-off value, diagnostic efficacy and the predictive parameters (sensitivity, specificity, positive predictive value and negative predictive value) were explored. RESULTS: (In the 912 general population group, the incidence of dry mouth in the snoring group (54.0%) was much higher than that in the non-snoring group (30.5%) (p<0.05). In 207 patients with suspected OSA, the incidence of dry mouth in the OSA group was much higher than that in the non-OSA group (p<0.05). The sensitivity and specificity of the STOP-Bang questionnaire were 88.8% and 23.7% for identifying OSA, and 92.2% and 23.1% for identifying moderate and severe OSA, respectively. Adding the option of dry mouth (dry mouth every morning) to the STOP-Bang questionare resulted in a new questionnaire (STOP-Bang-dry-mouth questionnarie) with 9 items. Its sensitivity and specificity were 81.70% and 42.10% for identifying OSA, and 89.10% and 42.30% for identifying moderate and severe OSA, respectively. CONCLUSIONS: The dry mouth symptom correlated with snoring and sleep apnea. The specificity of the STOP-Bang questionnaire can be improved by integrating dry mouth. The diagnostic accuracy of the STOP-Bang-dry mouth questionnaire is yet to be further verified in prospective studies.

Objective To evaluate the individualized 3D) printed drilling guide we developed and used for placement of C1/2 pedicle screws in the clinical treatment of fracture and dislocation of the atlantoaxial joint.Methods From January 2014 to June 2016,we treated a total of 17 patients with fracture and dislocation of the atlantoaxial joint.All the cervical CT data of the patients were imported into the digital orthopaedic workstation for 3D reconstruction,data modeling and 3D printing to design and manufacture individualized atlantoaxial vertebral guide templates.Intraoperatively,C1 and C2 pedicle screws were placed under the guide of individualized 3D printed drilling template.Cervical short-segment fixation and fusion were conducted for the patients.Postoperatively,regular clinical and radiographic follow-ups were carried out.Results No serious complications like injury to spinal cord and vertebral artery happened due to failed placement of C1/2 pedicle screws.The operation time ranged from 136 to 222 min (average,168.0 rmin);the intraoperative blood loss ranged from 260 to 556 mL (average,356.0 mL).The 17 patients were followed up for 6 to 36 months (average,13.5 months).The patients obtained bony union after 4 to 6 months (average,4.8 months).At the final follow-up,according to ASIA92 scoring system,the average sensory score was improved significantly from preoperative 7.4 + 3.2 to postoperative 13.1 + 5.9,and the average motor score was improved significantly from preoperative 5.3 + 3.1 to postoperative 11.7 + 5.1 (P < 0.05).No such complications as infection or implant failure occurred after operation.Conclusion Individualized 3D printed drilling guide for implantation of atlantoaxial pedicle screws can make the complicate and risky placement become accurate,safe and simple.

Objective To more intuitively understand the quality control for laboratory animals and further achie-ving a more scientific and reasonable management of laboratory animals, the infection index as evaluation criteria was intro-duced. Then the best way to calculate infection index was explored in order to more scientifically reflect the infection status of laboratory animals. Methods Infection index, also called the degree of infection, is a qualitative indicator of monito-ring laboratory animal quality. After arranging, analyzing, processing and gathering the data from laboratory animal quality monitoring, the index reflects synthetically the pathogen infection status or trend of a particularly investigated experimental animal population or the development of certain experimental animals. Results In general, the pathogen infection index of mice was slightly decreased, while the pathogen infection index of rats roughly increased year by year. In comparing infec-tion index by different pathogens, the parasite infection index of mice was found to be higher than bacteria and virus infec-tion indexes, while the bacteria infection index of rats was higher than parasite infection index and virus ones. Conclusions The infection index model intuitively reflects the quality control status of laboratory animals. The analysis also reveals that the parasite monitoring of the mice and the bacteria detection of rat needs to be reinforcement. In addition, the index of infection reveals that the pathogen infection of mice is well under control, while that of rats tends to be more serious year by year.

Objective To explore the developmental tendency of eco-migrant children's extroversive behavior and the relationship between extroversive behavior and personality and family environment.Methods 856 eco-migrant children(aged 6-16)participated in the present longitudinal study.At first time the Child behavior checklist(CBCL),Eysenck personality questionnaire (EPQ) and Family environment scale (FES) were used to assess their behavioral problems,personality and family environment.Participants' parents subsequently completed measures assessing behavioral problems every nine months for 27 months.Data were analyzed using hierarchical linear modeling analyses.Results The scores of extroversive behavior in eco-migrant children were(10.09±7.11) at first time,(7.66±7.56) at the second wave,(8.54±7.49)at the third wave and(8.11±7.33) at the last time.During the longitudinal period,the descending trend of eco-migrant children's extroversive behavior was significant (β=-0.51,P<0.05).The scores of psychoticism,neuroticism,family conflict,organization,cohesion and cultural factors were differently correlated with children's extroversive behavior (β=-0.67-0.32,P<0.05).Family conflict predicted the developmental trend of children's externalizing (β=-0.46,P<0.05).Conclusion During the longitudinal period,the level of eco-migrant children's extroversive behavior decreased,and personality and family environment have significant influences on it.

Objective To compare the efficiency of bacteria culture and PCR assays for detection of Staphylococcus aureus ( S.aureus) , Pseudomonas aeruginosa ( P.aeruginosa) and Klebsiella pneumoniae ( K.pneumoniae) in laboratory rats and mice.Methods Bacteria culture combined with biochemical identification and PCR assay were used to detect 78 SPF rats and 422 SPF mice and the results of the two methods were compared .Results All the 78 rats were negative .Of the 422 mice, the positive rate by culture was 7.11%(30/422), of which, 10 were S.aureus, 22 were P.aeruginosa, and 2 were K.pneumoniae.The positive rate by PCR was 7.58%(32/422), of which, 10 were S.aureus, 25 were P. aeruginosa, and 2 were K.pneumoniae.Conclusions The high sensitivity , rapid procedure and easy to operate of PCR assay makes it valuable for rapid bacteria diagnosis and large-scale screening in laboratory animals .

<p><b>OBJECTIVE</b>To investigate the detection rate and correlates of behavioral problems among eco-migrant children in Hui and Han ethnicities.</p><p><b>METHODS</b>Using multistage randomized sampling method, 2 653 eco-migrant teenagers, 3 174 indigenous and 2 334 homeland peers were selected. Their parents were asked to finish the Achenbach's Child Behavior Checklist (CBCL) and a self-developed correlates questionnaire.</p><p><b>RESULTS</b>765 eco-migrant children with behavioral problems were detected, with detection rate as 28.8%, higher than that of homeland group (24.2%) and native group (19.3%) (χ(2) = 73.547, P < 0.012 5). Ecological migration mainly influenced factors as bad contacts, depression, obsessive-compulsive, discipline, attack for boys aged 6-11, bad contacts for boys aged 12-16 and extraversion behavioral problems for girls aged 12-16, in Hui. These factors both associated to eco-migrant children's behavioral problems in Hui and Han:cesarean section (OR = 1.863, 95% CI:1.144-3.035, for Han) and (OR = 2.979, 95% CI:2.067-4.293, for Hui), physical defects (OR = 1.730, 95%CI:1.087-2.751, for Han) and (OR = 2.552, 95%CI:1.649-3.950, for Hui), arable as a main income way(OR = 1.272, 95%CI:1.066-1.518, for Han) and (OR = 1.212, 95%CI: 1.033-1.422, for Hui), parents' education level above middle school (OR = 0.709, 95% CI: 0.539-0.932, for Han) and (OR = 0.698, 95%CI: 0.542-0.898, for Hui), parents' frequently in good mood (OR = 0.710, 95%CI:0.613-0.821, for Han) and (OR = 0.826, 95%CI:0.718-0.949, for Hui). However, major diseases in the process of growth (OR = 1.794, 95% CI:1.163-2.767), and parents with chronic illness or disability (OR = 1.463, 95% CI:1.061-2.016) only associated with that in Han;single-parent or remarried families(OR = 1.583, 95% CI:1.078-2.325), parents often drinking (OR = 1.557, 95%CI:1.019- 2.379), the time of parents' work longer than half a year (OR = 1.494, 95% CI:1.197-1.864), parents' more content to life now(OR = 0.813, 95% CI:0.700-0.945), and children in older age (OR = 0.939, 95%CI:0.889-0.991) only related to that in Hui.</p><p><b>CONCLUSION</b>The eco-migrant children showed a low state in mental health, especially among Hui peers, and the correlated factors were different in Han and Hui, which was supposed to get great importance when taking effective intervention.</p>

Background and purpose:MicroRNAs (miRNAs, miR) are directly involved in cancer initiation, progression and metastasis. Alterations of miRNAs expression in cancer tissue may be reflected in circulation. We attempted to investigate the expression and clinical signiifcance of plasma miR-221 in patients with small cell lung cancer (SCLC). The plasma and tissues levels of miR-221 in 51 SCLC patients and 20 controls were evaluated and compared among various clinicopathological characteristics. Methods: Confirmation of higher miR-221 levels in primary SCLC tissues than normal lung tissues. Evaluation of plasma miR-221 concentrations by comparing results from 51 consecutive SCLC patients and 20 healthy volunteers. Evaluation of the assay for monitoring tumour dynamics in SCLC patients. Results:Expression of miR-221 was signiifcantly higher in SCLC tissues than in para-carcinoma tissues and normal lung tissues. Plasma miR-221 concentrations were signiifcantly higher in SCLC patients than those in normal people (P0.05), correlated with significantly chemosensitivity, overall survival and clinical stage (P<0.05). Cox regression analysis indicated that plasma miR-221 expression and disease stage were found to be significantly independent prognostic factors for the SCLC patients. Conclusion: Plasma miR-221 could be a useful biomarker for cancer detection, monitoring tumour dynamics and predicting malignant outcomes in SCLC patients, and may contribute to clinical decision making in treatments.

Objective To study the relationship between placenta HBsAg in HBsAg positive pregnant women and serum hepatitis B virus (HBV) markers,HBV DNA levels in newborns.Methods Placenta HBsAg was detected by immunohistochemical affinity hormone-biotin complex (ABC) method in 155 HBsAg positive pregnant women.Serum HBV markers in newborns were detected by enzyme-linked immunosorbent assay (ELISA).Serum HBV DNA levels of newborns were detected by real-time fluorescence quantitative polymerase chain reaction (PCR).The positive rates were compared using x2 test.Results HBsAg was expressed with different levels in various types of cells of placenta in 155 pregnant women.The total placenta HBsAg positive rate was 37.4% (58/155),and those in decidual cells,trophoblastic cells,villous mesenchymal cells and villous capillary endothelial cells were 37.4% (58/155),25.8% (40/155),18.7% (29/155) and 7.1% (11/155),respectively.The HBsAg positive rates of placenta gradually decreased from decidual cells of the maternal surface to villous capillary endothelial cells of the fetal surface (tendency x2 =43.01,P=0.00).The positivity of placenta HBsAg was associated with both HBsAg and HBeAg in newborns (x2 =4.88,P<0.05 and x2 =3.86,P<0.05,respectively),while that was not associated withanti-HBe and anti-HBc in newborns (x2 =3.36,P>0.05 and x2 =0.00,P> 0.05,respectively).The risk of HBsAg positive in newborns was higher when HBsAg was positive in villous capillary endothelial cells and villous mesenchymal cells (OR=5.31,95 %CI=1.38-20.40 and OR=3.33,95%CI=1.16-9.52,respectively).The risk of HBeAg positive in newborns was higher when HBsAg was positive in trophoblastic cells and villous mesenchymal cells (OR=3.04,95 %CI=1.45-6.39 and OR=3.05,95 % CI=1.32-7.03,respectively).However,placenta HBsAg positive was not associated with HBV DNA positive in newborns (x2 =0.09,P>0.05).Conclusion The risk of neonatal HBV serological markers positive is higher when the HBsAg positive placental cells are closer to fetal surface,which indicates that HBsAg enters fetal blood circulation by means of cell transferring layer by layer.

ObjectiveTo observe the changes of the levels of serum TNF-α and IL-6 in patients with hypertensive disorders in pregnancy(HDIP),to explore the significance of serum TNF-α and IL-6 in HDIP.MethodsThe levels of serum TNF-α and IL-6 in 30 patients with HDIP( 11 cases with gestational hypertension,11 casses with mild preeclampsia,8 cases with severe preeclampsia) and 18 healthy pregnant women were detected.ResultsThe levels of serum TNF-α in HDIP were( 1.88 ±0.48) μg/L,(2.32 ±0.40) μg/L and(2.35 ± 0.42) μg/L,The levels of serum IL-6 of HDIP were (99.54 ± 31.62) ng/L,( 120.53 ± 26.42)ng/L,( 140.83 ± 22.13 ) ng/L respectively,the serum levels of TNF-α and IL-6 in healthy pregnant women were( 1.63 ± 0.32)μg/L and(95.82 ± 30.65 )ng/L,there was significant difference among them ( all P < 0.05 ).ConclusionThe serum levels of TNF-α and IL-6 in HDIP were higer than those in healthy pregnant women,and vascular endothelium cell injury should play an important role in the pathogenesis and development of HDIP.

Objective To investigate the influence of long-term blood purification on atherosclerosis(AS) in maintenance hemodialysis(MHD) patients by measuring serum levels of homocysteine(Hcy) and carotid artery intima-medial thickness(IMT).Methods Seventy-two MHD patients visiting our hospital from June 2006 to December 2007,according to their blood purification mode,were assigned to three groups: hemodialysis(HD,n=25),hemodialysis+hemodiafiltration(HD+HDF,n=30),hemodialysis+continuous renal replacement therapy(HD+CRRT,n=17).Another 25 healthy subjects served as controls.Serum Hcy was detected before and after one-year treatment and meanwhile color Doppler ultrasonography was performed to check IMT of carotid artery and atheromatous plaques.Results The serum levels of Hcy,IL-6,MCP-1,hs-CRP in MHD patients were significantly higher than those in healthy controls(P

Objective:To investigate the therapeutic effect of sequential blood purification on rescuing acute grave intoxication.Methods:Twenty eight patients with acute intoxication were treated by HP+CVVH during 2002 and compared them with thirty patients without hemopurgation from 1998 to 2002.Results:The group of hemopurgation was better than non-hemopurgation group in the time of revive,the rate of recovery and death,and the total dose of atropine.Conclusion:Hemopurgation has decisively effect on rescuing all kinds of intoxication with toxins and drugs.It could quickly ameliorate symptom,raise success rate and reduce mortality;Hp combining with CVVH are suitable for the patients with toxicsis who had cardiac and respiratory failure,brain edema,and dielectric disorder.

<p><b>OBJECTIVE</b>To investigate the etiological agent of patients with diarrhea followed by acute kidney failure symptoms in China, 1999.</p><p><b>METHODS</b>Western blot was used to detect serum specific antibodies of patients against entero-haemorrhagic Escherichia coli hemolysin (EHEC-Hly) and lipo-polysaccharide of E. coli O157.</p><p><b>RESULTS</b>Twenty-one and 16 of 42 patients showed positive reaction of specific IgG or IgM antibodies against EHEC-Hly respectively. Eleven of 42 serum samples were positive for having both IgG and IgM antibodies while 26 of 42 samples were positive for IgG or IgM. For E. coli O157 LPS test, 24 and 24 of 42 samples showed positive for IgG or IgM antibodies respectively. In 42 samples, 20 were positive for IgG and IgM while 29 were positive for IgG or IgM.</p><p><b>CONCLUSIONS</b>Twenty-two of 42 samples were reacted with EHEC-Hly and E. coli O157 LPS, but 34 of 42 samples were positive for EHEC-Hly or E. coli O157. In combination of western blot results, bacterial isolation clinical symptoms and epidemiological investigation findings, it was reasonable to conclude that this cluster of patients with distinguish clinical symptoms was caused by E. coli O157:H7, which had never been reported in China. Hence serological methods with EHEC-Hly and E. coli O157 LPS are valuable for diagnosis of infections of E. coli O157:H7, when bacterial isolation is failed.</p>

RESUMEN

RESUMEN

Objective To investigate Hepatitis B Virus (HBV) infection status in placentas of Hepatitis B Surface Antigen (HBsAg) positive pregnant women during middle and late period of pregnancy. Methods One hundred and sixty seven placentas from HBsAg positive pregnant women were collected, including 158 term placentas and 9 aborted second trimester placentas. HBsAg and hepatitis B core antibody (HBcAg) in placental cytotypes were detected by immunohistochemistry. Results Of the 158 term placentas, HBV infection rates in decidual cells (DC), trophoblastic cells (TC), villous mesenchymal cells (VMC) and villous capillary endothelial cells (VCEC) were 66 46% (105/158), 58 23% (92/158), 27 22% (43/158) and 12 66% (20/158), respectively. The HBV infection rates gradually decreased from maternal side to fetal side of placentas. Both HBsAg and HBcAg were mainly located in the cytoplasm of infected cells. The positive cells were mostly in focal distribution. One out of 9 aborted second trimester placentas was infected. Conclusions HBV infection was found in different cells of term placentas. The positive rates for HBsAg and HBcAg in placentas were gradually decreased from maternal side to fetal side. The positive rate was low in aborted second trimester placentas.
